1.Effects of Coptidis Rhizoma on Gastrointestinal Motility Functions, MTL and GAS in Spleen-deficiency Rats

Objective To observe the effects of Coptidis Rhizoma on gastrointestinal motility functions, MTL and GAS in spleen-deficiency rats; To explore the dose-effect mechanism of “invigorating stomach” and “spoiling appetite”.Methods Fatigue and troublesome were used to establish exhausted spleen-deficiency models. 60 SD rats were randomly divided into blank control group, model group, positive medicine group, Coptidis Rhizoma high-, medium- and low-dose groups, with 10 rats in each group. Each medication group received relevant medicine for gavage, while the blank control group and model group received normal saline for gavage, once a day for 14 d. The general gastrointestinal motility functions, MTL and GAS of rats in each group were observed.Results Compared with blank control group, the gastric residual rate of model group was significantly higher; small intestinal propulsion rate, MTL, and GAS were significantly lower (P<0.05,P<0.01). Compared with model group, the gastric residual rate in Coptidis Rhizoma low-dose group was significantly lower; small intestinal propulsion rate, MTL and GAS increased significantly (P<0.05,P<0.01); small intestinal propulsion rate and MTL in Coptidis Rhizoma high-dose group were significantly lower (P<0.05).Conclusion High-doseCoptidis Rhizoma leads to “spoiling appetite”, and low-dose Coptidis Rhizoma leads to “invigorating stomach”, which may be related to adjusting gastrointestinal motility directly or adjusting the secretion of gastrointestinal hormones.

Objective To explore the role of plastic astrocytes in focal cerebral infarct. Methods Immunohistochemical and double immunofluorescence techniques were used in the study. Results The astrcytes positive for glial fibrillary acidic protein(GFAP) occurred in the periinfarct area appeared hypertrophy and proliferation,specially on their processes and formed a network,and the processes oriented toward the infarct center from the periinfarction.The expression of excitatory amino acid transporter 1( EAAT1) increased in the penumbra area of cerebral infarct and manifested spot and fibre.The confocal laser scanning microscopic analysis showed the double staining for EAAT1 and GFAP.Conclusion The plastic astrocytes might participate in the pathological process of cerebral infarct recovery by enhancing the expression of EAAT1.;

Objective To explore the cell frequency , phenotypes and in vitro cytotoxic effects of circulating CD56+T cells in the patients with chronic HCV infection .Methods Peripheral blood mononu-clear cells (PBMCs) were isolated from 33 patients with HCV chronic infection and 21 healthy subjects. Multi-color flow cytometry was used to analyze cell frequency , expressions of activating receptors ( NKG2C, CD16 and NKp46) and inhibitory receptors (NKG2A and CD158a) on CD56+T cells.The functional mark-er for cytotoxic effects (CD107a) on circulating CD56+T cells and their cytokines expression (IFN-γand TNF-α) with or without stimulation of K 562 human Leukemia cell line were also analyzed .Then the correla-tions among the expressing levels of CD 107 a, IFN-γand TNF-αwere investigated .Results The frequency of CD56+T cells in periphery lymphocytes were significantly decreased in the patients with chronic HCV in -fection as compared with that in healthy controls ( P=0.018 ).The expressions of activating receptors (NKG2C, CD16 and NKp46) on CD56+T cells from HCV infected patients were decreased (P=0.015 for NKG2C, P=0.036 for CD16 and P=0.001 for NKp46), while there was no significant change in the ex-pressions of inhibitory receptors (P>0.05 for both CD158a and NKG2A).The concentrations of IFN-γand TNF-αsecreted by CD56+T cells in the patients with chronic HCV infection were significantly decreased with or without K562 stimulation (P<0.0001).However, in the presence of K562 cells CD107a expression on CD56+T cells were sharply decreased in the patients (P<0.0001).In absence of K562 cells, there was no significant change in CD107a expression on CD56+T cells from patients and healthy controls (P>0.05). The expressions of CD107a, IFN-γand TNF-αwere closely related under the stimulation of K562(r>0.80, P<0.0001).Conclusion The frequency of CD56+T cells was reduced in patients with chronic HCV infec-tion.Moreover, cytotoxic effects and cytokines production mediated by CD 56+T cells were also significantly impaired, indicating that the dysfunction of circulating CD 56+T cells might be associated with the persist-ence of chronic HCV infection .
5.Establishment of a Rat Model of Ulcerative Colitis by Immunological Sensitization Combined with Local Acetic Acid Irritation

