1.EFFECTS OF NAPHTHYLMETHYL ISOQUINOLINE ON CONTRACTILE TENSION AND VOLUME OF OXYGEN CONSUMPTION IN HEART MUSCLES

The effects of naphthylmethyl isoquinoline (NI) and verapamil (Ver) on contractile tension and volume of oxygen consumption were compared on isolated guinea pig left atrips and cat papillary muscles. The results showed that NI and Ver diminished the Ca++-dependent contractile tension and volume of oxygen consumption by electrically driven preparations. Under the influence of increasing doses of NI (10 ~100?mol/L)and Ver(0.72~3.6?mol/L) linear reduction of contractile tension and volume of oxygen consumption were seen. These effects were reversed by increasing the extracellular Ca++ concentration. These results indicate that NI may be an effective calcium-antagonist very similar to Ver.
2.Effects of DP2 intervention on primary cultured rat hippocampal neuron treated with aluminum overload

Aim To establish primary cultured rat hip-pocampal neuron damage model induced by aluminum maltolate and study the effect of intervention for DP2 on primary cultured rat hippocampal neuron treated with aluminum overload. Methods The hippocampus was dissected out from fetal rat ( embryonic 18 d ) . After being cultured for 7 d, the hippocampal neuron was treated with Al( malt) 3 to establish the model of prima-ry cultured rat hippocampal neuron damage and mean-while treated with DP2 agonist DK-PGD2 and DP2 an-tagonist CAY10471, respectively. After treatment for 24 h, the cell viability was measured by MTT and LDH, Ca2+ fluorescence intensity. Neuronal pathomor-phology was observed by HE staining. Results The purity of hippocampal neuron was more than 95%. Compared with the control group, the number of hipp-ocampal neurons was reduced and neurons became chromatic agglutination and karyopyknosis in aluminum overload group. Treatment of aluminum caused a sig-nificant decrease in MTT value ( P<0. 01 ) and an in-crease in the LDH leakage rate (P<0. 01). The Ca2+fluorescence intensity significantly increased ( P <0. 01 ) in aluminum overload group. Compared with that of the aluminum overload group, treatment of DK-PGD2 , a selective DP2 agonist, significantly aggravated the primary cultured rat hippocampal neuron injury caused by aluminum overload accompanied with the significant decrease of MTT value ( P <0. 01 , P <0. 05 ) and an increase of the LDH leakage rate ( P<0. 01), significant increase of Ca2+ fluorescence inten-sity of neuron. Treatment of CAY10471, a selective DP2 antagonist, had opposite effects of DK-PGD2 . Conclusion The activation of DP2 can increase hipp-ocampal neural susceptibility to aluminum overload.
3.Comparison of curative effect of paracetamol and indomethaein suppository in eaneerons febrile

Objective To observe the curative effect of paracetamol and indomethacin suppository in cancer- ous febrile. Methods 108 patients of lung cancer with fever were divided into.two groups,the treatment group of 54 cases and the control group of 54 cases, administered with paracetamol and indomethacin suppository respectively. Results After the two groups took medicine, the difference in defervesceing was significant, with statistics signifi- cance( P < 0.05). Conclusion Indomethacin suppository shows better function of defervesceing than paracetamol.
4.Establishment of rat models of Parkinson’s disease by unilateral two-point injection with 6-hydroxydopamine

BACKGROUND:The establishment of animal models of Parkinson’s disease plays an important role in clinical and basic experiments of Parkinson’s disease, and its stability directly affected study results. OBJECTIVE:To evaluate the behavior and pathological changes of rats with Parkinson’s disease established by unilateral two-point injection with 6-hydroxydopamine.
METHODS:A total of 62 Sprague-Dawley rats were randomly assigned into experimental group (n=50) and normal group (n=12). Using stereotatic technique, 6-hydroxydopamine was injected into the right substantia nigra
RESULTS AND CONCLUSION:At 2 weeks, after induction with apomorphine, left rotation speed>7 r/min was observed in only 22 rats of the experimental group. No significant difference in the number of rotation between 2 and 4 weeks was detected (P>0.05). Tyrosine hydroxylase and dopamine content significantly reduced in the model group. Results indicated that unilateral two-point injection of 6-hydroxydopamine could be used to
successful y establish rat models of Parkinson’s disease with similar behaviors and pathologies.
5.STUDY OF BEAT TO BEAT SURFACE RECORDING OF HIS BUNDLE ACTIVITY WITHOUT AVERAGING IN MAN

