Objective To study whether eicosapentaenoate can decrease the apoptosis effects in-duced by palmitic acid in INS-1 or not. Methods Based on different condition, there were four groups in this study, including control group , EPA group, palmitic acid group, combination of EPA and palmitic acid group. The grow curve was detected by MTT, and the expressions of bax were detected by Western blot.Cell apoptosis was detected by caspase-3. ROS was detected by CM2H2DCFDA kit after 48h. Result The grow curve of combination of EPA and palmitic acid group was higher than that in plamitic group[24 h :(37.33±1.15)OD vs (30. 79 ± 1.55 )OD, P < 0. 01 ;48 h:(31.50 ± 1.56)OD vs (23.94 ± 1.10)OD, P<0. 01] . Caspase-3 activity and ROS and the expression of bax and SREBP1c in combination of palmitic acid and T0901317 group were lower than those in palmitic acid group[(3566. 67 ± 305.51 )OD vs (4233. 33 ±416. 33)OD]. Conclusion EPA can inhibit apoptosis in INS-1 induced by palmitic acid.

Chinese Journal of Hospital Administration 1996;0(04):-
The germs on the hands of medical personnel in medical institutions have become the chief pathogens for hospital infection. These pathogens can be acquired not only from infected wounds or drainage wounds but also from normal and whole skin regions. Hand washing and disinfection is one of the most important measures for preventing hospital infection. The paper describes the definitions of hand washing, hygienic hand disinfection and surgical hand disinfection and discusses the action mechanisms, effects and features of various hand hygiene products currently in use, such as alcohol, hibitane glucose saline and hexachlorophenol. It enumerates a number of problems inhand hygiene, including low rate of hand hygiene observance on the part of medical personel, insufficient attention to hand hygiene on the part of medical institutions, reduction of germs' sensitivity to disinfectants, and the occurrence of contact dermatitis in disinfectant usage. The paper also gives an account of some of the latest strategies and countermeasures aiming at improving the status of hand hygiene, These include ways of lowering the incidence of contact dermatitis, reducing the bad effects of germicides, and raising the rate of hand hygiene observance.

Objective To explore the diagnostic points and promote the checkout rate of fetal congenital cor-dis malformation. Method Analyzed the ultrasonic character by reviewing the fetal congenital cordis malformation from our hospital in recent 2 years. Result 3 cases was inaccuracy in the 28 cases of congenital cordis malformation diagnosed by antepartum. Diagnostic accuracy rate was 89.3%. Conclusion The inaccuracy was reduced greatly by gradual ultrasonic diagnosis by way of four cavity heart and three vas cross section,

Objective To explore the role of p16 gene methylation in fibroblasts in the occurrence and development of keloid. Methods Skin tissue specimens were resected from the lesions of patients with keloid and normal skin of healthy human controls.Fibroblasts were isolated from these tissue specimens and subjected a primary culture. An immunohistochemical analysis was performed to measure the expression of p16 protein in tissue specimens,real-time fluorescence-based quantitative PCR to determine the mRNA expression level (expressed as 2-△△Ct) of p 16 and DNA methyltransferases (DNMTs) in fibmblasts, and bisulfite sequencing PCR (BSP) to estimate the methylation status of p16 gene in the tissue specimens and primary fibroblasts. Results The keloid fibroblasts (KFbs) showed significandy lower mRNA expression of p16 gene (0.64 ± 0.18 vs.1.92 ± 0.23,t =10.54,P< 0.05),but significantly higher mRNA expressions of 3 DNMTs (DNMT1:2.58 ± 0.23 vs.1.13 ± 0.21,t =11.22,P < 0.05; DNMT3A:4.87 ± 0.46 vs.2.38 ± 0.32,t =10.81,P< 0.05; DNMT3B:1.57 ± 0.12 vs.0.57 ± 0.16,t =12.45,P< 0.05) compared with the normal fibmblasts (NFbs). The DNA methylation rate in the p16 gene promoter region was significantly increased in keloid tissue (1.81% ± 0.46%) and KFbs (3.15% ± 0.94%) compared with normal skin tissue (0.90% ± 0.35%,F =14.23,P< 0.01) and NFbs (0.17% ± 0.29%,F=37.62,P< 0.01).Conclusions The methylation and low expression of p16 gene in KFbs may be associated with the uncontrolled growth of keloid, and DNMTs may play a role in the pathogenesis of keloid.

Microthrombosis may be involved in the pathogenesis of cardiac microangiopathy due to diabetes. Recent studies have shown that fibrinogen-like protein 2 (fgl2) plays a pivotal role in microthrombosis in viral hepatitis, acute vascular xenograft rejection and cytokine-induced fetal loss syndrome. The current study was designed to examine the expression of fgl2 in microvascular endothelial cells and investigate the effects of microthrombi due to fgl2 on cardiac function and structure in rats with type 2 diabetes. Following induction of type 2 diabetes, 24 rats were observed dynamically. Fgl2 expression and related cardiac microthrombosis were examined. Local or circulating TNF-α was measured. Coronary flow (CF) per min was calculated as an index of cardiac microcirculation. Cardiac function and morphology were evaluated. It was found that Fgl2 was highly expressed in cardiac microvascular endothelial cells of rats with type 2 diabetes, which was promoted by local or circulating TNF-α. The Fgl2 expression was associated with cardiac hyaline microthrombosis. In parallel with the fgl2 expression, CF per min, cardiac diastolic or systolic function and cardiac morphology were aggravated to some extent. It was concluded that in rats with type 2 diabetes, microthrombosis due to fgl2 contributes to the impairment of cardiac diastolic or systolic function and morphological changes.

Autoimmune is involved in the pathogenesis of ventricular remodeling in acute myocardial infarction (AMI). In the present study, we investigated the effect of anti-cardiac myosin heavy chain antibodies (AMHCA) from patients with AMI on rat cardiomyocyte apoptosis. Cardiomyocyte apoptosis was observed and measured by DNA end labeling and Annexin-V/PI double-staining assay. The expression of apoptosis related p53 and Bcl-2 protein and the second messenger calcium were detected respectively by Western blotting, patch clamp and confocal calcium imaging. The results showed that AMHCA was able to induce cardiomyocyte apoptosis in a dose dependent manner. Apoptosis-accelerating nucleoprotein p53 was up-regulated, while apoptosis-inhibiting cytoplasmic protein Bcl-2 was down-regulated. In parallel, cytoplasmic calcium concentration was elevated. There was no effect on L-type calcium currents. It is concluded that AMHCA in patients with AMI as a novel triggering factor can induce cardiomyocyte apoptosis, which contributes to ventricular remodeling.
