1.PHARMACOKINETIC STUDY OF CEFUROXIME

Clinical pharmacological study of cefuroxime was carried out in 8 normal volunteers. The drug obeyed two-compartment model kinetics in serum. The mean peak serum concentrations were 32.14, 89.06 and 63.11 (.is/ml respectively after IM, IV injection and IV infusion of 0.75g cefuroxime. Serum concentrations after IV administration dec re- ased rapidly, the elimination half lives ranged from 1.05-1.48 hours Absorption after IM injection was rapid and complete, the bioava-ilability being 98.89%,The apparent volume of distribution. ranged from 0.34 L/kg to 0.37 L/kg after IM and IV administration which showed that cefuroxime distributed mainly in extracellular fluid.The mean urinary recovery of cefuroxime was more than 90% within 24 hours after the administration of the drug.Based on its pha.rmacokinetic characteristics and antimicrobial activity, a therapeutic regimen is suggested for use in the treatment of various infections.
2.Effects of Roots of Hawthornleaf Raspberry (Rubus crataegifolius)on Lipid Peroxidation in Tissues

Experiments in vitro demonstrated for the first time that an EtOAc extract of roots of Rubus crataegifolius Bge.can markedly inhibit the for mation of lipid peroxide in rat brain,heart. liver and kidney tissucs. When administered intragastrically for 15 successive days,the EtOAc extract markedly inhibited the formation of lipid peroxide in plasma and tissues of liver and brain of mice. The above results showed that the EtOAc extract from roots of R. crataegifolius Bge.had anti-lipid peroxidation effect.
3.COMPARATIVE STUDY ON HEMATOPOIETIC RECONSTRUCTION EFFECTS OF PERIPHERAL BLOOD STEM CELLS MOBILIZED BY ANTI-CD49d MONOCLONAL ANTIBODY AND rhG CSF IN MICE

The present experiment was to study the hematopoietic reconstruction effects of peripheral blood stem cell (PBSC) mobilized by anti-CD49d monoclonal antibody (McAb) and recombinant human granulocyte colony-stimulating factor(rhG-CSF) in mice. PBSCs from NS-treated mice (control group), rhG-CSF-mobilized mice (experimental group1), and anti-CD49d McAb-mobilized mice (experimental group 2), The changes in respectively, were transplanted to BALB/c mice pre-conditioned with high-dose chemotherapy and total body irradiation white blood cell (WBC) count, four-weeks survival rate, bone marrow nuclear cells (BMNC), granulocyte-macrophage colony-forming units (CFU-GM), and colony-forming unit-spleen (CFU-S) were observed. OBJECTIVEThis study aims to evaluate the optical data of the different sites of the cobalt-chrome (Co-Cr) alloy abutments covered by four different all-ceramic crowns and the color difference between the crowns and target tab using a digital dental spectrophotometer.
METHODSTen Co-Cr alloy abutments were made and tried in four different groups of all-ceramic crowns, namely, Procera aluminia, Procera zirconia, Lava zirconia (Lava-Zir), and IPS E.max glass-ceramic lithium disilicate-reinforced monolithic. The color data of the cervical, body, and incisal sites of the samples were recorded and analyzed by dental spectrophotometer. The CIE L*, a*, b* values were again measured after veneering. The color difference between the abutments covered by all-ceramic crowns and A2 dentine shade tab was evaluated.
RESULTSThe L* and b* values of the abutments can be increased by all of the four groups of all-ceramic copings, but a* values were decreased in most groups. A statistical difference was observed among four groups. After being veneered, the L* values of all the copings declined slightly, and the values of a*, b* increased significantly. When compared with A2 dentine shade tab, the ΔE of the crowns was below 4.
CONCLUSIONFour ceramic copings were demonstrated to promote the lightness and hue of the alloy abutments effecttively. Though the colorimetric baseline of these copings was uneven, veneer porcelain can efficiently decrease the color difference between the samples and thee target.