Objective To investigate the application of open beat to beat techniques recording His bundle activity without digital averaging. Methods Recording was performed with very low noise, high gain amplifiers and by analog filtering. 3 brigade amplifiers and 16 bite A/D were connected to a microcomputer by USB. The signal was processed by A pattern recognition plus digital filtering software. The subjects were in a supine position during the recording. Results A prominent waveform was observed between the atrial and ventricular complexes and corresponded to His bundle activity in 46 volunteers. Recordings in 8 patients were compared with records obtained by intracardiac electrode, and a good conformation was seen. Conclusion The noninvasive method of beat to beat surface recording of His bundle activity has broad clinical applicability and high utility for the patients with latent A-V block and eletrophysiologic study.
6.New breakthrough in beat to beat surface recording of his bundle potential

Objective To evaluate a noninvasive method which has been applied on human subjects to observe beat to beat activity of the His bundle potential. Methods Recording was performed with very low noise, high gain amplifiers and by analog filtering. 3-brigade amplifiers and 16-bite A/D were connected to a microcomputer by USB. The signal was processed by a combined pattern recognition and digital filtering software. Results The results of surface monitoring were compared with intracardiac method in 30 patients. The results of non-invasive method were consistent with that interventional method. A prominent waveform was observed between the atrial and ventricular complexes (A, H, V waves), and corresponded to His bundle activity in all patients. Conclusion The noninvasive method of beat to beat surface recording of His bundle activity has broad clinical applicability and high utility for the patients with latent A-V block and eletrophysiologic study.
7.Evidence-based medicine teaching reform practice of clinical graduate students

In view of the clinical graduate students' problems existing in the teaching of evidence-based medicine, such as having little knowledge of the course, lack of interest, not knowing how to retrieve, evaluate evidence and effectively use evidence, Xinjiang Medical University has car-ried on the reform in the curriculum content, teaching process, evaluation content and method of the teaching of EBM for the clinical graduate students. Practice shows that the clinical graduate students have significant improvement in objective examination average score, subjective examination average grades, total score and the students' satisfaction of the classroom teaching, and at the same time their interest in EBM courses and clinical thinking have also improved.
8.Experimental study on ovarian cancer immunotherapy by EpCAM activated dendritic cells induce antigen-specific CD8+ cytotoxic T lymphocytes

Objective:To observe the effect on ovarian cancer immunotherapy by dendritic cells (DC) which activated by Epithelial cell adhesion molecule (EpCAM) induce antigen-specific CD8+ cytotoxic T lymphocytes (CTL) and to provide some help to ovarian cancer immunotherapy.Methods: Interleukin (IL)-12,and IL-10 of DC were tested after inducing by EpCAM.Subsequently,EpCAM specific CTL CD8+ was induced by EpCAM-DC.The therapeutic effect and interferon (IFN)-γ of EpCAM-DC-CD8+ CTL on normal ovarian epithelial cells IOSE80 and ovarian cancer cell SKVO3 was detected.After treatment of EpCAM-DC-CD8+ CTL,the volume of ovarian tumor of bearing BALB/c-nu/nu mice was detected.Meanwhile,the morphology changes of tumor tissue were observed by HE staining.Results: Compared with PBS,EpCAM stimulation significantly inceased surface markers DC80,DC83,DC86 and HLA-DR levels,and added up to 4.79,4.85,4.60 and 10.91 times (P<0.05).EpCAM stimulation significantly increased the expression of IL-12 and reduced the secretion of IL-10 (P<0.05).Both of DC-CD8+ CTL and EpCAM-DC-CD8+ CTL resulted in minute amount of IOSE80 cell killing (P>0.05).However,the killing rate of EpCAM-DC-CD8+ CTL on SKVO3 cells was 6.82-folds as much as that of DC-CD8+ CTL.Animal experiments showed that ovarian cancer transplantation tumor volume ratio after EpCAM-DC-CD8+ CTL treatment,was significantly lower than PBS group and DC-CD8+ CTL group,which reached 0.27 and 0.28 times,respectively (P<0.05).HE staining showed that EpCAM-DC-CD8+ CTL treatment resulted in significant changes of tumor tissues in pathology.Conclusion: EpCAM protein stimulated the maturation of DC that induced the production of EpCAM specific CD8+ CTL.EpCAM-DC-CD8+ CTL can effectively kill ovarian tumor cells and delay the growth of tumor,which is of great significance for the immunotherapy of ovarian cancer.
9.Investigation and analysis of mental health of clinicians in a hospital in Xinjiang

