1.The effects of bFGF on the calicum influx in OHCs

AIM To examined the effect of bFGF on the calcium influx in outer hair cells (OHCs) and its antagonistical effects with streptomycin. The aim of the study is to explore the mechanism of acute ototoxicity of streptomycin and antagonism of bFGF. METHODSE The OHCs of guinea pig were isolated using an enzyme machine methods and loaded with 10 ?mol?L -1 Fluo 3/AM for 30 min at 37℃,then cultured 60 min at room temperature. Individual Fluo 3 loaded OHCs were examined with a confocal microscope (ACAS Ultima, USA) using a 20? objective lens and linear scan mean. The fluorescent images, collected every 5 sec for 300 s, were stored in a computer. The fluorescent intensity of the images were analyzed by the software cooperated with the confocal microscope, and a function curve showing the change trend of fluorescent intensity with time was obtained. RESULTS The OHCs [Ca 2+ ] i hold steady within the process of normal extracellular liquid perfusion. OHCs [Ca 2+ ] i increased when perfused 100 mmol?L -1 high potassium media(10/11) and normal media containing 1 nmol?L -1 bFGF(9/9), but the OHCs [Ca 2+ ] i don't changes in high potassium free calcium media(7/7) and free calcium media containing 1 nmol?L -1 bFGF(8/8). After treated by 1 mmol?L -1 streptomycin, the OHCs [Ca 2+ ] i increased 0/12, 4/8 respectively when perfused high potassium media and bFGF media. And after treated by 1 nmol?L -1 bFGF, high potassium media make OHCs [Ca 2+ ] i increased more obviously and keeping longer time. After treated by 1 mmol?L -1 streptomycin, respectively, 0 1,1 and 10 nmol?L -1 bFGF make 4/11,6/9 and 12/12 OHCs [Ca 2+ ] i increasing when perfused high potassium media. CONCLUSION High potassium media and bFGF perfusion can result in increasing of OHCs [Ca 2+ ] i ,and OHCs [Ca 2+ ] i increase rooted in the calicum influx, there is synergic effects between high potassium and bFGF. The streptomycin can block the process of calcium influx induced by high potassium media and the block effects can be antagonized by bFGF, and the antagonistical effects have bFGF concentration dependence.
2.Color Doppler Flow Imaging of Prostate

The hemodynamic examination of prostate was performed transperineally by HP SONOS 1000 or 2500 Ultrasound System with 7.5 MHz transducer.The results of 25 healthy persons showed that:1.In transverse section,two arteries locatced at 5 and 7 o'clock corresponded to the capsule branches of prostate artery with diameter(D)1.7±0.25mm,Vnlax 22.2±8.2cm/s,flow volume(V) 8.O±6.5ml/min;2.The sagittal view showed the urethra branch of prostate artery,red in colonr,D 1.8±0.2mm,Vmax 25.6±10.2cm/s,V 9.5±3.69mI/min.3.Blue streams with lOW velocity were seen near the internal orifice of urethra,were vesicoprostatic venous plexus with D 3.5±1.0mm,their velocity varied with respiration.In presence of prostatic hypertrophy,the velocity and flow volume were less than those in normal case.

SGCs[Ca 2+ ] i was increased when SGCs were perfused with 150mmol/L high potassium media(10/10) and normal media containing 1nmol/L bFGF(8/9), but the level of SGCs[Ca 2+ ] i did not change in high potassium free calcium media(9/9) and free calcium media containing 1nmol/L bFGF(10/11). After the treatment with 1 mmol/L streptomycin, the level of SGCs[Ca 2+ ] i was increased 1/11 and 5/12, respectively, when perfused high potassium media and bFGF media. And after being treated with 1nmol/L bFGF, high potassium media, SGCs[Ca 2+ ] i was increased more obviously and persisted for a longer time. After being treated with 1 mmol/L streptomycin, 0 1nmol/L, 1nmol/L and 10nmol/L bFGF, respectively, and 14/14 the level of SGCs[Ca 2+ ] i was increased 5/11, 9/12, and 14/14 when perfused high potassium media. High potassium media and bFGF perfusion could result in an increase of SGCs[Ca 2+ ] i ,and SGCs[Ca 2+ ] i increase was the result of calcium influx, and there was a synergic effect between high potassium and bFGF. Streptomycin could block the process of calcium influx induced by high potassium media, the blocking effect could be antagonized by bFGF, and the antagonistic effect was bFGF concentration dependent.
4.A study on correlation between contrast-enhanced sonography and microvessel density in peripheral lung tumor

