The 8p11 myeloproliferative syndrome is a rare hematologic malignancy that involves the fibroblast growth factor receptor 1 (FGFR1) gene at chromosome 8p11.The clonal change of the FGFR1 rearrangement is present in BM cells,and in the T-lymphoma cells in the LN.The hematological stem cells transplantation is the only method that can cure this rare hematologic neoplasma.
2.A dynamic observation on the levels of TNF-?、IL-6 and IFN-? produced by human dendritic cells infected with Dengue virus

AIM: To study the dynamic levels of tumor necrosis factor alpha (TNF-?), interleukin-6 (IL-6) and gamma interferon (IFN-?) produced by human dendritic cells infected with Dengue virus. METHODS: Monocytes isolated from healthy human peripheral blood were incubated in medium with GM-CSF and IL-4 for more than 7 days. DCs were then collected and identified by scanning electron microscope, immunohistochemistry and lymphocytes stimulatory ability. Human dendritic cells (DC) were infected with Dengue-2 virus (DV-2) in vitro, culture supernatants were collected in different time postinfection (6 h, 12 h, 24 h, 48 h and 72 h), DV antigen in human dendritic cells were demonstrated by an indirect immunofluorescent assay (IFA), production of TNF-?, IL-6 and IFN-? in the culture supernatants were evaluated by ELISA. RESULTS: After 7 days, typical dendritic cells could be obtained. Virus antigen were detected in infected DC by IFA. Dengue virus induces TNF-? and IL-6 secretion from DC and does not induce IFN secretion. CONCLUSION: Human dendritic cells are target of dengue virus infection. TNF-?, IL-6 production from DC are increased with DV infection. Dendritic cells may play an important role in DV pathogenicity and immunity.
3.Effect of Jiawei Glutinous decoction on gouty nephropathy

ObjectiveTo investigate the effect of Jiawei Glutinous decoction on gouty nephropathy. Methods54 patients with gouty nephropathy were randomized into two groups, 31 patients in the treated group were treated with Jiawei Glutinous decoction, and 23 patients in the control group were treated with allopurinol. Blood uricacid (BUA), renal function, β2-microglobulin (β2-MG) and 24 h urine protein of patiens in two groups were measured before and after treatment.ResultsImprovements of symptoms and renal function as well as decreasing of BUA, β2-MG and 24h PRO in the treated group was superior to those in the control(P<0.05).ConclusionJiawei Glutinous decoction present good effect in treating gouty nephropathy.
4.Establishment of experimental animal model of pristane induced rheumatoid arthritis in rats

Objective The animal model of rheumatoid arthritis was established in Lewis rats by a single intradermal injection of pristane. Methods The degree of pathogenicity and the pathological characteristics were assayed by histiopathological techniques and joint roentgenography. Results The incidence of arthritis in rats immunized by pristane was 62.5%. Peripheral joints were mainly involved. A chronic relapsing and progressive arthritis was developed.As showed by histopathology and roentgenography, typical arthritis pathology such as synovial proliferation, articular cartilage and bone erosien were found. Conclusion Pristane induced arthritis mirrors human rheumatoid arthritis and is a very good animal model for rheumatoid arthritis.
5.Micrometastasis of axillary lymph nodes and the nm23 expression in breast cancer

Objective To investigate micrometastasis in axillary lymph nodes and its relationship with nm23 expression of the tumor tissue in breast cancer patients. Methods Paraffin blocks of 832 surgically removed lymph nodes and carcinoma tissues from 52 lymph node negative cases comfirmed by routine pathology were re examined by immunohistochemical staining using monoclonal anti EMA and anti nm23 H1 respectively. Results Micrometastasis were observed in 12 of the 52 patients (23.0%) and in 34 of the 832 lymph nodes ( 4.1% ). The positive rate of micrometastasis that in the tumor larger than 2.0cm or poorly differentiated was significantly higher than in the tumor

respectively)was significantly higher than that for digital anterograde venography (0.684 and 0.587,P =0.026,respectively).Conclusion Diagnostic accuracy and detection rate of residuaI or recurrent varicose veins with CT phlebography are found to be more superior than that with digital anterograde venography which has high guidance value in preoperation and assesses the efficacy of post-surgery treatment.
7.An estimation of the 50% effective anesthetic volume of 0.5% ropivacaine in nerve stimulator-guided vertical infraclavicular brachial plexus block

Objective To research the 50% effective anesthetic volume ( EAV50) of 0.5% ropivacaine in nerve stimulator-guided vertical infraclavicular brachial plexus block. Methods Thirty patients scheduled for forearm or hand surgery were blocked using 0. 5% ropivacaine in nerve stimulator-guided vertical infraclavicular brachial plexus block. The EAV50, which is the anesthetic volume corresponding to 50% success and 50% failure was determined by up-and-down sequential test. The starting dose of 0. 5% ropivacaine was 0. 55ml/kg. Block failure resulted in a dose increase 110% , and block success in a reduction 110%. The sensory and motor blockade were accessed at 5- min intervals (up to 60 min). Results In nerve stimulator-guided vertical infraclavicular brachial plexus block, EAV50 for 0.5% ropivacaine was 0.417ml/kg. In up-and-down sequential test, there were significantly different in the block success rates of lateral, medial and posterior cord ( P <0.01). Conclusion In nerve stimulator-guided vertical infraclavicular brachial plexus block, enough anesthetic volume must be given to increase block success rates of all the cords of brachial plexus, and EAV50 for 0. 5% ropivacaine was 0.417ml/kg.
8.Therapeutic efficacy and safety of sublingual immunotherapy in treatment of adults with seasonal allergic rhini-tis:a meta-analysis

