1.The correlation between APOBEC3G mRNA in peripheral blood mononuclear cells and serum hepatitis C viral RNA level

Objective To study the relationship between APOBEC3G mRNA level in peripheral blood mononuclear cells (PBMC) and serum hepatitis C viral RNA level in patients with chronic hepatitis C infection. Methods TaqMan real-time fluorescence relative quantitative polymerase chain reaction (RT-PCR) was used to quantify APOBEC3G mRNA levels in PBMC from 49 patients with chronic hepatitis C (CHC) and 31 healthy subjects. The relationship between APOBEC3G mRNA level and hepatitis C virus (HCV) viral load was analyzed. SPSS11. 0 statistics software was used for t test and regression analysis. Results APOBEC3G mRNA level in CHC patients [(1.5×10-5±1.9×10-5 ) copy/mL] was significantly lower than that [( 5. 2 × 10-5 ± 5. 5 × 10-5 ) copy/mL] in the healthy control subjects (t=-3.005, P<0.01). While APOBEC3G mRNA level was not related with HCV viral loads (r=-0.082, P>0.05). Conclusion HCV has an inhibitive effect on APOBEC3G expression, whereas APOBEC3G doesn't affect HCV replication directly in vivo.
2.Analysis on Tn17/Treg cells in patients with pelvic tuberculousis

Objective To investigate the cellular immunologic response of TH 17/Treg cells in the peripheral blood of pelvic tuberculosis patients and explore their roles in the pathogenesis of pelvic tuberculosis.Methods The intracellular flow cytometry was performed to evaluate the expressions of TH 17 and Treg cells in 46 pelvic tuberculosis patients and 25 healthy controls in childbearing age.Twenty-eight of the 46 pelvic tuberculosis patients were followed up to monitor the variation of the TH17/Treg cells after 3 months and 6 months of anti-tuberculosis treatment.Results The percentage of TH 17 cells in the peripheral blood of pelvic tuberculosis patients was (3.26 ± 1.30) % which was significantly lower than that of healthy controls [(4.92 ± 1.71) %,P < 0.01].The percentage of Treg cells in the patients was (5.18 ± 1.53) % which was significantly higher than that of healthy controls [(3.26 ± 1.10) %,P < 0.01].The percentage of TH17 cells in the pelvic tuberculosis patients after 6 months of treatment was (4.67 ± 1.75) % which was significantly higher than that in the patients before treatment and after 3 months treatment [(3.26 ± 1.30) %,P < 0.01 and (3.70 ± 1.06) %,P <0.01,respectively].The percentage of Treg cells in pelvic tuberculosis patient after 6 months of treatment was (3.93 ±0.94)% which was significantly lower than that in the patients before treatment and after 3 months of treatment [(5.18 ± 1.53)%,P <0.01 and (4.94 ± 1.51) %,P < 0.01,respectively].The percentage of Treg cells in the patients after 6 months of treatment was still significantly higher than that of controls (P < 0.05).The TH 17/Treg ratio before treatment was significantly lower than that of healthy controls (P < 0.01),and the TH 17/Treg ratio was increased after 3 months of treatment but it did not show significant difference compared with that before treatment.The TH 17/Treg ratio after 6 months of treatment (1.18 ± 0.34) % was significantly increased in contrast to those after 3 months of treatment and before treatment [(0.77 ± 0.21) %,P < 0.01 and (0.55 ± 0.13) %,P < 0.01,respectively].The TH 17/Treg ratio could not rise to the normal level even after 6 months of treatment.Conclusion Both the TH 17 and Treg cells may involve in the immunologic responses of pelvic tuberculosis patients and the imbalance of TH1T/Treg cells may remain persistently.
3.Effects of Cook balloon cervical dilator on full-term nulliparous parturients under epidural analgesia

To explore the feasibility and effects of Cook balloon cervical dilator on full-term nulliparous parturients under epidural analgesia.The observation group (n =40) received Cook balloon cervical dilation under epidural analgesia.However, the control group (n =40) had no epidural analgesia.Latent phase of labor was shorter in observation group than that in control group [(198.7 ± 65.7) vs.(242.9 ± 78.9) min, P <0.05].And the incidence of cesarean section and perineal laceration was lower under epidural analgesia(8% vs.15%, 45% vs.50% respectively).There were no effects on success rate of induced labor in neither group.Cook balloon could shorten latent phase of labor, relieve the pain of parturients and increase no infection rate [leukocyte count : (8.82 ± 2.74) × 109/L vs.(9.10 ± 3.06) × 109/L, C-reactive protein: (4.73 ±0.87) mg/L vs.(4.82 ±0.90) mg/L respectively].It is worth wider clinical popularization.
4.The risk factors of deep vein thrombosis in the lower extremities among acute spinal cord injury patients

