1.The intervening effect of Qi-Boosting Toxin-Resolving Formula on migration potentiality of nasopharyngeal carcinoma cells and its association with the expression activities of cellular skeletal protein system

Objective To investigate the intervening effect of Qi-Boosting Toxin-Resolving Formula (QBTRF) on the migration potentiality of nasopharyngeal carcinoma (NPC) cells and its association with the expressive activities of cellular skeletal protein system based on an experimental study in vitro.Methods In this study,QBTRF medicine-containing plasma of rats was taken as the intervening factor to treat the targeted NPC cells of CEN2Z for evaluating the correlation of changes in migrating potentiality of CEN2Z cells with expression activities of cellular skeletal protein system and their response to the pharmacological intervening effects of QBTRF,compared with the normal plasma of rats as the control.Here,scratching test was taken to evaluate the migration potentiality of targeting cells,immunohistochemical SABC assays were used to determine the expression activities of β-tubulin,F-actin,p-Ezrin and Rac1,which were taken as the typical indicators of cellular skeletal protein system.Scanning electron microscopy was selected to observe the changes in cellular pseudopodia.Reverse transcription-polymerase chain reaction (RT-PCR) procedure was used to detect the transcriptional activity of p-Ezrin mRNA.Results QBTRF showed a very significant inhibitory effect on the migration potentiality of CEN2Z cells,with its migration distance reduced to (25.80 ±8.07) μm and an inhibitory rate reached at 46.59% at the concentration of IC50.Meanwhile,their cellular pseudopodia was shortened significantly,accompanied with very reduced extension length.Obviously decreased were the expression activities of β-tubulin,F-actin,p-Ezrin and Rac1 proteins,with their figure gray values being 0.857 ±0.059,0.900 ±0.040,0.812 ±0.050 and 0.785 ±0.022,respectively,with statistical significance compared with controls (P <0.01).Also seen was the same tendency of transcriptional level of p-Ezrin mRNA when cells cultured with QBTRE medicines-containing plasma together.Conclusions QBTRF holds significantly inhibitory effect on the migration potentiality of CEN2Z NPC cells.This kind of inhibitory effect may be associated with the reduced biological activity of cellular skeletal protein system and corresponding cellular pseudopodia changes.
2.Impact of palliative radiotherapy to the prognosis of patients with stage Ⅳ non-small cell lung cancer

Objective To investigate the impact of palliative radiotherapy to the prognosis of patients with stage Ⅳ non-small cell lung cancer (NSCLC).Methods Two hundred and forty-four cases of stage Ⅳ NSCLC were collected from our hospital.The study group (n =129) received the treatment of primary tumor and metastases palliative radiotherapy after chemotherapy.The control group (n =115) received only chemotherapy.Followed-up and compared 1-year and 2-year survival rate of the patients,and explored the factors affecting palliative radiotherapy.Results The 129 patients in the study group were followed up for 2 years,and 16 patients survived and 110 patients died.Three patients were lost to follow-up.The median survival time was 8 months.1-year and 2-year survival rates was 29.46% (38/129) and 12.40% (16/129) respectively.In the control group,7 patients survived,and 106 patients died.Two patients were lost to follow up.The median survival time was 6 months.1-year and 2-year survival rates was 11.30% (13/115) and 6.09% (7/115)respectively.Survival rates between the two groups were significantly different (x2 =8.451,P =0.014)Univariate regression analysis found that factors affecting the prognosis of stage Ⅳ NSCLC were KPS score (x2 =5.057,P =0.031),occurrence of brain metastases (x2 =4.781,P =0.029),number of organs of metastasis (x2 =6.341,P =0.010),and the primary stove radiotherapy dose (x2 =5.893,P =0.015) ; Cox regression model analysis showed that the number of organs of metastasis (HR =1.719,95% CI 为 1.172-3.126,P =0.008),the primary focal radiotherapy dose (HR =1.560,95% CI 为 1.082-2.761,P =0.022) were independent prognostic factors.Conclusion Palliative radiotherapy plays a role of prolonging survival time in the treatment of Ⅳ NSCLC.In palliative radiotherapy,we should pay attention to the control of the primary tumor and metastases radiotherapy dose,therefore prolonging the survival time of patients.
3.Study on the Relationship between Severity of Coronary Artery Lesion and the Level Cystatin C,Nitric Oxide and Superoxide Dismutase of Serum

