OBJECTIVE: To study the effect of compound glycyrrhizin on hepatic function and hepatic fibrosis of patients with chronic hepatitis B. METHODS: 33 patients with chronic hepatitis B were injected i.v with 80ml compound glycyrrhizin q.d for 4 weeks. The hepatic function, hepatic fibrosis of patients and the clinic effects before and after treatment were observed. RESULTS: The total effective rates for patients with mild, moderate and severe degrees were 100%, 92.3% and 91.7%, respectively. 2.Targeted monitor and comprehensive intervention of multidrug-resistant organism infection in a general hospital

Objective To evaluate the occurrence of multidrug-resistant organism (MDRO)infection in a general hospital,and take effective comprehensive intervention measures,so as to reduce MDRO infection.Methods Targeted monitoring on MDROs was performed from October 2012 to December 2013,comprehensive intervention measures were conducted,the occurrence of MDRO infection before and after intervention was compared.Results A total of 62 384 pa-tients were investigated,17.91%(n=11 176)were sent specimens for pathogen culture,606 times of culture for patients’ specimens were detected MDROs,292(0.47%)cases were MDRO healthcare-associated infection(HAI),314 were com-munity-acquired infection or colonization.MDRO infection case rate decreased from 0.65%(75/11 603)before intervention to 0.26%(36/13 875)after intervention (χ2 =21.08,P <0.05).Conclusion Comprehensive intervention measures can effectively reduce the occurrence of MDRO HAI.
3.The thalassemia screening analysis of 2 650 cases of childbearing women

Objective To investigate the incidence of poverty ,population carrying rate and its genetic type through the thalasse‐mia screening analysis of 2 650 cases of childbearing women .Methods Hematologic examination and hemoglobin electrophoresis were applied in people who were laboratory tested the crowd ,screening suspected people ,then use the genetic analysis confirm the diagnoses .Results Preliminary screening of the 536 cases which were suspected the thalassemia (or gene carriers) ,hematologic ex‐amination showed that the red blood cells ,reticulocyte counts in the cases were significantly higher than those in the controls ;and hemoglobin ,mean corpuscular volume (MCV)and mean corpuscular hemoglobin (MCH) were significantly lower .Hemoglobin elec‐trophoresis test showed that in 512 abnormal cases ,68 cases were diagnosed as α thalassemia and 438 cases were β thalassemia(506 cases in total ,accounting for 19 .09% of the total number) .In this area ,Southeast Asia(SEA) heterozygotes(‐SEA /αα) ,right hetero‐zygotes(‐α3 .7 /αα) ,left heterozygotes(‐α4 .2 /αα) ,Hb Constant Spring(‐αCS /αα) were common type of α thalassemia ;CD41‐42 hetero‐zygotes ,CD17 heterozygotes ,CD71‐72 heterozygotes and CD28 heterozygotes were common type of β thalassemia .Conclusion In Guangxi ,there was high incidence rate of thalassemia ,thus ,though premarital and prenancy anterior screening ,guide the rational marriage and pregnancy ,and through the crowd to intervene to prevent common human monogenic death and disabling genetic dis‐orders are of great importance for prepotency promotion and population quality improve .
4.Induced differentiation model of embryonic stem cell in vitro

