Journal of Central South University(Medical Sciences) 2012;37(5):469-473
OBJECTIVE:
To investigate the antioxidant effect of different solvent extracts from persimmon leaves (PL) in diabetic mice induced by streptozotocin (STZ).
METHODS:
The total ethanol-extracted fraction of PL was further extracted with chloroform, ethyl acetate and n-butanol, in that order, the residues after ethanol extraction were water-extracted and alcohol-precipitated, and concentrated. The hypoglycemic effects of different solvents extracts from PL were evaluated in diabetic mice induced by STZ. The experimental mice were randomly divided into groups: control group, model group, glibenclamide group, low and high dosage groups of the various solvent extracts. The drugs were administrated to mice in every morning for 15 days. During this time period, the contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were determined.
RESULTS:
The water-extracted and ethanol-precipitated fractions and the ethyl acetate-extracted fraction markedly reduced the content of MDA and increased the activity of SOD in the livers of STZ-induced diabetic mice (P<0.01 or P<0.05). The chloroform-extracted and n-butanol-extracted fraction did not markedly reduce the content of MDA nor increase the activity of SOD in liver of STZ-induced diabetic mice (P>0.05).
CONCLUSION
The ethyl acetate-extracted fraction, water-extracted and ethanol-precipitated fraction of persimmon leaves have potential value in the treatment of diabetes. The mechanism of action of the antioxidant is related to the hypoglycemic effects of extracts from persimmon leaves.

West China Journal of Stomatology 2012;30(4):364-367
OBJECTIVEThe purpose of this study was to investigate the regulatory role of tyrosine kinase 2 with immunoglobulin-like and epidermal growth factor homology domains (Tie2) on apoptosis and proliferation in the endothelial cells.
METHODSRNA interference (RNAi) technique was used to silence Tie2 gene expression by transfecting an expression vector containing short hairpin RNA(shRNA) for Tie2 into human umbilical vein endothelial cells (HUVECs). Real time quantitation reverse transcriptase polymerase chain reaction (QRT-PCR) and Western blot were used to monitor Tie2 mRNA, as well as protein expression. The proliferation of HUVECs was examined by methyl thiazolyl tetrazolium (MTT), and the apoptosis was detected under microscope. HUVECs transfected with pGenesil-hk was negative control, and HUVECs transfected with nothing was empty control.
RESULTSTie2 mRNA expression was down-regulated 24 h and 48 h after transfection, and Tie2 protein expression was significantly down-regulated at 24 h and 48 h (P< 0.05), especially 48 h after transfection. The apoptosis rate was conspicuously higher in experimental group than in negative control and empty control group after 48 h (P<0.05). The growth monitoring showed that proliferation was also markedly inhibited in experimental group (P<0.05) compared with two control groups.
CONCLUSIONDown-regulated expression of Tie2 by RNAi can promotes apoptosis of HUVECs and has an anti-proliferation activity effect on them.

Cystic echinococcosis (CE), a zoonotic disease caused by Echinococcus granulosus at the larval stage, predominantly develops in the liver and lungs of intermediate hosts and eventually results in organ malfunction or even death. The interaction between E. granulosus and human body is incompletely understood. Exosomes are nanosized particles ubiquitously present in human body fluids. Exosomes carry biomolecules that facilitate communication between cells. To the best of our knowledge, the role of exosomes in patients with CE is not reported. Here, we isolated exosomes from the sera of patients with CE (CE-exo) and healthy donors and subjected them to liquid chromatography-tandem mass spectrometry analysis. Proteomic analysis identified 49 proteins specifically expressed in CE-exo, including 4 proteins of parasitic origin. The most valuable parasitic proteins included tubulin alpha-1C chain and histone H4. And 8 proteins were differentially regulated in CE-exo (fold change>1.5), as analyzed with bioinformatic methods such as annotation and functional enrichment analyses. These findings may improve our understanding about the interaction between E. granulosus and human body, and may contribute to the diagnosis and prevention of CE.
