1.Photodynamic efficiency of new porphyrin-typed drug on HGC27 and MGC803 cells

Objective To explore photodynamic therapeutic efficiency and related mechanisms of a new porphyrin-typed photodynamic therapy (PDT) drug synthesized in our lab on HGC27 and MGC803 cells.Methods Two different kinds of gastric cancer cells, HGC27 and MGC803, were chosen as cell lines to evaluate the PDT efficiency of new porphyrin-typed PDT drug based upon four group experiments: control (no drug, no light) group, drug treated group (only drug, no light), light treated group (no drug, only light) and experiment group (with drug and light at the same time). Bleaching method was used to evaluate the photostability of new porphyrin-typed PDT drug upon repititive illumination. MTT assay was carried out to test the survival rate of tumor cells after PDT. Cofocal laser scanning microscope (CLSM) was applied to observe subcellular localization of drug in HGC27 and MGC803 cells (mitochondria and lysosome). Results New porphyrin-typed PDT drug has good stability to light, no toxicity on HGC27 and MGC803 cells without light (P>0.05), while intense lethal effect was observed upon light illumination (P<0.05). The peak efficacy appeared when concentration is 3.12 μmol/L for both HGC27 and MGC803 cells and the new porphyrin-typed PDT drug localizes in lysosome other than traditional mitochondrion. Conclusion New porphyrin-typed PDT drug is a good photodynamic therapeutic sensitizer for HGC27 and MGC803 tumor cells.
2.Effect comparison of different flow rate of oxygen inhalation on relieving scapulodynia after laparoscopic cholecystectomy with carbon dioxide pneumoperitoneum

Objective To compare the effect of different flow rate of oxygen inhalation on relieving scapuledynia after laparoscopic cholecystectomy(LC) with carbon dioxide pneumoperitoneum. Methods 100 patients adopted LC were divided into the 2 L/min group (n=50), the 4 Ldmin group (n=50), both re-ceived oxygen inhalation 6 hours after LC. The incidence and degree of scapulodynia and the blood- gas analysis were analyzed. Results The incidence of scapulodynia after LC in the 4 L/min group was signifi-cantly lower than that of 2 L/min group. PaCO2,PaO2 and pH in the 4 L/min group were different from those of the 2 L/min group. Conclusions Increasing flow rate of oxygen inhalation can relieve incidence and degree of scapulodynia after laparoscopic cholecystectomy.
3.Analysis on Neisseria gonorrhoeae infection status and related factors in outpatients of sexually transmitted diseases in Shenzhen

Objective:To understand the prevalence of Neisseria gonorrhoeae (NG) infection and related factors in outpatients of sexually transmitted disease (STD) in Shenzhen and provide scientific evidence for targeted intervention. Methods:A cross-sectional study was conducted in STD outpatients who were aged 18-49 years, had had sexual intercourse, had not received antibiotics in the last 2 weeks and sought medical cares in 22 medical institutions in 6 districts of Shenzhen from 15 April, 2018 to 16 May, 2018, their basic information and urine samples were collected, and NG in urine sample was detected by nucleic acid amplification test. Logistic regression analysis was conducted to identify the related factors for NG infection.Results:The information collection and sample collection were completed for 8 324 STD outpatients, among these STD outpatients, 196 were NG positive, with a positive rate of 2.4% (196/8 324). The positive rate of NG in men (5.8%, 148/2 567) was higher than that in women (0.8%, 47/5 742) ( χ2=189.43, P<0.05). Aged 24 years or below ( OR=2.11, 95% CI: 1.44-3.09), single/divorced/widowed ( OR=1.98, 95% CI:1.38-2.84), having casual sex in the last 3 months ( OR=1.77, 95% CI:1.29-2.43) were the related factors for NG infection. Conclusions:We found that NG infection rate was high in STD outpatients in Shenzhen. The infection rates in men and in women differed obviously. It is necessary to conduct gonorrhea screening and intervention in STD outpatients with high-risk behaviors, such as extramarital and casual sex behaviors, and standardize the treatment to reduce the incidence and spread of gonorrhea and other STDs.
4.Investigation of oxidised low-density lipoprotein as a risk assessment indicator in patients with type 2 dia-betes mellitus combined with pulmonary tuberculosis

