1.The correlation between RDW and different types of acute appendicitis

Objective the purpose of this study is to investigate the correlation of RDW and different pathological types of acute appendicitis(AA).Methods According to the pathological classification,AA can be divided into pure,suppurative and gangrenous AA.The RDW values were compared among the 3 kinds of AA patients,and compared with control group.Correlation analysis were performed in patients with the 3 types of AA respectively.ROC was used to analyse RDW′s the diagnosic value for different types of AA.Results In patients with pure and suppurative AA,RDW values were statistically different from those in control group(P <0.05),and weren′t correlated with WBC count(P >0.05);while in patients with gangrenous AA,RDW values were not statistically different from those in control group,and were positively correlated with WBC count(r=0.646,P <0.05).Conclusion The decline RDW is correlated with simple and suppurative AA,and can provide some reference for the diagnosis of simple and suppurative AA,but not be used as diagnostic indicator because the sample quantity is not enough in the study.
2.Converting rules from international classification of disease codes of trauma to abbreviated injury scale values

Objective To develop converting rule from international classification of disease (ICD) codes of trauma to abbreviated injury scale (AIS) values so as to provide clues for a simple and convenient trauma scoring way. Methods The corresponding set relations were made for ICD-9CM codes and AIS-90 codes with similar code meanings in the trauma diseases. Based on medical theories and knowledge, 8 indeces closely associated with injury severity were worked out for each ICD-9CM code of trauma diseases by using the database of discharge abstracts of trauma patients. Discriminant analysis was used to set up corresponding relations of ICD-9CM codes with AIS values. Results Seven indices were selected for discriminant function. Corresponding relations of all ICD-9CM codes of trauma diseases with AIS values was made through nonparametric discriminant method. Under deliberation and revision by several clinical experts on traumatology, the converting rule was established eventually. Consistency analysis between the sample's international classification of disease injury severity score (ICISS) values and injury severity score (ISS) values showed that ICISS values were closely correlated with 2-ICISS (Y=Ln), with correlation coefficient of 0.831 1 ( P

Objective To develop a new comprehensive trauma score method based on international classification of diseases(ICD) and evaluate the effectiveness of its use in cerebral injured patients.Methods The model of the new trauma score method was constructed by referring to those of some certain comprehensive trauma score methods generally used around the world.Receiver operating characteristics(ROC) analysis was employed to evaluate the effectiveness of outcome prediction for 355 cerebral injured patients using the new developed trauma score method.Statistical comparisons were performed between the AUC for the new trauma score values and that of NISS values derived from two professional raters.Results The area under the curve(AUC) for the new trauma score values was as large as 0.925 4,whose 95% CI ranged from 0.863 4 to 0.987 5,and is statistically different from 0.5(P
4.Chondrogenic differentiation of mouse bone marrow mesenchymal stem cells induced by cartilage-derived morphogenetic protein-2 in vitro.

To study the cartilage differentiation of mouse mesenchymal stem cells (MSCs) induced by cartilage-derived morphogenetic proteins-2 in vitro, the MSCs were isolated from mouse bone marrow and cultured in vitro. The cells in passage 3 were induced into chondrogenic differentiation with different concentrations of recombinant human cartilage-derived morphogenetic proteins-2 (0, 10, 20, 50 and 100 ng/mL). After 14 days of induction, morphology of cells was observed under phase-contrast microscope. Collagen II mRNA and protein were examined with RT-PCR, Western blotting and immunocytochemistry respectively and the sulfate glycosaminoglycan was measured by Alcian blue staining. RT-PCR showed that CDMP-2 could promote expression of collagen II mRNA in an dose-dependant manner, especially at the concentration of 50 ng/mL and 100 ng/mL. Immunocytochemistry and Western blotting revealed a similar change. Alcian blue staining exhibited deposition of typical cartilage extracellular matrix. Our results suggest that mouse bone marrow mesenchymal stem cells can differentiate into chondrogenic phonotype with the induction of CDMP-2 in vitro, which provides a basis for further research on the role of CDMP-2 in chondrogenesis.

