1.Changes of myoepithelial cells during regeneration of parotid gland.

OBJECTIVETo investigate the changes on the number and distribution of myoepithelial cells (MECs) during parotid gland regeneration.
METHODSA total of 54 Wistar rats were divided into eight experimental groups and one normal control group, with six rats in each group. The right parotid ducts of the rats in the experimental groups were ligated for 14 days and then reopened. The parotid tissue specimens were harvested at days 0, 1, 3, 5, 7, 10, 14, and 21. The histological changes of the regenerating gland were examined using hematoxylin-eosin staining. Immunohistochemical labeling was also performed to investigate the changes in the number and distribution of MECs at different time points of parotid regeneration. Both the histological and immunohistochemical changes observed in the experimental groups were compared with those in the normal control group.
RESULTSThe parotid gland showed marked atrophy 14 days after ligation. Most acinar cells disap- peared, but the number of duct-like structures obviously increased. MECs apparently increased in number and were mainly located at the periphery of the duct-like structures. Three days after duct reopening, the number of acinar cells significantly increased and the duct-like structures significantly reduced. Meanwhile, MECs also decreased in number and were mainly located at the periphery of the newly formed acini and duct-like structures. The number of MECs noticeably decreased 3 and 5 days after duct reopening. At 14 days after duct reopening, the glandular structures and the number and distribution of MECs returned to normal compared with those in the normal control group.
CONCLUSIONThe number and distribution of MECs returned to normal condition after parotid gland atrophy. Parotid regeneration mainly occurred within 5 days after duct reopening.

2.Research advances in naturally-derived biomaterials for tendon tissue engineering

The ideal scaffold material can reconstruct the morphology, structure and function of tissues and organs. Thus, it has received extensive attention in the field of tissue engineering. With the rapid development of the theories and technologies of naturally-derived biomaterials, it has gradually become a research hotspot in the field of regenerative medicine. Natural biomaterials have high bionics, good biocompatibility and a wide range of sources. Thus, they are very suitable to be used as scaffolds for tissue engineering. According to ingredients and source of raw materials, they can be roughly divided into protein-based biomaterials (collagen, gelatin, silk and fibrin), carbohydrate-based biomaterials (cellulose, chitin/chitosan, alginate and agarose), glycosaminoglycans (hyaluronic acid, chondroitin sulfate) and decellularized extracellular matrix (amniotic membrane, small intestinal submucosa, tendons). Different scaffold materials have unique natural structures and properties. Protein-based biomaterials can affect cell migration and differentiation through polymerization to form a network structure. They can be made into stents alone or used with other synthetic materials based on certain mechanical properties. Carbohydrate biomaterials can carry a large amount of liquid due to their high specific surface area. However, their mechanical properties are poor. Therefore, it is often used in the form of gel with other materials to control the release of cells and growth factors in tendon tissue engineering. Regularly, the anti-inflammatory, viscoelastic, lubricating and highly hydrating properties of glycosaminoglycan biomaterials can be combined with synthetic materials to increase their biocompatibility and hydrophilicity. Compared with the above materials, the decellularized extracellular matrix not only has a more similar extracellular structure and nutrients, but also has certain mechanical properties. Therefore, it can better reconstruct the morphology, structure and function of tissue and organ lesions. Ultimately, the above materials have been paid more and more attention in tissue engineering. The ingenious combination of different materials makes the repair of tendons show better results. In future, naturally-derived biomaterials could have broader clinical prospects and application value.
3.Histological observation of rat's parotid gland after ligation and reopening of the main duct

Objective:To investigate the histological changes of parotid gland after ligation and reopening of the main duct.Meth-ods:The right parotid main duct was ligated for 14 days and then reopened,the specimen were harvested at day 0,1,3,5,7,10,14 and 21 after reopening(n=5 )respectively.By HE staining and Morphology measurement,the morphological and volume fraction changes of acinus,duct and mesenchyma were observed and measured.Results:The parotid gland atrophied significantly 14 days af-ter the duct ligation.From day 3 after duct reopening,the acinar volume fraction continued to increase,both the duct and the mesen-chymal tissue volume fraction continued to decrease significantly(P<0.05).After 14 days of duct reopening,there was no signifi-cant difference in volume fraction of acinar,duct and mesenchyma,compared with those of normal control group(P>0.05).Conclu-sion:After reopening of the ligated main duct the atrophied parotid gland is able to regenerate to be normal.
4.Epidemiology of 10 000 mammary gland disease cases in Yangquan city