Objective To analyse the mental health status of doctors in the First Affiliated Hospital of Xinjiang Medical University. Methods The 673 doctors in different clinic character in the First Affiliated Hospital of Xinjiang Medical University were investigated by questionnaire and symptom checklist-90(SCL-90),and estimate the results through F-test and t-test . Results Doctors in SCL-90 in addition to interpersonal relationships(1.59±0.49, t=-1.02,P>0.05) and the rest of the factors:somatization (1.48 ± 0.55, t=2.32), force(1.75 ± 0.43, t=2.34),depress(1.67 ± 0.44,t=3.00), anxiety(1.55 ± 0.57, t=3,71), hostility(1.76±0.47,t=5.65),horrible(1.48±0.32,t=6.35),stubborn(1.65±0.39, t=4.04),insanity(1.43±0.46, t=3.39)are higher than the national norm, there is statistical difference (P<0.05). In different gender groups, in addition to the somatization and forced, female doctors other project factors were higher than male doctors, including interpersonal relationships(1.62 ± 0.37, 1.56 ± 0.41), hostility factor(1.79 ± 0.38, 1.73 ± 0.43) differences were statistically significant (t=2.15, t=2.07, P均<0.05). In different professional title ,Doctors with primary professional title in addition to the body of the rest are higher than the title doctor. Anxiety factor differences have statistical significance in different groups (F=2.73, P<0.05). In different department , Emergency department and pediatric clinicians in addition to depression, anxiety and other factors of points were higher than those of physicians and surgeons, the difference in interpersonal relationship factors have statistical significance in different groups (F=4.54, P<0.05). Conclusion The scores aboutdoctors, SCL-90 in the First Affiliated Hospital of Xinjiang Medical University in addition to interpersonal relationships, the rest of the factors are higher than the national norm ,showing that clinic doctors were in psy chological sub health state, especially in female doctors, primary professional title dortors and Emergency department and pediatric clinicians.
10.Protective effects of four alkaloids of embryo loti on H2O2-induced oxidative damage of vascular endothelial cells

Objective To observe protective effects of four active liposoluble alkaloids of a Chinese herb, lotusine (Lot), liensinine (Lien), isoliensinine (Iso) and neferine (Nef) of embryo loti (the green embryo), against H2 O2-induced oxidative damage on human umbilical vascular endothelial cell ECV-304.Methods The protective effects of Lot, Lien, Iso and Nef on the survival of normal and oxidatively damaged ECV 304 cells were studied by cell morphology observation and 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium (MTT) assay.The levels of nitric oxide (NO) and nitric oxide synthase ( NOS) were measured using colorimetric assay.Results Lot, Lien, Iso and Nef did not affect cell morphology and cell viability of normal ECV 304 cells.The survival of oxidative damaged vascular endothelial cells was rescued by incubating with Lot at 100μmol/L, and Lien, Iso and Nef at 0.1 μmol/L.The proliferative activity of medicated groups increased to 112.8%, 129.3%, 125.6 and 118.2%, respectively (P<0.01 or 0.05), relative to that of the group with H2O2 induced oxidative damage.The four alkaloids restrained oxidative injury of endothelial cells induced by H2 O2 and the protective influences were similar with captopril, which served as a positive control.Each alkaloid except Lot reduced intercellular space and increased the connections of oxidative damaged cells, concomitant with more recognizable cell borders.Lien, Iso and Nef also increased the concentration of NO ( P<0.05 ) .Besides, all of the four alkaloids activated NOS in damaged vascular endothelial cells ( P<0.05 ) . Conclusion The four alkaloids of embryo loti, especially Lien, Iso and Nef, have certain protective effects against H2 O2-induced oxidative damage on vascular endothelial cells.The protective mechanism may be promotion of NO release through the increase of NOS production.
