China Pharmacy 2001;0(08):-
OBJECTIVE:To evaluate the status quo and tendency of the use of nootropics in our hospital.METHODS:The nootropics used in our hospital from 2004 to 2006 were analyzed statistically.RESULTS:The consumption sum of nootropics in our hospital increased year by year,2.03 times in 2005 and 1.46 times in 2006 respectively over their previous year. The consumption sum of Ginkgo biloba extract for nootropics took the lead from 2005 to 2006,over 60% of which were medical insurance drugs.CONCLUSION:Leading the list of nootropics in our hospital is plant amedica, with its consumption sum and DDDs showing the equal importance in drug control, suggesting the great market demand for nootropics.

Objective To probe into the role of apoptotic cells in the disease development of systemic lupus erythematosus. Methods Apoptotic thymocytes injected intraperitoneally into syngeneic normal C57 mice and lupus prone BXSB mice, and then the level of anti dsDNA and anti ssDNA antibodies of IgM and IgG classes, and the concentration of urine protein and immune complex of IgG class deposited in kidney were detected. Results The apoptotic thymocytes induced the production of anti dsDNA and anti ssDNA antibody of IgG class in C57 mice, and their levels were higher in female BXSB mice than in male ones. The concentration of urine protein of male BXSB mice injected with apoptotic thymocytes was noticeably higher than that of the control male BXSB mice. Conclusion Apoptotic cells have immunogenicity, and may be the major source of autoantigens in lupus BXSB mice.Lupus genetic factors and Yaa gene can potentiate the immunogenicity of apoptotic cells or accelerate their inducing the lupus nephritis.

Objective To investigate the effect of IGF-1 on the TGF-β3 induced chondrogenesis of MSCs encapsulated in alginate beads and its application in cartilage tissue engineering. Methods MSC chondrogenesis in alginate beads was induced by TGF-β3 and/or IGF-1.Collagen type Ⅱ,aggrecan and Sox-9 expression was evaluated by immunostaining,RT-PCR and Western blot, respectively.Scanning electron microscope and laser confocal microscope were used to observe the differentiated chondrocytes when cultured on the chitosan-based scaffold. Results TGF-β3 with IGF-1 induced MSCs in alginate beads to express the higher level of collagen type Ⅱ,aggrecan and Sox-9 than any other growth factor alone (P<0.05).The correlation coefficient between Sox9 and collagen type Ⅱ or aggrecan was 0.95 and 0.91,respectively.The chitosan-based scaffold supported the cell's adhesion, migration and proliferation.Conlusion IGF-1 enhances the TGF-β3-induced MSC chondrogenesis via upregulating Sox9 expression. The chitosan-based scaffold is biocompatible with the differentiated chondrocytes.

Chinese Journal of Immunology 1985;0(02):-
T4 cells were isolated twice separately from peripheral blood of the same donor. After activation by lectin, the isolated cells were culturedwith the medium containing self-made IL-2 for a long period. The culturedcells were identified for OKT antigens by immuno-fluorescent technique onday 10 and 22 of the cultivation, and the appearance of both T4 and T8antigens on the same cell (double marker cell)could be detected in about60-80%of cultured cells. The double marker T cells decreased in numberand the percentage of single marker T cells, either T4 or T8 cells, increasedgradually along with the cultivation. The function of double marker T cellswere investigated and the suppressor activity of these cells on PWM indu-ced antibody production response by cooperation between B cells and fresh T4cells was found. These results suggest that human peripheral blood T4 andT8 cells are not terminaly differentiated cells, because T4 and T8 antigencan be coexpressed on the same cell during the earlier cultivation.Afterlong period of activation,T4 and T8 double marker cells may change intoeitherT4 or T8 cells.

Objective To observe the effects of Ketangte 2 on experimental diabetes in mice. Methods Adrenalin and streptozocin(STZ) were used to induce hyperglycemia mice model and diabetic mice model respectively. The mice were divided into five groups randomly:model group, glibenclamide control group,high-,middium-,low-dosage Ketangte 2 groups. The administration lasted consecutive 15 days. The blank control group and model group were treated with the same volume of saline. After treatment, the levels of blood sugar were measured in hyperglycemia mice induced by adrenalin, and the fasting blood-glucose,2-hour blood-glucose,insulin levels in diabetic mice induced by STZ were determined.Meanwhile the histopathological changes of pancreas were observed under light microscope. Results For STZ-induced diabetic mice, the hypoglycemic effect of Ketangte 2 was obvious(P

Objective:To study the mechanism of interleukin-10(IL-10)inhibiting the function of dendritic cells(DCs).Meth-ods:Cultured C57BL/6 mouse bone marrow-derived DCs were divided into 5 groups:control group,LPS stimulated group,IL-10 treated group,IL-10+Rapamycin treated group and Rapamycin treated group .The regulatory mechanism of IL-10 on dendritic cells were evalua-ted from DCs function ,Flow cytometry was used to analyse the expression of DCs surface co-stimulator CD80 ,CD40 expression ,the abil-ity of uptaking antigen and stimulating T cell to proliferate;ELISA was used to detect the cytokines IL-6 and TNF-α.Western blot was used to analyse the autophagy related protein LC3.Compared the differences between the groups.Results:(1)Compared to LPS stimu-lated group,IL-10 treated group,DCs surface co-stimulator CD40,CD80 were decreased,IL-6 and TNF-αsecretion level and the ability to stimulate T cell to proliferate were decreased ,the ability to capture OVA antigen was increased .Compared to IL-10 treated group ,the DCs surface co-stimulator CD80 was decreased ( P<0.05 ) ,IL-6 and TNF-αsecretion level and the ability to stimulate T cell to prolifer-ate were increased(P<0.0001)in IL-10+rapamycin treated group. In addition, autophagy related proteins LC3Ⅱ/LC3Ⅰwas decreased in IL-10 treated group.Conclusion:IL-10 may regulate functions of DCs through inhibiting the autophagy of DCs .

Objective:To detect the changes of tumor necrosis factor-α(TNF-α) in nasopharyngeal secretion of patients with nasopharyngeal carcinoma(NPC).Method: The content of TNF-α in nasopharyngeal secretion was determined by radioimmunoassay (RIA) in 20 controls,52 patients with NPC.Result:The average concentrations of TNF-α were (12.95±4.62)pmol/L in control group,(33.68±15.13) pmol/L in NPC group and (64.00±11.57)pmol/L in NPC with necrosis group. The content of TNF-α in nasopharyngeal secretion in control group was lower significantly than that of patients with NPC (P<0.01).Conclusion:The level of TNF-α in nasopharyngeal secretion of NPC patients was higher than that of normal subjects. To detect the content of TNF-α in nasopharyngeal secretion is a useful test for the study and diagnosis of NPC.