Objective To investigate the epidemiology of 10 000 patients with mammary gland disease.Methods 10 000 mammary gland disease cages during 1987~2006 in Yangquan Tumor Institute were collected,all patients had integrity case files and were diagnosed by molybdenum target radiography,near infrared ray,sonography,aspiration-needle biopsy or polyrrhea smear examination.Results In 8919 outpatient treated cases,7493 suffered cyclomastopathy(84%).1081 cases were inpatient cared(11%),the top five mammary gland disease are:breast cancer 342 cases,cyclomastopathy 252 cases,adenofibroma 104 cases,intraductal papilloma 86 cases and ductal ectasia 76 cages.For breast cancer patients,there were 125 cases during 1987~1996,3 were 21~30 years old and 23 were 31~40 years old.The number during 1997~2006 was 217,24 were 21~30 years old and 69 were 31~40 years old.Conclusion In 10000 cases,there are 7745 cyclomastopathy patients(77.45%)which takes the first place.Especially for 342(3.42%)breast cancer cases,the incidence grew up and patients age was much younger.
5.Effect of Guilong Kechuanning Tablets Combine with Antibiotic Drugs on Pulmonary Infection after Chemotherapy in Patients with Lung Cancer

Objective:To investigate the effect ofGuilong Kechuanning tablets combine with antibiotic drugs on pulmonary infection after chemotherapy in patients with lung cancer,so as to reduce the incidence of pulmonary infection in hospital.Methods:A total of 120 patients with lung cancer complicated with pulmonary infection,who underwent chemotherapy in Baoji Hospital of Traditional Chinese Medicine from August 2013 to August 2015,were selected and randomly divided into control group (n=60) and experiment group (n=60).The control group was treated with Ceftizoxime,based on which,the experiment group was added oral Guilong Kechuanning tablets.The course oftreatment of the two groups was 2 weeks.Inflammation indicators of the two groups were compared before and after treatment,and the clinical effect,hospitalized time,antifebrile time,time significantly reduced of cough and expectoration,disappearance time of lung rales and the pulmonary bacterial clearance rate were observed between the two groups.Results:The total effective rate (91.67%) of experiment group was significantly higher than that(73.33%) of control group (x2=13.121,P=0.004).Before treatment,there were no significant differences in WBC,CRP and NEUT levels between the two groups (P>0.05);while after treatment,the levels of WBC,CRP and NEUT were all lower than those before treatment in both groups (all P<0.05),and the levels of WBC,CRP and NEUT in the experiment group were lower than those in the control group(P<0.05).After treatment,the bacterial clearance rate (99.09%) of the experiment group was significantly higher than that (86.94%) of the control group (x2=54.876,P=0.000).Conclusion:Guilong Kechuanning tablets combined with antibiotic drugs can effectively control the pulmonary infection after chemotherapy in the patients with lung cancer,and can improve the clinical symptoms and inflammatory reaction,which is worthy of clinical application.
6.The effects of nourishing lung and kidney formulas on inflammatory response of alveolar epithelial cells stimulated by monocytes conditioned medium

