1.Role of mucins secretion in the process that bifid triple viable capsule attenuates the recurrence of murine experimental colitis

Objective To investigate the role of mucins secretion in the process that bifid triple viable capsule attenuates the recurrence of murine experimental colitis. Methods Mice were randomly allocated into BIFICO group (gavage with bifid triple viable capsule), 2,4,6-trinitrobenzenesulfonic acid (TNBS) group (gavage with phosphate-buffered saline), and normal control group. Intrarectal TNBS/ethanol challenge was given to mice on Day 0 followed by rechallenge on Day 5 ( 125 mg/kg and 180 mg/kg, respectively). On Day 5 several mice in both groups were randomly sacrificed before the second TNBS challenge and the remaining mice were sacrificed on Day 7. Disease activity index (DAI) was measured throughout the experiment. Inflammatory score was evaluated and MUC2 expression was determined using immunohistochemistry method. Results Recurrence of colitis was successfully simulated by challenging mice with two times of TNBS/ethanol solution. Compared with the TNBS group, DAI ( 1.39 ± 0. 68 vs. 2.39 ± 0.73, P = 0.003) and inflammatory score (2. 11 ± 0.78 vs. 3.22 ± 0. 97,P =0.03) were significantly decreased in BIFICO group after the second TNBS challenge. BIFICO administration significantly increased MUC2 expression (8. 67 ± 0. 87 vs. 7.86 ± 1.10, P = 0. 042 ) in BIFICO group compared with TNBS group before the second TNBS challenge. Conclusion Bifid triple viable capsule may enhance mucosal barriers by promoting mucins secretion, and thus attenuates the recurrence of murine colitis.
2.A phantom study of dose reduction potential in pelvic CT with advanced iterative reconstruction algorithms

Objective To assess the dose reduction potential of adaptive statistical iterative reconstruction(ASiR)and model-based iterative reconstruction(MBIR)in pelvic CT with a standard male phantom.Methods A Fluke Biomedical RANDO standard male phantom was scanned with discovery CT750 HD using different tube currents.CT images were reconstructed with FBP,50%ASiR and MBIR.The CT value,the image noise and the contrast-to-noise ratio(CNR)for the sacral vertebra relative to muscle were measured.The volume CT dose indexes(CTDIvo1)and dose-length product(DLP)were recorded.Results Compared with FBP,using 50%ASiR and MBIR had significant reduced image noise and greater CNR.The effective minimal tube currents for displaying sacral vertebra were 250 mA(FBP),180 mA(50%ASiR),and 100 mA(MBIR).With the similar image quality using FBP,the dose was reduced by 28.0% and 59.9% using 50%ASiR and MBIR,respectively.Conclusions Using advanced iterative algorithms can reduce image noise,improve CNR,and reduce the radiation dose in pelvic CT examination.
3.Long-term health effect of 25 persons overexposed to ionizing radiation

Objective To explore the long-term health effects of overexpose to radiation.Methods The 32 -41-year medical followed-up observations were performed for 25 persons exposed to ionizing radiation in the dose range of 0.10 -0.33 Gy. Observations were made of clinical symptom,eye lens,cytogenetics,immune function and endocrine function for these persons. Results The incidence of neurasthenic symptom was higher in exposed group than that in control group.2 cases suffered from liver cancer and esophageal cancer,respectively. Posterior capsule punctate phacoscotasmus of the lens wereobserved in 7 cases.There was I case of cataract in the exposed group. The frequencies of chromosomeaberration and micronucleis in peripheral blood lymphocyte in the exposed group were significantly higherthan those in control group (x2 =8.88,8.71,40.60,45.63,P < 0.05 ).The average value of serum IgG was higher(t =2.16,P <0.05 ),while that of IgM was obviously lower in the exposed group (t =2.03,P <0.05).The average values of serum triiodothyronine (T3),thyroxine (T4 ) and thyroid-stimulating hormone (TSH) in the exposed group were obviously lower than those in the control group (t =2.40,3.54,2.13,P <0.05 ).Conclusions Overexposure has effect on lens,immune function,cytogenetics to some degree.It is important to observe the long-term health effect on overexposed persons.
4.Expression changes of microRNA array in mesencephalon of rats with traumatic coma

Objective To set up a rat model with acute traumatic coma and identify the variation of microRNA in mesencephalon. Methods After rats were injured moderately by central fluid percussion system, tissues of the mesencephalon were removed one hour after injury. RNA of brain tissue of the mesencephalon was isolated for microRNA array by using the exiqon microarray system. The data were analyzed statistically by Genepix Pro 6.0 after hybridization results were scanned and fluorescence intensity standardized. Resets Hybridization results showed 33 microRNAs with up-regulated expressions but 38 microRNAs with down-regulated activity. Conclusion Expression of microRNA array shows marked changes in the tissues of the mesencephalon in rats with traumatic coma, as may be injury mechanism of traumatic coma and also a way of neurobiological protection of coma.
5.The endoscopic and clinical features of Indigo Naturalis-associated ischemic lesions of colonic mucosa

