Objective To investigate the effect of donor specific immature dendritic cells infusion in combination with CsA on differentiation of liver recipient′s Th1/Th2 cell in rats. Methods Allograft models of orthotopic rat liver transplantation were established, 88 LEW rats (recipient) and SD rats (donor) were randomly divided into three groups: (1) in control group no immunosuppresive drug was used after liver transplantation. (2) in CsA group 1 mg/100 g body weight of CsA was injected intraperitoneally every the other day for 3 times beginning the 2nd day after liver transplantation. (3) in CsA+DC group additional 1? 10 6 immature DC from donor bone marrow was injected through penile vein at the 8th day after operation, in combination with CsA as in CsA group. Rats were sacrificed for immunopathological examination and detection of INF ?mRNA, IL 6 mRNA of celiac lymph node on 5th, 10th, 15th, and 25th day after transplantation. 2.Renal Tuberculosis CT Diagnosis and Mis-diagnostic Analysis

Objective To evaluate the CT values in diagnosis of renal tuberculosis. Methods 26 cases were collected , the CT findings of renal tuberculosis were analysed and compared with the data of operation-pathology and clinical followed up. Results Of 26 cases, 22 cases diagnosed by CT as renal tuberculosis of one side were confirmed by operation-pathology but one (pseudaneurysm). Bilateral renal tuberculosis and renal cysts were diagnosed in 2 cases respectively by CT, while these 4 cases were confirmed as tuberculosis by other auxiliary checking methods and followed-up. The multiple tuberculoses cavities appeared as a petal-shape low-density area and other untypical signs on CT. The single cavity only showed as a low-density area should be distinguished with cystonephrosis. At early renal tuberculosis, a great quantity of effusion beneath the perinephrium was the only sign on CT.Conclusion CT scan has a special value for diagnosis renal tuberculosis. Typical renal tuberculosis can be made closely combining with the clinical information.
3.Effects of SWH solution on hepatic energy metabolism in rats

Objective To investigate the effects of SWH solution on hepatic energy metabolism in SD rats compared with UW solution. Methods Levels of ATP, ADP, and AMP in hepatic tissues were examined by high performance liquid chromatography (HPLC) while ALT, AST, and LDH were examined and the 7-day survival rate of animals was observed with orthotopic liver transplantation model in rats. Results Levels of ATP, TAN, and EC in 18-h group were significant lower than those in 8-h group. No significant difference was found between 12-h group and 8-h group. AST, ALT, and LDH were significantly increased after preservation for 18-h than preservation for 8-h. No significant difference was found between 12-h group and 8-h group. The 7-d survival rate of animals in SWH group was higher than that in UW group. The change tendency of ATP, TAN, and EC was the same as the tendency of 7-d survival rate of animals. Conclusion These results suggest that levels of ATP, ADP, and AMP in hepatic tissues are closely associated with the survival rate, and SWH solution is effective in protecting hepatic energy metabolism.
4.Vascular endothelial growth factor in improving liver function of rats following partial liver transplantation

Objective To determine the impact of exogenous vascular endothelial growth factor (VEGF) on rat liver function after partial liver transplantation. Methods Male SD rats which had undergone 30% partial liver transplantation were divided into VEGF treatment group (n=60) and control group (n=60). Animals in each group were injected with solution of VEGF or saline, respectively. Serum ALT, AST, and hyaluronic acid (HA) levels were assessed at 0, 12, 24, 72, 168 hours after operation. Results Serum ALT, AST and HA levels were significantly decreased at 72 hours after transplantation in VEGF treatment group compared with controls (t =4.681, 4.252, 2.853, P<0.05). In accordance with liver function, treatment with VEGF minimized the damage to the liver sinusoidal endothelial cells and decreased the infiltration of inflamatory cells. Conclusion Administration of VEGF may improve the liver function of rats after partial liver transplantation by maintaining the integrality of the sinusoidal structure and reducing the inflamation reaction.
5.Effects of cold preservation and reperfusion injury on the regeneration of donor liver and associated mechanisms

Objective To investigate the effects of cold preservation and reperfusion injury on the regen-eration of donor liver and to study the mechanisms. Methods Male SD rats were divided in to sham group (6 rats), UW 1 h group (48 rats) and UW 12 h group (48 rats). Liver tissue specimens were collected at different time points after orthotopic liver transplantation or sham operation. The morphology of liver tissue was observed via light microscope and transmission electron microscope. Proliferation of hepatocytes and sinusoidal endothelial cells (SECs) were assessed by a double immunostaining technique using antibodies against rat endothelial cell antigen-1 and bromodeoxyuridine (BrdU). Expression of vascular endothelial growth factor (VEGF) and its receptors, fins-like tyrosine kinase-1 (flt-1) and fetal liver kinase-1 (flk-1) was evaluated by immunohistochemistry. The mRNA expression of flt-1 was detected by a RT-PCR method. Mean comparison in groups was conducted by one-way ANOVA or t test. Results BrdU labeling indexes of hepatocytes and SECs in UW 12 h group was significantly higher than those in UW 1 h group (F = 61.45,41.4, P < 0.05). The proliferation of hepatocytes peaked at 48 h after operation in both UW 1 h group and UW 12 h group. However, the proliferation of SECs was fallen behind compared to hepatocytes, with a peak appeared at 72 h in UW 1 h group and at 96 h in UW 12 h group, respec-tively. The expression of VEGF was up-regulated in both UW 1 h group and UW 12 h group compared to sham group. Furthermore, expression of flt-1 and flk-1 was found to be mainly limited in SECs, with a peak in expres-sion occurring between 72 h and 96 h, coinciding with the peak in SECs proliferation in UW 1 h group. Conversely, flt-1 was found to be reduced significantly on mRNA level at any time throughout the experiment in UW 12 h group compared to sham group (F = 141.67, P < 0.05). Conclusion Reduced expression of flt-1 results in a retarded regeneration of SECs, and then the recovery of rat donor liver function is delayed after cold-preserved transplantation.
6.Prognostic effect of number of lymph nodes sampled in patients with stage Ⅰ non-small cell lung cancer

