AIM To study the therapeutic effects of Deketoprofen trometamol (D KPT) on adjuvant arthritis (AA) and its side effects on gastroduodena. METHEDS The change of primary and secondary inflammatory and the injury effect of gastroduodena treated with D KPT were observed. The effect of D KPT on prostaglandin E 2 (PGE 2) produced by peritoneal macrophage (PM) on AA rats in vitro was also observed. RESULTS D KPT (2 5, 5, 10 mg?kg -1 ) significantly inhibited primary inflammatory, secondary inflammatory and multiple arthritis of AA rats. D KPT(10 -8 ,10 -7 ,10 -6 ,10 -5 ,10 -4 mol?L -1 )inhibited the elevated PGE 2 released from PM of AA rats in vitro . D KPT (10 mg?kg -1 ) significantly injured gastroduodena on AA rats, but the degree of injuries was less than that of ketoprofen (KP) (10 mg?kg -1 ). CONCLUSION D KPT has therapeutic effects on AA rats, and its injury effect on gastroduodena is less than that of KP.

Objective:To investigate the effects of different doses of rosuvastatin on lipid levels in elderly patients with CHD complicated with hyperlipidemia.Methods:120 elderly patients with coronary heart disease admitted in our hospital were retrospectively chosen.By lipid examination,they were all hyperlipidemia.According to the dose of rosuvastatin,they were divided into three groups,and 40 cases in each group.They were given 5 mg,10 mg and 20 mg treatment,respectively.After 2 months,the changes of blood lipid level and the clinical effective rate were compared before and after the treatment.Results:①The blood lipid levels HDL-C [(1.17 ± 0.62) mmol / L] was significantly higher than the other two groups.LDL-C [(3.67 ± 0.83) mmol / L],TC [(4.36 ± 0.96) mmol / L] and TG [(1.68 ± 0.94) mmol / L] were significantly lower than those of the other two groups (P ＜0.05).②The effective rate of treatment in the 20mg group (97.5％) was significantly higher than that in the 5mg group (87.5％) and the 10mg group(85.0％).Conclusions:Rosuvastatin can treat coronary heart disease complicated with hyperlipidemia in elderly patients.The dosage of 20 mg can improve the blood lipid level of the patients,is an effective and convenient method,and with high therapeutic efficiency.

OBJECTIVE: To discuss the new work mode of clinical pharmacists participating in ward round and to promote rational use of antibiotics. METHODS: The status quo of clinical pharmacists participating in analysis of antibacterials used before and after ward round was analyzed statistically. RESULTS: After pharmacists participating in ward round, the cost ratio of antibiotics to total drugs was decreased by 5.56%. Rationality of antibacterial used in perioperative period or by infectious patients was improved significantly. CONCLUSIONS: The work mode of clinical pharmacists participating in ward round is effective and feasible, which plays a key role in promoting rational use of antibacterial and medical quality.

Objective To investigate the effect of long hairpin RNA ( lhRNA) expression vector targeting HBV X gene ( HBx) on replication of hepatitis B virus ( HBV) and gene expression.Methods Four kinds of small interference RNAs ( siRNAs) were synthesized and lhRNA expression vectors targeting HBx were constructed.Four siRNA oligonucleotides and two lhRNA expression vectors were transfected into HepG2.2.15 cells.HBsAg, HBV DNA in culture supernatants and HBx mRNA in HepG2.2.15 cells were detected by time-resolved immunofluorometric assay, real-time quantitative PCR, and reverse transcription PCR, respectively.Negative sequence group or empty vector group was taken as the control.Independent-samples t test was performed to evaluate the inhibition effect on replication of HBV and gene expression. Results Compared with the negative control, HBsAg, HBV DNA level in culture supernatants and HBx mRNA in HepG2.2.15 cells were significantly decreased after siRNA-1 and siRNA-4 transfected at high concentrations (60 nmol/L or 90 nmol/L) (P<0.05), especially the HBsAg and HBV DNA levels in the siRNA-1 transfection group, which were significantly decreased at 24, 48 and 72 h after transfection ( P<0.05 or P <0.01 ) . Two lhRNA expression vectors ( pMD-HBxlh1 and pMD-HBxlh4 ) were successfully constructed and transfected into HepG2.2.15 cells, HBsAg and HBV DNA level in transfected cells was significantly lower than those in negative control (P<0.05).Conclusion The novel siRNA-1 is confirmed to target HBx gene and lhRNA expression vector targeting HBx can effectively inhibit the replication of HBV and expression of HBV gene.

AIM: To study the antipyretic, analgesic and antiinflammatory actions of dexketoprofen.　METHODS: Hot-plate and tail-flick models in mice, fever model in rats and rabbits induced by carrageenin or typhoid vaccine respectively, ear swelling induced by dimethylbenzene in mice; foot pad swellings induced by carrageenin and granuloma induced by cotton in rats were adopted.　RESULTS: Dexketoprofen (7.5,15,30 mg*kg-1) could significantly prolong latency of analgesic in hot-plate and tail-flick models in mice and reduce carrageenin-induced fever in rats, and inhibit dimethylbenzene-induced ear swelling in mice. Dexketoprofen (1.9,3.8,7.5 mg*kg-1) could significantly inhibit typhoid vaccine-induced fever in rabbits; Dexketoprofen (3.8,7.5,15 mgkg-1) could also significantly inhibit the swelling of foot pad induced by carrageenin, and the granuloma induced by cotton in rats.　CONCLUSION: Dexketoprofen has antipyretic, analgetic and antiinflammatory effect.