OBJECTIVE: Embryonic stem cells (ES cells) is a special kind of cell population with totipotency and uninhibited self-renewal property. Researches on induced differentiation of ES cells were received and summed up to provide reasonable and constructive suggestions for clinical application of ES cells.DATA SOURCE: Papers published in Highwire press and Medline database were retrieved with keywords "Embryonic stem cell, differentiation, and induce" from 2000 to 2004. Additionally, papers published in Wanfang database were also retrieved with keywords "Embryonic stem cell" in Chinese from 2000 to 2004.STUDY SELECTION: Data were analyzed firstly in order to select papers related to induced differentiation. The inclusion criteria were selection of the original works, but the summaries and Meta analysis were excluded.DATA EXTRACTION: A total of 369 papers were chosen according to correlation with keywords; among them, 364 papers were in foreign language. After reading through summaries of these papers, 15 papers thoroughly discussing induced differentiation of ES cells were chosen for further intensive reading. Papers discussed various kinds of differential models, neural cells, cardiac muscle cells, epithelial cells and hematopoietic cells.DATA SYNTHESIS: The induced differential models of neural cells, cardiac muscle cells, epithelial cells and hematopoietic cells from ES cells were introduced comprehensively in the seven literatures. Based on these references mentioned above and other references which introduced cell model separately, all data were generally studied, sorted out and summed up.CONCLUSION: ES cells can be induced to be various kinds of cell models such as neural cells, epithelial cells, cardiac muscle cells and hematopoietic cells, which may be used to clarify the mechanism of cellular development and differentiation to provide good respects for clinical cellular therapy and screening of medicines.
5.Protective effect of granulocyte-colony-stimulating factor against chronic myocardial ischemia in rabbits

Objective To investigate the protective effects and mechanisms of granulocyte-colony-stimulating factor (G-CSF)on a rabbit model of chronic myocardial ischemia.Methods Myocardial ischemia models were created by partial ligation of the left anterior descending coronary artery in Japanese white male rabbits.Rabbits were subcutaneously injected with G-CSF (G-CSF group)or saline (control group)for 6 days after myocardial ischemia.The percentage of CD34-positive cells in the peripheral blood was evaluated by flow cytometry,and CD34-positive cells homing and vWF expression in the ischemic myocardium were determined by immunohistochemistry.Results Rabbits in G-CSF group had a higher survival rate than those in control group (P <0.05).Immunohistochemistry of the ischemic myocardium showed that compared with control group,G-CSF group had increased homing of CD34-positive cells on day 7 post-surgery,and more vessels on day 28 post-surgery by anti-von Willebrand factor staining.In addition,we observed an increase in the percentage of CD34-positive cells in the peripheral blood in G-CSF group.Conclusion G-CSF produces an obvious protective effect against chronic myocardial ischemia in rabbits by increasing stem cell mobilization,homing to ischemic myocardium and accelerating neovascularization.
6.Reflection on medical genetics teaching for lower grade undergraduates

In order to improve the quality of teaching in medical genetics,we adopted a lot of measures in medical genetics teaching program according to the requirements of basic course and the characteristics of lower grade undergraduates.Firstly,cultivating students' awareness of genetic diseases; Secondly,integrating related courses and selecting teaching contents seriously; Thirdly,guiding the class by introducing cases of illness.The results showed that these methods can promote the understanding of medical genetics and improve the quality of study in lower grade undergraduates.
7.Correlation between T help lymphocyte and autonomic nervous system in patients with congestive heart failure

