1.Research on the clinical value of tumor deactivation after radiofrequency ablation of liver cancer assessed by MDCT

Objective:To explore the clinical value of tumor deactivation after radiofrequency ablation of liver cancer assessed by MDCT.Methods: By CT perfusion combined with CT angiography and CT enhanced scan in 91 cases of liver cancer patients before and after radiofrequency ablation treatment, the change of tumor size, density change, scope of coagulation necrosis area and comparison sensitivity, specificity and accuracy of two methods diagnosis of recurrent or residual tumor were observed.Results:In the diagnosis of tumor recurrence or residual degree of sensitivity, specificity and accuracy CTP combine CTA were 100%, 95.3%, 96.2%, conventional enhanced CT were 33.3%, 88.4% and 78.8%. The diagnosis of recurrence or residual tumor after radiofrequency sensitivity and accuracy CTP combine CTA is higher than conventional enhancement CT, The difference was statistically significant(x2=6.912, x2=7.121;P<0.05). Both no statistical difference between specificity(x2=0.615,P<0.05).Conclusion: CTP combination CTA can accurately determine the tumor after radiofrequency ablation for inactivated and degree, and superior to the conventional enhanced CT, is an effective evaluation method of radiofrequency ablation with a high clinical value.
2.Toxicological assessment on safety of Fangyouling, a cercaricide of preparation

Objective To evaluate the safety of Fangyouling, a cercaricide of preparation. Methods All of the tests including the acute oral and dermal toxicity, acute eye and skin irritability and the dermal sensitivity reaction were carried out according to the toxicological test methods of pesticides for registration (GB 15670-1995). Results Fangyouling belonged to slight grade of acute toxicity through both oral and skin tests in rats. Fangyouling was not acute irritant for the skin and eyes in rabbits and weak allergic for the skin in guinea pigs. Conclusion Fangyouling may be a safe ceraricide preparation for prevention of Schistosoma japonicum cercariae infection by skin administration.
3.Flow injection chemiluminescence determination of loxoprofen and naproxen with the acidic permanganate-sulfite system

A novel flow injection chemiluminescence(CL)method for the determination of loxoprofen and naproxen was proposed based on the CL system of KMnO4 and Na2SO3 in acid media.The CL intensity of KMnO4-Na2SO3 was greatly enhanced in the presence of loxoprofen and naproxen.The mechanism of the CL reaction was studied by the kinetic process and UV-vis absorption and the conditions were optimized.Under optimized conditions,the CL intensity was linear with loxoprofen and naproxen concentration in the range of 7.0×10^-8-1.0×10^-5g/mL and 2.0×10^-7-4.0×10^-6g/mL with the detection limit of 2.0×10-8g/mL and 3.0×10-8g/mL(S/N=3),respectively.The relative standard deviations were 2.39% and 1.37% for 5.0×10^-7g/mL naproxen and 5.0×10^-7g/mL loxoprofen(n=10),respectively.The proposed method was satisfactorily applied to the determination of loxoprofen and naproxen in pharmaceutical preparations.

5.Isolation of glioma stem cells from human glioma cell lines U87 with serum deprivation method

BACKGROUND: Cell cycle has proved that about 90% stem cells were in resting state in the G0 phase; therefore, serum deprivation method which did not contain any growth factors and related nutrition additive was beneficial for separating glioma stem cells.OBJECTIVE: To explore the method of serum deprivation to culture glioma cell lines U87, and to isolate and identify the cancer stem cells which are in the U87 cell lines. METHODS: U87 cells were cultured in the medium containing both DMEM and L-glutamine for six days; cancer stem cells would be screened out; then the medium was changed for the neural stem cell culturing medium. The formation of tumor spheres and their differentiation characteristics were observed when they were inoculated onto serum-containing medium. The immunofluorescence staining of cells was employed to identify the surviving cells cultured in serum deprivation medium, tumor spheres and differentiated cells. RESULTS AND CONCLUSION: The serum deprivation method was used to select tumor stem cells successfully which expressed CD133 and could form tumor spheres. The tumor spheres had an ability of multi-differentiations, and the daughter cells expressed glial fibrillary acidic protein and neuron specific enolase. The U87 cell lines exist glioma stem cells which have the capacities of self-renewal and multi-differentiation.
6.Dithothreitol (DTT) treating lymphocytes highly sensitized antibodies in renal pretransplantation patients

72 sera of highly sensitized antibody patients were detected by 35 samples of undonor lymphocytes before and after the dithothreitol (DTT) treating.The results showed that 72 sera reacted completely to B lymphocytes in both 4℃ and 37℃,but incompletely to T lymphocytes.After removal of the transplant kidney due to rejection,the sera antibody levels of T or B lymphocytes were higher than those of the patients'sera after blood transfusion or pregnancy.The serum T or B antibody levels of pregnancy were the lowest.In DTT-treated sera of highly sensitized patients,the antibodies were mixed types of IgG and IgM.

