Objective To investigate the effect of NS-398, a selective cyclooxygenase-2 (COX-2) inhibitor, on the expression of P-glycoprotein(P-gp) in a gastric cancer cell line SGC-7901. Methods SGC-7901 cells were treated with NS-398 in different concentrations (0, 10 ?mol/L and 100 ?mol/L) respectively. Prostaglandin E2(PGE2) was detected by ELISA. Twenty four hs and 48 hs later, the expression of mdr1 mRNA were detected by RT-PCR. P-gp protein expression in SGC-7901 cells was detected by immunocytochemical technique after NS-398 treatment.Results NS-398 can inhibit the expression of PGE2 in a dose-dependent manner(P
2.Correlated study for the levels of NO,NOS and PRL with neonatal h ypoxic-ischemia en cephalopathy

To explore th e role of nitric oxide(NO), nitric oxide synthetase (NOS)and prolactin (PRL) in the pathogenesis,progress and prognosis of neonatal hypoxic-ischemia encephalop athy (HIE),plasma NO, NOS and serum PRL were determined by radioimmunoassay(RIA) and colorimetry in 68 newbom infants with HIE and 30 normal control neonates, r espectively. The results showed that the levels of NO, NOS and PRL were signific antly higher in all neonates with HIE than that in the control group (P<0.05).There were in particular more higher levels of NO,NOS an d PRL in infants with either moderate or severe HIE(P<0 .01). It was noticed that the level of PRL was much higher in HIE infants with seizures than that in HIE infants without seizures. There was a positive relati onship between the levels of PRL and the duration of seizure attack( P<0.01). It is suggested that the levels of NO, NOS and PRL is ass ociated with HIE.The determination of these parameters will be valuable clinical ly for the diagnosis, judgement of prognosis and estimation of seizure attack in newborn infants with HIE.
3.Analysis on 1162 ADR Case Reports in Our Hospital during 2002~2004

OBJECTIVE:To investigate the characteristics of adverse drug reactions(ADR)of our hospital so as to promote rational drug use in the clinic.METHODS:1162ADR cases collected during2002~2004in our hospital were analyzed retrospectively.RESULTS:The occurrence of ADR induced by combined use of drugs accounted for33.6%of the total(390cases),chiefly through IV.Altogether531kinds of drugs involved ADR,in which the anti-infective agents(110kinds)took the lead which amounted to51.3percent of the total,then came the antineoplastic agents,cardiovascular system medicines and Chinese drugs preparations.The first3drugs that involved highest frequency of ADR occurrences were levofloxacin,cefo?peazone/sulbactam and azithromycin.The main clinic manifestations of ADR were skin damage,then gastro-intestinal damage and etc.There were28serious ADR cases.CONCLUSION:The cultivation of professional ability and quality of professional staff should be strengthened and an omnibearing ADR monitoring work should be carried out so as to reduce and avoid the occurrence of ADR.
4.Antagonist of leukotriene B4 receptor 1 attenuates cisplatin induced acute kidney injury in mice and its associated mechanism

Objective To investigate the effect of pretreatment with U75302,antagonist of leukotriene B4 receptor 1 (BLT1),on cisplatin induced acute kidney injury in mice and its immunoregulatory mechanism.Methods Healthy C57BL/6 mice were randomized into four subgroups:1.healthy control group;2.cisplatin group;3.U75302 control group;4.cisplatin + U75302 group,n=6.Group 2 and 4 received intraperitoneal injection of cisplatin (20 mg/kg) on day 0,group 3 and 4received intraperitoneal injection of U75302 (5 μg/mouse) on day 0 and day 2.Mice were sacrificed on the 3rd day and blood and kidney were collected.Renal function and histological changes were estimated,the infiltration of immune cells were determined by flow cytometry,the level of peroxidase (MPO) in kidney were determined by colorimetry,relative expression of TNF-α,IL-1β,CXCL1,CXCL2 were detected by Real-time PCR.Results Compared with healthy control group,levels of BUN,Scr were higher in cisplatin group with serious tubular structural damage.There were more neutrophils,macrophages,CD4+ T lymphocytes,CD8+ T lymphocytes in kidneys of cisplatin group,the level of MPO and relative expression of TNF-α,IL-1β,CXCL1,CXCL2 were also higher in cisplatin group.Compared with cisplatin group,lower BUN [(17.75±1.80) mmol/L vs (42.6±6.66) mmol/L,P <0.05],Scr were found in cisplatin+ U75302 group with less tubular structural damage.Meanwhile,U75302 reduced infiltration of neutrophils [(146±13)×103/g vs (296±66) ×103/g,P < 0.05],macrophages [(245± 13)× 103/g vs (420±78)× 103/g,P < 0.05] in the kidney.Levels of MPO [(1.756±0.283) U/g vs (3.308±0.577) U/g,P<0.05] and relative expression of TNF-α,IL-1β,CXCL1,CXCL2 were also lower.Conclusions BLT1 antagonist U75302 protects mice against AKI induced by cisplatin,and the mechanism is associated with reduced infiltration of inflammatory cells in kidney and the inhibition of kidney inflammation.
5.Analysis of Adverse Drug Reactions Reported in Our Hospital

