m-Nisoldipine ( m-Nis ) , isobutyl methyl 1 , 4-dihydro - 2, 6-dimethyl-4-( 3-nitrophenyl )-3 , 5-pyridine dicarboxylate, is a new calcium antagonist. Its effects on hemodynamics and regional blood flow as well as the distribution of cardiac output were evaluated in comparison with Nis with radio-biomicrospheres in conscious rabbits. ( 1 ) m-Nis ( 1 ?g/kg ? min-1 ? 10 min, iv ) increased cardiac output and cardiac index significantly (P0.05) ; ( 2 ) m-Nis increased stroke volume and left ventricular systolic work ( per beat ) while Nis slightly increased the former and decreased the later, and these differences were statistically significant (P
2.Application of 6 sigma nursing management model in rescue of patients with severe trauma

Objective To investigate the effect of 6 sigma nursing quality management model in rescue of patients with severe trauma.Methods According to the admission sequence,268 patients with severe trauma were selected as control group and treated with traditional first aid pattern; another 292 with severe trauma were chosen as the observation group and treated with 6 sigma nursing management mode(definition,measure,analysis,improvement and control).The two groups were compared in terms of rescue success rate and effective rescue time.Result The rescue success rate in the observation group was significantly higher than that in the control group and the effective rescue time in the observation group was significantly shorter than that in control group(P<0.05). Conclusion The application of 6 sigma nursing management mode in rescue of patients with severe trauma can improve the success rate,shorten the effective rescue time,so it enhances the nursing quality of severe trauma.
3.Pharmacodynamic experiment of the antivirus effect of houttuynia cordata injection on influenza virus in mice.

It is to investigate the effect of two kinds of Houttuynia Cordata Injection on preventing and treating H1N1 influenza virus infection in mice. Pneumonia model was set up by intranasal infection of the normal and immunocompromised mice with influenza virus FM1 and PR8. The two injections were administered before and after the administration of virus, separately, and the lung index was observed. The results showed that the two preparations have obvious therapeutic effect on normal mice infected with influenza virus FM1 and PR8. And to FM1, the new injection's effect is better at small dosage. The results also showed that the two preparations have obvious prophylactic effect on immunodepressed mice infected with influenza virus FM1 and PR8. And to PR8, the old injection's effect is better at small dosage. Houttuynia Cordata Injection can improve the mice pneumonia caused by influenza virus H1N1 and decrease the lung index markedly. It has a remarkable preventive and therapeutic effect on H1N1 influenza virus in mice.
4.YAP silencing reverses doxorubicin resistance in lung cancer cell line PC9 and its mechanism

Background and purpose:Drug resistance is a major cause of failure in lung cancer chemotherapy. This study aimed to investigate the effect of YAP on doxorubicin resistance in lung cancer and its underlying mechanism. Methods:Doxorubicin resistant lung cancer cell clones were established from parental sensitive cancer PC9 cell line via in vitroinduction, and the expression of YAP was analyzed. YAP was down-regulated via shRNA to different levels. MTS assay was employed to determine cell proliferation and drug sensitivity. Flow cytometry was used to determine cell cycle distribution, apoptosis and uptake of Rh-123. Western blot and quantitative real-time polymerase chain reaction (QRT-PCR) assay were used to determine the expression of ABCB1, ABCC1, p53, Runx2, ITGB2 and ErbB4. The phosphory-lation of serine/threonine kinase (AKT) was determined as well.Results:Doxorubicin resistant PC9/Adr cell clone was obtained with over-expressed YAP. Expression of YAP in PC9/Adr cells was down-regulated to different levels via shR-NA. After YAP silencing, cell proliferation was reduced, while sensitivity to doxorubicin was increased. The cell cycle was significantly halted by G0/G1 phase. Doxorubicin induced-apoptotic rate and cellular uptake of Rh-123 were increased,with positive correlation to YAP silencing level. Western blot and QRT-PCR results showed that after YAP silencing, ABCB1, ABCC1, Runx2, ITGB2, and ErbB4 proteins were down-regulated, while the expression of p53 was up-regulated. Phosphorylation of AKT was inhibited as well.Conclusion:Over-expression of YAP is involved in doxorubicin resistance in PC9/Adr cell line. Silencing of YAP could restore doxorubicin sensitivity. The mechanism involves regulation of drug resistance-related genes and promotion of apoptosis.
5.Effect of the Gardenia extracts T9 on VP16 mRNA in herpes simplex virus type-1 infected mice's brains

Objective To investigate the effect of the Gardenia extracts T9 on herpes simplex virus (HSV) viral protein 16(VP16) in HSV-1 infected mice's brains and its potential anti-viral mechanism. Methods HSV-1 infection was induced in BALB/c mice, the expression of HSV VP16 was detected by RTPCR on the 4th, 7th, 10th, 14th, and 21st day. Results The expression of VP16 mRNA decreased in both groups of large and low doses compared with virus group at the same time. Conclusion Gardenia extracts T9 can down-regulate the expression of VP16 mRNA in HSV infected mice's brains, and it may be one of its anti-HSV mechanisms.
6.Effects of jin chai antiviral capsule on IFITM3 expression in mice.

