Objective To get the antigen of CD7.Methods The extracellular domain of human CD7 was cloned from a plasmid containing the full length of human CD7 cDNA and expressed in pinpoint-xa3 prokaryotic system. Results Analysis with SDS-PAGE and Western-blotting showed that the expressed protein could bind to the anti-CD7 mAb specifically and is about 30 000 u in molecular weight. Conclusion These results paved the way for preparing anti-CD7 engineering antibody.
2.Super-early Minimal Invasive Evacuation of Hematoma via Keyhole for Treating Hypertensive Basal Ganglionic Hemorrhage

Objective To estimate the efficacy of super-early minimal invasive evacuation of hematoma via keyhole for treating hypertensive basal ganglionic hemorrhage. Methods 36 patients with hypertensive basal ganglionic hemorrhage(9 cases in grade Ⅱ, 17 in grade Ⅲ and 10 in grade Ⅳ) were treated by minimal invasive evacuation of hematoma via keyhole at super-early stage, and their efficacy was statistically compared with 35 patients with hypertensive cerebral hemorrhage underwent conservative therapy or lateral ventriculopuncture. Results In the 36 patients underwent minimal invasive evacuation of hematoma via keyhole, 22 cases had a good prognosis(61.1%), 9 had a poor prognosis(25.0%), and 5 died(13.9%). Among the 35 patients underwent conservative therapy or lateral ventriculopuncture, 5 cases had a good prognosis(14.3%), 22 had a poor prognosis(62.9%), and 8 died(22.9%). Objective: To examine the distribution of IL-1B+3953 genotypes of Han nationality people with different periodontal health status and to evaluate if there is an association between the genotype of IL-1B+ 3953 and the severity of periodontitis. Methods: 271 subjects of Han nationality were selected, among them there were 85 cases with severe chronic perioodontitis(group①), 97 cases with light and mild chronic periodontitis (group ②) and 89 subjects of periodontal healthy control(group ③). Full periodontal examinations were taken including clinical periodontal tissue attachment loss measurements of full-mouth, periodontal pocket depths and bleeding on probing. DNA samples were obtained with buccal swabbing technique and were further analyzed for IL-1B+3953 genotype polymorphisms using PCR-RFLP-based method. 4.Effects of alprostadil and low dose dopamine in treatment of hepatorenal syndrome

Objective To investigate the effects of alprostadil (Lipo-PGE) combined low-dose dopamine in the treatment of hepatorenal syndrome (HRS). Methods 61 patients with hepatorenal syndrome were randomly di-vided into two groups, treatment group 31 cases, 30 cases of the control group. Treatment group in the control group based on the 20μg soluble in 10% glucose infusion 100 ml in combined treatment with Lipo-PGE a day, one time,and combined with low-dose dopamine 3 μg/(kg·min) intravenous infusion, 1 week course of treatment. Result The treatment group liver function improvement in renal function are superior to the control group (P <0.01), to-ted effective rate was 67.7%. Conclusion The united alprostadil and low-dose dopamine satisfied with liver and kidney syndrome, there is synergy.
5.Expression and significance of p38 mitogen-activated protein kinase in oral lichen planus and oral squamous cell cacinoma

Objective:To assess the differences among the expressions of p38 mitogen activated protein kinases (MAPK),phospho-p38MAPK and nuclear factor kappa B (NF-κB)in oral lichen planus (OLP)and oral squamous cell carcinoma (OSCC ).Methods:In the study,53 cases of OLP,45 of OSCC,and 18 controls were obtained and 4-μm-thick histological sections were prepared from formalin-fixed paraffin-embedded tissue blocks.The expressions of p38MAPK,phospho-p38MAPK and NF-κB were detected by immunohistochemistry staining.Furthermore,the expressions of p38MAPK and phospho-p38MAPK were detected using Western blotting analyses in the fresh tissues from 1 1 cases of OLP,5 ca-ses of OSCC,and 7 cases of the controls.Results:p38MAPK was over-expressed in the lamina propria, but lowly expressed in the epithelium in OLP group.Phospho-p38MAPK was lower expressed in OLP group than in OSCC and control groups.NF-κB was found over-expressed in the lamina propria in OLP group.p38MAPK was found expressed in all the samples in the 3 groups.The expression of phospho-p38MAPK was observed in 8 (8/11)OLP samples,5 (5/5)OSCC samples and 4 (4/7)controls by Western blotting,but no significant differences were found within the 3 groups.Conclusion:p38MAPK can be detected in normal oral mucosa,OLP and OSCC.phospho-p38MAPK may be related to the onset and progression of OSCC.The role of p38MAPK in OLP is yet to be revealed.
6.Effect of cetuximab combined with radiation on the expressions of Ku80 and ATM in CNE-2 nude mice xenograft tumor model

