2.Change of interleukin-23 level in mouse asthma model and its action mechanism

Objective To measure the change of serum interleukin-23(IL-23) level before and after treatment with glucocortcoid and analyze the correlations between levels of IL-23 and IL-4,IFN-?,IL-17,and explore the role of IL-23 in bronchial asthma.Methods Thirty femal BALB/c mice were randomly divided into three group(n=10): group A(asthma model),group B(corticosteroid treatment) and group C(normal control).All mice were killed 24 h after final OVA challenge.The blood samples were obtained for measurements of serum IgE,IL4,IFN-?,IL-17and IL-23 levels by enzyme-linked immunoabsorbent assay(ELISA).Results The mouse asthma model appeared ethological changes specific to asthma.The level of OVA specific IgE in serum increased.The level of serum IL-4 increased and the level of IFN-? decreased.Correlation analysis showed negative relationship between IL-4 and IFN-?(r=-0.859,P
3.Effect of Deep-brain Magnetic Stimulation on Learned Helplessness Behavior in Rats with Chronic Restrained Stress

Objective To explore the curative effect of deep-brain magnetic stimulation (DMS) on learned helplessness behavior in the chronic restrained stress (CRS) rat model. Methods Twenty-nine Sprague-Dawley rats were randomly divided into control group (n=8) and CRS group (n=21). CRS group was exerted chronic restrained stress, while the control group did not receive any stress, for three weeks. Then learned helplessness behavior was tested using Forced Swimming Test (FST) and the hopeless rats of the CRS group were divided ran-domly into sham group (n=6), DMS group (n=8) and citalopram group (n=7), that received corresponding treatment respectively. They were evaluated with FST again after one-week treatment. Results The immobile time in FST was longer in CRS group than in the control group after three-week stress (F=11.260, P=0.002). After one-week treatment, no significant improvement was found in the citalopram group (F=1.565, P=0.235), however, the immobile time in DMS group decreased (F=6.277, P=0.025), and was shorter than that in the sham group (F=5.560, P=0.036). Conclusion CRS could result in learned helplessness behavior, which could be alleviated with one-week DMS.
4.Comparation of immunohistochemistry and fluorescence in situ hybridization for detection of Her-2 expression in gastric cancer

Objective To investigate the association of Her-2 protein expression and gene expression in gastric cancer by different methods,and to explore the effective approaches of Her-2 in gastric cancer detection.Methods Immunohistochemistry (IHC) was used to mark Her-2 protein in 68 cases of gastric cancer,and fluorescence in situ hybridization(FISH)wasused to detect the Her-2 gene amplification.Results Among 68 cases,IHC Her-2 protein in 12 cases(17.65 %)was positive expression,including ++ 3 cases ++ 9 cases,+ 7 cases and-49 cases.By FISH,Her-2 gene in 11 cases(16.17 %)was overexpression.Corresponding to the IHC results,FISH results was 3 cases(100 %),8 cases(88.89 %),0 cases,0 cases,respectively.Conclusion Her-2 protein expression and gene overexpression occur in gastric cancer.Her-2 protein expression in gastric cancer shows significant heterogeneity.Her-2 gene overexpression and Her-2 protein expression is correlated (P<0.05).The differences of Her-2 expression in gastric cancer by IHC and FISH mostly occur in IHC++ groupe.Suggestion of Her-2 gene test should be given for patients with IHC++ who will be undertaken Her-2 gene therapy.
5.Open reduction and internal fixation for treatment of Lisfranc joint injury at early stage

Objective To summarize the clinical experience in treatment of the Lisfranc joint injury with open reduction and internal fixation at early stage. Methods Twelve patients ( including ten males and two females at average age of 34 years) with early stage Lisfranc joint injury received open reduction and screw/wire fixation from 2005 to 2010. According to the Myerson classification, there were two patients with type A, eight with type B and two with type C. All the patients received open reduction and internal fixation with screw or Kirschner wire within 17 days after injury. The post-operative function was estimated by mid-foot scoring scale of AOFAS. X-ray and CT scan were used in radiography estimation. Results All the patients were followed up for average 33 months ( range, 6-60 months). The mean score of post-operative mid-foot scoring scale of AOFAS was 74.5 points ( range, 53-96 points), with excellent result in eight patients, good in two and fair in two. The anatomical reduction was observed in eight patients and all the patients obtained bony union according to the results of X-ray and CT scan.There was no any complication found.Conclusions Open reduction and internal fixation is a good choice for the treatment of Lisfranc joint injury at early stage. A preoperative comprehensive analysis combined with clinical X-ray and CT scan is necessary.
6.Study on the metabonomics of vitamin A deficiency urine of left-behind children under 1 year old in Zunyi area based on hydrogen-nuclear magnetic resonance

