1.Role of protein tyrosine phosphatase SHP-2 in group I metabotropic glutamate receptors-induced allodynia

Aim To investigate whether the pain modi-fication by group I metabotropic glutamate receptors (mGluRs)required the involvement of Src homology-2 domain-containing phosphatase-2 (SHP-2 ).Methods Co-immunoprecipitation was performed to examine the possible interaction between SHP-2 and group I mGluRs in spinal cord dorsal horn of mice.By measur-ing the paw withdrawal thresholds,the effects of SHP-2 inhibitor NSC-87877 or its catalytically inactive SHP-2 (C459S ) mutant on allodynia induced by group I mGluRs agonist DHPG (50 nmol)were observed.Re-sults Anti-mGluR5 antibody was able to co-immuno-precipitate SHP-2 from spinal dorsal horn of mice, while no SHP-2 was precipitated by anti-mGluR1 anti-body.Inactivation of SHP-2 by NSC-87877 (6 nmol) or SHP-2 (C459S ) effectively attenuated allodynia caused by DHPG.Conclusion SHP-2 can physically interact with mGluR5.The activation of SHP-2 may be necessary for group I mGluRs to process the nocicep-tive information.
2.HLA-B alleles and nasopharyngeal carcinoma in Xinjiang, China:relationship and clinical significance

Objective To explore the relationship between HLA?B allele polymorphisms and nasopharyngeal carcinoma ( NPC) in Xinjiang, China and its clinical significance. Methods A total of 226 patients were assigned to NPC group, while 207 healthy volunteers were assigned to control group. PCR amplification with sequence?specific primers was used to determine HLA?B alleles. Comparison of HLA?B allele frequency between the above two groups, between Han and Uygur populations, and between patients with various clinical characteristics of NPC was made by chi?square test. The Kaplan?Meier method was used to calculate the survival rates and the log?rank univariate analysis was used to explore the relationship between survival rates and HLA?B allele frequency. Results In all the subjects or Han population alone, the allele frequency of HLA?B?46 in the NPC group was significantly higher than that in the control group ( P=0. 000;P=0. 000 ) . In Uygur population, however, there was no significant difference in the allele frequency of HLA?B?46 between the NPC group and the control group (P>0. 05). In the patients with NPC, those less than 30 years old had a significantly higher allele frequency of HLA?B?44 than those no less than 30 years old (P=0. 029);those with differentiated non?keratinizing carcinoma had a significantly higher allele frequency of HLA?B?48 than those with undifferentiated non?keratinizing carcinoma ( P=0. 029);those with stage T1+T2 disease had a significantly higher allele frequency of HLA?B?48 than those with stage T3+T4 disease ( P=0. 029) . The 5?year overall survival, disease?free survival, distant metastasis?free survival, and local relapse?free survival rates had no relationship with the expression of HLA?B?46, HLA?B?44, or HLA?B?48 in NPC patients ( all P>0. 05) . Conclusions HLA?B?46 allele is probably a NPC susceptibility gene in Han population in Xinjiang. HLA?B?44 is probably associated with early age of onset, while HLA?B?48 is probably associated with the pathological type and T stage of NPC. Therefore, HLA?B alleles are probably associated with the development and progression of NPC.
3.Expression and significance of p38 mitogen-activated protein kinase in oral lichen planus and oral squamous cell cacinoma

Objective:To assess the differences among the expressions of p38 mitogen activated protein kinases (MAPK),phospho-p38MAPK and nuclear factor kappa B (NF-κB)in oral lichen planus (OLP)and oral squamous cell carcinoma (OSCC ).Methods:In the study,53 cases of OLP,45 of OSCC,and 18 controls were obtained and 4-μm-thick histological sections were prepared from formalin-fixed paraffin-embedded tissue blocks.The expressions of p38MAPK,phospho-p38MAPK and NF-κB were detected by immunohistochemistry staining.Furthermore,the expressions of p38MAPK and phospho-p38MAPK were detected using Western blotting analyses in the fresh tissues from 1 1 cases of OLP,5 ca-ses of OSCC,and 7 cases of the controls.Results:p38MAPK was over-expressed in the lamina propria, but lowly expressed in the epithelium in OLP group.Phospho-p38MAPK was lower expressed in OLP group than in OSCC and control groups.NF-κB was found over-expressed in the lamina propria in OLP group.p38MAPK was found expressed in all the samples in the 3 groups.The expression of phospho-p38MAPK was observed in 8 (8/11)OLP samples,5 (5/5)OSCC samples and 4 (4/7)controls by Western blotting,but no significant differences were found within the 3 groups.Conclusion:p38MAPK can be detected in normal oral mucosa,OLP and OSCC.phospho-p38MAPK may be related to the onset and progression of OSCC.The role of p38MAPK in OLP is yet to be revealed.
4.Effect of Chaihu Shugan San and its components on expression of ERK1/2 mRNA in the hippocampus of rats with chronic mild unpredicted stress depression