Objective to explore the risk assessment potential of oxLDL in patients with T2DM combined with PTB.Methods A prospective study was conducted,which included 60 cases of simple hyperlipidemia,100 cases of PTB,100 cases of T2DM,and 100 cases of T2DM combined with PTB.These patients visited the outpatient department of our center from June 2022 to June 2023.The PTB group,T2DM group,and T2DM combined with PTB group were further divided into subgroups based on normal blood lipids(40 cases)and hyperlipidemia(60 cases),totaling 360 cases in the case group.Additionally,a control group consisting of 60 healthy individuals was included.The age range for inclusion in the study was between 35 to70 years old.Venous blood samples were collected from each group to detect HbA1c,INS,FSG,CHOL,TG,HDL,LDL,ApoA I and Apo B.OxLDL levels were measured using the ELISA method.Differences in levels between groups were compared.Multivariate logistic regression analysis was applied to assess the association between oxLDL levels and PTB as well as T2DM combined with PTB.Results There were no statistically significant differences in BMI,blood sugar,blood lipids,and insulin resis-tance between the T2DM hyperlipidemia subgroup and the T2DM combined with PTB hyperlipidemia subgroup.The oxLDL level in the T2DM hypertipidemia subgroup was more than double that of the control group,while the oxLDL level in the subgroup with normal blood lipids was significantly higher than that of the control group.Moreover,both the T2DM combined with PTB hyperlipidemia subgroup and simple hyperlipidemia group exhibited significantly elevated levels of oxLDL compared to the control group;however,there were no statistically significant differences when compared to the PTB hyperlipidemia subgroup.Correlation analysis revealed a significant positive linear corre-lation between TG and LDL with oxLDL in both the T2DM hyperlipidemia subgroup and the T2DM combined with PTB hyperlipidemia subgroup(R=0.352,P<0.05).Additionally,CHOL and LDL levels in the PTB hyperlipid-emia subgroup also showed a significant positive correlation with oxLDL(R=0.441,P<0.05).Multivariate logistic regression analysis indicated that having oxLDL levels more than double that of the control group was an independent risk factor for both PTB and T2DM combined with PTB(P<0.05).Conclusion The significantly elevated levels of oxLDL may serve as a potential risk factor for the comorbidity of T2DM and PTB.It is recommended to consider oxLDL levels exceeding twice those of the control group as a clinically meaningful pathological threshold for further assessment.
5.A preliminary study on the dynamic imaging of chest high resolution CT in patients with mild COVID-19

Objective:To explore the evolution of imaging findings on dynamic chest high resolution CT(HRCT) in patients with mild COVID-19.Methods:The data of epidemiology, clinical data and continuous dynamic chest high-resolution CT images of the patients with mild COVID-19 were analyzed retrospectively. Twenty-five patients (including 24 common type and 1 mild) were enrolled in the group, including 14 males and 11 females, with age of (42±12) years and hospital stay of (19±5) days. The basic images and dynamic images of HRCT were analyzed and compared by the radiologists.Results:The clinical manifestations were fever (22 cases), cough (18 cases), expectoration (8 cases), pharyngeal pain (6 cases). Most laboratory tests lacked specificity. There were no significant abnormalities on chest CT of one mild patient. HRCT findings of the common type were as follows: (1) the distribution of the lesions: most of the multiple lesions involved both lungs (19 cases), with average of 3 (3±1) lobes, located in the peripheral pulmonary zone near the pleura (22 cases); (2) the morphology and density of the lesions: most of the lesions were ground glass density foci (22 cases), which were patchy and massive (18 cases), nodular (10 cases) and arc broadband (7 cases); with the development of the disease, the density of some lesions increased with localized pulmonary consolidation (10 cases), accompanied by air bronchus sign (5 cases) and halo sign (5 cases). Dynamic changes of HRCT images in the chest: the positive manifestations were found on the 5th (5, 6) day after the onset of the disease, the progressive time of CT lesions was 5 (5, 7) days, the peak time of CT lesions was 11 (10, 13) days, and the turning time of CT lesions was 9 (8, 11) days.Conclusions:Dynamic chest HRCT can monitor the basic evolution process of the disease in patients with mild COVID-19, and provide a more intuitive basis for clinical early diagnosis and treatment.
6.Hepatitis E virus seroprevalence among blood donors in Liuzhou, China

