1.Expression and significance of plasma circulation microRNA155 in ulcerative cilitis

Objective To explore the diagnostic value of the concentration of plasma circulation microRNA155 (miRNA155) in ulcerative cilitis (UC) and its correlation with clinical characteristic of UC.Methods From October 2010 to August 2012,a total of 136 patients diagnosed as UC were enrolled,and at same time,170 healthy individuals were set as healthy control.The blood samples of all participants were obtained and plasma was isolated.The adsorption column was used for RNA extraction according to miRNeasy kit instruction.RNA was reverse-transcribed into cDNA with miScript reverse transcription kit.cDNA was a template and miRNA155 real-time quantitative polymerase chain reaction (PCR) was performed with miScript SYBR Green PCR kit.The relative quantity of miRNA155 expression was calculated with 2-△△Ct method.Analysis of variance were performed for comparison between groups.Receiver operating characteristic curve analysis was used for the diagnostic value of miRNA155 concentration in UC.Multiple linear regression analysis was used for the correlation between miRNA155 concentration and clinical characteristics of UC.Results The concentration of plasma circulation miRNA155 of patients with UC ((1357.43±326.15) fmol/L)was higher than that of healthy controls ((1140.70 ± 312.47) fmol/L) and the differences were statistically significant (F=35.56,P<0.01).The area under receiver operating characteristic curve of the concentration of plasma circulation miRNA155 of patients with UC was 0.847,and the 95 %CI was 0.806 to 0.888 (P<0.01).When the concentration of plasma circulation miRNA155 was 1404.51 fmol/L,its specificity in the diagnosis of UC was 94.7%,and sensitivity was 40.4%.There was correlation between the concentration of plasma circulation miRNA155 and the disease activity in patients with UC (F=12.91,P<0.05).However there was no correlation with the severity and location of the disease (both P>0.05).Conclusion Plasma circulation miRNA155 highly expressed in patients with UC,and its concentration is correlated with the disease activity.
2.Serologic Presentation of Helicobacter Pylori Infection in Patients With Inflammatory Bowel Disease

Objective To investigate serologic manifestation of Helicobacter Pylori (H. pylori) infection in patients with inflammatory bowel disease (IBD) and the role of H. pylori infection in pathogenesis of IBD.Methods We measured anti-H.pylori-IgG and anti-H.pylori-CagA of 45 IBD patients and their with sex and age matched 45 chronic gastritis(CG) control patients during 4 years, and analyzed the relationship between seroprevalence of H.pylori and inflammatory range of ulcerative colitis(UC).Results There were 40 UC patients and 5 Crohn's disease(CD) patients; the positive anti-H.pylori-IgG patients in IBD was 40 0% and in chronic gastritis was 66 7% respectively(? 2=6 43,P0 05).Conclusions The anti-H.pylori-IgG positive rate is in high level in IBD patients, but that is lower than chronic gastritis patients.H. pylori infection could play a inhibition role in the inflammation range of UC. The anti-H.pylori-CagA positive rate of IBD patients and pan-colorectal UC are lower than their controls respectively, but there is no obviously different in statistics.

ObjectiveTo know about the correlation analysis of the physical and mental status of organ transplanted patients' family members during the patients' rehabilitation period and the long term survival of the transplanted organs.MethodsA total of 600 organ transplanted patients' family members were investigated by Zung self-rating anxiety scale (SAS) and simple coping style questionnaire.The results underwent subsequent analysis.ResultsAnxious state of organ transplanted patients' family members was more severe than that of domestic norm (P<0.01).There were some differences in terms of anxious state of family members with different genders,education backgrounds,income and the resource of medical expenditure (P<0.05).Family members most took positive coping styles,whereas fewer adopted negative coping styles (P<0.01).Positive coping styles were negatively correlated with the anxiety of family members (P<0.01) and positively correlated with long term survival of transplanted organ(P<0.05).However,negative coping styles of family members were significantly positively correlated with their anxiety (P<0.05) and were negatively correlated with long term survival of transplanted organ.ConclusionsThe anxiety generally exists in organ transplanted patients' family members.There are differences in terms of anxious state among family members of different genders,different education backgrounds,income or with the resource of medical expenditure.The more they adopt positive coping styles,the lower anxiety level they show and the longer the transplanted organ survive.Conversely,the more they adopt negative coping styles,the higher anxious level they show and the shorter the transplanted organ survive.
7.The initial study on microRNA expression profile in human colorectal cancer with liver metastasis

Objective To investigate the microRNAs expression profile in human colorectal cancer with or without liver metastasis and try to screen miRNA associated with liver metastasis in colorectal cancer. Methods Twenty five surgical resected colorectal cancer specimens were collected and frozen in liquid nitrogen. Three without liver metastasis and three with liver metastasis were selected, from which total RNAs were isolated. The expressions of miRNAs in these two types of specimens were detected by illumine microRNA microarray, and the difference of miRNA expression was screened. The biochip results were verified with real-time RT-PCR in all colorectal caner specimens. Results The miRNA expression was significantly different in colorectal cancer with liver metastasis and without liver metastasis. Compared with colorectal cancer without liver metastasis, 28 miRNA expressions was different in colorectal cancer with liver metastasis, 4 up regulated and 24 down regulated. The quantity of miR-139-3p expression in colorectal cancer with liver metastasis was 1.75±0.40, up regulated compared with that incolorectal cancer without liver metastasis(0. 69 ±0.58,P<0.05). The quantity of miR-19a expression in liver metastasis was 0. 39±0. 20, downregulated compare with no liver metastasis( 1.38 ± 0.98, P<0. 05). The result of miRNA biochip was consistent with that of RT-PCR. Conclusion The difference of miRNA expression might relate to liver metastasis of colorectal cancer. The specific miRNAs expression profile might provide new target for diagnosis and treatment of colorectal cancer with liver metastasis.
8.Anti-inflammatory and anti-nociceptive effects in mice of water and ethanol extracts of roots and rhizomes of Asarum heterotropoides var. mandshuricum.