Objective To explore the antidepressant effect and mechanism of Chaihu Shugan San (CHSGS) composition, a compound traditional Chinese herb medicine and its components. Methods Rats were randomly divided into 6 groups: normal control group, model control group, a CHSGS group, a component I group, a component II group and a fluoxetine control group. The depression model was replicated by chronic unpredictable mild stress and single house for 28 d. Behavioral scores of the rats were detected by Open-field test and sucrose solution consumption test, and ERK1/2 mRNA expression in the hippocampus tissue was assayed by real-time fluorescent quantitative PCR. Results ERK1/2 mRNA expression was down-regulated in the depression model group compared with the normal control group (P<0.01). Compared with the model group, ERK1/2 mRNA expression in the CHSGS group and fluoxetine group was both up-regulated (P<0.05 or P<0.01); and only ERK1 mRNA expression in the component I group was up-regulated (P<0.05). No significant difference existed between the component II group and the model group (P>0.05). Conclusion Isolated-living condition and chronic mild unpredictable stress can down-regulate the expression of ERK1/2 mRNA in the hippocampus tissue. CHSGS may exert an antidepressant effect through increasing the expression of ERK1/2 mRNA in the hippocampus, component I may play an important role in its antidepression effect, while compatibility of the use of component II can enhance the antidepressant efficacy.
5.Development of the Wounded Information Collection and Management System

Objective 70 realize the fast collection and management of the basic wounded information in the field treatment. Methods Two-dimensional electronic medicine tag was used as a massage loader. The wounded information was analyzed with a program reader, and the basic personal and cure information were shared with the information management system. Result The system was used in the massive discipline for the first time, fulfilled the wounded information record, analysis, transfer, and the management of the cure information with 80 items. Conclusion The correctness of the basic massage record and analysis is full percent. The manage progress of the system accord with the field condition.
6.Development and Application of the Medical Treatment Information Management System in Field Battle

Objective To realize medical information collection and digital management in field battle and the fundamental information and cure of medical staff of emergency medical, equipment and the wounded were included in it. Methods The collecting information were transmitted to computer through wireless connection and manage the information with Delphi7 procedure. Results The system was used firstly in large-scale practice and completed the information transcription and management on elementary and cure in hundred of wounded. Conclusion The information is up to the mustard of field and it has established base on cure information in digital management.
7.Effect of cetuximab combined with radiation on the expressions of Ku80 and ATM in CNE-2 nude mice xenograft tumor model

Objective To investigate the effect of cetuximab (C225) combined with radiation on the expressions of Ku80 and ATM in CNE-2 nude mice xenograft tumor model.Methods The CNE2 nude mice xenograft tumor model was established and divided into control group,conventional radiotherapy (CRT) group,simulated intensity modulated radiation therapy (sIMRT) group,C225 group,CRT + C225 group and sIMRT + C225 group with 8 mice in each group.The dose of radiation was 20 Gy,and 1mg C225 per mice in abdominal cavity was applied for the drug treatment.The transcriptional levels of Ku80 and ATM were assayed by RT-PCR and Western blot.Results Compared with control group,the transcriptional and translational levels of Ku80 and ATM in CRT group,sIMRT group,CRT + C225 group and slMRT + C225 group were decreased (P < 0.05).Compared with CRT group,the down-regulation of transcriptional and translational ATM level in sIMRT group was obvious (P < 0.05),and further strengthened in the CRT + C225 group and sIMRT + C225 groups (P <0.05),but there was no significant difference of ATM expression between sIMRT + C225 group and CRT group (P > 0.05).Conclusions C225 alone can restrain the transcriptional and translational level of ATM,and the transcriptional and translational level of Ku80 and ATM are also restrained after megadose.ATM is down-regulated by sIMRT but up-regulated by the combination of C225 and radiation.
8.Investigation of six-degree-of-freedom image registration between planning and cone beam computed tomography in esophageal cancer