Objective To identify the risk factors for deep vein thrombosis(DVT) in the lower extremities during hospitalization for acute spinal cord injury(SCI).Methods Retrospective data analyses were performed for 49 SCI patients admitted within a week after they were diagnosed.The patients were divided into a DVT-free(n =40) group and a DVT group(n =9) according to their lower limbs' color Doppler ultrasound results.And the subjects' gender,age,chronic diseases (diabetes,hypertension and hyperlipidemia,etc.),complications (pulmonary infection,urinary tract infections and pressure sores,etc.),the history of cancer,smoking history and lower extremity fractures were recorded.Intermittent pneumatic compression treatment,the early rehabilitation intervention,antithrombotic therapy,history of venous catheterization,operative intervention,blood transfusion history,muscle strength of lower extremity and time from onset to being transfered to rehabilitation department were also recorded Single factor analysis,was conducted,followed by Binary logistic regression to pick out the risk factors for DVT.Results The result of the binary logistic regression analysis showed that gender (OR 6.87 ; 95% CI 1.09-43.22 ;P < 0.05) and intermittent pneumatic compression treatment (OR 12.47 ; 95% CI 1.09-142.36 ; P < 0.05) were significant predictors of DVT.Conclusion Gender and intermittent pneumatic compression treatment are related with DVT risk for patients with acute SCI.
5.Surface functional electrical stimulation combined with electroacupuncture in treating neurogenic bladder after the spinal cord injury

Objective To observe the therapeutic effect of surface functional electrical stimulation combined with electroacupuncture on neurogenic bladder after the spinal cord injury.Methods Forty cases of neurogenic bladder after the spinal cord injury were randomly divided into a control group (n =20) and a combination group (n =20).The combination group was treated with surface functional electrical stimulation combined with electroacupuncture on the basis of internal urethral catheterization,while the control group was treated with surface functional electrical stimulation on the basis of internal urethral catheterization.The bladder urination was tested using urodynamic study before and after two months of treatment.Results There was no significant difference between the 2 groups in the average maximum bladder capacity,detrusor pressure at filling time,maximal urethral closure pressure,maximum urine flow rate and residual urine volume before the treatment.After the treatment,however,the average maximum bladder capacity,maximal urethral closure pressure and maximum urine flow rate in the combination group were significantly higher than those of the control group,while the average detrusor pressure at filling time and the residual urine volume of the combination group were significantly lower than the control group.Conclusion Surface functional electrical stimulation combined with electroacupuncture could remarkably improve bladder function of patients with the neurogenic bladder after the spinal cord injury.
7.Study on Metabolic Differences of Cajanonic Acid A in Different Species of Liver Microsomes by UPLC-MS/MS

OBJECTIVE: To establish a determination method for the concentration of cajanonic acid A (CAA) in liver microsome incubation system, and to compare the metabolism characteristics of it in different species of liver microsomes. METHODS: CAA was dissolved in liver microsome incubation system of rat, Beagle dog and human initiated by reduced nicotinamide adenine dinucleotide phosphate (NADPH), and was incubated in water at 37 ℃. The reaction was terminated with acetonitrile at 0, 5, 10, 15, 30, 45 and 60 min, respectively. Using genistein as internal standard, the concentration of CAA in different incubation systems was determined by UPLC-MS/MS. The determination was performed on Waters BEH C18 column with mobile phase consisted of water (containing 0.1% formic acid)-acetonitrile (containing 0.1% formic acid) (45 ∶ 55, V/V) at the flow rate of 0.25 mL/min. The column temperature was 30 ℃, and the sample size was 2 μL. The electrospray ionization source was used to the select reaction monitoring mode for negative ion scanning. The ion pairs for quantitative analysis were m/z 353.14→309.11 (CAA), m/z 269.86→224.11 (internal standard) respectively. The residual percentage and enzymatic kinetic parameters of CAA in different incubation systems were calculated according to the mass concentration of CAA at 0 min. RESULTS: The linear range of CAA was 0.05-20 μg/mL; the limit of quanti- tation was 0.05 μg/mL, and the lowest detection limit was 0.01 μg/mL. RSDs of intra-day and inter-day were lower than 10%; relative errors ranged -4.83%-8.94%; extraction method and matrix effect did not affect the determination of the substance to be measured. At 60 min of incubation, residual percentages of CAA in rat, Beagle dog and human liver microsomes were(62.79±9.99)%,(64.07±11.59)%,(96.66±5.71)%, respectively. The half-life period (72.19, 68.61 min) of CAA in rat and Beagle dog liver microsomes were significantly shorter than human liver microsome (364.74 min). The clearance rates [0.019 2, 0.020 2 mL/(min·mg)] were significantly higher than human liver microsome [0.003 8 mL/(min·mg)] (P<0.05). CONCLUSIONS: Established UPLC-MS/MS method is simple, rapid, specific and sensitive, and can be used for the determination of CAA concentration in liver microsome incubation system and the study of metabolism stability in vitro. The stability of CAA metabolism in rat and Beagle dog liver microsomes are poorer than human liver microsome.