Objective To systematically investigate the efficacy and safety of sublingual immunotherapy( SLIT) in the treatment of a-dults with seasonal allergic rhinitis( SAR) .Methods Literature was retrieved online in CNKI,CBM,VIP,Wanfang data,PubMed,EMBase, Medline,Cochrane Library,et al.After screening literature,data collection and assessment quality,randomized controlled trials relevant to our study were selected independently based on inclusion and exclusion criteria by two researchers.Statistical analyses were conducted by RevMan 5.2 software.Results A total of 13 RCTs were enrolled.The meta-analysis revealed that SLIT provided a significant improvement in aspect of reducing Symptom Scores(SS) and Medication Scores (MS)of adults with SAR,the difference had statistical significance(P<0.000 01). Compared to placebo,SLIT could increase the rate of adverse events( AEs) in the treatment of adults with SAR compared to placebo.The fre-quently reported AEs during clinical trials were oral pruritus,tongue edema,ear pruritus,throat irritation,et al.Conclusion Sublingual im-munotherapy could improve the symptoms significantly and reduce the SS and MS in the treatment of adults with SAR.And its safety could be well tolerated.
9.Effect of microRNA-101 on atrial fibrosis in human chronic atrial fibrillation

Objective To investigate the effect of microRNA-101 (miRNA-101) on atrial fibrosis in human chronic atrial fibrillation (AF). Methods Right atrial appendages were obtained from 59 patients (30 with AF) undergoing cardiac surgery, including 47 patients with valve heart disease and 12 patients with congenital heart disease. The expression of miRNA-101 was determined by quantitative real-time PCR in the right atrial appendages of patients with and without AF. The cell-specific localization of miRNA-101 was detected by in situ hybridization assay. The mRNA and protein expression levels of transforming growth factor β typeⅠreceptor (TGFβRⅠ) and collagen type I (COL1) were determined by quantitative real-time PCR and Western-blot assay, respectively. Collagen in the right atrial appendages was observed by Masson staining assay. Results The expression of miRNA-101 was found to be significantly down-regulated in AF patients compared with patients with sinus rhythm (SR) (P < 0.05). The result of miRNA-ISH showed that miRNA-101, which was highly distributed within the connective tissues of heart, was down-regulated at about 24.9% in patients with AF compared with patients with SR. No significant differences at the mRNA expression level of TGFβRI was found between patients with AF and patients with SR (P > 0.05). But the protein expression of TGFβRI in patients with AF was significantly higher than that of patients with SR (P < 0.05). The mRNA and protein expressionsl of COL1 were significantly higher in patients with AF than thoset of patients with SR (P < 0.05). The collagen was significantly increased in patients with AF than that of patients with SR (P < 0.05). Conclusions Downregulation of miRNA-101 may contribute to atrial fibrosis in human atrial fibrillation by targeting TGFβRⅠ.
10.Anti-tumor effect of 153Sm-DTPA-c (CGRRAGGSC) on nude mice model bearing MHCC97-H human liver cancer

Objective To observe the antitumor effect of 153Sm-DTPA-c (CGRRAGGSC) on MHCC97-H human liver cancer using a nude mice model.Methods 153Sm-DTPA-c(CGRRAGGSC) was indirectly synthesized by the reaction of 153SmC13 with DTPA-c(CGRRAGGSC).Western blot was used to detect the expression changes of IL-11 receptor of eight different cell lines.20 MHCC97-H tumor-bearing nude mice were randomly divided into four groups by random number table method with five mice in each group,administered with different activities of 153 Sm-DTPA-c (CGRRAGGSC) by tail vein injection (5.5,11.0,22.0 MBq/0.2 ml and physiological saline/0.2 ml in turn).All nude mice were terminated after 16 d then the tumor tissue was observed and analyzed with microscopy,electron microscopy and immunohistochemistry for Ki-67,Bc1-2 and IL-11 receptor.Western blot was used to evaluate the expression changes on IL-11 receptor of tumor tissue at different doses.Results After administration with 153 Sm-DTPA-c(CGRRAGGSC),the central region in the early stage of tumor became dry,necrosis,and crusting.In the late stage,tumor continually grew surrounding necrosis tissue.The inhibition rates in the low,middle and higher dose group were (22.72 ±2.76)%,(34.65 ±2.36)% and (85.13 ±5.78)%,respectively(F =89.32,P < 0.05).The morphologic results showed that tumor structures were severely damaged along with radiation dose.The results of immunohistochemistry showed that the IL-11 receptor-positive cells in the control group and 5.5 MBq group was (84.13 ± 5.71) % and (61.57 ± 5.98) % (t =13.62,P <0.05),respectively.The Ki-67 and Bcl-2 positive cells were decreased in the injection group (t =20.91,6.68,P <0.05).Western blot analysis showed that the expression of IL-11 receptor protein decreased with antitumor-effect of radiation.Conclusions 153Sm-DTPA-c (CGRRAGGSC) effectively depresses the growth of MHCC97-H human liver cancer and suppresses the expression of IL-11 receptor,and hence could be a promoting agent for the treatment of MHCC97-H human liver cancer.
