1.SIMULTANEOUSLY DEMONSTRATION OF TYROSINE-HYDROXYLASE(AND SOMATOSTATIN)GENE EXPRESSION AND PEPTIDE STORAGE IN SINGLE NEURON OF THE HEART

Objective To demonstrate simultaneously on transcription and translate levels the tyrosine-hydroxylase(and somatostatin)gene expression and peptide storage in same intrinsic cardiac ganglionic neuron of bufo bufo gargarizans. Methods Using immunohistochemical method,in situ hybrodization method and in situ hybrodization/immunohistochemical double labeling method on freeze-sections of sinus venanum cavanum of bufo bufo gargarizans atrial posterior wall (30*!?m)to observe THmRNA and /or TH-IR and SSmRNA and /or SS-IR dyeing neurons of intrinsic cardiac ganglia. Results There are kinds of positive neurons:1.TH-IR positive neurons;2.THmRNA positive neurons;3.TH mRNA/TH-IR double positive neurons and 1.SS-IR positive neurons;2.SSmRNA positive neurons;3.SSmRNA/SS-IR double positive neurons.Conclusion 1.TH mRNA and TH appearing in same intrinsic cardiac ganglionic neuron demonstrate that a part of intrinsic cardiac ganglionic neurons are possibly adrenergic sympathetic neurons;2.SSmRNA positive neurons and SSmRNA/SS-IR double positive neurons indicat that SS could be a neurotransmitter of intrincic cardiac ganglinic neurons.It provides neuroanatomy evidence of regulating cardiac function(excitatory conduct、cardiac muscle secrete and contract)by intrinsic cardiac SS-nergic neuron.

Monocyte chemotactic peptide 1(MCP 1) is a specific chemotactic and activating factor for monocytes in acute renal transplant rejection. 3.The effect of rotation constancy static magnetic field on the expression of BMP-2 and PDGFs during fracture healing

Objective To evaluate the effect rotation constancy static magnetic field(RCSMF) on the expression of bone morphogenetic protein-2 (BMP-2) and platelet-derived growth factors (PDGFs) in fracture healing, and elucidate the possible mechanism of RCSMF promoting fracture healing.Methods A total of 80 rats with femur closed fracture were randomly divided into a magnetic treatment group (M) and a control group (C).The M group was given RCSMF treatment 30 minutes per day, 6 times per week, using 0.4 T 6 Hz magnetic field, while the C group was not given any intervention.Femurs and calluses were subjected to BMP-2, PDGF-A and PDGF-B immunohistochemistry assay at 2, 3, 4 and 8 weeks after the treatment.The integrated optical density (IOD) of positive staining was calculated.Two-factor variance analysis was used to compare the main effect of time, treatment and that of their interaction.Results For both groups, the IOD of immunoreactivity of BMP-2 at 2, 3 and 4 weeks after the treatment was significantly higher than that 8 weeks after the treatment, and the value at four weeks after the intervention was also significantly higher than that at 2 weeks after it.There was a significant interaction effect of time × treatment of BMP-2 (F =3.17, P < 0.05).Significant differences were observed in the IOD of positive staining PDGF-A at different time points for both groups : the value at 2 weeks after treatment was significantly higher than that at 3, 4 and 8 weeks after the treatment, and that at 3 and 4 weeks after treatment was also significantly higher than that at 8 weeks after the treatment.The IOD of positive staining PDGF-B of the M group (57.6 ± 2.1) was significantly higher than that of the C group (50.11 ± 2.22, P < 0.05).There was significant difference in the IOD of positive staining PDGF-B (F =50.06 ,P < 0.01) at different time points : the IOD of positive staining PDGF-B at 3 weeks after treatment was significantly higher than that at 2 and 8 weeks group after the treatment, and the value at 2 and 4 weeks after treatment was also significantly higher that at 8 weeks after the treatment.Conclusion RCSMF may elevate the expression BMP-2 and PDGF-B to promote fracture healing.
4.Construction of pcDNA3.1-myc/PINK1 and its expression in COS-7 cells