5.Effect of the Wnt/LRP5/β-catenin signaling pathway on the pathogenesis of postmenopausal osteoporosis

Objective To investigate the role of the Wnt/LRPS/3-catenin signaling pathway in the pathogenesis of postmenopausal osteoporosis.Methods Fifty female Wistar rats aged 6-month-old,were randomly divided into control group ( NS,n =24 ) and ovariectomized group ( NOVX,n =26),NOVX underwent bilateral ovariectomy.At 0,4 and 8 weeks,all of rats were measured blood estrogen ( E2 ) and bone mineral density (BMD),4 and 8 weeks,low density lipoprotein receptor-related protein 5 (LRP5),3-catenin and Runx2 mRNA in bone were measured respectively by reverse transcription (RT)-PCR.Results In 4 and 8 weeks,compared with NS which had (117±29) and (114+15) pmol/L in E2 level,(0.098 ± 0.016 ) and (0.095 ± 0.028 ) g/cm2 in BMD,NOVX had significantly decreased to ( 92±15 ) and(95±22) pmol/L in E2 level ( P<0.05 ),( 0.076 ± 0.016) and ( 0.052 ± 0.013 ) g/cm2 in BMD values ( P < 0.01 ).And bone tissue LRP5,β-catenin and Runx2 mRNA expression was 1.02 ± 0.06,1.04 ±0.05,1.07 ±0.21 in NS,NOVX was significantly reduced to 0.97 ± 0.04,0.58 ± 0.05,0.86 ±0.03 (P<0.05).Conclusion Wnt/LRP5/β-catenin signaling pathway may be important in the pathogenesis of postmenopausal osteoporosis.
6.Clinical analysis of 37cases of clavicular midpiece fracture treated by open reduction and internal fixation with TMZF reconstruction plate

Objective To investigate the clinical efficac y of internal fixation with Titanium Alloy(TIA)reconstruction plate in treatment o f clavicular midpiece fracture.Methods 37cases of clavicular midpiece frac-ture underwent open reduction and in ternal fixation with TIA.All the 37c ases were followed up and their clini cal efficacy were analyzed.Results All the clavicle fractures achieved clinical union and satisfactory fun ctional re-covery.Conclusion Open reduction and internal fixatio n with TIA reconstruction plate is on e of the best choices for clavicular midpiece fractures,for it can free the patients from distress of reoperation.[
7.Relationship of Th17 cells as well as interleukin (IL)-17A and IL-23R mRNA with psoriasis vulgaris

Objective To assess the number of peripheral blood Th17 cells and mRNA expressions of IL-17A and IL-23R and their correlations with disease severity in patients with psoriasis vulgaris (PV).Methods Tissue specimens were resected from the lesions of 25 patients with PV and normal skin of 10 human controls,and venous blood samples were obtained from 20 of the patients and all of the normal human controls.Reverse transcription PCR and flow cytometry were carried out to measure the mRNA levels of IL-17A and IL-23R in these tissue specimens and quantify the number of peripheral blood CD4+IL-17+ T lymphocytes.Psoriasis area and severity index (PASI) was calculated for these patients.Results A significant increase was observed in the IL-17A and IL-23R mRNA levels in the patients compared with the controls (0.996 ± 0.231vs.0.437 ± 0.096,t =10.572,P < 0.05; 1.006 ± 0.339 vs.0.491 ± 0.196,t =6.015,P < 0.05).The levels of both IL-17A mRNA and IL-23R mRNA were positively correlated with PASI (r,=0.67,0.70,respectively,both P < 0.05).The number of CD4+IL-1 7+ T lymphocytes in peripheral blood showed no significant differences between the patients and controls.No statistical correlation was observed between the counts of CD4+IL-17+ T lymphocytes and expression levels of IL-17A or IL-23R mRNA in psoriatic lesions.Conclusions In patients with PV,there is an increase in the expressions of IL-17A and IL-23R mRNA in lesions,which are correlated with disease severity,while no significant change is observed in the number of peripheral blood CD4+IL-17+ T cells.
8.Nursing and effect of cyclogest in threatened abortion treatment

