Objective:To approach the role of lymphangiogenesis in oral squamous cell carcinoma(OSCC). Methods: Immunohistochemical double staining technique was utilized to distinguish lymphatic vessels from blood vessels. MLVD was determined by manual counting to analyze the relationship with lymph node metastasis. Results: MLVD was significantly higher in 40 OSCC than 14 normal mucosa(P<0.01). MLVD in OSCC showed a statistically significant relationship with regional lymph node metastasis, depth of invasion(P<0.01). Conclusion: Lymphangiogenesis might occur in growth, infiltration and metastasis of OSCC. Detection of MLVD in OSCC might be important to evaluate tumor metastasis and judge prognosis.
3.Three-dimensional reconstruction and display of intraocular and orbital tumors based on multi-modality image fusion theory

Objective: To study the application of CT and MRI fusion technique in the diagnosis and treatment of intraocular and orbital tumors. Methods: 2D-images of 13 patients with intraocular and orbital tumors were fused by special-point registration and Iterative Local Closest Point(ILCP) method; 3D-fusion images were reconstructed by Ray Tracing method. Results: A 3D-CT-MRI fusion images of intraocular and orbital tumors were reconstructed and displayed. The CT and MRI data of intraocular and orbital tumors were displayed on the same image as a comprehensive whole,which provided a stereogram of 3D-structure of the normal and abnormal orbital tissues. Anatomical structure of the orbit was clearly visualized by 3D-CT-MRI image. Conclusion: The multi-modality fusion technique can provide more accurate and comprehensive information for clinical diagnosis of intraocular and orbital tumors, which is helpful for doctors' planning of surgical operations,clinical education and doctor-patient communication.
4.Expression of Twist and relation with epithelial-mesenchymal transition in oral squamous cell carcinoma.

OBJECTIVEThe objective of this paper was to study the expression of related protein and Twist transcription factor of epithelial-mesenchymal transition in oral squamous cell carcinoma (OSCC) tissue and the correlations of OSCC and oral squamous cell carcino-metastasis. The paper also investigated the clinical significance of expression on OSCC.
METHODSThe labels of epithelium materialization (E-cadherin and cytokeratin), stromal labels (N-cadherin), transcription factor Twist protein, and mRNA expression in 30 OSCC tissues were investigated via immunohistochemistry and in situ hybridization. The paper also conducted contrast analysis with clinicopathology.
RESULTSImmunization result showed that the expressions of Twist and N-cadherin in the OSCC group were more significant than those of the normal group (P<0.05). The expressions of E-cadherin and keratin in OSCC were significantly lower than those of the normal group (P<0.05). In the moderate- and low-differentiated group of OSCC, the expressions of Twist and N-cadherin were higher than those of the high-differentiated group (P<0.05). The expressions of E-cadherin and keratin were lower than those in the high-differentiated group (P<0.05). In the lymphatic metastasis group, the expressions of Twist and N-cadherin were higher than those of no-lymphatic metastasis group (P<0.05). The expressions of E-cadherin and keratin were lower than those of the no-lymphatic metastasis group (P< 0.05). Results of in situ hybridization showed that the expression of Twist mRNA in the moderate- and low-differentiated groups of OSCC, T3, and T4 groups as well as that of the lymphatic metastasis group were higher than those of the high-differentiated, T1 and T2 groups, and no-separate lymphatic metastasis group, and the differences were statistically significant (P<0.05).
CONCLUSIONEpithelium materialization exists in OSCC tissue. Twist can enhance the invasiveness of tumor cell and promote the infiltration and metastasis of OSCC. The combined detection of Twist, E-cadherin, and N-cadherin expressions can effectively predict and estimate OSCC metastasis.

5.Retroperitoneal laparoscopic simple nephrectomy: Reports of 6 cases

Objective To discuss the clinical value of retroperitoneal laparoscopic simple nephrectomy. Methods Retroperitoneal laparoscopic simple nephrectomy was performed in 6 patients, including 5 cases of severe hydronephrosis with non-functioning kidney resulted from ureteral calculi and 1 case of renal tuberculosis. The operation was conducted via retroperitoneal approach. After the upper ureter and the renal pedicle were exposed, the renal artery and vein were clipped and severed. The renal pedicle was occluded only with titanium clips before the removal of the kidney. Results All the operations were performed successfully without complications. The operation time was 130~220 min (mean, 150 min) and the intraoperative blood loss, 80~150 ml (mean, 120 ml). The postoperative hospital stay ranged 5~7 days. Follow-ups for 3 months in the 5 patients with ureterolithiasis revealed normal renal functions. The patient with renal tuberculosis was given anti-tuberculosis therapy and followed for 6 months. No recurrence was seen and the patient’s serum creatinine level was 120 ?mol/L, which was slightly higher than the normal. Conclusions Retroperitoneal laparoscopic simple nephrectomy has advantages of minimal invasion, short hospital stay and rapid recovery. The procedure should be regarded as the “golden standard” for simple nephrectomy.

Objective To identify the metabolites of loganin in rat urine through using ultra high performance liquid chromatography combined with triple TOF mass spectrometry (UPLC-Q-TOF/MS/MS) after oral administration of loganin;To summarize the metabolic pathway of loganin. Methods Rats were given a gavage with loganin. The urine samples were prepared by use of solid phase extraction (SPE). The chromatographic separation was performed on an UPLC C18 column with gradient elution program of acetonitrile and 0.1% formic acid aqueous as mobile phase. Dynamic background subtraction (DBS) technology was employed to trigger information dependent acquisition (IDA) to obtain the characteristic fragment ions of metabolites. Results MetabolitePilot software including several data processing methods were employed to speculate the structures of liganin metabolites and three metabolites were identified. Then, the metabolic pathway of loganin in rats was summarized. Conclusion The analytical method was simple, credible and highly-sensible, which is useful for screening and identifying trace constituents in vivo.