Objective To construct eukaryotic expression vector of PINK1 (pcDNA3. 1-myc/PINK1) and verify PINK1 expression in pcDNA3.1-myc/PINK1 transfected COS-7 cells. Methods The cDNA fragment encoding human PINK1 gene was amplified by PCR method from human cDNA library. After nucleotide sequencing, this cDNA fragment was inserted into an eukaryotic expression vector pcDNA3. 1-myc-his( - )B using gene cloning and DNA recombination. The recombinant was then transferred into COS-7 cells by liposome. The expression of the target molecule was assayed by western blotting. Results The sequence of DNA fragment amplified by PCR was consistent with that published in Genbank, and digestion of the recombinant plasmid with EcoR Iand BamH Iliberated DNA fragments with expected size. PINK1 was expressed and synthesized in transfected cells after 48h culture. Conclusion An eukaryotic expression plasmid containing human PINK1 gene was successfully constructed, and it can express out objective protein, which has laid a concrete foundation for future study on PINK1.
5.A Study on the Named Entity Recognition Method on Symptom Names in the History of Present Illness in Traditional Chinese Medical (TCM) Clinic

Clinical cases of TCM are used as important clinical data to record the whole process of the interaction between doctors and patients in the form of text.However,in the context of big data,there is a lack of research on the use of information covered in clinical cases.Therefore,we studied the method of extracting the symptom term from the history of present illness in TCM clinic in this paper,in order to lay the foundation for the further use of clinical cases.First,twelve thousand,three hundred and sixty-seven history data of present illness were obtained by random selection and expert review.According to the different disease types,they were divided into the two groups of the experiments:4,838 data in the diabetes group,7,529 data in the spleen and stomach disease group and 12,367 data in the mixed or combined group.A glossary of symptom terms covering 22,996 words were compiled.Then,five feature templates,such as sliding window feature,prefix and suffix character and lexical features,were selected.CRFs model was adopted to carry out named entity extraction experiment.As a result,in the open test,the performance of diabetes,spleen and stomach disease and mixed group were (0.83,0.8,0.82),(0.9,0.9,0.89) and (0.88,0.87,0.87),respectively,while the results were (0.83,0.82,0.83),(0.95,0.95,0.95) and (0.93,0.92,0.92) in the ten-fold cross validation.In conclusion,the results showed that the CRFs algorithm was an excellent sequence labeling algorithm and applied to the named entity extraction task of symptom history.

Objective In this study ,we investigated the apoptosis of hair cell in the cochlea of age -related hearing loss(AHL) generated by ENU mutagenesis ,and to study a pan caspase inhibitor (z-VAD -FMK) which is to protect the cochlea hair cells from hearing loss induced by age-related hearing loss(AHL) .Methods Through z-VAD-FMK intraperitoneal injection and round window membrane (RWM) drug were delivered into the Cdh23 nmf308 nmf/nmf mice 5(postnatal days 2 -32) inner ear .ResuIts The results showed that the nmf308 mice with progressive hair cell loss along a base to apex gradient with age-related hearing loss .The cochlear OHCs reduced from 5% ~10% at 1 month to 100% at 3 month in the basal region .Substantial amounts of TUNEL -positive OHCs nuclei appeared at 1 month age ,and activated caspase-3 labeling demonstrated that most OHCs appeared at 2 months age .These suggested that DNA single strand break was attributed primarily to apoptosis of cochlear le_sions ,whereas in the later stage of lesion ,the expansion led to activation of caspase-3 activity reduced with further progression of nuclear condensation in age-related hearing loss .ConcIusion The addition of a pan caspase inhibitor (z -VAD -FMK ) significantly protected the cochlea against the hair cell loss induced by apoptosis .Our study showed that aspase inhibitor ,Z-VAD-FMK appeared to play a prominent role in age-related hearing loss media_ted hair cell death loss induced by apoptosis .Our study showed that aspase inhibitor ,Z-VAD -FMK appeared to play a prominent role in age-related hearing loss mediated hair cell death .
7.The application of septum cartilage in correcting of the unilateral cleft lip nasal deformity.