Objective To explore the effect of cyclogest and femotroneinthreatened abortion treatment and summerize key points of nursing. Methods Forty-two cases of first trimester threatened abortion women were selected from department of gynecology and obstetrics, which were randomly divided into the control group (n=21) and experiment group (n=21)using random digit table. The control group was given femotrone 40mg intramuscular injection each day while the experiment group was given 90 mg of vaginal progesterone suppository (Cyclogest) each day. Compare the improvement of symptoms, the success rate of pregnancy, changes in serum human chorionic gonadotropin (HCG) and progesterone. Results The improvement of symptoms, curative effects and change of HCG of both groups were no significantly difference. But elevatorconcentration of serum progesterone in femotrone group was higher than that of the cyclogest groupwith statistically difference. Conclusions Although cyclogest cannot appear significantly in increasing serum progesterone levels, it has the same effect as femotrone. Recommendations and notes about the use and side effects of cyclogest and femotrone should be provided for education to alleviate concerns of patients.
9.Effect of lipopolysaccharide on expression of ATP-binding cassette transporter A1 in alveolar macrophage cells of rats

Objective To evaluate the effect of lipopolysaccharide (LPS) on the expression of ATP-binding cassette transporter A1 (ABCA1) in alveolar macrophage cells of rats.Methods The alveolar macrophage cells of rats NR8383 were seeded in 6-well plates at a density of 1 × 106 cells/ml (2 ml/well) and randomly divided into 6 groups:control group (group C,n =24),0.2 μg/L LPS group (group L0.2,n =12),2.0 μg/L LPS group (group L2.0,n =12),20.0 μg/L LPS group (group L20.0,n =60),200.0 pg/L LPS group (group L200.0,n =12),and 20.0 μg/L LPS + ABCA1 siRNA group (group L20.0 + siRNA,n =12).The cells were routinely cultured in group C.LPS with the final concentrations of 0.2,2.0,20.0 and 200.0μg/L was added to the culture medium in L0.2,L2.0,L20.0 and I200.0 groups,respectively.In group L20.0 + siRNA,siRNA 50 nmol/L was added to the culture medium and 12 h later LPS 20.0 μg/L was added.In group C,6 wells were chosen for determination of ABCA1 mRNA and protein expression.At 24 h of incubation with LPS 0.2,2.0 and 200.0 μg/L,or at 2,6,12 and 24 h of incubation with LPS 20.0 μg/L,6 wells were chosen and the cell suspension was obtained for measurement of ABCA1 mRNA expression (by real-time fluorescent quantitative PCR),and ABCA1 expression (by flow cytometry).At 12 h of incubation with 20.0 μg/L LPS or with 20.0 μg/L LPS + 50 nmol/L siRNA,6 wells were chosen and the cell suspension was obtained for measurement of TLR4 mRNA expression (by real-time fluorescent quantitative PCR) and TLR4 expression (by flow cytometry).Results Compared with group C,the expression of ABCA1 mRNA and protein was significantly down-regulated in L0.2,L2.0,L20.0 and L200.0 groups,and the expression of ABCA1 mRNA and protein was up-regulated in L20.0 and L20.0 + siRNA groups.In L20.0 group,the expression of ABCA1 mRNA and protein was gradually down-regulated with the prolonging time of incubation with LPS.Compared with group L20.0,the expression of TLR4 mRNA and protein was significantly up-regulated in group L20.0 + siRNA.Conclusion LPS can down-regulate the expression of ABCA1 in alveolar macrophage cells of rats,however,ABCA1 can inhibit the synthesis of TLR4.
10.Experiment research of contrast-enhanced ultrasonography in diagnosis of pancreatic laceration

Objective To approach the diagnostic value of contrast-enhanced ultrasonography(CEUS)for the detection of traumatic laceration of pancreas. Methods Sixty cases of pancreatic traumatic model were made in twelve healthy swines after the animals were anesthetized and laparotomized. Then the conventional ultrasonography(US) and CEUS were performed in each case to diagnose the traumatic region,immediately. The results were compared with surgical findings. Results Among sixty injuries,the detection rate of conventional ultrasonography was 66. 7%,the detection rate of CEUS was 88.3%. Conclusions CEUS shows higher detection rate than conventional US in diagnosing pancreatic laceration,and it also can improve the diagnostic value of ultrasound for the detection of pancreatic laceration.