Objective To observe the effects of nourishing lung and kidney formulas on inflammatory response of alveolar epithelial cells stimulated by monocytes conditioned medium and study the anti-inflammatory mechanism of the formulas for the treatment of chronic obstructive pulmonary diseases (COPD).Methods The reproduction of inflammation models of A549 cells were stimulated by monocyte THP-1 cell strain conditioned medium.A549 cells were randomly divided into blank control group (20% blank rabbits serum),model group (20% blank rabbits serum+25.0% THP-1 cell conditioned medium),nuclear transcription factor activator protein-1 (AP-1) pathway inhibitor T-5224group (20% blank rabbits serum+25.0% THP-1 cells conditioned medium+100 μmol/L T-5224),nourishing lung and kidney group (20% rabbits serum with nourishing lung and kidney formulas+25.0% THP-1 cells conditioned medium).The contents of interleukins (IL-6,IL-8),tumor necrosis factor-α (TNF-αα),matrix metalloprotein-9 (MMP-9) in cell culture supernatant were detected with enzyme linked immunosorbent assay (ELISA),the supernatant content of malondialdehyde (MDA) was detected with thibabituric acid (TBA) method,the total activity of superoxide dismutase (T-SOD) was detected with hydroxylamine method,and the activity of AP-1 pathway was detected with electrophoretic mobility shift assay (EMSA) method.Results Compared with the blank control group,the A549 cell proliferation were significantly increased at 24 hours,48 hours stimulation by 25.0% cell conditioned medium (A value:24 hours was 0.41 ± 0.02 vs.0.37 ± 0.04,48 hours was 1.30 ± 0.09 vs.1.15 ± 0.19).Compared with the blank control group,the contents of IL-6,IL-8,TNF-αα,MMP-9,MDA,AP-1 expression were significantly increased in model group [IL-6 (ng/L):35.00±3.63 vs.23.15±1.72,IL-8 (ng/L):273.09± 164.36 vs.231.45±33.90,TNF-α(ng/L):51.61 ± 9.51 vs.28.87 ± 3.34,MMP-9 (ng/L):442.85 ± 78.86 vs.235.60 ± 14.62,MDA (μmol/L):6.90 ± 0.11 vs.6.01 ± 0.12,AP-1 expression (A value):2.260 ± 0.062 vs.1.000 ± 0.000],MDA/T-SOD ratio was increased (4.43 ± 0.05vs.3.96 ± 0.06).Compared with model group,the levels of IL-8 (ng/L:100.29 ± 17.03),TNF-α (ng/L:25.13 ± 0.46),AP-1 expression (A value:1.38 ± 0.02),and the MDA/T-SOD ratio (4.23 ± 0.23) in T-5224 group,and MMP-9 (ng/L:195.44±9.80),MDA (μmol/L:5.86±0.30),MDA/T-SOD ratio (3.56±0.41),AP-1 expression (A value:0.76 ± 0.01) in nourishing lung and kidney group were all reduced significantly (all P < 0.05).Conclusion Nourishing lung and kidney formulas can suppress the inflammatory response through regulating the alveolar epithelial cells AP-1 signaling pathways.
7.The expression of Bcl-2 and Bax proteins and their significance with the atrophy of parotid gland cells in rats

Objective:To investigate the expression and correlation of Bcl-2 and Bax in the parotid gland after leading duct ligation in rats. Methods:Atrophy of the right parotid was induced by ligating the right main duct of 72 rats. Immunohistochemical labelling was performed to study the expression of Bcl-2 and Bax 1, 3, 5, 7, 14, 21, 30, 60, 90, 150 and 180 days after duct ligation. Results:7 d after duct ligation most acinar cells disappeared. The distribution of Bcl-2 and Bax protein in normal parotid was in cytoplasm with unifo-maity. Bcl-2 and bax higher expression was identified in the gland at all time points. The expression of Bcl-2 protein was significantly in-creased and reached the peak at 21 d after duct ligation. More Bax-positive acinar cells on day 3 were observed, then the expression of Bcl-2 and Bax protein was descended. Higher Bcl-2/Bax ratio was identified at 1-21 d, then descended. Conclusion:The expression of Bcl-2 and Bax is associated with the atophy of the parotid glad after rat parotid duct ligation.
8.Balloon dilatation plus support tube for treatment of biliary stricture after orthotopic liver transplantation by using the endoscope technique

BACKGROUND:Biliary stricture following liver transplantation is mainly focus on biliary stoma stricture; while, balloon dilatation temporarily keeps biliary tract open but not works out a solution at all.OBJECTIVE: To discuss the diagnosis and treatment of postoperative biliary stricture after orthotopie liver transplantation by the endoscope technique.DESIGN, TIME AND SETTING: A case analysis, which was performed at Dalian Liver and Gall Surgical Institute. Ten patients hospitalized from the Department of Liver and Gall Surgery of Dalian Friendship Hospital and four patients hospitalized from the Department of Organ Transplantation of Tianjin First Central Hospital were diagnosed as biliary stricture after orthotopic liver transplantation.PARTICIPANTS: Among 14 patients, 10 males and 4 females with mean age of 46 years provided end-to-end biliary anastomose.METHODS: Fourteen cases of postoperative biliary stricture after orthotopic liver transplantation were analyzed and diagnosed by endoscope technique. And by endoscope technique, the stricture was supported with tube after balloon dilatation.MAIN OUTCOME MEASURES: Bile duct mucous membrane under T-tube radiography and endoscope; calculary distribution and bile duct mucous membrane at stoma; healing of biliary stoma of donors and recipients; inflammatory edema and stricture; recheck of above-mentioned parameters after stricture expansion by endoscopic stone extraction technique.RESULTS: Thirteen cases of postoperative biliary stricture after orthotopic liver transplantation were analyzed and diagnosed by endoscope technique, including one was induced by calculus, and one non-stoma stricture. One case was treated with balloon dilation; biliary infection and jaundice occurred in 2 cases after endoscopic sphincterotomy (EST) + basket lithotripsy + endoscopic nasobiliary drainage (ENBD), so operations or fibrocholedochoscope treatments had to be carried out. By T tube radiography, in 1 case there was strip-like negative simulacrum or no stricture, well-healed anastomosis and good mucous membranel transition; poor or no intrahepatic visualization were found in 2 cases, so anastomosis dilation was processed after the calculi removal by fibrocholedochoscope, stricture disappeared in 3 or 4 months; in 8 cases there were blur extrahepatic or intrahepatic biliary visualization, cord-like, column or branch-like negative simulacrum in biliary ducts and sign of non-anastomosis stricture, after removal of calculi, anastomosis stricture and congestion, edema were found, all these disappeared after average 2.5 months of dilation; the other 1 case was found stricture by T the radiography, but no calculi was found with fibrocholedochoscope, finally the Ttube was removed after 2 months of stricture dilation.CONCLUSION: Endoscopy is significant to directly reflect and reliably diagnose postoperative biliary stricture and effectively treat biliary stricture by anastomosis dilation.
9.Correlation between the residual amount of decellularization reagents in common tissue materials and cytotoxicity