OBJECTIVE:To understand the situation of adverse drug reactions occured and the harm to pa?tients.METHODS:298cases reported in our hospital were classified,analysed and evaluated.RESULTS:165kinds of drug caused adverse drug reactions,of which anti-infective agents occupied the first place,anticancer agents the second,cardiovas?cular and herbal agents induced ADRs very often too.The combined use of drugs accounted for28.5%in patients with ADRs.Most ADRs were induced by drugs given via intravenous route.The main types of ADRs were skin damage,gastro-intestinal damage,fever,etc.20cases had severe ADRs(6.7%).CONCLUSION:The adverse drug reactions difficult to observe and chronic ones should be paid more attention to.
6.Comparison of rabbit degenerative intervertebral disc models constructed by anulus puncture and annulus incision

BACKGROUND:Constructing animaI model of intervertebraI disc degeneration which more faithfully mimics the pathologic hallmarks of human intervertebral disc degeneration can be a beneficial assistance for further intervertebraI disc degeneration therapy.However,there is not an accepted optimal model for intervertebral disc degeneration study OBJECTIVE:To compare the rabbit model of degenerative intervertebral disc constructed by anulus puncture and anulus incision.METHODS:Totally 32 New Zealand white rabbits were randomly divided into the anulus puncture group and annulus incision group.Intervertebral disc of L_(3-6) 6was exposed by extro-peritoneal approach,and the discs were injured by puncturing the anulus or cutting the anulus The deep and direction were controlled.Pathological change of intervertebral disc was checked with MRI and histopathological examination at weeks 2,4,12,and 20 after operation.RESULTS AND CONCLUSION:At week 4 after operation.the area of nucleus gelatinosus was deflated with enlarged anulus fibrosus,T_2-weighted image(T_2WI)declined,blurred,and the height of intervertebral space was also decreased,the grade of T2 value in the anulus puncture group was lower than that of the annulus incision group(P<0 05):with time prolonged,T2 scores increased,and the intervertebraI space narrowed.which reached a peak at week 20 after operation.The differences had no significance.The histological sections demonstrated that the cell content in nucleus pulposus was increased gradually.The rabbit model of intervertebraI disc degeneration can be successfully constructed by the methods of anulus puncture and annulus incision.The degeneration of incision modelis more severe than that of puncture model.Anulus puncture method can faithfully mimic intervertebral disc degeneration after damage in human being.
7.Investigation on mechanical property of the compound scaffold of Sol-Gel bioactive glass/collegan

Objective To investigate the mechanical prosperity and degradation rate of the scaffolds with compounding collagen and the nano sol-gel derived bioactive glass were studied,and provide the theoretical basis for the further application of collagen based scaffolds.Method The scaffold with compounding collagen and the nano sol-gel derived bioactive glass(58S)were prepared using the freeze-drying techniques with the bioactive glass as phase addition.By affecting the aggregation state of the collagen fibers with adjusting the supplementation of bioactive glass to change the microstructures of the compound scaffolds and finatly the compound scaffolds with different mechanical properties were prepared.Results (1)As the aggregation state of the collagen fibers changes,the scaffolds with the coarser collagen fibers is prepared with the diameters 400-600 nm approximately.The coarser collagen fibers will play an important role in improving the mechanical property and slowing down the degradation rate of the collagen based scaffolds.(2)The interactions between bioactive glass and collagen are studied by FTIR and Raman technologies.When the quality of content of collagen in the compound scaffolds is lower than 20%,the secondary structure of collagen is damaged severely.Conclusion The composite scaffolds with the mass ratio of collagen to bioactive glass 40:60 has the best performance in mechanical property and degradation,which will be helpful for further applications.
8.Constituents and bioactivities of Lamiophlomis rotata.

