2.Study of chromosome aberrations OH the workers occupationally exposed to thorium and rare earth mixed dust

Objective To study the effect of thorium and rare earth mixed dust on chromosome aberrations in the lymphocytes of occupational exposed worker8.Methods Analyses of unstable chromosome aberrations on 53 occupational exposed worker8 and 58 control workers were carried out by the conventional Giemsa staining method.Fluorescence in situ hybridization method Wills performed to analyze the chromosome stable aberrations on 10 occupational exposed workers and 10 control workers.Results The frequencies of chromosomal aberration cells,dicentfics plus rings,total aberrations in exposed workers were significantly higher than those in controls.No significant difference was found in the frequency of acentric aberrations between exposed and non-exposed workers.No significant difference was found in the frequency of translocations between exposed and non-exposed workers.Conclusions Chronically occupational exposure to thorium and rare earthmixed dust can increase the induction of unstable chromosome aberration,but the increase of stable chromosome aberrations(translocation)can not be observed.
3.Infrastructure and contents of clinical data management plan.

Establishment of quality management system (QMS) plays a critical role in the clinical data management (CDM). The objectives of CDM are to ensure the quality and integrity of the trial data. Thus, every stage or element that may impact the quality outcomes of clinical studies should be in the controlled manner, which is referred to the full life cycle of CDM associated with the data collection, handling and statistical analysis of trial data. Based on the QMS, this paper provides consensus on how to develop a compliant clinical data management plan (CDMP). According to the essential requirements of the CDM, the CDMP should encompass each process of data collection, data capture and cleaning, medical coding, data verification and reconciliation, database monitoring and management, external data transmission and integration, data documentation and data quality assurance and so on. Creating and following up data management plan in each designed data management steps, dynamically record systems used, actions taken, parties involved will build and confirm regulated data management processes, standard operational procedures and effective quality metrics in all data management activities. CDMP is one of most important data management documents that is the solid foundation for clinical data quality.
4.Resistance reversal effect of a novel taxane compound NPB304 and its collaboration with verapamil.

The tumor multidrug resistance reversal effect of NPB304, a novel taxane, was studied. MTT assay was used to determine the IC50 of chemotherapy drugs. Western blotting assay was applied to analyze the expression of P-glycoprotein (P-gp). The effect of compounds on the P-gp function and P-gp ATPase activity was determined by rhodamine 123 (Rh123) accumulation assay and analysis kit, respectively. Molecular docking was employed to predict the binding force between compounds and P-gp. Transmembrane transport of NPB304 was analyzed using MDCK II and MDR1-MDCK II cell model. NPB304 displayed multidrug resistance reversal effect on KBV cells and MCF-7/paclitaxel cells, NPB304 collaborative with P-glycoprotein (P-gp) inhibitors verapamil enhanced the reversal activity, specifically, 10 μmol x L(-1) verapamil in combination with paclitaxel reversed resistance by 56.5-fold, while combined with NPB304 increased the reversal fold; NPB304 synergistically increased Rh123 accumulation in the resistant cells when combined with verapamil, and NPB304 at 0-1 μmol x L(-1) enhanced the ATPase activity activated by verapamil was observed. NPB304 existed the hydrophobic interactions with the TM regions of P-gp, and the binding force between NPB304 and the A chain of the TM region was stronger. P-gp ATPase activity assay demonstrated NPB304 at lower concentrations (0-1.5 μmol x L(-1)) could activate the P-gp ATPase, playing a role on inhibition of P-gp function. However, NPB304 did not have an obvious feature of P-gp substrate. NPB304 exerted itself and synergy with verapamil activity on reversing tumor resistance via inhibiting the P-gp function.

6.Effects of low-dose interleukin-2 on peripheral regulatory T cells counts of patients with psoriatic arthritis and its short-term clinical efficacy

Objective To study the expression of peripheral blood lymphocyte subsets in psoriatic arthritis (PsA) patients and the short-term efficacy of low doses of interleukin-2 (IL-2). Methods Ninety-five patients with PsA were enrolled as study subjects and 106 healthy people as control group. On the basis of conventional treatment, a total of 22 PsA patients were randomly selected and treated with low dose IL-2 (5 ×105 U/d, continuously used for 5 days, IH), and the disease condition and lymphocyte changes were observed. Flow cytometry was used to detect the absolute count of T subsets dominated by regulatory T cell(Treg) and T helper cell 17(Th17). Wilcoxon rank sum test, χ2 test and Spearman correlation analysis were used for statistical analysis. Results The absolute number of Th17 cells of PsA patients [7.2 (4.0, 12.8) cells/μl] was higher than that of the control group [5.9(4.0, 8.6) cells/μl] (Z=-1.997, P=0.046), the number of Treg cells [25 (17, 36) cells/μl] decreased compared with the control group [30 (23, 40) cells/μl] (Z=-2.957, Z=0.003), T/Treg [50 (36), 76)], B/Treg [7.6 (5.4, 11.5)], CD4+T/Treg [27 (21, 42)], CD8+T/Treg [20 (12, 30)], Th17/Treg [0.34(0.13, 0.51)], Th1/Treg [4.4(2.3, 7.2)], Th2/Treg [0.34(0.20, 0.53)], compared with control group T/Treg [40 (32, 54)], B/Treg [6.5 (4.2, 8.1)], CD4+T/Treg [20 (17, 25)], CD8+T/Treg [16 (11, 24)], Th17/Treg [0.19 (0.13, 0.31)], Th1/Treg [3.5 (1.8, 5.8)], Th2/Treg [0.24 (0.15, 0.39)] were significantly increased (Z=-3.365, -3.217,-5.285, -2.097, -1.69, -1.482, -2.304, P<0.05). Treg cells were negatively correlated with disease activity indexes TJC (r=-0.213, P=0.038), VAS (r=-0.299, P=0.003), PHGA (r=-0.287, P=0.005), DLQI (r=-0.208, P=0.043). Th17 cells increased from [6.3 (2.3, 11.5) cells/μl] to [11 (7, 20) cells/μl, Z=-2.808, P=0.005] after low-dose IL-2 treatment, Treg cells increased from [27 (15, 30) cells/μl] to [71 (37, 98) cells/μl, Z=-3.945, P<0.01]. Because the growth rate of Treg was much higher than Th17, Th17/Treg [before IL-2 treatment: 0.26 (0.11, 0.44), after IL-2 treatment: 0.14 (0.1, 0.35)] returned back to the normal range. After the treatment with IL-2, the patient's activity indicators were significantly improved, and there were no reversible adverse reactions. Conclusion The reduction of Treg cells may be involved in the occurrence and devel-opment of PsA. Low-dose IL-2 treatment can effectively promote the proliferation of Treg cells and the recovery of Th17/Treg balance, which is conducive to the improvement of the condition and good safety.
7.Pathologic changes, mechanisms and diagnosis in renal bone disease