This study is to investigate the treatment of Jin Chai antiviral capsule for influenza virus FM1/47 (H1N1) infection. The model of pneumonia was established by dropping influenza virus into the nose of normal mice, real-time PCR and Western blot technique were used to detect the virus load and the interferoninducible transmembrane protein3 (IFITM3) in lung of mice at the 1st day, 3rd day, 5th day and 7th day after affected. The results showed that Jin Chai antiviral capsule in large, middle, small dose groups can decrease virus load significantly at each time point, after being affected (P<0.05, P<0.01), Jin Chai antiviral capsule can increase the interferoninducible transmembrane protein3 in lung of mice, large dose groups are significantly higher in expression of IFITM3 compared with model group at each time point (P<0.05, P<0.01). Middle dose groups are significantly higher in expression of IFITM3 compared with model group at the 3th day and the 5th day (P<0.05), small dose groups are significantly higher in expression of IFITM3 compared with model group at the 3th day (P<0.05). It can be concluded that Jin Chai antiviral capsule exerts antiviral effects against influenzavirus by raised expression of IFITM3.
7.Establishment and applicability evaluation of animal model which was suitable to evaluate immediate hypersensitivity induced by injections of traditional Chinese medicine in BN rats.

Qingkailing injection, Shuanghuanglian injection, baicalin, chlorogenic acid as sample, guinea pig as control, to observe the specificity of allergic response to traditional Chinese medicine (TCM) injection in BN rats and establish a suitable animal model to evaluate applicability of allergic response in BN rats and guinea pigs induced by TCM. BN rats were sensitized by TCM injection, the symptoms, the rate and degree of allergic response were observed, the level of histamine in serum and tissues were determined by ELISA assay, the rate and degree of pathological changes in target organs were observed by HE staining under light microscope. There were significant symptoms of allergic response can be in BN rats, the level of histamine in serum, lung and trachea tissues increased significantly and there were significant pathological changes in lungs and tracheas. Meanwhile, the similar symptoms of allergic response can be induced by penicillin and trichosanthin. The rate and degree of allergic response, the rate and degree of pathological changes was higher in BN rats than in guinea pigs. Compared with guinea pig, BN rat is probably more suitable animal model in evaluating allergic response to injection of TCM.

8.Construction of Human Cytomegalovirus Han?ΔUS12?BAC Strain and Studies on Its Replication in Human Embryonic Lung Fibroblasts

Objective To construct human cytomegalovirus(HCMV)Han?ΔUS12?BAC strain and to study the role of US12 in HCMV replica?tion in human embryonic lung fibroblasts(HELF). Methods Kanamycin?resistant gene was amplified with primers containing homology arms se?quence flanking US12 and then electroporated into E.coli DY380?Han?wt?BAC competent cells. Successfully recombinant Han?ΔUS12?BAC clones were identified by PCR,sequenced for confirmation Han?ΔUS12?BAC plasmids were electroporated into HELF to produce infectious viri?ons. Han?ΔUS12?BAC and Han?wt?BAC were inoculated onto HELF at the multiplicity of infection of 0.1 pfu/cell. The viral titer in the supernatant was measured by TCID50 assay and growth kinetics of the viruses in HELF was studied. Results Han?ΔUS12?BAC clone was successfully con?structed. Han?ΔUS12?BAC was reconstructed in HELF to generate infectious virions. Growth kinetics assay indicated that Han?ΔUS12?BAC and Han?wt?BAC showed no differences in their growth and dissemination in HELF. Conclusion US12 in HCMV clinical strain Han is dispensable for HCMV growth and dissemination in HELF.
9.The risk factors of normal fasting blood glucose population with high postprandial glucose