Objective:To study the correlation between the residual amounts of commonly used tissue decellularization reagents like sodium dodecyl sulfate (SDS), Triton X-100, sulfobetaine 10 (SB-10) and cytotoxicity, and to find the safety threshold for controlling the residual of the reagent.Methods:SDS, Triton X-100, SB-10 solutions of different concentrations were prepared, and the cytotoxicity of solutions of different concentrations was detected by L-929 cells and MC3T3-E1 cells according to the method of GB/T 16886.5. The mass concentration range of the above three decellularization reagents was refined, and the threshold for the residual non-cytotoxicity of a single reagent was determined. Mix the three reagents in pair or three blends, and determine the threshold of non-cytotoxicity after the reagents are mixed by cytotoxicity test.Results:SDS, Triton X-100 and SB-10 caused L-929 cells to produce cytotoxic concentration values of 4×10 -3, 2×10 -2 and 6×10 -1 mg/ml, and MC3T3-E1 cells 6×10 -3, 4×10 -2 and 2 mg/ml. The toxicity trend of the three reagents is SDS>Triton X-100>SB-10. When three reagents at critical concentrations are mixed in pairs or three, they have no cytotoxicity to L-929 cells and MC3T3-E1 cells. Conclusions:When using the above three decellularization reagents to prepare decellularized scaffolds, a variety of reagent cleaning procedures should be used to efficiently clean the tissues so that the residual amount of reagents is below the critical value to ensure the biological safety of the decellularized scaffolds.
10.Measurement agreement of 25-hydroxyvitamin D results derived from Roche immunoassay and liquid chromatography tandem mass spectrometry

Objective:To evaluate the measurement agreement of Roche 25(OH)D immunoassay(evaluation method) with LC-MS/MS (reference method).Methods:A total of 909 residual serum samples from routine health check participants were collected from May to June in 2019. 25(OH)D concentrations were measured by evaluation method and LC-MS/MS, respectively. Passing-bablok regression, intraclass correlation coefficient (ICC), Bland Altman plots and Kappa test were used to analyze the consistency and bias on the results derived from the two measurement methods.Results:The 25(OH)D concentration derived from evaluation method was significantly different from those from LC-MS/MS method ( P<0.001). Slope of regression for evaluation method and LC-MS/MS was 0.962(95% CI 0.919-1.007), while intercept was -0.185 (95% CI -1.191-0.745). The ICC was 0.765 (95% CI 0.735-0.792). Altman plot showed that the average deviation between evaluation method and LC-MS/MS was -0.902 ng/ml (0.300%). The coincidence rate of evaluation method′s judgment of vitamin D sufficiency, insufficiency and deficiency with LC-MS/MS was 83.39%, and the weighted Kappa values was 0.790. Conclusion:Roche automatic 25(OH)D immunoassay shows acceptable correlation and agreement with LC-MS/MS, however, it is to note that the deviation between immunoassay and LC-MS/MS may lead to wrong judgment of vitamin D nutritional status. It is recommended that each laboratory should establish own corresponding reference values for 25(OH)D concentrations derived from these two methods.
