Objective To investigate the expression of vascular endothelial growth factor (VEGF)、matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9 (MMP-9) in tongue squamous cell carcinoma and the clinical significance.Methods The expression of MMP-2,MMP-9 and VEGF in 60 cases of tongues quamous cell carcinoma tissues and in 20 cases of normal tongues tissues were examined by immunochemistry.Results The expression of MMP-2,MMP-9 and VEGF was higher in tongues quamous cell carcinoma than that in normal pancreatic parenchyma.In cases with lymph nodes metastasis and TNM stage Ⅲ + Ⅳ,the expression of two markers were significantly higher than those without lymph nodes metastasis and TNM stage Ⅰ + Ⅱ (P ＜ 0.05).A positive correlation was found between the expression of VEGF and MMP-2,MMP-9 in tongues quamous cell carcinoma ( P ＜ 0.05 ).Conclusion The expression of MMP-2,MMP-9 and VEGF was closely correlated with lymph nodes metastasis and TNM stage of tongues quamous cell carcinoma(P ＜ 0.05 ).VEGF and MMP-2,MMP-9 play an important role in the development of tongues quamous cell carcinoma,may cooperate with each other.

Objective To screen 8 series of LY compounds, c-Met tyrosine kinase inhibitors, and evaluate their anti-tumor effects in vitro and in vivo. Methods Preliminary screening was carried out by detecting the c-Met kinase phosphor?ylation inhibition activity of the compounds. CCK-8 assay was adopted for secondary anti-tumor screen of the selected com?pounds using MKN-45, U87MG, Caki-1 and PC-3 cell lines in vitro. The transplanted tumor model of U87MG cells in nude mice was established to evaluate the antitumor activity in vivo. Results Four compounds (LY22, LY25, LY28 and LY32) with better activities were selected by HTRF method, in which LY28 had better inhibitory effect on c-Met than that of Crizo?tinib. The above active compounds showed different degrees of inhibition on the four kinds of target cells (MKN-45, U87MG, Caki-1 and PC-3) detected by CCK-8 method, and the inhibitory effect of LY28 showed the most obvious. Antitumor activi?ty in vivo showed that LY28 can significantly inhibited tumor growth in a dose-dependent manner. The tumor inhibitory rate in high-dose of LY28 was 78.13%. Conclusion The compound LY28 has good antitumor activity in vitro and in vivo, which will be a new tyrosine kinase inhibitor.

Objective To evaluate the effect of sevoflurane on myocardial injury induced by hemorrhagic shock and resuscitation (HS/R) in pigs.Methods Twenty-four Bama miniature pigs (12 males, 12 females) , weighing 20-25 kg, aged 3-5 months, were randomly divided into 3 groups (n=8 each) using a random number table: sham operation group (group S) , HS/R group and sevoflurane group (group Sev).The left and right femoral arteries and right femoral vein were cannulated for blood pressure monitoring, blood-letting, blood sampling and fluid infusion.HS/R was induced by blood-letting maintaining for 1 h, followed by resuscitation with autologous blood reinfusion and infusion of lactated Ringer's solution 2 times the volume of the blood withdrawn.The pigs in group Sev were exposed to 2％ sevoflurane for 30 min before resuscitation.After cannulation, at 30 min after hemorrhagic shock, before resuscitation, and at 30 min, and 1.5, 2.5 and 3.5 h after resuscitation, blood samples were collected from the femoral artery for determination of creatine kinase-MB (CK-MB) activity and cardiac troponin Ⅰ (cTnI) concentration in serum using an automatic biochemical analyzer.Myocardial specimens were obtained at 3.5 h after resuscitation for detection of tumor necrosis factor-alpha (TNF-ot) and interleukin-6 (IL-6) contents (by ELISA) , and phosphor-signal transducer and activator of transcription 1 (p-STAT1) expression (by Western blot), and for examination of the pathological changes (with light microscope).Results Compared with S group , the CK-MB activity and cTnI concentration in serum and contents of TNF-α and IL-6 were significantly increased, and the expression of p-STAT1 was up-regulated in HS/R and Sev groups (P＜0.05).Compared with HS/R group, the CK-MB activity and cTnI concentration in serum and contents of TNF-α and IL-6 were significantly decreased, and the expression of p-STAT1 was downregulated (P＜0.05) , and the pathological changes of myocardia were alleviated in Sev group.Conclusion Sevoflurane can alleviate HS/R-induced damage to myocardia of pigs, and inhibited STAT1 activity and attenuated inflammatory responses in the myocardium are involved in the mechanism.