Objective To investigate the association of T help (Th) lymphocyte and heart rate variability (HRV) in congestive heart failure (CHF) patients. Methods Ninety-six patients with CHF and thirty healthy persons were enrolled in the study. Time-domain HRV analysis was performed based on 24 hour Holter Electrocardiogram (ECG) monitoring. Interferon-γ (IFN-γ) was used as markers for the differentiation of Th1 subsets and interleukin-10 (IL-10) for the Th2 subsets. IFN-γand IL-10 in CD4 + T lymphocytes were quantified by 3-color flow cytometry. Results The frequency of IL-10-Producing T Cells in the CHF group was significantly lower than those in the healthy control ( ( 16.4 ± 5.8 ) % vs. ( 26.8 ± 3.7 ) %, t = 9. 243, P < 0. 001 ). The frequency of IFN-γ in the CHF group ( ( 18.4 ± 7.3 )% ) was significantly lower than that in the healthy controls ( (7.3 ±4.6) % ,t =7. 917, P < 0. 001 ). The following index of HRV in the CHF groups were all significantly lower than those in the healthy controls: (98. 6 ± 21.3) ms vs. ( 145. 1 ± 42. 6) ms for SDNN, (83. 9 ± 22.4) ms vs.(136.5 ±39.6)ms for SDANN, (40.6 ± 14.5) ms vs. (55.8 ± 17.9) ms for SDNNI, (20. 7 ± 12.9) ms vs.(29.1 ± 12.6) ms for RMSSD, (5.6 ± 3.7 ) % vs. ( 11.8 ± 4.4) % for PNN50 ( Ps < 0.05 ). In CHF patients, the frequency of IL-10 were positively associated with SDNN, SDANN, SDNNI, RMSSD and PNN50 ( r = 0. 49,0. 57,0. 58,0.47 and 0. 52 ,respectively,Ps < 0.01 ). In the CHF patients, the frequency of IFN-γand IFN-γ/IL-10 ratio were negatively associated with SDNN ,SDANN ,SDNNI, RMSSD and PNN50 ( r = - 0. 49, - 0. 54, - 0. 57, - 0.52,- 0.53, - 0. 52, - 0.64, - 0.57, - 0. 58, - 0. 67, Ps < 0. 01 ). Conclusions Autonomic nervous system is involved in the regulation of the balance of Th1/Th2 in patients with CHF. Sympathetic nerve system enhances the effect of Th1 ,whereas parasympathetic nervous system enhances the effect of Th2.
9.Role of PTEN on Proliferation of Airway Smooth Muscle Cells in Asthmatic Rats

Objective:To investigate the role of phosphatase and tensin homolog deleted on chromosome 10 (PTEN) in regulating proliferation of airway smooth muscle cells in ovalbumin(OVA)-induced asthma rats. Methods:Male Wistar rats (n=16) were randomized into OVA-induced asthma group and control group(8 rats each). The histomorphological changes of bronchia and lung tissues were observed by H-E staining. The expressions of proliferating cell nuclear antigen(PCNA) and PTEN were assayed by immunohistochemistry. Reverse transcription-polymerase chain reaction(RT-PCR) was carried out to determine the changes in the expression of PTEN mRNA. Results: The typical pathological features of asthma were revealed in the OVA-exposed rats including numerous inflammatory cells infiltrated around the bronchia and in the lung tissues, the thickened airway smooth muscle and the narrowed airway. The levels of PCNA were distinctly increased in OVA-induced asthma group than that of control(P < 0.05),while the levels of PTEN and PTEN mRNA were significantly decreased in lung tissues of OVA-exposed rats(P < 0.05). Conclusion:The gene inactivation of PTEN may play a pivotal role in proliferation of airway smooth muscle cells in asthma rats, and the most probable mechanism is associated with the functions of PI3K signaling pathway.
10.The influence of IL-2 on the apoptosis of activated and inactivated lymphocytes

Objective:To investigate the influence of IL-2 on the apoptosis of activated and inactivated lymphocytes.Methods:Selected an appropriate system as the model of activated lymphocytes by detecting the expression of Fas and FasL in two systems of one way MLR using RT-PCR. Detected the apoptosis of activated and inactivated lymphocytes after the stimulation of IL-2 with the methods of FITC-Annexin-V/PI staining.The resultaint change of membrane Fas was evaluated by FACS,and the lymphocytes from doner and acceptor were distinguished by CFSE staining.Results:To the inactivated spleen lymphocytes, exogenous IL-2 inhibited their apoptosis and the expression of Fas; while to the spleen lymphocytes activated by mixed lymphocyte reaction, exogenous IL-2 promoted their apoptosis and upregulated the expression of Fas, which did the same both to the doner cells and the acceptor cells.Conclusion:IL-2 can inhibit the apoptosis of inactivated lymphocytes, and promote the apoptosis of activated lymphocytes.These effects have something to do with the change of expression of Fas.
