1.Perioperative nursing of patients undergoing off-pump coronary artery bypass grafting

Objective To summarize the experience of nursing patients undergoing off-pump coronary artery bypass grafting. Methods Fifity patients undergoing off-pump coronary artery bypass grafting were nursed by preoperative measures including metal nursing, blood pressure management, oxygen therapy, training of respiratory function, nursing of prepared bloods and postoperative ones including monitoring of cardiovascular and respiratory systems, medication of vasoactive agents, nursing of drainage tubes, monitoring of blood sugar and sustaining of electrolyte balance.Results Among the 50 cases, 6 had infections of incisional wound and 5 had the complication of rapid atrial fibrillation, all improved after medication. All other patients were cured and discharged except two deaths. No nursing-related complications occurred.Conclusion The off-pump coronary artery bypass grafting is highly risky and perioperative delicate nursing is critical for the success of the operation.
2.Superficial siderosis of the center nervous system

Superficial siderosis of the central nervous system is a seldom disease in clinical practice. Its most common clinical manifestations are hearing loss and ataxia. It is a hemosiderin deposition in the surface of the brain and spinal cord after chronic subarachnoid hemorrhage. Magnetic resonance imaging can make the diagnosis. However, further research and clinical verification are needed.
3.Test methods of visual alarm signal for medical alarm systems.

This paper introduces several test methods of characteristics of alarm indicator lights which can be used in the medical alarm system to verify the compliance of YY 0709-2009. These methods include the direct measurement of the pulse current supply of the alarm signals, image analysing of a video and reception by photoelectric sensor. After the comparation of the advantages and disadvantages among these methods, this paper proposes a new method of signal reception. This method which used a silicon photocell as the sensor with light filter in its front, and amplified circuit in the output end then used an oscillograph as the reviewer, has resolved the testing difficulties of the flashing signal simulating by LCD, and can be applied to test the alarm indicator lights in any medical electrical equipment.

7.Construction and primary characterization of cDNA expression library of psilgramma menephorn

Objective To construct a cDNA expression library of psilgramma menephorn and provide the basis for screening the major allergen and producing recombination allergen of psilgramma menephorn. Methods Total RNA was extracted from the whole body of psilgramma menephorn with TRIZOL mRNA purified with Oligo (dT) Spin-Column. ds cDNA was synthesized through reverse transcription. EcoRI adapters were ligated to the blunt ends and the ends were phosphorylated. The XhoI digestion released EcoRI adapter. The fragments smaller than 400 bp were removed by GHROMA SPIN-400 column and the fragments longer than 400 bp were ligated to the Uni-ZAP XR vector. The lambda library was packaged in vitro and is plated on the E.coli cell line XL1-Blue MRF. The content and recombination rate of cDNA library were tested. The length of the inserted fragments was analyzed by PCR. Results The cDNA expression library contained 5?10 5 recombinants and the recombination rate was 67%. The average length of inserted cDNA fragments was about 1.49 kb. Conclusion The constructed cDNA expression library contains appropriate contents and size of cDNA fragments for screening target cDNA to clone.
8.A preliminary study on ASPP1 mRNA expression level in tumor cell line

Objective To establish a reverse transcription-PCR(RT-PCR) assay for detecting ASPP1 mRNA expression levels in normal blood monocyte and leukemia jurkat,HL-60 and K-562 cell lines.And to analyze the differences in relative expression levels of ASPP1 in respective cell line by calculating the value of ASPP1/?-actin for evaluating the effects of ASPP1 on inhibiting the malignant transformation of normal cells.Methods The monocyte isolation reagent was used to isolate normal blood monocyte,and the RPMI 1640 medium was used to culture jurkat,HL-60 and K562 cell lines.The cell RNA was extracted with TripureTM reagent and amplified by RT-PCR.The amplified products were E.B.stained and 10?l of the products were used for agarose gel electrophoresis.The expression levels of ASPP1 mRNA and ?-actin in respective cell line were measured and then compared.Result The ASPP1 mRNA was expressed both in normal blood monocyte and all the leukemic cells.The value of ASPP1/?-actin was 0.545?0.019 in normal blood monocyte,and it was 0.155?0.081,0.199?0.016 and 0.191?0.015,respectively,in the leukemic cells jurkat,HL-60,and K-562.The expression level of ASPP1 mRNA was higher in normal cells than that in the three leukemic cell lines.Conclusion On the process of tumor development,an ASPP1-p53 compound is formed by collaborated effects of ASPP1 and p53,which may act on the arch-apoptosis gene promoter,specifically enhance the ability of p53 binding DNA,and reinforce the function of p53 in enhancing apoptosis.The declined expression and/or deprived function of ASPP1 might lead to tumorigenesis,implying that the function of ASPP1 in enhancing p53 apoptosis may play an important role on inhibiting the malignant transformation of normal cells.

With the aggravation of population aging and the change of social convention,especially after the introduction of two-children policy,some families wish to have two children can not be achieved due to various reasons.Therefore,they use assisted reproductive technology to achieve fertility aspirations.Surrogate is a controversial topic in the assisted reproductive technology,the contradictions between the normal reproductive rights of the infertile woman and the women who have lost their children but passed the age of childbearing and the bad ethical consequences and rights disputes of surrogacy.How to avoid the bad ethical consequences and meet the demand of reasonable surrogate is the major issue worthy to be discussed.
10.Effects of isoflurane inhalation during reperfusion on different degrees of global cerebral ischemiareperfusion injury in rats

Objective Isoflurane preconditioning has been shown to protect against cerebral ischemia-reperfusion(I/R)injury.The purpose of this study was to investigate if isoflurane inhalation during reperfusion hasany protective effects.Methods Fourty-two SD rats weighing 318-365 g were randomly divided into 3 groups:sham group(n=6),control group(n=18)and isoflurane group(n=18).Control group and isoflurane groupwere further divided into 10,15 and 20 rain ischernia subgroups(subgroup A,B,C,n=6).In isoflurane group1.4% isoflurane in air was inhaled immediately after reperfusion was started for 30 min.Two days before theexperiment the animals were anesthetized with intraperitoneal chloral hydrate 300 mg?kg~(-1).Microdialysis catheterwas inserter into right hippocampns using stereotactic technique and fixed.BIS microelectrodes were placed in thebrain.Vertebral arteries were permanendy occluded by electric coagulation.Bilateral common carotid arteries wereexposed and atranmatic sutures were placed around them.Globol cerebral ischemia was produced by tighteningcarotid sutures and maintained for 10,15 or 20 min(subgroup A,B,C).Cerebral iscbemia was confirmed by lossof righting reflex,dilated pupils,loss of light reflex,BIS=0 and isoelectric potential on EEG.Carotid sutureswere then released for reperfusion.Isoflurane inhalation was started right after the beginning of reperfusion andmaintained for 30 min.Neurologlc outcome was assessed by motor performance according to Combs(0-10,0=severe dysfunction,10=no dysfunction)at 24 h,48 h and 72 h of reperfusion.Microdialysis samples werecollected before during and 0-15,15-30,30-45 and 45-60 min after ischemia for determination of glutamateconcentration.Three days after ischemia the animals were sacrificed and brains were removed for microscopicexamination of hippocampns CA1 region.The number of apoptotic(TUNEL positive)neurons were counted and thepercentage(the number of TUNEL positive neurons/the total number of neurons)was calculated.Results Theglutamate content in hippocampus was significantly lower in isoflurane group than in control group duringreperfusion(P
