Pointing to the contradiction features existent in teaching of clinical practice in the beginning stage of establishing affiliated hospital of university,it primarily discusses the management method in teaching of clinical practice,which gets first achievements after 3-year trial.

Objective To study the effect of different iodine nutrition on thyroid function in adult and pregnant women.Methods A random sampling method was used to select healthy adult and pregnant woman from the communities of coastal city,coastal rural and inland rural areas in Liaoning Province.Drinking water,urine and salt samples were collected to measured urinary iodine (U I),salt iodine (SI) and water iodine content.Fasting venous blood was collected to measured thyroid stimulate hormone (TSH),freethyroxine (FT4),free triiodothyronine (FT3),thyroglobulin antibody (TgAb) and thyroid peroxidase antibody (TPOAb) with the method of immunoassay chemical luminescence.Results A total of 150 salt samples were collected,means of SI was (30.1 ± 6.0)mg/kg.A total of 72 pregnant woman and 271 adults were investigated in iodized salt supplied regions.median UI of pregnant woman and adults were 176.3,203.2 μg/L.Iodine nutrition of pregnant women and coastal region adults was in an adequate level.Means of SI of inland adults(244.4 μg/L) was higher than appropriate level but not reached the excessive level.FT4 of the adults (11.7 pmol/L) and pregnant women (10.7 pmol/L) in inland regions were slightly higher than that of coastal city,rural adults(11.2,8.6 pmol/L) and pregnant women (10.9,9.6 pmol/L).TSH,FT3 and FT4 were not statistically different between regions (all P ＜ 0.05).But UI,FT4 and FT3 levels of pregnant women(176.3 μg/L,9.5 pmol/L,4.3 pmol/L) were significantly lower than that of the adults(203.2 pμg/L,11.3 pmol/L,4.7 pmol/L,all P ＜ 0.05).Hypothyroxinemia(4.4％,10/173) was higher than that of the inland adults (2.0％,2/98,P ＜ 0.05).And all hypothyroxinemia were found in women of childbearing age.Hypothyroxinemia prevalence of pregnant women(16.7％,12/72) was higher than that of adults(4.4％,12/271,P ＜ 0.05),The prevalence of hypothyroidism,hyperthyroidism and subclinical hyperthyroidism between the 3 regions adults and pregnant women were not statistically different (all P ＞ 0.05).Conclusions Under appropriate supply conditions of iodized salt,iodine nutrition and thyroid function are closely related.Pregnant women and women of childbearing age are at risk of iodine deficiency.The thyroid function of these people should be strengthen detect.

A method was evaluated for determination of twenty-one doping (including nandrolone, boldenone and methandienone) in human urine by gas chromatography-high resolution mass spectrometry. Samples were prepared by liquid-liquid extraction, concentrated, TMS derivatization and limit of detection at ng x mL(-1) by MID/GC/HRMS. According to the code of the World Anti-Doping Agency (WADA), precision and recoveries of the procedure were evaluated by replicate analysis (n = 6), the recoveries in the range of 66%-103%, with the RSD below 10.0%. The precision within the day of the method with three different concentrations was also determined RSD were less than 9.5%, 10.0% and 9.7%.

Objective To clone and express the new gene R049 of uropathogenic Escherichia coli 132.and to investigate the immunopmtective effects of the R049 recombinant protein on mice.Methods The pmkaryotic expression system of gene R049 was constructed by directed cloning.Thereafter,the R049 recombinant protein Was expressed and purified by Ni affinity chromatography.Polyclonal antibody was pre-pared by immunizing BALB/c mice with R049 recombinant protein.The R049 recombinant protein and whole bacterial proteins of UPEC132 were analyzed by SDS-PAGE and Western blot.BALB/c mice were im-munized with R049 recombinant protein before challenged by UPECl32 through urinary tract.Then the differences of urine and renal colony counts between immunization group and control group were compared.Results The recombinant strain E coli BL21(DE3)/pET32a-R049 ORF was constructed successfully,and the relative molecular mass of the R049 recombinant protein was 66.9×103 and its purity was up to 95% af-ter purification.The titer of polyclonal antibody wag≥1:102 400 analyzed by indirect ELISA.Both of the R049 recombinant protein and whole bacterial proteins of UPECl 32 were confirmed to show specffic reactions on the antiserunl throughh Western blot.The animal experiments showed the urine and renal colony counts of immunization group were significantly lower than that of the control group(P<0.01,P<0.05).Conclu-sion The new gene R049 of uropathogenic E.coli 132 had immunopmtective effects on mice and the defini-tive mechanism would be needed to further study.