Objective To study the correlation between coronary lesion and Cystatin C (CyC),Nitric Oxide (NO)and Super oxide dismutase,accumulate the clinical experience in prevention and diagnose of Coronary Heart Disease (CHD)by analy-zing the contents of serum Cystatin C (CyC),Nitric Oxide and Superoxide Dismutase in patients with coronary lesion.Meth-ods According to their severity of coronary artery lesion,all cases were divided into four groups:Normal compared group (40 cases);Simple branch pathological group (44 cases);Double branch pathological group (43 cases):Multi-branch patho-logical group (35 cases).All cases were tested CyC,NO,SOD and hs-CRP in serum.Results The level of CyC and hs-CRP were increased gradually from normal group to multi-branch group.The level of NO and SOD were decreased gradually from normal group to multi-branch group.There were significant differences the levels of CyC,NO,SOD and hs-CRP among the four groups.Coronary lesion Gensini in the CHD patients had significant positive relationship to the level of CyC or hs-CRP respectively (r=0.473,0.429),had significant negative relationship to the level of NO or SOD respectively (r=-0.356, 0.384)(P<0.05).CyC in the CHD patients had significant positive relationship to the level of hs-CRP (r=0.458),had sig-nificant negative relationship to the level of NO or SOD respectively (r=-0.426,0.484,P<0.05).Conclusion By aggra-vation of coronary artery lesion,the level of CyC increased,reduced the contents of NO,SOD,aroused endothelial dysfunc-tion,induced occurrence and aggravation of coronary artery lesion.CyC,NO and SOD play important mark in the assistant diagnosis of the disease.
4.Bibliometric Analysis on Antidepressive Agents in PubMed Database During 1998~2008

OBJECTIVE:To evaluate the research of antidepressive agents from the literatures perspectively in order to develop antidepressive agents in China.METHODS:The literatures about antidepressive agents recorded in PubMed database from Jan.1st,1998 to Oct.31th,2008 were analyzed with bibliometric methods.RESULTS:A total of 17784 literatures on antidepressive agents were recorded and the total number increased slowly.A total of 96 literatures were collected from Chinese literatures which increased rapidly in recent years.34 literatures among 74 literatures during 1998~2007 are in English and 40 in Chinese involved 56 institutes and 51 magazines.Literatures collected from Beijing took up the greatest proportion.CONCLUSION:It is needed to enlarge investment and scale of scientific research on antidepressive agents continuously.

6.Effects of total flavonoids from astragalus complanatus and endoplasmic reticulum stress-induced cell apop-tosis in acute lung injury following paraquat poisoning in rats

Objective Paraquat(PQ) is an effective herbicide which is widely used in agricultural production .PQ poisoning is frequently seen in humans with the lung as the target organ ,but the poisoning mechanisms is not very clear .Studies show that endoplasmic reticulum stress(ERS) is closely associated with poisoning , but there are few reports on the relationship between ER stress and PQ poi-soning.This article was to investigate the effects of ERS-induced apoptosis and total flavonoids from astragalus complanatus (FAC) in a-cute lung injury(ALI) following paraquat poisoning in rats . Methods A total of 30 adult healthy Sprague-Dawley (SD) rats were ran-domly divided into 3 groups:control group, ALI group, ALI+FAC group and ALI+saline group.Biochemical method was applied to de-tect superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) in lung tisssue,TdT-mediated dUTP nick end labeling (TUNEL) assay in observing lung apoptosis, Western blotting and real-time PCR(RT-PCR) in detecting the changes in expressions of C/EBP homologous peotein (CHOP), activating transcription factor 4 (ATF4) and X-box binding protein 1 (XBP) following ALI, and HE staining in observing the pathological changes of lung tissue . Results Compared with control group , the expression of MDA content was enhanced in ALI group ([3.26 ±0.24] vs [5.04 ±0.36],P<0.01), along with significantly decreased activity of SOD and CAT ([300.26 ±35.69] vs [187.21 ±25.66]), ([5.78 ±1.28] vs [2.15 ±1.12],P<0.01), increased cell apoptosis , upregulated pro-tain level of CHOP ([0.74 ±0.20] vs [0.23 ±0.07],P<0.01) and mRNA expression of ATF4, XBP1 and CHOP.However, FAC sig-nificantly attenuated ALI following PQ , as showed by reduced MDA content , enhanced activity of SOD and CAT , decreased cell apopto-sis, inhibited protain level of CHOP and mRNA expression of ATF 4, XBP1 and CHOP ([5.04 ±0.36] vs [3.99 ±0.27],P<0.01). Furthermore, the activity of SOD and CAT were higher in FAC pretreatment group than those in ALI group ([ 0.74 ±0.20 ] vs [0.42 ±0.11],P<0.01). Conclusion From the research, ERS-induced cell apoptosis is involved in ALI following PQ , and the protec-tive role of FAC in lung tissue following PQ is due to its effect in atten-uating ERS-induced apoptosis .
7.Whole Body Simulation and Visualization of Metabolic Process of ~(18)F 2-fluoro-2-deoxy-D-glucose