Objective To establish a rat model of ulcerative colitis by immunological sensitization combined with local acetic acid irritation.Methods Thirty healthy 2-3-month-old Wistar rats were randomly divided into 3 groups:normal control group,model group and SASP-treated model group(n=10 rats per group).The ulcerative colitis in the model group and positive group was developed by immunological sensitization with rabbit colon mucosal proteins and local irritation with 5%acetic acid.The rats of the model and positive groups were fed with sulfasalazine,and rats of the control group fed with normal saline.The rats of positive control group were treated with sulfasalazine.Two weeks later,all rats were killed and colon mucosal and blood samples were collected.The concentrations of NO,NOS,MDA,SOD and GSH-Px in the colonic mucosal homogenate were measured by biochemistry,the contents of IL-4 and TNF-α were measured by RIA,and the serum concentration of IFN-γ was measured by ELISA.Results Compared with the normal control group,the concentration of MDA,NO,NOS in the colonic mucosa was increased,and SOD and GSH-Px levels significantly decreased in the model group.Compared with the model group,the concentrations of MDA,NO,and NOS in the colonic mucosal tissue were decreased,and the levels of SOD and GSH-Px decreased in the SASP-treated model group.The contents of IL-4 in the serum and colonic mucosal homogenate of model group were significantly lower, and the contents of TNF-α in the serum and colonic mucosal homogenate were significantly increased than those of the normal control group. The IL-4 levels in the serum and colonic mucosal homogenate was increased, and that of TNF and IFN-γ significantly decreased in the SASP-treated model group, compared with those of the normal control group.Conclusion The rat model of ulcerative colitis established by immunological sensitization combined with local acetic acid irritation, as described in the present study, can better simulate the characteristic chronic alterations in human ulcerative colitis,and may better serve studies and evaluation of treatment of this disease.
6.Comparative study of the clinical effect of locking plate and anatomical plate in the treatment of Pilon fracture

Objective To compare the clinical effect of two kinds of internal fixation of locking plate and anatomical plate in the treatment of Pilon fracture.Methods 64 patients with Pilon fracture undergoing elective surgery were randomly divided into study group and control group by digital table method,32 cases in each group.The study group received locking plate internal fixation for the treatment,the control group was treated with anatomical plate internal fixation.The clinical efficacy and safety of two kinds of internal fixation methods were compared.Results After treatment,the excellent rate of ankle joint function in the study group(96.88%)was significantly higher than that in the control group (81.25%),and the difference had statistical ssignificance (χ2 =4.01,P <0.05 ).The operative time,hospitalization time and fracture healing time[(57.32 ±10.98)min,(13.72 ±591 )d,(15.72 ± 2.82)weeks]in the study group were shorter than those in the control group[(68.11 ±11.85 )min,(16.62 ± 5.21)d,(17.81 ±3.42)weeks],and the differences had statistical significance(t =3.98,3.11,321,all P <0.05). The amount of bleeding (153.32 ±21.36)mL in the study group was less than that in the control group [(188.65 ± 26.24)mL](t =5.46,P <0.05).The incidence rate of delayed fracture healing of the study group (3.13%)was significantly lower than that in the control group(28.13%),the difference had statistical significance(χ2 =7.59,P <0.05).There were no significant differences in the incidence of other complications between the two groups(χ2 =1.95,102,102,0.35,all P >0.05).Conclusion Locking plate fixation in the treatment of Pilon fractures could significantly promote the recovery of joint function,and has the advantages of short operation time,fast fracture healing,less complications and other advantages,it is better than anatomical plate internal fixation.
7.Diagnostic utility of WT-1, AR and Ki-67 in Sertoli cells and germ cells for testicular biopsy