Abstract
Survival analysis has been widely used in the field of medical research. The Cox proportional hazard model is commonly used, but its practical application is limited. Machine learning method can compensate for the shortcomings of the Cox proportional hazard model in terms of nonlinear data processing and prediction accuracy. This article reviewed the advance of machine learning methods represented by neural networks, within the field of survival analysis, and highlighted the principles and benefits of three machine learning methods that DeepSurv, Deep-Hit and random survival forest, providing methodological insights for the analysis of complex survival data.
8.Repairing flexor tendons defect in Zone Ⅲ digitorum superficial tendons by e-stage transferring near flexor digitorum superficial tendons

This study is to evaluate the curative effect of repairing the flexor tendons defect in the fibroosseous sheath by transferring the near flexor digitorum superficial tendons. Thirty-two cases (39 fingers) with acute flexor tendon defect in Zone I were repaired by transferring the flexor digitorum superficial tendons. Early protected passive motion was advocated postoperatively. All cases were followed up for 11 to 23 months. The outcome of fingers were excellent in 24,good in 12 and fair in 3 by TAM score. The percentage of excellent and good outcome was 92. 3%. Microrepair of fresh flexor tendons defect in Zone II by transferring flexor digitorum superficial tendons combined with early protected passive motion can prevent the adhension of tendons effectively and restore finger function earlier.
9.Flow-injection-enhanced chemiluminescence method for the determination of three β-blockers

Objective To develop a rapid, simple and sensitive chemiluminescence method for the determination of three β-blockers (bisoprolol, atenolol and propranolol). Methods The chemiluminescence of cerium (Ⅳ)-sulfite system was obviously sensitized by adding anyone of three β-blockers in acid media. A new chemiluminescence method was set up by combining with flow-injection technique and used to determine the three β-blockers. Results Good linear ranges were obtained at the concentrations of 2.0×10-7g/mL-4.0×10-5g/mL, 1.0×10-7g/mL-3.0×10-5g/mL and 7.0×10-7g/mL-1.0×10-5g/mL, respectively, with the detection limits of 5.0×10-8g/mL, 7.0×10-8g/mL and 5.0×10-8g/mL (S/N=3), respectively, and the relative standard deviations for 11 times consecutive injections of 1.0×10-6g/mL bisoprolol, atenolol and propranolol were 3.57%, 2.21% and 2.26%, respectively. Conclusion The developed method is sensitive, accurate, rapid and of low cost. And it can be applied to determine bisoprolol, atenolol and propranolol in pharmaceutical preparations.
10.Sensitive determination of 4-O-methylhonokiol in rabbit plasma by high performance liquid chromatography and application to its pharmacokinetic investigation

A novel high performance liquid chromatographic method was developed for the determination of 4-O- methylhonokiol in rabbit plasma and was applied to its pharmacokinetic investigation. Plasma samples were treated by one-fold volume of methanol and acetonitrile to remove the interference proteins. A reverse phase column of SHIM- PACK VP-ODS (150 mm ×4.6 mm, 5.0 μm) was used to separate 4-O-methylhonokiol in the plasma samples. The detection limit of 4-O-methylhonokiol was 0.2 μg/L and the linear range was 0. 012 - 1. 536 mg/L. The good extraction recoveries were obtained for the spiked samples (84.7%, 89.3% and 87.7% for low, middle and high concentrations of added standards, respectively). The relative standard deviation of intra-day and inter-day precisions was in the range from 0.6% to 13.5%. The pharmacokinetic study of 4-O-methylhonokiol was made and the results from the plasmaconcentration curve of 4-0-methylhonokiol showed a two-apartment open model. This work developed a sensitive, stable and rapid HPLC method for the determination of 4-O-methylhonokiol and the developed method has been successfully applied to a pharmacokinetic study of 4-O-methylhonokiol.