Objective To explore six-degree-of-freedom (6-DF) registration methods between planning and cone beam computed tomography (CBCT) during image-guided radiation therapy (IGRT) in esophageal cancer.Methods Thirty pairs of CBCT images acquired before radiation and the corresponding planning computed tomography (CT) images of esophageal cancer were selected for further investigation.Registration markers for 6-DF image registration were determined and contoured in those images.The results of registration as well as time cost were compared among different registration methods of bone match, gray value match, manual match, and bone plus manual match.Results Contouring bone and spinal canal posterior to the target volume of esophageal carcinoma as registration marker could make 6-DF registration quick and precise.Compared with manual match, set-up errors of v rotation in bone plus manual match (-0.55° vs.-0.88°, t=2.55, P=0.020), of x-axis and v rotation in bone match (0.12 mm vs.-2.33 mm, t=5.75, P=0.000; -0.35° vs.-0.88°, t=3.00, P=0.007), and of x-axis and w rotation in gray value match (7.20 mm vs.-2.33 mm, t=3.10, P=0.006; -0.10° vs.-0.59°, t=2.81, P =0.011) were significantly different.Compared with manual match, the coincidence rate of bone plus manual match was the highest (85.55%), followed by bone match and gray value match (74.45% and 74.45%).The time cost of each registration method from longest to shortest was:6.00 -10.00 minutes for manual match, 1.00 - 5.00 minutes for bone plus manual match, 0.75 - 1.50 minutes for gray value match, and 0.50 - 0.83 minutes for bone match.Conclusions Registration marker is useful for image registration of CBCT and planning CT in patients with esophageal cancer.Bone plus manual match may be the best registration method considering both registration time and accuracy.
9.Effects of Chaihu Shugan San on behavior and CRHmRNA expression in the brain of mice with chronic mild unpredicted stress depression

Objective To investigate the effects of Chaihu Shugan San(CHSGS) ,a compound traditional Chinese herbal medicine,on the behavior and plasma levels of corticosterone level and corticotrophin releasing hormone ( CRH) mRNA expression in the prefrontal cortex and the hypothalamus of mice with chronic mild unpredicted stress depression. Methods 40 adult male Kunming mice were randomly divided into 4 groups: the normal control group,the model control group,the fluoxetine group and CHSGS group. Except the normal control group, mice were exposed to an unpredicted sequence of mild stressor for continuous 21 days to induce depression. Since the 8th day, mice were intragastrically administered with equal volume agents respectively for 14 days (normal saline for the normal control group and the model group,fluoxetine(2. 6 mg · kg-1 ) for fluoxetine group and CHSGS (8.45 g · kg-1) for CHSGS group). Body weight and behaviors were investigated,the plasma levels of CORT in different groups were detected by radioimmunoassay, and the mRNA expression in the prefrontal cortex and the hypothalamus were semiquantified by the RT-PCR method. Results In comparision with the model group, body weight changes ( (13.22 ±3.57)g) and consumption of sucrose solution((1. 90 ±0. 74) ml) of CHSGS groups were significantly increased, and the duration of immobility during the forced swimming ((62.20±13.73)s) and tail suspension((58. 10 ± 11.71 )s) ,the levels of plasma CORT((12.61 ±4.63)ng/L) and the CRHmRNA expression in the prefrontal cortex(0.34 ±0.10) and the hypothalamus(0.70 ±0.09) were significantly decreased. Conclusion CHSGS can reverse the depressive behavior, reduced the levels of plasma CORT and down-regulated CRHmRNA expression in the prefrontal cortex and the hypothalamus of the depressive mouse caused chronic stress.