Objective To understand the condition of iodine deficiency disorders,and the iodine nutritional status of population before adjustment of iodine level in edible salt in Liaoning Province.Methods Thirty countries (cites,districts) which were divided into coastal,inland,city,rural areas according to location and population characteristics were sampled by population probability sampling method in the whole province; one township (town,street office) was sampled from each country (city,district); one village (neighborhood) and one school were chosen from each township(town,street office).Forty children aged 8-10 were selected from each school to measure their thyroid volumes and household salt samples were collected to detect their iodine content; fourteen of those 40 children were selected to detect their urinary iodine content.In each village (neighborhood),five drinking water samples were collected in the east,the west,the south,the north and the center positions.Two tap water samples were collected to detect their iodine content if the water supply was centralized.Around each school; three townships were selected,in each township,random urine samples were collected from 5 pregnant women and 5 lactating women to detect their iodine content.Thyroid volume was examined by ultrasound method; the salt iodine was tested by the method of direct titration; iodine content of urine and drinking water was tested by arsenic cerium catalytic spectrophotometry.Results Totally one thousand two hundred and nineteen children aged 8-10 were examined; twenty-nine children were diagnosed goiter,and the goiter rate was 2.4％(national standards:＜ 5％).One thousand two hundred and nineteen edible salt samples were tested,and the median salt iodine level was 30.1 mg/kg.The iodized salt coverage rate was 99.3％(1 211/1 219),and the consumption rate of qualified iodized salt was 97.9％(1 194/1 219).Four hundred and eighty urine samples were tested,and the median urinary iodine level was 189.0 μg/L(suitable content of salt iodine was 100-199 μg/L) ; urinary iodine ＜ 20 μg/L accounted for 0.6％(3/480).Ninety-nine drinking water samples were selected,and the average iodine content of the drinking water was (5.9 ± 5.7)μg/L.Four hundred and fifty-one urine samples of pregnant women and four hundred and fifty urine samples of lactating women were selected,and the median urinary iodine level of pregnant women and lactating women was 163.2,151.0 μg/L(suitable contents of urinary iodine in pregnant women and lactating women were 150-249 μg/L,＞ 100 μg/L).The median urinary iodine of pregnant women was 135.4 μg/L in coastal city,138.0 μg/L in coastal rural,168.0 μg/L in inland city,171.1 μg/L in inland rural.The difference of urinary iodine between coastal region and inland region was significant(H =14.287 6,P ＜ 0.05).Conclusions The iodine nutrition conditions of pregnant women,lactating women and children are adequate in Liaoning Province,but the iodine nutritional status is insufficient in pregnant women from the coastal areas.

BACKGROUND:Diffusion tensor imaging aad fiber tractography(DT-FT)is the only method to identify cerebral white matter fiber organization in vivo.OBJECTIVE:To observe the changes in brain areas with language function and the fiber stucture in patients with Broca aphasia.DESIGN,TIME AND SETTING:Comparative observation was performed at First Hospital of Kunming Medical College and Tiantan Hospital of Capital Medical University between November 2003 and February 2005.PARTICIPANTS:Thirty patients(9 females,21 males,aged 17-63 years)with Broca aphasia of different degree were selected.They were diagnosed by a set of Chinese aphasia examinations of Research Laboratory of Neuropsychology,First Hospital of Beijing Medical University,following by further classification according to the subitems.METHODS:3T super-high field magnetic resonance scanner was used for the data collection and processing of the entire brain.Brodmann areas 45,44,areas 22 and 39(Wernicke area),and the corresponding brain areas in the right hemisphere are selected as areas with language function.The primary commissure,commissural fibers(arcuate fasciculus,internal capsule,external capsule)that are closely correlated with language function were regarded as region of interest of fiber tracking.The neural fiber tracking was conducted.MAIN OUTCOME MEASUIURES:The tracking and distribution of neural fiber tracts,and its correlation with other brain areas,and changes under pathological conditions.All data were compared with normal brain.RESULTS:Thirty patients with Broca aphasia were included in final analysis.The mean fractional anisotropy of fiber tracts in left Brodmann areas 45 and 44 was significantly lower than that in normal Brodmann areas(t=-2.683 65,-5.300 55,P＜0.05).The inferior fiber tracts in Broca area showed interruption or displacement,loosely connected even separate from the frontal arcuate fasciculus.The fibers of precentral gyrus and medial frontal lobe were decreased.The separation of arcuate fasciculus from Broca area caused by the integrity disruption and displacement of fiber tracts of arcuate fasciculus was the main alteration of arcuate fasciculus in Broca aphasia.The number of fiber tracts of left arcuate fasciculus and the mean fractional anisotropy in Broca aphasia were significantly less than normal people(P＜0.05).CONCLUSION:The lesions of cerebral cortex and its fiber pathway in Broca area can result in Broca aphasia.