Objective By analysing the clinical features of Indigo Naturalis-associated ischemic lesion of colon mucosa to improve the precautionary and therapeutic level of the disease.Methods Thirteen patients diagnosed as Indigo Naturalis-associated ischemic lesion of colon mucosa in Peking University Third Hospital from 2005 to 2010 were reviewed.The endoscopic and clinical features were analysed.Results The 13 patients with an average age of(60.6±14.1)years old were prescribed Chinese traditional medicine containing Indigo Naturalis for psoriasis or idiopathic thrombocytopenic purpura(ITP).The ratio of males to females was 1:1.6.The typical manifestations were abdominal pain and bloody stool with watering diarrhea before bloody stool in 61.5%patients.Endoscopic and pathological characteristics were coincident with ischemic lesion and more like a chronic index.Vasodilatic medicine was effective and the average hemostatic time was(1.7±0.8)days.The prognosis was well and no recurrence was found during 3 months follow-up.Conclusions Patients having psoriasis or ITP treated with Chinese traditional medicine containing Indigo Naturalis have an inclination to colon mucosa lesions, even ischemic lesion. Careful assessment and observation before prescribing arc necessary in these patients.
6.Clinical features of severe H1N1 influenza in pregnant women

Objective To investigate the clinical characteristics of severe patients with influenza A H1N1. Methods Fifteen prenant pneumonia patients with influenza A H1N1 were selected from November 26 to December 20,2009. Results The average age of all patients was 24 years old,with an average gestational age of 32 weeks. Leukopenia was observed in 13. 3% of IS patients,and lymphopenia in 86. 7%. Data on the ratio of CD4 cells to CD8 cells were available for 12 patients,54. 5% of whom had an abnormal CD4:CD8 ratio(< 1. 4). Ten of the 14 patients(71. 4%)had increased serum lactate dehydrogenase levels,which were above 245 U per liter. Four patients (26. 6%) had elevated creatine kinase levels at admission. 4 cases of 15 patients (26. 7%) had decreased serum potassium levels,which were below 3.5 mmol per liter. Four patients (33. 3%)had C4 levels higer than 36 g per liter,and 4 cases had C3 less than 0.75 g per liter. All 15 patients had radiologically confirmed pneumonia with bilateral patchy alveolar opacities, affecting three or four lung quadrants. Findings on chest radiographs were consistent with the acute respiratory distress syndrome in all patients requiring mechanical ventilation. 4 cases were found a small amount of pleural effusion, of which 1 case was combined a small amount of pericardial effusion. Respiratory distress requiring intubation and mechanical ventilation developed in 9 patients within the first 24 hours after admission, who were all pregnant women. Two of them in the third trimester died, and 7 cases who were timely terminated pregnancy were in stable condition. Conclusions Pandemic influenza A(HIM) may pose an increased risk of severe illness in pregnant women, and it is easy to develop rapidly into adult respiratory distress syndrome. The pregnancy immunological tolerance may be involved in the severe lung injury process of H1N1 influenza pneumonia.
7.Analysis of chromosomal aberration in seven medical staffs suspected with exposure to ionizing radiation

Objective To investigate the chromosomal aberrations in seven medical staffs suspected with exposure to ionizing radiation and to speculate its possible causation.Methods The hospital staffs,including 6 females and 1 male,worked in a clinical laboratory where a CT room was located downstairs.The thickness of precast slab between these two rooms was 6 cm.Peripheral blood lymphocytes of seven staffs were examined for conventional chromosomal aberrations.Results The frequencies of dicentrics in the peripheral lymphocytes of 4 females were from 0.40% to 1.60% that was significantly higher than the spontaneous frequency of dicentrics (0.03%,x2 =36.79,P < 0.05).The translocation was observed in the lymphocytes from all subjects with frequencies from 0.33% to 1.20%,obviously higher than its spontaneous frequency of 0.01% (x2 =42.90,P < 0.05).Conclusion These staffs suffer from ionizing radiation.
8.Berberine promotes epirubicin-induced G0/G1 phase arrest in T24 bladder cancer cells

AIM:To observe the effects of the combination of berberin and epirubicin on the cell cycle of T24 bladder cancer cells and the underlying mechanisms.METHODS:The cancer cells were exposed to epirubicin in the presence or absence of different concentrations of berberin.The viability of the cancer cells was determined by MTT assay.The cell cycle distribution was detected by flow cytometry, and the protein levels of cyclin D1, CDK2, CDK4, P21 and P27 were detected by Western blot.RESULTS:Berberine markedly enhanced the inhibitory effect of epirubicin on the viability of T24 cells and promoted epirubicin-induced cell cycle arrest at G0/G1 phase as compared with the negative control cells.Epirubicin increased the protein expression of P27 and P21, both of which were enhanced by treatment with berberin.In contrast, berberin exposure further decreased the protein expression of cyclin D1, CDK2 and CDK4 in epirubicin-treated T24 cells.CONCLUSION:Berberine significantly promotes epirubicin-induced G0 /G1 phase arrest in human bladder cancer cells by up-regulating P27 and P21 expression and inhibiting the expression of cyclin D1, CDK2 and CDK4.