Objective To investigate the prognostic effect of quantity of lymph node(LN)resected in operations of patients with stage Ⅰ non-small cell lung cancer(NSCLC).Methods The clinical, pathological and follow-up data of 74 patients with stage Ⅰ NSCLC who were treated with surgery from January 1998 to December 2002 Beijing Friendship Hospital, Affiliated to Capital Medical University were reviewed retrospectively.Grouping the patients, according to the quantity of lymph node resected, the Kaplan-Meier method and Cox proportional hazards model was used for univariate analysis and multivariate analysis of factors with prognostic effect.Results The five year survival rate and disease-free survival(DFS)rate of these 74 patients were 64.9% and 47.3%.The univariate analysis showed that tumor size(P =0.016),T-staging (P =0.008)and extent of lymph node dissection(P =0.013)could influence the survival rate.The 5-year OS and DFS rates of patients with less than 6 LNs resected were less than the other group(more than 6 LNs)apparently.The multifactorial analysis indicated that other than staging, the quantity of lymph node resected was also an influence factor of prognosis.Conclusions The OS rate of patients has positive correlation with quantity of lymph node resected in operations.Six LNs must be resected leastways in operations of patients with stage Ⅰ NSCLC.
7.A TENTATIVE STUDY ON THE MECHANISM OF INHIBITION OF PLATELET AGGREGATION BY FISH OIL RICH IN EPA

The mechanism of decreasing platelet aggregation by fish oil rich in n-3 fatty acids was studied in rabbits and rats. These animals were randomly divided into four groups. In rats, group Ⅰ was fed a basal diet; group Ⅱ was fed a high fat diet containing 7.5% lard, 0.25% bile salts, 0.5% cholesterol, group Ⅲ and Ⅳ received the diet with 5% lard, 0.25% bile salts,0.5% cholesterol and 0.5 ml fish oil or 0.5ml olive oil respectively. In rabbits, group Ⅰ was fed a basal diet; group Ⅱ received 20g diets containing 10.4% lard, 2.5% cholesterol, then, some basal diet; group Ⅲ and Ⅳ received 20g diets with 4.2% lard, 2.5% cholesterol and 2 ml fish oil or olive oil respectively, then, some basal diet. Fish oil contained 20% EPA and 50% DHAThree months later, plasma TXB2/6KPGF1? ratios were 1.4, 2.2, 1.0, 1.7. Plasma TXB2, cholesterol, serotonin, calcium, fibrinogen and blood viscosity were significantly lowered in group Ⅲ as compared with group Ⅱ. Platelet cAMP was greatly increased in group Ⅲ than that in group Ⅱ n-3 fatty acids in platelet, plasma and tissues such as liver and heart were definitively higher in group Ⅲ.These experiments showed that fish oil inhibited platelet aggregation and thrombosis formation as a consequence of changing the fatty acids composition in tissue membrane and decreasing TXA2/PGI2 ratio which may stimulate platelat cAMP synthesis and then decrease some agglutinating factors such as serotonin and calcium etc, released from platelet. On the other hand fish oil lowered blood cholesterol, which may change blood cell membrane fluidity, lowered blood fibrinogen and viscosity, so the situation of blood platelet and its surroundings could be improved.

Objective To determine the effects of exogenous VEGF on the serum levels of HGF and IL-6 in rats following partial liver transplantation.Methods Totally 180 SD rats were subjected to 50 percent partial liver transplantation,then each 60 rats were treated either with VEGF,anti-VEGF or normal saline.HGF,IL-6 and hepatic biochemical markers were assessed at 0,12,24,72,168 h after operation.Results In the VEGF group,the serum levels of HGF and IL-6 were significantly increased as compared with those of anti-VEGF group or control group.Blockage of endogenous VEGF led to a marked increase of ALT and AST.Conclusion VEGF can markedly increase the secretion of HGF and IL-6,which may be related to its enhancing liver regeneration and improving liver function.
10.The establishment of an arterialized liver transplantation model in mice

Objective To establish a stable orthotopic liver transplantation(OLT)model with hepatic artery reconstruction in mice.Methods Of the 66 OLT mice,33 recipient mice underwent conventional 'two cuff' technique without hepatic artery reconstruction(control group),33 with hepatic artery reconstruction using 'stent' technology(experimental group).Results The 2 week posttransplant survival rate was 84.85%(28/33)and 81.82%(27/33)in the arterialized and nonarterialized groups respectively.Serum γ-GT and ALP level of arterialized recipient were much lower than those of nonarterialized animals.The pathology of the donor liver showed that rearterialization helps to relieve bile duct injury of liver allograft.Conclusions The hepatic artery supply plays a central role in preventing bile duct injury after transplantation.This mouse OLT model may meet the need for the study of problems associated with liver transplantation.