OBJECTIVETo investigate the chemical constituents from Lamiophlomis rotata and the bioactivities of 8-epideoxyloganic acid.
METHODThe constituents were isolated by using a combination of various chromatographic techniques including column chromatography over ployamide, silica gel and Sephadex LH-20. Structures of the isolates were identified by spectroscopic data analysis. Bioactivities were screened by using models in vivo.
RESULTFive constituents were isolated. 8-epideoxyloganic acid was isolated for the first time in L. rotata and also in lamioplomis genus. 8-epideoxyloganic acid could significantly inhibit aectic acid-induced twisting times and slower the time of homeostatsis, also inhibit xylene-induced ear edema in mice.
CONCLUSION8-epideoxyloganic acid possesses bioactivities of analgesia, homeostasis and anti-inflammatory.

9.Clinical application of jejunal feeding tube for early enteral nutrition after surgical treatment of upper digestive tract malformation in newborns

Objective:To study the clinical application of jejunal feeding tube (J-tube) for early enteral nutrition after surgical treatment of upper digestive tract malformation in newborns.Methods:From January 2019 to December 2021, newborns with upper digestive tract malformation received stage Ⅰ small bowel resection and anastomosis in our hospital were enrolled in this prospective randomized controlled study. According to different types of postoperative nutritional support, these patients were randomly assigned into J-tube group and control group using block randomization method. The J-tube group were given enteral nutrition vis J-tube within 48-72 h after surgery and the control group were given oral feeding after the recovery of gastrointestinal function. Calories and proteins intake, growth indicators, duration of hospital stay and parenteral nutrition, time needed for full oral feeding and complications were compared between the two groups.Results:A total of 24 patients were in J-tube group and 28 in controlled group. No significant differences existed on the general status between the two groups ( P>0.05). The average daily intake of calories and proteins in j-tube group in the first week after surgery were significantly higher than control group [(108.7±8.3) kcal/(kg·d) vs. (97.9±7.0) kcal/(kg·d), (3.4±0.3) g/(kg·d) vs. (3.1±0.2) g/(kg·d)] ( P<0.05). No significant differences existed in the average daily intake of calories and proteins during the second postoperative week between the two groups ( P>0.05). Compared with control group,J-tube group showed increased growth velocity in head circumference and weight over time ( P<0.05), while the trend over time in length growth was not significant ( P>0.05). No significant differences existed in the duration of hospital stay and parenteral nutrition, time needed for full oral feeding and complications between the two groups ( P>0.05). Conclusions:Enteral nutrition via J-tube 48-72 h after surgery is safe and feasible in the postoperative nutritional management of newborns with upper digestive tract malformation. This strategy may promote physical growth after surgery without increasing the incidences of complications.
10.Glutathione S-transferase M1 and T1 genotypes and endometriosis risk: a case-controlled study.

OBJECTIVETo investigate the correlation between glutathione S-transferase (GST) M1 and T1 genotypes and endometriosis risk (EM).
METHODSPolymerase chain reaction (PCR) technique was used to detect the presence or absence of the GSTM1 and GSTT1 genes in genomic DNA isolated from the blood samples of 68 Han Chinese women with endometriosis and 28 without endometriosis.
RESULTSThe frequencies of GSTM1 and GSTT1 null genotypes in women with endometriosis were 0.721 (49/68) and 0.779 (53/68), respectively, and in women without endometriosis were 0.429 (12/28) and 0.321 (9/28), respectively. There was a significant difference with regard to the frequencies of GSTM1 and GSTT1 null genotypes between the women with and without endometriosis (P < 0.01). Furthermore, the frequencies of GSTM1 and GSTT1 null genotypes were significantly higher in the patients with stage III and IV endometriosis [0.731 (38/52) and 0.788 (41/52), respectively] than in women without endometriosis (P < 0.01), and the frequency of GSTT1 null genotype was statistically higher in patients with stage I and II endometriosis [0.75 (12/16)] than in the women without endometriosis (P < 0.01). No correlation between GSTM1 and GSTT1 null genotypes and age, induced abortion or dysmenorrhea was detected in this study (P > 0.05).
CONCLUSIONGSTM1 and GSTT1 null genotypes may be risk factors for the development of endometriosis.