Objective To investigate the effect of cetuximab (C225) combined with radiation on the expressions of Ku80 and ATM in CNE-2 nude mice xenograft tumor model.Methods The CNE2 nude mice xenograft tumor model was established and divided into control group,conventional radiotherapy (CRT) group,simulated intensity modulated radiation therapy (sIMRT) group,C225 group,CRT + C225 group and sIMRT + C225 group with 8 mice in each group.The dose of radiation was 20 Gy,and 1mg C225 per mice in abdominal cavity was applied for the drug treatment.The transcriptional levels of Ku80 and ATM were assayed by RT-PCR and Western blot.Results Compared with control group,the transcriptional and translational levels of Ku80 and ATM in CRT group,sIMRT group,CRT + C225 group and slMRT + C225 group were decreased (P < 0.05).Compared with CRT group,the down-regulation of transcriptional and translational ATM level in sIMRT group was obvious (P < 0.05),and further strengthened in the CRT + C225 group and sIMRT + C225 groups (P <0.05),but there was no significant difference of ATM expression between sIMRT + C225 group and CRT group (P > 0.05).Conclusions C225 alone can restrain the transcriptional and translational level of ATM,and the transcriptional and translational level of Ku80 and ATM are also restrained after megadose.ATM is down-regulated by sIMRT but up-regulated by the combination of C225 and radiation.
7.Construction and identification of expression of superantigen SED mutants

Objective To construct staphylococcal enterotoxin D (SED) mutants expressed in Escherichia coli ( E. coli ). Methods The expected mutants were introduced into the SED DNA by megaprimer PCR method. The PCR products ligated to plasmid pTrcHis B were transformed into E. coli DH5? for IPTG induced expression. The target protein was purified by Ni NTA metal affinity chromatography and analyzed by SDS PAGE and immunoblotting. Results The sequencing results showed that mutant nucleic acids were successfully introduced at the expected sites of SED gene. SDS PAGE and immunoblotting confirmed that the proteins of SED mutants were obtained by Ni NTA metal affinity chromatography. The mutants were named as SEDN23A, SEDN23A/H26R, SEDF45A, SEDL59A, SEDN61A, SEDI92A, and SEDF203A, respectively. Conclusion Several SED mutants are successfully constructed, which lays a foundation for subsequent studies of immune recognition of SED.

Objective To explore the possible correlation of the clinical parameters(such as neck circumference,waist circumference,Hipc,WHR,BMI,ESS scores)with the severity of obstructive sleep apnea/hypopnea syndrome(OSAHS).Methods There were 111 patients with diagnosis of OSAHS.All patients underwent polysomnography and the physical examination.The meatured body parameters and AHI were analyzed.Results A statistically significant correlation between neck circumference,waist circumference,Hipc,WHR,BMI with the AHI was detected.No correlation was found between ESS and AHI.Except for Hips and ESS,the difference of neck circumference,waist circumference,WHR,BMI were significant between the different serious patients(P
9.Effects of Chaihu Shugan San on behavior and CRHmRNA expression in the brain of mice with chronic mild unpredicted stress depression

Objective To investigate the effects of Chaihu Shugan San(CHSGS) ,a compound traditional Chinese herbal medicine,on the behavior and plasma levels of corticosterone level and corticotrophin releasing hormone ( CRH) mRNA expression in the prefrontal cortex and the hypothalamus of mice with chronic mild unpredicted stress depression. Methods 40 adult male Kunming mice were randomly divided into 4 groups: the normal control group,the model control group,the fluoxetine group and CHSGS group. Except the normal control group, mice were exposed to an unpredicted sequence of mild stressor for continuous 21 days to induce depression. Since the 8th day, mice were intragastrically administered with equal volume agents respectively for 14 days (normal saline for the normal control group and the model group,fluoxetine(2. 6 mg · kg-1 ) for fluoxetine group and CHSGS (8.45 g · kg-1) for CHSGS group). Body weight and behaviors were investigated,the plasma levels of CORT in different groups were detected by radioimmunoassay, and the mRNA expression in the prefrontal cortex and the hypothalamus were semiquantified by the RT-PCR method. Results In comparision with the model group, body weight changes ( (13.22 ±3.57)g) and consumption of sucrose solution((1. 90 ±0. 74) ml) of CHSGS groups were significantly increased, and the duration of immobility during the forced swimming ((62.20±13.73)s) and tail suspension((58. 10 ± 11.71 )s) ,the levels of plasma CORT((12.61 ±4.63)ng/L) and the CRHmRNA expression in the prefrontal cortex(0.34 ±0.10) and the hypothalamus(0.70 ±0.09) were significantly decreased. Conclusion CHSGS can reverse the depressive behavior, reduced the levels of plasma CORT and down-regulated CRHmRNA expression in the prefrontal cortex and the hypothalamus of the depressive mouse caused chronic stress.
10.Pharmacokinetic Studies of Quercetin in Semen Cuscutae

Objective To establish the method of measuring quercetin content in semen cuscutae(SC) and to explore its in- vivo pharmacokinetics features.Methods The rapid and sensitive HPLC- UV method was adopted with carbamazepine acting as internal standard. After the extract of SC was given, serum samples of rats were collected and measured on C18 reverse- phase chromatographic column after liquid- liquid extract. Results The linear range of calibration curve were 0.05~ 9? g/mL (r=0.9998). The method showed good precision and recoveries for serum with 86.65~ 99.07% , and extracting recoveries 79.91~ 84.56% .The limit of detection(LOD) as 0.05? g/mL. Conclusion The pharmacokinetic process of quercetin in vivo manifestated an opening two- compartment model with t1/2(Ab)=0.13h, t1/2(? )=0.19h, t1/2(? ) =1.22h, tmax=0.333h. The results may provide evidence of safety and effectiveness for clinical use of SC.