Objective:Explore the hydrogen-nuclear magnetic resonance ( 1H-NMR) application potential of metabolomics in early screening of vitamin A deficiency(VAD). Methods:From January to June 2019, 40 cases of left-behind children with confirmed VAD <1 year old were enrolled in Zunyi area as VAD group, and 40 healthy children were selected as healthy control group.Urine from the 2 groups of children was collected and 1H-NMR spectrum was measured.By combining metabonomics technology and multivariate statistical analysis, the 2 groups of metabolites were compared, and the differential metabolites of children with VAD were screened out. Results:Compared with VAD group [(0.56±0.10)μmol/L], the difference in the serum retinol concentration of healthy control group [(1.35±0.18)μmol/L] was statistically significant ( t=23.93, P<0.001). Compared with healthy group, VAD group showed higher level of methylmalonate, 2-ketoisovalerat, N-acetyl glycoprotein signal, dimethylglycine and betaine, with statistically significant differences (| r|>0.602, all P<0.05). The level of Isovalerylglycine, 2-hydroxyisobutyrate, creatine, phosphate choline/glycerophosphocholine, 4-PY and Hippurate were significantly decreased, with statistically significant differences (| r|>0.602, all P<0.05). Conclusions:Compared with healthy children, children with VAD have differences in urine Metabolites.Differential metabolites are closely correlated with intestinal microecological ba-lance, digestive system diseases, respiratory system diseases, immune-related diseases, energy metabolism, growth and development.Urine 1H-NMR metabolomics analysis has potential application value in the early screening of VAD.
7.Expressions of Twist transcription factor and E-cadherin in anaplastic thyroid carcinoma and their relationship with the prognosis

Objective To investigate the expressions of Twist transcription factor and E-cadherin in anaplastic thyroid carcinoma and their correlation with the prognosis. Methods The clinicopathological data of 20 patients with anaplastic thyroid carcinoma in Shanxi Provincial Cancer Hospital from May 2016 to April 2018 were retrospectively analyzed. The expressions of Twist protein and E-cadherin protein in anaplastic thyroid carcinoma tissues and corresponding paracancerous tissues were detected by using immunohistochemistry. The relationship between the expressions of the two proteins and the clinicopathological features of the patients was analyzed, and the correlation among the expressions of the two proteins as well as the clinicopathological factors and the prognosis of the patients was analyzed. Results The positive expression rate of Twist protein in anaplastic thyroid carcinoma tissues was 65% (13/20), while it was 20% (4/20) in paracancerous tissues, and the difference was statistically significant (P= 0.004). The positive expression rate of E-cadherin in anaplastic thyroid carcinoma tissues was 60%(12/20), while it was 100%(20/20) in paracancerous tissues, and the difference was statistically significant (P = 0.001). There was a negative correlation between the expression of Twist protein and E-cadherin protein in anaplastic thyroid carcinoma (r=-0.685, P=0.001). The expression of Twist protein in anaplastic thyroid carcinoma was correlated with lymph node metastasis and TNM stage (both P< 0.05), while it had no correlation with gender, age and tumor diameter (all P> 0.05). The expression of E-cadherin protein in anaplastic thyroid carcinoma had no correlation with clinicopathological factors (all P> 0.05). The results of Kaplan-Meier survival analysis showed that lymphnode metastasis (P< 0.01), TNM stage (P = 0.002) and the expression of Twist protein (P = 0.017) were prognostic factors in patients with anaplastic thyroid carcinoma. Conclusions The expression of Twist transcription factor is upregulated in anaplastic thyroid carcinoma, and the expression of E-cadherin is downregulated. The expression of Twist protein is correlated with the prognosis of the patients. The epithelial-mesenchymal transition may be involved in the progress of anaplastic thyroid carcinoma.
8.Clinical value of preoperative 18F-FDG PET-CT imaging in staging of colorectal cancer