AIM To establish the model of type Ⅱ collagen(CⅡ) induced arthritis in mice. METHODS The type Ⅱ collagen induced arthritits(CIA) mice were induced by intradermal injection with CⅡ and complete Freund adjnvant emulsion. RESULITS In comparison with control mice, the arthritic swelling and index were significant increased. The responses of CⅡ induced delayed type hypersensitivity were remarkably positive, and IgG anti bodies to CⅡ in serum could be detected in CIA mice. CONCLUSION The model CIA mice was successfully established.

AIM To study the antiinflammatory and immunomodulatory effects of actarit(Acta). METHODS To abserve the change of primary and secondary inflammation,immune function on adjuvant arthritis(AA) rats treated with Acta and to detect the effect of Acta on T cell subgroup in mice in vitro. RESULTS Acta(10,30,90 mg?kg -1 ) intragastric injection(ig) did not significantly inhibit primary reaction of AA in rats. But Acta(10,30,90 mg?kg -1 ?11 d) ig signifcantly inhibited secondary reaction (secondary inflammatory swelling、 multiple arthritis) of AA rats. The lowered response of ConA induced splenolytes and the decreased IL 2 synthesis were restored to normal in AA rats treated with Acta, while the elevated IL 1 released from peritoneal macrophate (PM?) was reduced. Acta(1～100 ?mol?L -1 ) could inhibit ConA induced T h cells and enhance ConA induced T s cells culture in vitro . CONCLUSION Acta do not show obvious antiinflammatory action. But Acta has therapeutic action on AA rats, which may be related to its immunomodulatory activities,mainly through modulate cell mediated immunity.

Objective To compare the detection performance of the modified serum test (TRUST) method and the traditional method in syphilis screening.Methods A series of TRUST high titer syphilis serum was diluted,and the positive rate of each method was calculated by using the improved method and the traditional method.Comparison of two detection methods of C50,C5 ～ C95 interval,as well as the accuracy of the density curve,and the consistency of the two methods were compared,and diagnostic performance were compared.Results The improved method of C50 was less than the traditional method of C50,and the improved method of C5 ～ C95 range was narrower than the traditional method,compared with the traditional method.The improved method of the non precision density curve was steeper than the traditional method,and the two confidence interval of the consistency degree of the 95％ methods was 73.4％ to 95.8％.The diagnostic sensitivity (SEN),diagnostic specificity (SPE),positive predictive value (PPV),negative predictive value (NPV) and diagnostic efficiency(DF) of the improved method were 64.29 ％,99.1％,85.71％,97.05 ％ and 96.39 ％,respectively.The SEN,SPE,PPV,NPV and DF of the traditional methods were 3.75 ％,98.49 ％,75 ％,96.18 ％ and 95 ％,respectively.The improved method was superior to the traditional methods in the two aspects of SEN and PPV (x2 =8.25,10.03,all P＜0.05),with the statistically significant difference.The improved method was slightly higher than the traditional method in SPE,NPV and DF (x2 =2.39,3.45,4.03,all P＞0.05),with the no statistically significant difference.Conclusion The precision,diagnostic sensitivity and diagnostic specificity of the improved method was higher than that of the traditional method,and it can be applied to the detection of large batch samples with the aid of the full automatic enzyme immunoassay instrument.The improved method can be used to replace the traditional method for syphilis screening.

Objective To explore the dilution regression method of coagulation detection(PT ,APTT) in fat blood samples . Methods We collected 40 normal blood coagulation specimens (no fat blood ,no jaundice ,no hemolysis) in Yan′an hospital of Kun-ming ,then we detected the PT and APTT of the original plasma and 3-fold diluted plasma and 5-fold diluted plasma ,the we used the data both of before dilution and diluted to do the linear regression analysis ,and finally we got the regression equations of each index .we also collected 33 fat blood samples in Yanan hospital of Kunming ,which be divide into three groups through the severity of triglycerides :mild fat blood group(1 .7 mmol/L≤TG<11 .0 mmol/L) and moderate fat blood group(11 .0 mmol/L≤TG<20 .0 mmol/L)and severe fat blood group(TG≥20 .0 mmol/L) ,then we detected the PT and APTT after 3-fold diluted plasma and high-speed centrifugation plasma ,and then we brought diluted results into the normal regression equations and the results were compared with the high-speed centrifugation results .Results Because most of the 5-fold diluted plasma can not get the effective results ,so we use 3-fold diluted plasma to get the regression equations .The results of 3-fold diluted plasma was calculated by the regression equa-tions ,which then compared with high-speed centrifugation results ,after the analysis of statistical software ,two results had not sta-tistically significant (P>0 .05) .Conclusion Dilution regression method can be used to detect the fat blood samples in the clinical coagulation detection .