OBJECTIVETo compare the anti-inflammatory and anti-nociceptive effects of the different water extracts which were prepared by regular decoction time or removing volatie oil and ethanol extracts which were prepared in different concentration ethanol of Xixin (the roots and rhizomes of Asarum heterotropoides var. mandshuricum), and then to investigate the anti-inflammatory and antinociceptive mechanisms.
METHODXylene-induced ICR mouse ear edema and hot plate test were utilized to evaluate the anti-inflammatory and anti-nociceptive effects of Xixin at different dose level (water extracts of regular decoction time or removing volatie oil at the dose of 0.8 g x kg(-1) and 1.6 g x kg(-1); 95% ethanol extracts at the dose of 0.91 g x kg(-1) and 1.82 g x kg(-1); 50% ethanol extracts at the dose of 0.76 g x kg(-1) and 1.52 g x kg(-1)). Xylene-induced adrenalectomized mouse ear edema model was used to study the anti-inflammatory mechanisms. To reveal the anti-nociceptive mechanisms, mice were pretreated with naloxone in the hot plate test.
RESULTThe water extracts significantly reduced the weight of ear edema as compared with control group. Inhibition ratios were 43.20% and 63.69% at the higher dose, respectively. The ethanol extracts also significantly reduced the weight of ear edema and the inhibition ratios were 61.86% and 52. 56% at the higher dose, respectively. Mice administered water extracts delayed the latency times in the hot plate test. The anti-nociceptive effects of water extracts peaked at 2.0 h after i.g. administration. The hot plate latency times were increased by 51.27%, 62.78%, 60.08% and 68.00% at peak time, respectively. Regular decoction time group showed more significant effects in both models. The anti-inflammatory effect of 95% ethanol extracts was similar to 50% ethanol extracts. The water extracts were not effective in reducing xylene-induced adrenalectomized mouse ear edema. The anti-nociceptive effect of water extracts was blocked by naloxone.
CONCLUSIONBoth the water extracts and ethanol extracts of Xixin showed considerable anti-inflammatory potency against xylene induced inflammation. The water extracts produced anti-nociception in thermal model. The water extracts prepared in regular decoction time showed better anti-inflammatory and anti-nociceptive effects. Both the 95% ethanol and 50% ethanol extracts showed similarly anti-inflammatory effects. The anti-inflammatory effect of water extracts related to adrenal gland. The anti-nociceptive effect of water extracts was involved in activating opioid receptor.

9.Effects of Gaultheria yunnanensis on adjuvant arthritis in rats.

OBJECTIVEGaultheria yunnanensis. are used widespreadly in the south of China to treat rheumatoid arthritis. The aim of this study was to provide an experimental basis for G. yunnanensis to therapy rheumatoid arthritis.
METHODWe prepared water extracts, ethanol extracts, n-butanol extracts, ethyl acetate extracts and the rest of ethanol extracts from G. yunnanensis. Then, the n-butanol extracts were applied to macroporous resin and eluted with water, 30% ethanol, and 95% ethanol. Rheumatoid arthritis was induced by Freund's complete adjuvant injected into right postpedes in Wistar rats which was utilized to elucidate the anti-inflammatory effect of different extracted liquid of G. yunnanensis. Rats were intragastric injected (ig) with extracts as experimental group or normal saline as control group.
RESULTFreund's complete adjuvant induced arthritis was successfully established: paw edema were increased after Freund's complete adjuvant injection, peaked at 2 or 3 day, then decreased, the paw edema were increased again at 7 or 8 day, and persisted 15 d. Water extracts, n-butanol extracts or ethyl acetate extracts could a significantlly decrease the paw edema as compared with the control group (P < 0.05, P < 0.01). The effect of n-butanol extracts was the most powerful. Further, n-butanol extracts eluant with water and 30% ethanol decreased the paw edema. The activity of extracts eluant with 30% ethanol was stronger than that of eluant with water.
CONCLUSIONG. yunnanensis displays considerable effects against Freund s complete adjuvant induced arthritis in rats, which is in concordance with clinical practice. n-Butanol extracts and both of the eluants with water and 30% ethanol produce a significant decrease in the paw edema. 30% ethanol eluants show a stronger activity than others. The effects against rheumatoid arthritis of different parts of G. yunnanensis differ in degree. It is deserved to explore the potential mechanisms of anti-inflammtion of the G. yunnanensis, especially the n-butanol extracts eluant with 30% ethanol.

10.Clinical observation the effect of Tongue Manipulation of Pulling and massage on stroke in acute stage with allolalia

Objective To analyze the clinical effect of tongue traction massage in the improvement of speech deficit in acute stage of ischemic stroke. Methods A total of 60 cases of acute ischemic stroke aphasia patients from June 2015 to June 2016 were selected and divided into experimental group (30 cases)and control group(30 cases)by random digits table method,the control group was treated with drug therapy, experimental group was treated with drug plus tongue traction massage therapy. The curative effect of tongue body massage and traction safety was compared between two groups. Results After 2 courses of treatment, there was no significant change in vital signs in the experimental group, the total effective rate was 90.00%(27/30), and the total effective rate of the control group was 70.00% (21/30), there was significant difference between two groups(Z=-2.05,P<0.05). Conclusions In the treatment of acute ischemic stroke aphasia,the tongue traction massage,good clinical effect,can effectively improve the patient′s speech function,simple operation,safety,compliance is good,worthy of promotion.
