1.Studies of the Active Component Parts of APN on the Antiplatelet Aggregation

Objective:This paper was designed to reveal the active ingredients to inhibit platelet aggregation in Andregraphis Paniculata Ness (APN). Methods:Separating the active component parts of APN on the antiplatelet aggretion by extraction of organic solvent, chemical separation and silica column chromatography step by step, simultaneously comparing the effect of samples in APN on the antiplatelet aggretion induced by ADP, so that we can find the more active components. Results:We find the acid components in chlorlform parts (F021) show more active on the antiplatelet aggretion, farthermore we think we can obstain pure chemicals on the antiplatelet aggretion in F0212 and F0214. Conclusions:The active components on antiplatelet aggretion of APN can be prepared by separation step by step.
2.Research on the Extraction and Purification of Organic Acids from Folium Isatidis with Macroporous Adsorptive Resin

OBJECTIVE:To extract and purify organic acids with macroporous resins from Folium Isatidis.METHODS:Folium Isatidis was extracted with 70% aqueous ethanol(pH=2)by Soxhlet extraction.The process was optimized by employing orthogonal test design with the content of anthranilic acid as the index.RESULTS:The resin HPD100 had much better adsorption capacity and desorption ratio by using 3 BV 60% aqueous ethanol to extract organic acids in Folium Isatidis than other resins.The results showed that HPD100 possessed the better ecomic under the concentration of sample 0.18mg?mL-1,the condition of pH 3.5,and flowing rate of 2BV?h-1.CONCLUSION:Macroporous resin HPD100 may be value of isolating and purifying the organic acids from Folium Isatidis.
3.Screening of anti-endotoxin components from Radix isatidis.

To find a component with the strongest anti-endotoxin effect from the four components (F021, F022, F023, and F024) of chloroform extract (F02) of Radix Isatidis, the protective effects of the five components on endotoxin-challenged mice and their effects on the production of TNFalpha and IL-6 by macrophages from the endotoxin-challenged mice were compared. It was found that the five components all had protective effects on endotoxin-challenged mice in terms of mortality. The mortalities of the mice challenged by 2.42 mg/kg endotoxin were 30 %, 50 %, 20 %, 50 % and 60 % after treatment with F02, F021, F022, F023, and F024. The 5 components all had inhibitory effects on the production of TNFalpha and IL-6 by macrophages from the mice. At a concentration of LPS of 50 ng/mL, the inhibitory rates of F02 , F021 1, F022, F023, and F024 for the production of TNFalpha and IL-6 were 76.54 %, 30.86 %, 78.60 %, 36.63 %, 2.06 % and 75.85 %, 18.45 %, 77.68 %, 24.41 %, 3.64% and at the dose of 100 ng/mL, the inhibitory rates were 49.65 Y, 16.86 %, 66.97 %, 13.39 %, 7.16 % and 59.78 %, 1.28 %, 61.86 %, 2.08 %, 1.44 %, respectively. It is concluded that the component F022 of Radix Isatidis has the strongest anti-endotoxin effect.
4.Effect of Radix Isatidis on the expression of moesin mRNA induced by LPS in the tissues of mice.

To investigate the effect of the anti-endotoxic part of Radix Isatidis on the expression of moesin mRNA in murine tissues induced by lipopolysaccharide (LPS), the sample solution of F(022) part from Radix Isatidis was intraperitoneally administered to experimental mice, and the lipopoly-saccharide (LPS) were injected into the tail vein, and then the tissues of liver, kidney and spleen were colleted and cut into slices. The mRNA was detected by moesin mRNA hybridization in situ. The staining results were observed under microscope. It was found that moesin mRNA expression was increased in the tissues of liver, kidndy and spleen in mice treated with LPS, while in the mice pre-treated with F(022) part from Radix Isatidis, the LPS-induced moesin mRNA expressions in these tissues were inhibited in a dose-dependant manner. Our study showed that F(022) part from Radix Isatidis can inhibit the LPS-induced expression of moesin mRNA in the tissues of liver, kidney and spleen in mice.
5.The effect of intervention on blood pressure and incidence of cerebrovascular diseases