Objective To simulate the metabolic distribution process of 18F 2-fluoro-2-deoxy-D-glucose(FDG) in human body and to visualize this distribution process through 3D images with high resolution and high quality. Methods The model parameters of FDG metabolism in tissues were estimated through clinical experiments, and the curves which represent the FDG metabolic process in tissues were calculated using the model parameters and blood input function. This FDG distribution process in human body was visualized basing on the high-resolution anatomical structure. Results The simulation and visualization results directly and clearly displayed the FDG metabolic distribution process after injection to human body. The properties of the FDG distribution process represented by our simulation were consistent with that represented by clinical experiment. Conclusion The method presented in this study is effective to simulate and visualize human functional information of metabolism, and it may provide a useful tool for education and research on nuclear imaging.

9.Study on experimental autoimmune encephalomyelitis with oxymatrine on rats model

Objective To establish experimental autoimmune cerebral spinal cord inflammation ( EAE) model rats, and observe the pathological changes and effect of oxymatrine on EAE model.Methods 30 rats were randomly divided into control group,EAE ( model group) group and oxymatrine group.The EAE symptom score was used to evaluate the rats after the model, and to observe the changes of its behavior.By HE staining and Kluver &Barrera myelin dyeing to observe the inflammation of the brain and spinal cord demyelinating changes.Results The animals in control group had no change in behavior and pathological.In model group, all animals occured behavioral changes, accompanied by varying degrees of demyelination and inflammatory infiltration of the brain and spinal cord.In oxymatrine group,6 rats did not appear EAE clinical manifestations and behavioral change, and the myelin structure was intact.Conclusion Oxymatrine can extend the incubation period experimental autoimmune encephalomyelitis in rats of the disease, relieve symptoms and protect nerve.
10.Posterior reversible encephalopathy syndrome:report of six cases

ObjecfiveTo investigate clinical features and imaging manifestation in patients with posterior reversible eneephalopathy syndrome (PRES) to improve its recognition.MethodsSix patients with PRES were enrolled,four women with history of end-stage renal disease,kidney transplantation,eclampsia,or systemic lupus erythematosus (SLE) and two men with history of chemotherapy or hypertension.All of them underwent multi-serial MR imaging (T1 WI,T2 WI,FLAIR) and post-contrast T1 WI.Three cases also underwent CT scan and gadolinium enhancement. ResultsAll the six cases of PRES had different inducing causes such as acute hypertension,preeclampsia or eclampsia, taking immunosuppressive agents or steroids.and their clinical symptoms were characterized by sudden occurrence of headache,eclampsia or seizure of epilepsy,altered melltal status,visual disturbances.Clinical symptoms were died out in about one week after prompt and appropriate treatments for high blood pressure.or removal of precipitating factors,or treatment for epileptic seizures or status epilepticus.MRI and CT scanning demonstrated multifocal subcortical white lesions in bilateral parieto-occipital lobes (six cases), bilateralfrontal lobes (two cases),bilateral post temporal lobes (two cases) and left cerebellum (one cases).and cortical involvement (two cases).All lesions appeared unenhanced with gadolinium enhancement. FoHow-up by MRI showed decreased abnormal signs and small infarct foci were left in the cortex-subeortex of one case.ConclusionsPRES is a clinico-neuroradiologieal transient condition, usually benign and reversible in nature.Completely clinical and radiographic recovery Can be achieved with prompt antihypertensive treatment or removal of precipitating factors and supportive care,but delayed diagnosis and therapy Can result in cerebral infarct with neurological sequelae.
