Objective To investigate the clinical effect of Gonadotropin-releasing hormone agonist (GnRHa) combined with addback therapy after laparoscopic surgery on 153 cases with severe endometriosis patients. Methods 153 cases with severe endometriosis patients who underwent laparoscopic surgery from 2011 to 2013 were retrospectively analyzed in the Second Affiliated Hospital of Zhengzhou University. According to GnRHa or addback therapy three groups were divided:GnRHa group,GnRHa and addback therapy group, control group. The curative effects of different treatment group, adverse reactions, recurrence rates, and fertility were analyzed. The follow-up time was 6-24 months with an average time of 17.15 months. Results 51 patients suffered from obvious menopausal symptoms and received addback therapy (Progynova,0.5~1 mg/d). The VAS score and Kupperman score in GnRHa group,GnRHa and addback therapy group were significantly superior to the control group (P <0.05). And no statistical significance was found in the interval to relapse, recurrence and postoperative pregnancy rate (P>0.05). The VAS scores in the GnRHa group and addback therapy group were 2.14±2.02 and 1.73±2.17 , respectively (P > 0.05). The main side effect of GnRHa group is hot flashes, diaphores, insomnia, sexual dysfunction, etc. The incidences were higher in the GnRHa group than that in the addback therapy group. But the addback therapy can significantly relieve the symptoms of low estrogen introduced by the application of GnRHa (P<0.05). Conclusions Laparoscopic combined with GnRHa treatment can significantly improve clinical curative effect of endometriosis. Addback therapy could abate the side reaction of GnRHa and do not affect the clinical curative effect. The clinical curative effect in the GnRHa and addbacktherapy group were superior to the GnRHa group.

Objective To analyze the immunological characteristics of Trichinella spiralis secretory antigen P53 and to evaluate its value in diagnosis of trichinellosis. Methods An open read frame of secretory antigen P53 was cloned from Trichinella spiralis by reverse transcriptasepolymerase chain reaction (RT-PCR) and then sequenced. Bioinformatics analysis was performed to search for its homologues in other helminths and predict its potential linear B cell epitopes and T cell epitopes. The sequence coding mature peptide was inserted into prokaryotic expression vector pET28a(+) and the purified recombinant product was identified by Western blot using serum samples of patients infected with Trichinella spiralis or other helminth. Results Bioinformaties analysis results showed that there was no P53 homologue in other helminths, which indicated that there were many linear B cell epitopes and T cell epitopes in TsP53. The recombinant P53 antigen only reacted with the serum samples of patients infected with Trichinella spiralis without any cross-reaction with the serum of patients infected with other helminths. Conclusion P53 has strong immunogenicity and immunoreactivity, which may be a promising candidate for developing Trichinella spiralis specific diagnostic method.

OBJECTIVE:To establish a method for the content determination of saccharide in polysaccharides containing galact-uronic acid based on phenol-sulfuric acid method combined with correction factor method. METHODS:The determination condi-tion of phenol-sulfuric acid was optimized. A series of standard curves were drawn with glacturonic aid-glucose mixed control with different mass ratio. The content of saccharide in Codonopsis pilosula polysaccharides CPP1b samples containing galacturonic acid was determined. According to regression equation of galacturonic acid-glucose ratio of 0-100%,the correction factor was calculated by using C. pilosula polysaccharides CPP1b as the reference polysaccharide, and the results of content determination of saccharide were corrected. The rationality of this method was verified by using mixed monosaccharide control with same composition as C. pi-losula polysaccharides CPP1b. RESULTS:The correction factor of C. pilosula polysaccharide CPP1b to glucose was 3.33;in vali-dation test, the content of saccharide in mixed monosaccharide control with same composition as C. pilosula polysaccharides CPP1b was 103.47%. RSDs of precision and stability tests was <1%. The recoveries ranged 93.52%-107.35%(RSD=5.09%,n=6). CONCLUSIONS:The established method can accurately determine the content of saccharide in C. pilosula polysaccharides CPP1b containing galacturonic acid.