5.Three-dimensional CT reconstruction for analysis of normal acetabular spatial contour and its curvature transformation

BACKGROUND: The determination of the normal acetabular spatial contour, the pattern of its curvature transformation, and adaptability between acetabulum and femoral head plays an important role in the description of spatial relationship of the hip joint the management and evaluation of hip disorders, especially developmental dysplasia of the hip.OBJECTIVE: Based on spiral threa-dimensional CT data of normal hip joints, to create isoline map of each acetabulum and femoral head by a well-developed and cooperating programmed computer software so as to determine the normal acetabular spatial contour and the pattern of its curvature transformation.METHODS: Three-dimensional CT data of 56 normal hip joints, aged from 6 to 17 years, were collected. After three-dimensional simulation and surface shade display (SSD) reconstruction by a well-developed VxWork software, the isoline map was created using contours map in Gecmorphology, and each isoline map of the normal acetabulum and femoral head was analyzed via a specific computer software (Terrain). RESULTS AND CONCLUSION: The isoline array of normal acetabulum revealed the modality of some concentric "C" or oval "O",with at least one close "O" at the central zone, indicating that there existed dome shape at the superior zone of acetabulum. The modality of sparse isolines around dome illustrated larger curvature radius and more slowgoing change of spatial contour transformation of this zone, while the dense isolines on the bottom zone Of acetabulum represented a small curvature radius,steep shape, and rapid changing profile of this zone. The isoline array of normal femoral head was multiple concentric "O" with different space between each line. The congruence isolines of acetabular dome and superior surface of femoral head emerged as the similar trend of geomorphological characteristics, revealing that both the spatial contour and the curvature pattern between acatabulum and femoral head are well-adaptable to each other. Results show that the isolines of acatabulum and femoral headcan stersoscopicly visualize the spatial contour and curvature pattern of the topographic surface of the hip joint.
6.Analysis of cognitive function and its influenced factors in patients with prostate cancer after maximal androgen blockade therapy

Objective To investigate the cognitive function of patients with prostate cancer after maximal androgen blockade therapy and its influenced factors,and to provide a new way for early prevention strategy.Methods Montreal cognitive assessment (MoCA),hospital anxiety depression scale (HAD),social support rating scale (SSRS) and self-designed questionnaire were used in 56 cases treated with maximum androgen blockade therapy for more than six months and 37 cases who underwent radical prostatectomy treatment to evaluate their cognitive function and collect the observation indexes between January 2013 and October 2015.Based on MoCA score,all patients were divided into cognitive dysfunction group (n =40) and normal cognitive function group (n =53).The observation indexes in two groups were compared and cognitive function with different treatment in two groups were analyzed.The changes on the influencing factors of cognitive function in patients were filtered using multivariable logistic regression analysis.Results In the cognitive dysfunction group and normal group,the proportion of MAB treatment was 80.0% (32/40) vs.45.3% (24/53),the age was 73.7 vs.73.7 years,the proportion of solitary was 32.5% (13/40) vs.13.2% (7/53),the proportion of depressive symptoms was 87.5% (35/40) vs.62.3% (33/53),the social support level was 32.5 vs.41.1 and the proportion of testosterone decreased was 95.0% (38/40) vs.45.3% (24/53).All events showed statistically significant differences (P <0.05).Compared MAB treatment group with radical surgical treatment group,the testosterone level was (0.27-±O.15) vs.(12.14 ± 1.86) nmol/L,visual space and executive function score was 4.18 ±0.79 vs.4.54 ±0.56,attention score was 4.73 ±0.99 vs.5.16 ±0.79,delayed memory score was 3.75 ± 1.21 vs.4.30 ± 1.05 and MoCA score was 26.13 ± 1.48 vs.27.27 ± 1.39,which all showed the statistically significant difference (P < 0.05).The results of multiple regression analysis showed that age (OR =1.183,95% CI 1.135-1.223),depressive symptoms (OR =1.092,95% CI 1.047-1.149),social support (OR =0.897,95% CI O.838-0.956),testosterone (OR =2.105,95% CI 1.369-4.083) were the influenced factors of cognitive dysfunction.Conclusions The incidence of cognitive dysfunction in patients with prostate cancer after maximal androgen blockade therapy more than six months was higher than others.Age,depression,social support level and testosterone levels were related to the occurrence of cognitive dysfunction.

