2.Study on the drug resistance of 45 hacemophilus par lainfluenzae in respiratory tract infection

Objective To investigate the infectious state and drug resistance of Haemophilus par lainfluenzae in respiratory tract infection, so as to instruct the rational use of antibiotics. Methods Haemophilus par lainfluenzae were isolated from the pa-tients with respiratory tract infection and performed by K-B methods. Nitrocefin slip test was applied into detecting β-lactamase. Re-sults A total of 45 Hacmophilus par lainfluenzae were isolated from 760 specimen, with the isolation rate being 5.92%. Amoxicil-lin/claulanic acid,Azithromycin, Imipenem,Ceftazidime and Cefuroxime had the better antibiotic function and the average drug re-sistance was lower than 5 % ,The β-lactamase positive rate was 37.78 %. Conclusion The high drug resistance and β-lactamase pos-itive rate of Haemophilus par lainfluenzae isolated from respiratory tract characterized by multi-resistance must be taken seriousely.

8. The effect of a small interfering RNA targeting STAT3 gene on the biological function of human endometrial cancer cell line HEC-1B

Objective: To investigate the effect of a small interfering RNA (si-RNA) targeting signal transducer and activator of transcription-3 (STAT3) on the biological function of human endometrial cancer cell line HEC-1B. Methods: The HEC-1B cells were transfected with chemically synthesized si-RNA targeting STAT3 (si-STAT3). Then the expression of STAT3 mRNA was detected by real-time fluorescence quantitative-PCR, the proliferation ability was determined by cell counting kit-8 (CCK-8) assay, the cell cycle distribution and apoptosis were measured by flow cytometry, the migration and invasion were examined by Transwell chamber, and the expression levels of STAT3, phospho-STAT3 (p-STAT3), and cyclin D2 (CCND2) and metalloproteinase-2 (MMP-2) were detected by Western blotting. Results: As compared with the blank control group (only liposome was added) or transfection with si-control group, the expression level of STAT3 mRNA in HEC-1B cells after transfection with si-STAT3 was down-regulated (P < 0.01), the proliferation ability was inhibited (P < 0.01), the percentage of the cells in G1-phase was increased and which in S1-phase was decreased (P < 0.01). The abilities of migration and invasion of HEC-1B cells after transfection with si-STAT3 were inhibited (P < 0.01), and the expression levels of STAT3, p-STAT3, CCND2 and MMP-2 proteins were down-regulated (P < 0.01). Conclusion: Si-RNA targeting STAT3 gene can inhibit the expressions of STAT3 mRNA and protein in HEC-1B cells, and also inhibit the cell proliferation, the abilities of migration and invasion, and apoptosis. 9.Observation of clinical efficacy of S-1 combined with oxaliplatin in neoadjuvant chemotherapy for local advanced gastric carcinoma

Objective To observe the clinical efficacy of S-1 plus oxaliplatin (SOX) in neoadjuvant chemotherapy for patients with local advanced gastric carcinoma.Methods Clinicopathological data of 68 patients with local advanced gastric cancer were retrospectively analyzed and enrolled as treatment group.All the patients in the treatment group achieved 3-4 cycles chemotherapy of SOX regimen preoperatively.Outcomes of tumor remission after neoadjuvant chemotherapy were evaluated according to RECISI1.1 criteria.Meanwhile,70 patients who underwent operation directly after pathological diagnosis of local advanced gastric cancer during the same period were recruited as control group.Resection rate,R0 resection rate,postoperative complications rate and postoperative progression free survival(PPFS) time were compared between the two groups.Results In the treatment group,after neoadjuvant chemotherapy,4 cases (5.9%) achieved complete renission,36 cases (52.9%) achieved partial remission.Meanwhile,the numbers of cases with stable disease and progressive disease were 15 cases(22.1%) and 13 cases (19.1%) respectively.The objective remission rate and disease control rate in the treatment group were 58.8% and 80.9% respectively.The tumor resection rate,R0 resection rate,postoperative complications rate in the treatment group were 92.6%,85.3% and 14.7% respectively.The R0 resection rate in the treatment group was higher than that in the control group,the difference was statistically significant(x2 =8.042,P < 0.05).However,the differences of tumor resection rate and postoperative complications rate between the two groups were not statistically significant (x2 =1.711,0.673,all P > 0.05).The median time of PPFS in the treatment group and the control group was 36 months and 30 months respectively,there was statistically significant difference in PPFS between the two groups (x2 =4.049,P < 0.05).Conclusion The neoadjuvant chemotherapy of S-1 plus oxaliplatin for local advanced gastric carcminoma has satisfactory short-term effect and can also improve PPFS.However,further investigation is required to observe the effect of this neoadjuvant chemotherapy regimen on postoperative overall survival.
10.EFFECT OF EXOGENOUS HORMONES ON LYMPHATIC STOMATA IN OVARIAN BURSA AND LYMPHATIC DRAINAGE IN MICE

Objective To investigate the effect of steroid hormones on lymphatic stomata of ovarian bursa and fluid adsorption. Methods The lymphatic adsorption was traced by trypan blue,and the ultrastructure of the lymphatic stomata and estrogen receptor level of mesothelial cells on ovarian bursa was observed by scanning electron microscopy,transmission electron microscopy and immuno-colloid golden technology. Results The absorption and opening area of lymphatic stomata were enhanced in estrogen group mice,and decreased in androgen group as compared to control group mice.Estrogen receptor was first identified in the nuclei of mesothelial cells on the inner layer of mice ovarian bursa and the level was down-regulated by estrogen and slightly up-regulated by androgen.Conclusion The absorption and the opening area of lymphatic stomata can be regulated by steroid hormones in mice,and the machanism may be related to the expression levels of estrogen receptor in the nuclei of mesothelial cells.