Objective To study the distribution of the specific fragment R049 of uropathogenic E. coli(UPEC) 132 in UPEC and fecal E. coli strains. Methods The specific fragment R049 was amplified by PCR from 20 UPEC strains and 40 fecal E. coli strains, and 5 genes encoding virulence factors (papC, fimH, hly, aer, cnf1) were detected from fragment R049 positive strains. Pulse field gel electronphoresis (PFGE) was applied for isolating the Xba Ⅰ restriction fragments of the genomes of fragment R049 positive strains, and then Southern blot was applied for analyzing the distribution features of the positive hybridization bands by digoxin-labeled R049 ORF probe. Results The specific fragment R049 was amplified from 8 of 20 UPEC strains (40%) and 3 of 40 fecal E. coli strains (7.5%), and statistics analysis showed significant difference (P<0.01). The specific fragment 11049 was closely related with 5 virulence factors of UPEC in the fragment R049 positive strains. Southern blot showed the sizes of positive bands were 150 kb, 15 kb and 240 kb in 3 fecal E. coli strains, 350 kb in 6 of 8 UPEC strains, and 280 kb and 25 kb in the rest two UPEC strains. Conclusion The specific fragment R049 of UPEC132 possessed the feature of clustering distribu-tion in domestic isolated UPEC strains.

[ ABSTRACT] AIM:To investigate the biological characteristics of newborn rabbit tracheal chondrocytes in vitro. METHODS:Newborn rabbit tracheal chondrocytes were obtained by the method of enzyme digestion, and then cultured in monolayer in vitro.Morphological and growth observations were performed under inverted phase contrast microscope.The ultrastructures of the cells were observed under scanning electron microscope and transmission electron microscope.The bi-ological characteristics of secreted extracellular matrix components were detected by real-time PCR, immunocytochemistry staining and toluidine blue staining.RESULTS: Newborn rabbit tracheal chondrocytes isolated and cultured in vitro showed short triangular or irregular shapes, and adherent growth very well.The ultrastructures of the cells showed pore and abundant cytoplasm and organelles, with a lot of protein secretions in the cells.The chondrocytes expressed the mRNA of collagen I, collagen II and proteoglycans, mainly collagen II and proteoglycans.Immunocytochemistry staining showed col-lagen II and SOX9 positive, and collagen I weakly positive.Toluidine blue staining was also positive.CONCLUSION:Enzyme digestion and monolayer culture are suitable method to obtain newborn rabbit tracheal chondrocytes.These cells, secreting extracellular matrix components, are able to be selected as seed cells for tissue engineering of trachea in vitro, and used to study the therapeutic method for neonatal rabbit tracheal stenosis.

Objective\ To investigate apoptosis of Hepatoma BEL\|7402 cells induced by allicin. Methods\ The inhibit effect of allicin on BEL\|7402 cells was investigated with MTT assay. Apoptosis was ascertained by cell morphology under light microscope, electron microscope and flow cytometry. Results\ BEL\|7402 cells were suppressed with dose and time dependent relationship after exposed to allicin of 10,20,40 mg/L for vairous lengths of time. Exposed to allicin at 60 mg/L for 3 hours, BEL\|7402 cells showed typical morphology characters of apoptosis: the microvilli of the cell surface disappeared, the cell shrink and decreased in volume and blebs or ball\|like bodies appeared. Typical nuclear condensation, margination and fragmentation were observed. Apoptotic cells were of trypan blue\|rejected staining. Percentage of apoptotic cells of control group and inductive group was 2\^02?0\^37%,78\^48?3\^15% respectively by trypan blue, and 1\^78?0\^48%, 74\^07?3\^94% by flow cytometry. Cells in G\-0/G\-1, S, G\-2/M phases of control group before induction were 47\^66?2\^72%, 22\^06?2\^04%, 30\^25?3\^78% respectively.Each of them is lower than the percentage of apoptotic cells. Conclusion\ Allicin induced apoptosis of hepatoma BEL\|7402 cells.Apoptotic cells were supposed to be initiated in some phases of cell cycle.\;[

Objective:To evaluate the impression accuracy of different impression methods for flabby ridge.Methods:5 patients with flabby ridge were included.Conventional technique,perforation technique and window opening technique were performed to make im-pressions and then final models were prepared.The optical models of the flabby ridge area were obtained both in patients'mouths and on the master casts by intraoral optical scanner.Data handling and computations were made by using 3D inspection and metrology soft-ware,regarding SD,Mean +and Mean -,with subsequent analysis.Results:The 3D deviation of the models obtained by conventional technique was the biggest,that obtained by window opening technique was the least.Conclusion:Window opening technique can de-crease the deformation of flabby ridge impression.