Objective To investigate the protective effect of ischemic postconditioning on lung injury in situ during lung ischemic reperfusion.Methods 24 SD rats were randomly divided into 3 groups,sham-operated group(S),ischemic-reperfusion group(I/R) and ischemic postconditioning group (IpostC).IpostC was established by several brief reperfusion-ischemia (30 s,5 cycles) before continuous reperfusion.AKT,p-AKT,p70S6K,p-p70S6K protein expression,apoptotic cells were tested by Western blotting and TUNEL,wet to dry weight ratio(W/D) in lung tissue were determined respectively.The lung pathological changes were also observed.Results Compared with S group,expression of Akt、p-Akt、p70S6K、p-p70S6K and the ratio of W/D,apoptotic index in lung tissue all markedly increased in I/R and IpostC group (P < 0.05).Compared with I/R group,expression of Akt、p-Akt、p70S6K、p-p70S6K markedly increased in IpostC group(P <0.05),while the ratio of W/D and apoptotic index significantly reduced (P < 0.05),the pathological injury in IpostC group also reduced significantly.Conclusion IpostC has a protective effect on lung ischemic reperfusion injury via PI3K/Akt pathway.
8.Effect of inulin-type hexasaccharide on forced swimming tests in mice and rats and DRL 72 s in rats

AIM To determine whether O-β -D-fructofuranosyl-〔(2→1 )-O-β-D-fructofuransyl〕4α-D-glucopyranoside (inulin-type hexasaccharide, IHS), a monomer extracted from the roots of Morinda of ficinalis How, has antidepressant action. METHODS Fo rced swimming tests in mice and rats and differential-reinforcement-of-low-r ate 72 second schedule (DRL 72 s) in rats were used. RESULTS In the forced swimming test in mice, IHS (80 mg*kg-1, po), like the effe ct of clinically effective antidepressant desipramine (10 mg*kg-1, ip), produced significant decrease in immobility time. IHS (20 mg*kg-1,po ) also elicited significant decrease in immobility time in forced swimming test in rats, which was comparable to the effect of desipramine (40 mg*kg-1, po). Moreover, in the DRL 72 s in rats, IHS (5~10 mg*kg-1, ip), s imilar to desipramine (5 mg*kg-1, ip), elicited significant increase in reinforcers. CONCLUSION These findings demonstrate that IHS has antidepressant action and is an effective component extracted from the root s of Morinda officinalis How.
9.Antiarrhythmic effect of κ-opioid on Cx43 in rat heart during mycardial ischemia and reperfusion via inhibiting β-adrenergic receptor pathway

Aim To investigate the effect of U50488H(a selective κ-opioid receptor agonist)and isoproterenol(ISO,a β-adrenergic receptor agonist)on ventricular arrhythmias and Cx43 during myocardial ischemia and reperfusion in rats.Methods 60 rats were randomly divided into five groups,ie,normal control group,I/R group,ISO+I/R group,U50488H+ISO+I/R group,Nor-BNI+U50488H+ISO+I/R group.The incidence of ventricular arrhythmias and arrhythmia score were determined. The expression of Cx43mRNA was tested by RT-PCR.The expression of Cx43 protein in myocardial cell was tested by an immunohistochemical approach with a quantitative imaging system.Results ① Compared with the I/R group,arrhythmia score was increased with administration of ISO(P<0.05).U50488H intravenously injected before ISO significantly decreased the arrhythmia score(P<0.05).② Compared with the normal control group,the expression of Cx43 mRNA was decreased in the I/R group(P<0.05).With administration of ISO,the amount of Cx43 mRNA was not significantly increased.③ Compared with normal control group,total and phosphorylated Cx43 proteins were significantly decreased in the I/R group(P<0.05),and the phosphorylated Cx43 was also decreased with administration of ISO.Compared with ISO+I/R group,phosphorylated Cx43 was increased with administration of U50488H (P<0.05).Conclusion κ-opioid receptor agonist U50488 H antagonizes the arrhythmias through the regulation of Cx43 during myocardial ischemia and reperfusion via inhibiting β-adrenergic receptor pathway.