Objective To observe the correlation between contrast-enhanced sonography and microveesel density (MVD) in peripheral lung tumor.Methods Contrast-enhanced sonography was performed in 55 patients with peripheral lung tumor.Among of them,malignant tumor was 40 cases,benign tumor was 15 cases.Analyzed the quantitative parameters (peak intensity,enhanced strength index) by using time-intensity curve (TIC) with computer.MVD was measured by immunohistochemistry after operation.Results The level of MVD in malignant tumor was significantly higher than that in benign tumor [(24.57 ± 5.93)/HP vs.(15.50 ± 2.82)/HP] (P < 0.05).The level of MVD had significant correlation with peak intensity and enhanced strength index (P < 0.01).Conclusions The peak intensity and enhanced strength index have significant correlation with MVD.They are valuable indexes in evaluating angiogenesis in peripheral lung tumor.
5.Evaluation of TDP in Treatment of pneumonia in Children

110 patients under three years of age with pneumonia were admitted to this hospital in a period from August of 1982 to March of 1983. There were 68 males and 42 females. The patients were randomly divided into Group A and Group B .The symptoms and signs, the laboratory data, and the severity and duration of the illness of the patients of both groups were similar.After admission, laboratory examinations including WBC counts, throat swab cultures and determinations of immunity function were performed and chest x-ray films were taken for all the patients. Regular treatments were given to all of them but the patients of Group A received TDP radiation in addition.After comparing the clinical courses of the two groups, the authors found that TDP could cause more rapid disappearance of cough and moist rales in the lungs, shorten the time of recovery and the whole course of hospitalization, and hasten the absorption of pulmonary infiltrations as seen from the x-ray films. Furthermore, TDP is helpful to promote the immunity function by raising many immune indices. And the rate of lymphocyte transformation was also increased. Its influence on the bringing down of the fever to normal is not very remarkable.It is concluded that TDP radiation is a simple, safe and effective treatment for pneumonia and it is suitable to be used in children.
6.On_line Preconcentration for Sensitive Analysis of PAHs in Cosmetics by Reversed Microemulsion Electrokinetic Chromatography

A sensitive analytical method based on reversed microemulsion electrokinetic chromatography ( MEEKC) combined with on_line preconcentration technique was developed for the determination of polycyclic aromatic hydrocarbons ( PAHs ) in cosmetics. For six lipophilic PAHs analytes which are difficult to be separated under conventional conditions, three stacking techniques including large volume sample stacking ( LVSS) , dynamic pH junction and sweeping ( LVSS_DypH_sweep ) were combined to realize the efficient preconcentration and separation. Under the optimum conditions, including the microemulsion buffer with the composition of 2. 4%(w/w)SDS_0. 6% (w/w) octane_6. 6% (w/w)n_butyl alcohol_20 mmol/L NaH2PO4 ( pH 2 . 2 ) , HCB injection time of 20 s ( 16 kPa ) and sample injection time of 80 s ( 16 kPa ) , good enrichment effect was reached with the enrichment factors ranged from 25 to 80 , and the PAHs were analyzed successfully within 27 min. The developed method was used to analyze the PAHs in cosmetics. The recoveries ranged from 90 . 6% to 95 . 9%. The RSD values ( n=5 ) were less than 5 . 1%.
7.Correlation Factors of Lower Respiratory Tract Infection after General Anesthesia with Laryngeal Mask Airway:A Survey