Objective To discuss the value of preoperative 18F-FDG PET-CT imaging in the staging of colorectal cancer. Methods Eighty-three patients with colorectal cancer were studied who underwent 18F-FDG PET-CT examinations without any previous treatment and subsequent operations within one week. The results of PET-CT and pathology were analyzed with chi-square test and Kappa consistency test. Results The diagnostic accuracy of primary colorectal lesions was 100 %, the average of maximum standardized uptake value(SUVmax) was 14.54±8.10(4.43-48.19). The diagnostic accuracy of local infiltration depth, lymph node metastasis, distant metastasis and TNM staging before operation were 73.49 %(61/83), 90.36 %(75/83), 97.59 % (81/83) and 85.54 % (71/83), respectively, and the Kappa values were 0.481, 0.797, 0.950, 0.788 (P <0.05).The diagnostic sensibility of staging with T1-2,T3and T4were 30.00 %(3/10),68.42 %(13/19) and 83.33 % (45/54), respectively, the diagnostic accuracy of staging with T1-2, T3and T4were 91.57 % (76/83), 74.70 % (62/83) and 80.72 % (67/83). Conclusion Preoperative 18F-FDG PET-CT imaging is an effective staging method for colorectal cancer, and has high accuracy in the detection of primary colorectal lesions, lymph node metastasis, distant metastasis and T4stage lesions, but it is difficult for diagnosing of the T1-2and T3lesions.
9. Clinical analysis of lung transplantation in eight patients with obstructive bronchiolitis syndrome after hematopoietic stem cell transplantation

Objective:
To investigate the efficacy and prognosis of lung transplantation (LT) for end-stage bronchiolitis obliterans syndrome (BOS) after allogeneic hematopoietic stem cell transplantation (allo-HSCT) .
Methods:
The clinical data of eight cases with end-stage BOS after allo-HSCT who were treated by LT in our hospital were retrospectively analyzed.
Results:
Eight patients with hematological malignancy underwent allo-HSCT, and the median age was 23 (12-40) years. The donors are parents or siblings. Severe BOS occurred in 8 patients after allo-HSCT, the median age for LT was 27.5 (13-47) years. The median interval between allo-HSCT and LT was 69 (21-132) months. The median follow-up time for 8 patients after LT was 15 (6-63) months, 7 patients survived, 1 patient died of pulmonary hemorrhage 15 months after LT treatment. Of the survivors, three had BOS again, and one of them received reduplicated lung transplantation.
Conclusion
LT is an effective treatment for patients with severe BOS after HSCT.
10.Atypical MSCT pleomorphic signs in metastatic adenocarcinoma and squamous cell carcinoma of the lung

Objective:To investigate the correlations between multi-slice spiral CT (MSCT) atypical pleomorphic signs and pathological findings of lung metastases.Methods:From January 2012 to July 2019, the MSCT chest imaging data of 168 metastatic tumor of lung from the General Hospital of Central Theater Command of the Chinese People′s Liberation Army and Shaanxi Provincial Tumor Hospital were collected. According to the pathological type, they were divided into metastatic adenocarcinoma group ( n=88) and metastatic squamous cell carcinoma group ( n=80). The atypical imaging signs of MSCT of the two groups were observed and recorded, and classified after labeling one by one. The difference of atypical MSCT imaging features between the two groups was compared, and the correlations between lesion size and atypical imaging features of MSCT in the metastatic adenocarcinoma group and metastatic squamous cell carcinoma group were analyzed. Results:The spicule sign in metastatic adenocarcinoma and metastatic squamous cell carcinoma were 61 (69.32%) and 28 (35.00%), with a statistically significant difference ( χ2=19.811, P<0.001). The pleural depression sign in the two groups were 48 (54.55%) and 16 (20.00%), and there was a statistically significant difference ( χ2=21.206, P<0.001). The vacuole/cavity sign in the two groups were 10 (11.36%) and 61 (76.25%), and there was a statistically significant difference ( χ2=72.303, P<0.001). The air bronchial sign in the two groups were 43 (48.86%) and 13 (16.25%), with a statistically significant difference ( χ2=20.057, P<0.001). The halo sign/ground glass shadow in the two groups were 58 (65.91%) and 37 (46.25%), with a statistically significant difference ( χ2=6.591, P=0.010). The results of the Spearman rank correlation analysis indicated a positive correlation between the size of metastatic adenocarcinoma and spicule sign, pleural depression sign ( r=0.270, P=0.011; r=0.226, P=0.035). There was no correlation between the nodule size and atypical MSCT imaging features in metastatic squamous cell carcinoma (all P>0.05). Conclusion:The atypical MSCT of metastatic lung adenocarcinoma are mostly spicule sign, pleural depression sign, air bronchial sign and halo sign/ground glass shadow. The characteristic atypical imaging of metastatic squamous cell carcinoma is vacuole/cavity sign. The spicule sign and pleural depression sign are related to the size of metastatic lung adenocarcinoma nodules.