Objective To study the biological effects of grape seed proanthocyanidin extract (GSPE) on human ovary cancer cells and to explore the molecular mechanism. Methods Human ovary cancer cell line SKOV3 were co-cultured with GSPE solution of the terminal concentrations of 25,50,100,200,400 ?g/ml respectively in 96-well plate. At 24,48,72 h after coculture,the following parameters were detected: cell growth curve,the inhibition rate of SKOV3 cells by MTT assay,DNA cycle by FCM,the apoptosis of SKOV3 by TUNEL and Annexin-V labeling method,the mRNA and protein expressions of survivin by semi-quantitative RT-PCR and Western blotting,respectively. Results GSPE dose-dependently inhibited the proliferation of the SKOV3 cells. Treated by GSPE,the progress of SKOV3 cells at S stage into G/M stage was inhibited. TUNEL showed that treated by 25 ?g/ml GSPE for 24,48 h,the apoptotic rates of SKOV3 cells were 31.98%,45.78% respectively. Annexin-V showed that after incubation with 25 ?g/ml GSPE for 24 h,the apoptotic rate was 14.68%. The survivin mRNA and protein expressions were both down-regulated. Conclusion GSPE inhibits the proliferation of malignant human ovary cancer cells and induces their apoptosis. Expression of survivin mRNA and protein may be related to cell growth inhibition and to the apoptosis mediated by GSPE in vitro.

Objective To observe the oncololigcal and cosmetic outcomes and complication of breastconserving surgery and radiotherapy for early breast cancer. Methods A total of 78 early breast cancer patients, who were treated in the First People's Hospital of Pingdingshan between January 2001 and September 2008, were identified and followed. Among them, 36 were in stage Ⅰ and 42 in stage Ⅱ. Tangential irradiation was used with a dose of 50 Gy. Tumor bed received a boost dose of 10 Gy electron beam. For axillary lymph node-positive patients, ipsilateral supraclavicular field was hybrid irradiated with 50 Gy X and electron beam.Results The total follow-up ranged from 6 to 98 months. Local recurrence was observed among 4.9% of the patients. The 3-year and 5-year overall survival rate were 96.6% and 92.7% ,respectively. The breast cosmetic satisfaction rate was 94.9%. The main complications reported were acute skin reaction and upper limb edema.Conclusion Breast-conserving surgery and post-operative radiotherapy in early breast cancer patients can lead to satisfactory survival rates and cosmetic effects and hence improved life quality.

Objective To investigate the effects of methylthioadenosine phosphorylase (MTAP) on invasion and migration in breast cancer cells.Methods Human breast cancer cell line MCF-7 cells were treated with MTAPtargeted siRNA to diminish MTAP mRNA.MCF-7 cells proliferation was evaluated by cell counting kit-8,the analysis of cells invasion and migration was performed using Transwell chamber.The expressions of MTAP and matrix metalloproteinase 1 (MMP1) in cell extracts were detected by Western blotting.The experimental divided into blank contrd group,negative control group,MTAP-siRNA experimental group.Results The MCF-7 cells growth was promoted after knockdown the MTAP mRNA.MATP-siRNA experimental group 450 nm absorbance values at 24h,48 hand72 h of the control group were (112.3±11.9)％,(144.4±8.4)％,(169.3±9.4)％ respectively.Cell invasion analysis by Transwell chamber showed 570 nm absorbance values were 0.49 ± 0.06 (control),0.45 ± 0.07 (negative control) and 0.87 ± 0.07 (MTAP-siRNA) respectively.Cell migration analysis by Transwell chamber showed 570 nm absorbance values were 0.46 ± 0.06 (control),0.49 ± 0.08 (negative control)and 0.75 ± 0.07 (MTAP-siRNA) respectively.The expression of MMP1 in MCF-7 cells was upregulated after knockdown the MTAP mRNA.Conclusion The knockdown of MTAP in MCF-7 cell can increase the cells invasion and migration,and this may involve the the MMP1.