OBJECTIVE To analyze the correlation factors and preventive measures of lower respiratory tract infection after general anesthesia with laryngeal mask airway.METHODS In surgery 158 cases of general anesthesia with laryngeal mask ventilation in gynecology and orthopedic wards were analyzed retrospectively.RESULTS Forty-three cases were found in hospitals with lower respiratory tract infection,accounting for 83% of the total lower respiratory tract infection in the wards at the same period.CONCLUSIONS The anesthesia way,the age,the operation time,whether gastric tube,the abuse of antibiotics and whether the anesthesia equipments strictly disinfected and managed may be the important factors which can cause the lower respiratory tract infection.

Objective To evaluate the clinical and imaging features of the sellar region lesions in children.Methods The clinical and imaging features of 112 cases with sellar region lesions were analyzed retrospectively,which were confirmed by pathology.Results 37 cases were craniopharyngiomas, which had two main symptoms of intracranial hypertension(57%)and diabetes insipidus(11%).On imaging it demonstrated as a calcified cystic tumor(81%).18 cases were gliomas, the main clinical feature of which was decreased visual acuity,13 of them were pilocytic astrocytoma, which manifested as a solid tumor with significantly enhancement(94%).16 cases were germ cell tumors,the main complaint was diabetes insipidus (75%),13 of them were germinoma, which showed iso-high density on CT and moderate enhancement after administration of contrast.And it showed high signal intensity on DWI.7 cases were hamartoma,86% patients of which showed gelasmus epilepsy,the imaging showed iso-signal masses with no enhacement in the hypothalamic papillary region.5 cases were LCH,80% of which had the complaint of diabetes insipidus,imaging findings manifested as thinkened pituitary stalk and loss of hyperintensity of posterior pituitary on T1WI.3 cases were pituitary tumors.23 cases were Rathke's cleft cysts,3 cases were arachnoid cyst.Most of the patients presented with headache.Conclusion The clinical and imaging features shows some specific features, which is helpful to improve the correct rate of diagnosis and provide the basis for further treatment.
10.Clinical application of genotyping detection of human Mycoplasma urealyticum based on parC gene sequences

Objective To establish a novel PCR method that can differentiate the two biotype of Mycoplasma urealytium rapidly based on the disparities of parC gene sequences for clinical routine examination.Methods Design two pairs of specific primers and probes for the targeted gene according to the differences of the parC gene sequences from 14 standard serotypes.The specificity of this amplification was verified by detecting two Mycoplasma urealyticum standard strains including serovar1 and serovar4,50 clinical isolated strains ( 12 are Uu strains and 38 are Up strains) and 7 common bacterias from vagina.Collected 70 swab specimens from patients of Sexually Transmit Department with urogenital inflammation symptom and 71 swab specimens from the gynecology health examination population.All those specimens were detected by culture and our method respectively.The sensitivity of this method was evaluated by comparing with the culture.Differences of the infection rate and distribution of biotypes between different populations were analyzed using statistical software.Results Standard strains of Mycoplasma urealyticum and clinical isolates can be typed into two species by the PCR method without nonspecific amplification.The sensitivity of PCR method is much higher than that of culture ( P<0.05).The infection rates of Uu,Up and the mixing were 8.57%,61.40%,24.30% respectively for the patients with vaginitis.However,it was 8.80%,67.60%,8.45% respectively for the gynecology health examination population.There is a significant difference of the total infection rate between the two population(P<0.05).It showed no significant difference with the distribution of the two types of simple infection in the two groups ( P>0.05).But the rate of mixing infection is much higher in the patients with vaginitis ( P<0.05 ).Conclusion The pathology of Mycoplasma urealyticum may be related to the mixed infection of different biotypes.
