Objective:To establish the sensitive, specific, stable and convenient sgp130 ELISA kit. Methods:The mAb T2 against human gp130 was used as coating antibody;the other mAb T12 recognized different epitope with T2 was labeled by biotin, then a ELISA kit for detecting sgp130 was set up.Results:sgp130 ELISA kit is successfully established and its sensitivity is 10 ng/ml.After the kit is placed in 4℃ for 3 months, the kit's CV is less than ?7.6% and the retrievable rate is 95%~111%.These indicate that it has highly sensitivity, stability and accuracy. The normal serum concentration of sgp130 in healthy donors is 536.92~287.88(ng/ml),but there is higher in patients with hyperthyroidism(937.16?217.5) and chronic nephritis(806.45?138.47).Significant difference is found comparing with normal control(P

Objective To investigate the effect of implanting uncovered self-expandable metal stent for treatment of distal malignant biliary obstruction through endoscope. Methods The effect of therapy about implanting uncovered self-expandable metal stents to 16 patients who had unsectable malignant tumors companing with obstructive jaundice through endoscope was reviewed. Results Fifteen of the studied patients were implanted uncovered self-expandable metal stents successfully (94%), for their internal drainage were patent. At the seventh and fourteenth day after implantation, liver function and B-ultrasound were rechecked. Compared to the data before operation, total bilirubin, direct bilirubin and transaminase declined respectively (P

Objective To report 6 cases of congenital heart disease with high origin of coronary artery and to evaluate the imaging method for diagnosis of congenital high origin of coronary artery. Methods Six patients with congenital high origin of coronary artery underwent angiocardiography ,echocardiography ,and 2 patients also underwent magnetic resonance examination. All 6 cases were confirmed by operation. Results All 6 cases were congenital high origin of right coronary artery. Angiocardiography made correct diagnosis in all 6 cases; MRI made the correct diagnosis in 1 of the 2 cases; echocardiography made 1 correct diagnosis. Conclusion Correct diagnosis of congenital high origin of coronary artery was very important for patients with congenital heart disease. Angiocardiography was a very reliable imaging method and MRI can play an important role in preoperative diagnosis of congenital high origin of coronary artery.

Chinese Journal of Radiology 2001;0(07):-
Objective To understand the incidence of renal involvement of children with non Hodgkin′s lymphoma and to recognize its different CT findings. Methods The thoracic and abdominal plain and contrast enhanced CT of 30 cases of NHL in children were reviewed and all cases were confirmed by pathology. The changes in both pre and post chemotherapy were analysed in the 10 selected cases with renal involvement. Results CT demonstrated 6 cases of multiple masses and 1 case of multiple patchy lesions in bilateral kidneys. Two cases of single mass and 1 case of multiple masses were detected in single kidney. Conclusion The incidence of renal involvement of children with non Hodgkin′s lymphoma is relatively high. CT can clearly demonstrate the renal involvement of NHL, which is helpful for clinical stage, especially in the evaluation of the therapeutic effects. Hence, abdominal plain and contrast enhanced CT scan should be done in children with NHL.

Objective To evaluate the clinical diagnostic value and limitation of MRI and contrast enhancement MRA (CE-MRA) in congenital heart disease of children. Methods Three hundred patients with congenital heart disease underwent CE-MRA. 173 were confirmed by operation. The results of MRA and CE-MRA were compared with operation data. Results 196 (86.0%) intracardiac malformation including septal defect and valve anomaly were diagnosed correctly by MRI within 228 abnormalities which were confirmed by operation. 101 (96.2%) extracardiac malformation including great vessels stenosis and anomalous connection were diagnosed correctly by MRI and MRA within 105 abnormalities which were confirmed by operation. Conclusion MRI and CE-MRA are very accurate diagnostic method for extracardiac malformation. CE-MRA is the best sequence for congenital heart disease.

This study aimed to investigate the method of conserving the mice skin for long time in vitro, and forty clean male KM mice of 8-week old were divided into two groups by random digit table, namely vitrified skin transplantation group(n =30) and fresh skin transplantation group(n =10) .The skin was vitrified quickly with freezing solution EFS40 by the two-step method, and then was thawed with 0.5 mol/L glucose, which was followed by self-transplanted.The vitrified skin transplantation group was self-transplanted after the skin was vitrified for 2 weeks.A piece of skin which was smaller than the vitrified one was cut off on the other side of the spine, and the thawed skin were laid smoothly to coincide with the edge of the cut skin which was sutured then.In fresh skin transplantation group, fresh skins were prepared to put in Dulbecco phosphate-buffered saline for 15 minutes and then be transplanted to original position. The outcomes of transplantation were valuated on day 14 and day 28 after transplantation. And the results indicated that the survival rate of the fresh skin transplantation group was 100%(10/10) .The survival rate of the vitrified skin transplantation group was 77%(23/30) on day 14 and 70%(21/30) on day 28,respectively.But the surfaces of the mice skin and part of the hair follicle cells were damaged during vitrification. However, the method of vitrifying the mice skin quickly in this paper may be used to conserve the skin in vitro.

Objective To detect systematic oxidative stress in preeclampsia. Methods (1)Morphological features of placenta hypoxia were observed by histological method ; (2) Level of granulocyte intracellular reactive oxygen species was monitored by dyeing full blood with 2' ,7'-dichlorodihydrofluorescein diacetate (H2DCFDA) ; (3) Level of H_2O_2 in sera was detected by special kits. Results Compared to normal pregnancy, placentas from preeclampsia showed distinct features of hypoxic stress injury, such as more syncytial knots formation, fibrosis emerged, vein in-jury and loss its normal configuration; Fluorescence values of ROS probe in neutrophils from different women were 45.61±12.20(n =49),51.02 ± 13.60(n =56,P <0.01)and 85.10 ± 16.30(n =47,P <0.01); Concentra-tions of H_2O_2were (24.57±5.17)μmol/L(n =49),(26.61±3.25)μmol/L(n =56,P 0.01) and (39.84±9.67)μmol/L(n=47,P<0.01) respectively. Conclusion With the help of histological method, flow cytometry and special kits, systematic oxidative stress can be detected through checking placentic tissues, netrophils and sera of preeclampsia.

Objective To Explore the therapy of elderly patients with hypoproliferative acute myeloid leukemia. Methods Twenty-six elderly patients with hypoproliferative acute myeloid leukemia had received induction of CAG regimen and 15 cases were treated intensive chemotherapy after complete remission, then if achieving complete remission had received subsequent intensive chemotherapy: stand or reduce AA, DA or HA regimen. Results In 26 cases patients who received CAG regimen, the complete remission rate, partial remis sionrat and the totle total effective rate were 53.85% (14/26), 19.23% (5/26) and 73.08% (19/26), respectively.Accordance with response to CAG regimen and the therapy after complete remission, 26 patients were divided into 3 groups:intensive chemotherapy after complete remission(CR1), non-intensive chemotherapy after complete remission(CR2) and non-remission(NR, including partial remission patients). The median overall survival of CR1 ,CR2 and NR were 13.5 months, 8.2 months, and 4.5 months, respectively. There was higher median overall survival of CR1 group than CR2 and NR groups (P =0.041,0.001, respectively). There was higher OS of CR2 group than NR group (P =0.064).No serious bleeding events were found in chemotherapy. Conclusion CAG regimen was an effective and little adverse effect regimen for the treatment of elderly H-AML,and with well tolerance in patients.Advisable intensive chemotherapy can prolong the patient's survival time after complete remission.