BACKGROUND: Blood pressure of children increases with ages, and is related to anthropometric parameters, diet, sports, and other factors. To analyze relative factor of a little high blood pressure of children provides a scientific evidence for prevention and cure of primary hypertension. OBJECTIVE: To investigate correlation between blood pressure and anthropometric parameters among children. DESIGN: Cross-sectional study. SETTING: Department of Hygiene of Weifang Medical College. PARTICIPANTS: According to cluster sampling, 1 546 pupils in grade three or four were selected from a primary school for physical examination and questionnaires. Totally 1 508 pupils with complete data were regarded as the subjects.METHODS: Partial correlation analysis was used to analyze correlation between blood pressure and anthropometric parameters among children.MAIN OUTCOME MEASURES: Correlation among anthropometric parameters, systolic pressure and diastolic pressure.RESULTS: ① Correlation coefficients of systolic pressure with sex, age, height, body mass, chest circumference of Peason were 0.078, 0.166, 0.337, 0.313 and 0.304 (P ＜ 0.01). ② Correlation coefficients of diastolic pressure with sex, age, height, body mass, chest circumference of Peason were 0.047, 0.120, 0.268, 0.271 and 0.251 (P ＜ 0.01). ③ After controlling two effect of anthropometric parameters on each other, systolic pressure and diastolic pressure were only correlated with height and chest circumference (P ＜ 0.01), but was not correlated with age, sex and body mass (P ＞ 0.05). CONCLUSION: After controlling the effect of anthropometric parameters on each other, systolic pressure and diastolic pressure of children were positively correlated with height and chest circumference.

AIM: To investigate the antagonistic effect of zinc on hepatic ischemia- reperfusion (HIR) injury and the effect of zinc on the expression of adhesion molecules in rat liver to clarify the mechanisms involved. METHODS: After zinc supplementation (5 ?mol/kg bw,po) , the changes in hepatocellular morphology,ALT and AST activities in serum,MDA levels in serum,and expression of ICAM-1,VCAM-1 in liver of the animals subjected to HIR were examined. RESULTS: The results showed that HIR (30 minutes of ischemia followed by 90 minutes of reperfusion) significantly increased ALT, AST activities and MDA levels in serum. The destruction of hepatic structure was observed in HIR rats. In the mean time, the expression of adhesion molecule was enhanced.After zinc administration, ALT and AST activities in serum and MDA levels in serum were all decreased. The structure of hepatocyte was nearly normal.The further experiment showed that adhesion molecule expression was suppressed.CONCLUSION: These results indicate that zinc may protect liver against ischemia-reperfusion injury by inhibiting the production of free radicals and the expression of adhesion molecule.

Objective To evaluate the effect of sevoflurane on the expression of aquaporin 8 (AQP8) in the intestinal mucosa in a pig model of hemorrhagic shock.Methods Twenty-four Bama miniature pigs of both sexes, weighing 22-25 kg, were randomly divided into 3 equal groups using a random number table: sham operation group (group S), hemorrhagic shock group (group HS) and sevoflurane group (group PS).The femoral artery and jugular vein were cannulated for blood pressure monitoring, blood-letting, and blood sampling in anesthetized pigs.Hemorrhagic shock was induced by withdrawing blood from the right femoral artery.Hemorrhagic shock was induced after cannulation in group HS.In group PS, 2％ sevoflurane was inhaled for 30 min after the model of hemorrhagic shock was successfully established.Before anesthesia, and at 0.5, 1, 1.5, 2, 3 and 4 h after hemorrhagic shock, blood samples were collected from the jugular vein for determination of serum D-lactic acid and intestinal fatty acid-binding protein (I-FABP) concentrations.The animals were sacrificed at 4 h after hemorrhagic shock, and the intestinal specimens were obtained for microscopic examination and for determination of AQP8 expression in the intestinal mucosa (by enzyme-linked immunosorbent assay).The intestinal water content was calculated.Results Compared with group S, the serum D-lactic acid and I-FABP concentrations, AQP8 expression, and intestinal water content were significantly increased in HS and PS groups (P＜0.05).Compared with group HS, the serum D-lactic acid and I-FABP concentrations, AQP8 expression, and intestinal water content were significantly decreased in group PS (P＜0.05).The pathological changes of intestinal tissues were significantly reduced in group PS as compared with group HS.Conclusion Sevoflurane can decrease the intestinal mucosal edema through inhibiting AQP8 expression, thus reducing hemorrhagic shockinduced damage to the intestinal mucosa in pigs.