Purpose To explore the specific diagnostic markers of Sertoli cells and germ cells for testicular biopsy. Methods Normal testis from 3 patients who suffered from carcinoma of the prostate and treated with testis castration, and 15 testicular biopsy tissues were stained by EnVision two steps with WT-1, AR and Ki-67. Results The expression of WT-1 and AR protein were found in Sertoli cells from the 3 normal testis and 15 testis biopsy, the positive rate were all 100%, and non was positive in germ cells. The germ cell was positive for Ki-67, and the positive rate was 100%. Conclusions WT-1, AR and Ki-67 may be the specific diagnostic markers of Sertoli cells and germ cells for testicular biopsy, which may contribute to the diagnosis of testicular biopsy.
8.Cortical vein thrombosis: three cases for the clinical, neuroimaging and pathological analysis

Objective To analyze the clinical,imaging and pathological features of cortical vein thrombosis (CoVT).Methods Three cases of cortical vein thrombosis were diagnosed in our hospital from February 2010 to October 2012.We reviewed and summarized their clinical manifestation,radiological feature and pathological characteristics.Results All patients were young with acute onset.The main clinical manifestations included headache,epilepsy or limbs weakness.Two cases had intracranial hypertension.One case had decreased activity of protein S.One had increased plasma homocysteine.Brain computed tomography scan showed hypodensity lesion with some hyperdensity inside.Cranial routine magnetic resonance imaging showed long signal in T1-weighed image and T2-weighed image,with occasional short T1 signal inside.Contrast-enhanced magnetic resonance imaging revealed heterogeneous enhancement.All of 3 cases underwent brain biopsy because of the suspected diagnosis of brain tumor.Brain pathology showed the local necrosis and hemorrhage,dilated small vein with congestion or thrombosis.Neuronal degeneration,hyperplasia of gliocyte,hyperplasia of endotheliocyte in small blood vessels with reaction of histiocytes was also displayed.Duration from initial visit to final diagnosis was from 14 days to 2 months.Conclusions CoVT has various clinical and radiological manifestations and it is easy to misdiagnose as brain tumor.Careful analysis of clinical and imaging data could improve its diagnostic accuracy.Brain biopsy would also be helpful for diagnosis.

Objective To evaluate a protocol of early treatment for dogs suffering from open abdominal injury and seawater immersion. Methods A canine model of open abdominal injury was made with 30 adult health dogs. After 2 hours seawater immersion, the dogs were divided into two groups: treatment group( n =18) and control group(untreated group, n =18). The changes of survival rate, body liquid metabolism and hemodynamics were observed respectively. Objective To investigate the differences of X-ray findings of skeletal fluorosis between coal-burning type endemic fluo-rosis and industrial fluorosis.Methods The patients were randomly selected as research objects including 60 cases of coal-burning type endemic osteofluorosis and 60 cases of industrial osteofluorosis.The X-ray findings on the left forearm,crus and pelvic radio-graphs of these patients were analyzed retrospectively to find out the differences between skeletal fluorosis of coal-burning type endemic fluorosis and industrial fluorosis.Results X-ray features are no significant statistical differences between coal-burning type endemic fluorosis and industrial fluorosis,except these of interosseous membrane ossification of forearm and crus (forearmχ2=10.909,P<0.05;crusχ2=8.547,P<0.05),obturator membrane ossification of pelvis (χ2=36.554,P<0.05),periosteal proliferation outside bone of crus (χ2=4.937,P<0.05),and ossification of soleus (χ2=4.904,P<0.05).Conclusion The X-ray signs of endemic osteofluorosis and industrial skeletal fluorosis are almost similar,but there are some differences between them.