Objective To study the effect of intervention on blood pressure and incidence of cerebrovascular diseases. Methods In 1987, two cohorts population were selected in urban areas of Changsha . One was intervention group , another was control group. Baseline blood pressure levels were investigated and the events of stroke were collected. Results After 14 years, the cumulative stroke events were 89 in the intervention group and 128 in the control group; the mean blood pressure increased with statistical significance in each group except diastolic pressure in intervention group, but the control group increased more significantly; the analysis of Kaplan-Meier displayed that the rate of non-stroke events were higher in intervention group than that in control group and the analysis of COX regression indicated that the risk for stroke-events were 1.4 times higher in control group. Conclusions The intervention of risk factors can delay the increase of blood pressure by aging and reduce the risk of stroke-events.
6.Anti-endotoxic effects of syringic acid in Radix Isatidis

Object To study the anti endotoxic effect of syringic acid (SA) which was isolated from Radix Isatidis (Banlangen in Chinese, BLG). Methods SA was extracted and isolated from BLG and made to 1% solution. The content of SA pretreated ET was quantitatively determined using Limulus Test. Then the ability of fever induction of endotoxin (ET) pretreated with SA was measured using endotoxin induced fever test in rabbits. At last, the lipopolysaccharide (LPS) induced death in mice pretreated with and without SA was compared. Results Being pretreated with SA, 83.16% ET was destroyed, the ET induced fever in rabbits relieved markedly and the LPS induced death rate in mice dropped from 68% to 20%. Conclusion SA isolated from BLG has anti endotoxic effects.

8.Expression of CEA,ICAM-1 on lymphatic endothelial cells in rectum cancer of human

Objeetive:To study the expression of cell adhesion molecules on the lymphatic endothelial cells In the rectum cancer and peritumoral lymph nodes as well as their relationship with the diffusion of cancer cellS. Methods: In this Study the immunhistochemistry method was employed to observe the expression of CEA, ICAM-1 and CD31 on rectum cancer lymphatic endo-thelial cells. Results :The results demonstrated that CEA and lCAM-1 were highly expressed on the lynphatio endothelial cells in the peritumoral rectum tissues and lymph nodes. It was, howev-er,negative in normal lymphatic endothelial cells. CD31 was expressed on both lymphatic endothe-Ilal cells in the periturmral rectum tissues and lymph nodes and vessel endothelial cells of the normal human. Conelusion: The study suggests that there are relationships between the dis-semination of cancer cells and excession of CEA, lCAM-1 and CD31 in lymphatic endothelial cells.The CAE and ICAM-1 seem to induce stable adhesion between oancer cells and endothelial cells.
9.The expression of ICAM-1,CEA and CD31 in peritumoral tissues of rectal cancer

Objective:In order to study mechanism of hematogenous metastasis of rectum cancer.Methods:8 specimens of human rectum cancer and 6 specimens of rectum in normal human were examined.The immunohistochemical SP method was employed in study of the expression of ICAM1,CEA and CD31 in the peritumoral rectum tissues and lymphy nodes.Results:The intercellular role in the adhesion molecule-1(ICAM-1)and carcinoma embryonic antigen(CEA) were expressed on the vascular endothelial cells of peritumoral rectum tissues and peritumoral lymph nodes in the rectum cancer.CD31 are expressed on the vascular endothelial cells of rectum tissues from normal human with the same intensity of cancer peritumoral rectum tissues.Conclusion:This study showed that ICAM-1 and CEA seemed to play a stable role in the adhesion effect between cancer cells and endothelial cells.It is not clear whether CD31 plays a role in the interaction between cancer cells and endothelial cells.
10.Identification of a novel HLA allele, HLA-B*4086, and investigation of its family pedigree

Objective To identify a novel HLA allele, HLA-B*4086, in Chinese population and to investigate its pedigree. Methods An exceptional reaction pattern was detected in routine HLA typing of a CMDP (China Marrow Donor Programme) sample by PCR-sequence specific oligonucleotide primer (PCR-SSOP) assay. A new HLA-B allele was confirmed by sequence-based typing. Then family investigation was performed. Results DNA sequencing confirmed a new HLA allele. Compared with the closest macthing allele HLA-B*40060101, the novel allele has a difference at nt419 (A→T) in exon 3, which resulted in an amino acid change from Tyr to Phe at codon 140. Family investigation indicated the new allele derives from mother of the carrier. Conclusions One novel HLA allele was confirmed by sequencing based typing and it had been designated as HLA-B*4086 by the WHO Nomenclature Committee. This novel allele was inherited from mother of the carrier.