Objective:To analyze the blood glucose screening condition for individuals receiving physical examinations. Specifically, to examine the rates of abnormal postprandial glucose at different levels of fasting blood glucose and the characteristics of people who had normal fasting blood glucose with high postprandial glucose.Methods:Participants were individuals who received physical examinations at the Health Management Centre, Tianjin Medical University General Hospital, from January 2016 to December 2016. Data collection included general information, physical examination, and laboratory tests including fasting blood glucose, HbA1c, postprandial glucose, blood fat, and blood uric acid. The blood glucose indicators for the study population were analyzed. Among participants without diabetes, the rate of abnormal postprandial glucose at different fasting blood glucose levels or with a HbA1c screening were compared. For participants with complete information, chi-square tests and multivariate logistic regressions were used to analyze the characteristics of individuals who had normal fasting blood glucose with high postprandial glucose.Results:A total of 45 447 participants were included, with 23 001 males (50.61%) and 22 446 females (49.39%). For blood glucose, the most frequently completed indicator was fasting blood glucose (97.04%), then HbA1c (56.17%) followed by postprandial glucose (17.51%). There were 7 351 participants without diabetes who had all three glucose indicators. As fasting blood glucose increased, the abnormal rate of postprandial glucose increased gradually. When fasting blood glucose was lower than 5.6 mmol/L, the rate of abnormal postprandial glucose was 11.59%. When fasting blood glucose was between 5.6 mmol/L and 6.1 mmol/L, the rate was 33.9%. When fasting blood glucose was higher than 6.1 mmol/L, the rate was 68.73%. When the cutoff for fasting blood glucose was 5.6 mmol/L, the rate of abnormal postprandial glucose decreased compared with a cutoff of 6.1 mmol/L (11.59% vs. 14.32%, respectively). Combined with HbA1c screening, the rate of postprandial glucose abnormalities were both reduced (8.83% vs.11.59% for 5.6 mmol/L; 10.08% vs. 14.32% for 6.1 mmol/L). A total of 5 872 individuals had complete information and were included in the analysis. Participants who were men, >45 years old, and were overweight or obese had higher risk for abnormal postprandial glucose [ OR(95% CI): 2.85(2.33-3.48), 2.15(1.76-2.62), 1.82(1.45-2.27), and 2.64(2.04-3.42), respectively, P<0.05]. Hypertension, hyperlipidemia and hyperuricemia were also risk factors [ OR(95% CI): 1.80(1.51-2.15) and 1.52(1.27-1.82), respectively, P<0.05] Conclusions:The completion rate was highest for fasting blood glucose and lowest for postprandial glucose among the study population. The rate of abnormal postprandial glucose was high when fasting blood glucose was normal. Screening combined with HbA1c decreased the rate of postprandial glucose abnormalities. Postprandial glucose testing should be recommended for those who are male, older, overweight or obese, and have other risk factors including hypertension, hyperlipidemia, and hyperuricemia.
10.Effect of fecal microbiota transplantation on intestinal flora in mice with acute-on-chronic liver failure

ObjectiveTo investigate the protective effect of fecal microbiota transplantation (FMT) on mice with acute-on-chronic liver failure (ACLF) and its effect on intestinal flora. MethodsA total of 40 mice were randomly divided into control group (CON group), model group (MOD group), FMT group (feces of the mice in the CON group were used as fecal microbiota donor), and FMT model group (ANFMT group, with feces of the mice in the MOD group as fecal microbiota donor), with 10 mice in each group. All mice were observed in terms of body weight, death, liver histopathology, and changes in aspartate aminotransferase (AST), alanine aminotransferase (ALT), and intestinal flora. A one-way analysis of variance was used for comparison of normally distributed continuous data between multiple groups, and the SNK-q test was used for further comparison between two groups. ResultsCompared with the CON group, the MOD group had a significant reduction in body weight and significant increases in AST and ALT (all P<0.05), as well as large patchy necrosis of hepatocytes, significant increases in Verrucomicrobia, Akkermansia, and Erysipelatoclostridium, and significant reductions in Dubosiella and Duncaniella (all P<0.05). Compared with the CON group, the ANFMT group had a significant increase in AST (P<0.05), hepatocyte swelling and mild ballooning degeneration, significant increases in Unclassified and Faecalibaculum, and significant reductions in Patescibacteria, Deferribacteres, Muribaculum, Candidatus_Saccharimonas, Rikenella, Odoribacter, Mucispirillum, and Lachnospiraceae_unclassified (all P<0.05). Compared with the MOD group, the FMT group had significant reductions in AST and ALT (both P<0.05), mild hepatocellular necrosis and marked ballooning degeneration, significant increases in Paramuribaculum and Bilophila, and significant reductions in Firmicutes, Rikenella, and Absiella (all P<0.05). ConclusionIntestinal flora disturbance is observed in ACLF mice, and dysbacteriosis may lead to liver injury. FMT can alleviate liver inflammation in ACLF mice and thus exert a protective effect.