【Objective】 To investigate the hepatitis E virus(HEV) infection among voluntary blood donors in Liuzhou, Guangxi. 【Methods】 Qualified blood donations in Liuzhou from October to November 2019 were tested for anti-HEV IgG and IgM and.HEV antigen. HEV RNA test was performed on samples positive for HEV antigen and/or anti-HEV IgM. and the test results were analyzed statistically. 【Results】 The seroprevalence rates of HEV antigen and anti-HEV IgG and IgM among 5 751 qualified donations were 0, 26.03% (1497/5 751), and 1.67% (96/5 751), respectively.None of the 91 anti-HEV IgM positive samples was positive for HEV RNA, whereas 60.41% of anti-HEV IgM positive samples were anti-HEV IgG positive. The HEV antibody prevalence varied significantly by gender, age, and nationality (P<0.05). 【Conclusion】 The prevalence of anti-HEV IgG / IgM among blood donors in Liuzhou was higher than that in other domestic regions. Significant increase in HEV antibody prevalence was observed among ethnic minorities, such as Miao and Dong, and age group of more than 45 years.Follow-up studies with larger sample size could be conducted to estimate potential risks of HEV transmission by blood transfusion, which may provide references for selective screening of HEV infection marker among high-endemic population.
7.Inhibitory Effect of Sinomenine on Human Glioblastoma and Its Pharmacokinetic Characteristics

ObjectiveTo observe the inhibitory effect of sinomenine on human glioblastoma and its pharmacokinetic characteristics in glioblastoma. MethodA human glioblastoma U87 cell line stably expressing luciferase was constructed, and a mouse glioma model was established for use in both pharmacodynamic and pharmacokinetic studies. Pharmacodynamics: Model mice were randomly divided into model group and sinomenine low-, medium-, and high-dose groups (50, 100, 150 mg·kg-1). Sinomenine was administered intraperitoneally for 14 days. The fluorescence value of brain tumors was observed to analyze its inhibitory effect on glioblastoma proliferation. Brain tumors and the surrounding brain tissue were collected, and the expression levels of vascular endothelial growth factor A (VEGFA), P-glycoprotein (P-gp), breast cancer resistance protein (BCRP), and Occludin were detected by Western blot. Pharmacokinetics: Mice were divided into a normal group (50 mg·kg-1) and model groups (50, 100, 150 mg·kg-1). After a single intraperitoneal injection of sinomenine, extracellular fluid from brain tumors was collected in vivo by microdialysis every 15 min for 6 h. Sinomenine concentrations in the dialysate were detected by HPLC-MS/MS, and pharmacokinetic parameters were calculated to analyze pharmacokinetic characteristics of sinomenine in the brain and glioblastoma. ResultCompared with model group, after 14 days of sinomenine administration, the fluorescence value of brain tumors significantly decreased (P<0.05) in a dose-dependent manner. Sinomenine inhibited the increase in VEGF and the degradation of Occludin in the tissue surrounding the tumor and inhibited the expression of VEGF, P-gp, and BCRP in glioblastoma. After a single administration, sinomenine was detected in brain and tumor tissues within 7.5 min. Compared with normal group, the Cmax and AUC in the tumor significantly increased, Tmax shortened (from 1.63 h to 0.71 h), and CLz/F decreased. In the dose range of 50-150 mg·kg-1, sinomenine exhibited a linear pharmacokinetic process in glioblastoma. ConclusionSinomenine has a significant inhibitory effect on glioblastoma, which can inhibit VEGF elevation and drug transporter efflux, reduce tumor invasion, and maintain the integrity of the blood-brain barrier. Sinomenine can rapidly cross the blood-tumor barrier, reach peak concentration, and exhibit linear pharmacokinetic characteristics in the tumor.