OBJECTIVETo explore an ideal method for comprehensive correcting of the unilateral cleft lip nasal defomity.
METHODSThe operation includes replacement of the displaced tissue, excision of the deviated septum, and correction of the deviated septum and the deformity of the lower nose. The excised nasal septum was grafted on the alar base, the nasal columella and the nasal tip.
RESULTS32 patients were treated with this method from 1994 to 1999. Postoperative fellow-up for 1-3 years demonstrated satisfactory results.
CONCLUSIONThrough excision and grafting of the nasal septum, the deviated septum and the deformity of the lower nose are effectively corrected. Complete undermining and replacement of the displaced structure are the basis of correcting the cleft lip nasal deformity.

8.Experimental study on assistant designing of computer for new Chinese herbal compounds with anti-infectious function

Objective To study the feasibility of artificial immune system(AIS)in assistant designing of computer for new Chinese herbal compounds with anti-infectious function.Methods A group of Chinese herbal drug compounds from Treatise on Exogenous Febrile and Miscellaneous Diseases were selected as training samples.Serum was obtained from mice which were infected by Staphylococcus aureus and ig administrated with the various Chinese herbal compounds.The inhibition ability against S.aureus of the serum was tested and input into an artificial neural network(ANN)together with the characters of the every Chinese herb included in those compounds.The ANN was optimized using AIS so that it can predict the inhibitory ability against S.aureus of the Chinese herbal compounds according to the characters of Chinese herbs included in those compounds.After that the computer generated a number of Chinese herbal compounds randomly,calculated their inhibitory ability against S.aureus and output those among the top levels,so as to verify whether the predictions were correct or not.Results After verification,the Chinese herbal drug compounds designed by computer could significantly raise the serum's inhibitory ability against S.aureus.Conclusion The computer can be an assistant for designing new Chinese herbal compounds with anti-infectious function
9.Clinical Research on Detoxification with Acupuncture

Purpose To confirm the effects of acupuncture, and Chinese medicines in controlling the withdrawal symptoms from the opium-like drugs. Method 96 heroin-dependent subjects were divided into four groups,which were treated respectively by western medicine (Agroup), acupuncture (B group), Chinese herbs (Cgroup), and acupuncture & Chinese herbs (D group).Before, during and after treatment, the concentration of serum testosterone and prolactin, and immune functions (serum CD3+ 、CD4+ 、CD8+ and CD4+/CD8+ ) were tested.Results After treatment, the concentration of serum testosterone in A and B group were higher than before and during treatment, and in C and D group, during treatment were higher. In the four groups, the concentration of serum prolactin before treatment was the highest. The levels of CD3+、CD4+、 CD8+ and CD4+/CD8+ were lowest before treatment and highest after treatment. Conclusion Acupuncture and Chinese medicines effective in relieving spasm and pain can control the opium-like drug withdrawal symptoms to different degrees, especially acupuncture. However, acupuncture cannot ease the withdrawal symptoms completely. Acupuncture does not strikingly cooperate with the Chinese medicines effective in relieving spasm and pain (including M-receptor antagonists). In detoxification, the Jiaji points are the primary ones and symptom-based points the secondary ones.
10.Comparison of the Detoxification Effects of Acupuncture, Chinese Herbs and Acupuncture Plus Chinese Herbs on Heroin Addiction

Heroin addicts were treated with acupuncture, Chinese herbs, and acupuncture plus Chinese herbs respectively, their effects were compared with that of Western medicine.The scores of withdrawal symptoms were observed continuously for 20 days. The effect of acupuncture was better than that of Western medicine 48 hours after the withdrawal treatment (P< 0.05), and better than that of other three methods 72 hours after the withdrawal treatment, (P < 0.01). Acupuncture, Chinese herbs, and acupuncture plus Chinese herbs have the detoxification effects of varying degrees, in particular acupuncture' effect was the best. But all the therapies could not completely improve the withdrawal symptoms in the first 3 days of treatment, and acupuncture plus Chinese herbs had no synergetic effect in treating withdrawal symptoms.