Objective To investigate the incidence rate, pathologic changes, mechanisms and diagnostic methods in renal bone disease.Methods The blood levels of carboxyterminal parpthyriod hormone (C-PTH), 1,25(OH)2D3, calcium and phosphate, aluminum in serum and bone tissue were measured. The bone biopsy and bone scan with 99m technetium methylene diphosphonate (99m TC-MDP) were performed in 51 uremic patients.Results One hundred per cent of the patients had varying degree of pathologic changes in bone, in which 50.9% of the patients presented high-turnover bone disease, 7.8% of the patients presented lowe-turnover bone disease and 41.8% of the patients had mixed-type bone disease. The levels of serum C-PTH were predominently high in high-turnover bone disease while the levels of serum 1,25(OH)2D3 were significantly decreased in low-turnover bone disease. There was a high positive rate for the diagnosis of renal bone disease by bone scan with 99m TC-MDP.Conclusions The examination of bone pathology is the most valuable method for the diagnosis of renal bone disease. Bone scan with 99m TC-MDP has reference value when clinical conditions do not allow to make bone biopsy.

The results show that: the particles falling off the edge of the polypropylene fiber Double-sided liquid filtration membrane are very important and must be considered off the evaluation of particle pollution.

9. Effect of mitogen-activated protein kinase signaling pathway on apoptosis induced by chloroacetic acid in human normal bronchial epithelial 16HBE cells

Objective:
The aim of this study was to investigate the effect of mitogen-activated protein kinase (MAPK) signaling pathway on apoptosis induced by chloroacetic acid in human normal bronchial epithelial 16HBE cells.
Methods:
16HBE cells were exposed to 0.5, 1.0, 1.5, 2.0, 2.5, 3.0 and 3.5 mmol/L chloroacetic acid for 24 h in vitro. The cytotoxicity induced by chloroacetic acid was assessed by CCK-8 and LDH assays. Cell apoptosis was detected by Annexin V-FITC and PI staining. The protein expression levels of phosphorylation of p38, ERK1/2 and JNK were determined by western blotting. 16HBE cells were pretreated with MAPK signaling pathway specific inhibitors including SB203580, U0126 and SP600125 for 1 h, and these cells were subsequently treated with 2.5 mmol/L chloroacetic acid for 24 h. The expressions of p-p38, p-ERK1/2 and p-JNK as well as the changes of cell viability and apoptosis were measured after pretreated with inhibitors for 1 h.

10.Correlation between sensory characteristics and behavioral problems in children with autism spectrum disorder

Objective:
To understand sensory characteristics and its correlation with behavioral problems among children with autism spectrum disorder (ASD), so as to provide reference basis for early diagnosis and comprehensive intervention of ASD children.
Methods:
Using cross sectional survey, 193 ASD children trained by Nantong rehabilitation institutions were recruited from September 2021 to February 2022. All children were investigated with the Short Sensory Profile (SSP) and Parent Symptom Questionnaire (PSQ).
Results:
Among the 193 ASD children, 106 (54.9%) children had abnormal sensory characteristics, including 45 (23.3%) with possible abnormalities and 61 (31.6%) with obvious abnormalities. The abnormal rate of auditory filtering was the highest in the seven dimensions, reaching 85.0%, followed by low strength/weakness, motor sensitivity, taste/smell sensitivity, low response/seeking sensation, vision/hearing sensitivity and tactile sensitivity. The total incidence of sensory abnormalities was 93.3%. There were significant differences in six dimensions of PSQ scale scores of ASD children with different degrees of sensory abnormalities, including conduct problems, learning problems, psychosomatic disorders, impulsive hyperactivity, anxiety and hyperactivity index ( H =38.89, 38.90, 19.53, 46.34, 34.54, 54.90, P <0.01). With the aggravation of sensory abnormalities in children with ASD, the scores of all dimensions of the PSQ scale increased significantly. The total score of SSP was negatively correlated with the six dimensions of conduct problems, learning problems, psychosomatic disorders, impulsive hyperactivity, anxiety and hyperactivity index in PSQ ( r =-0.53, -0.50, -0.32, -0.55, -0.43, -0.61, P <0.01).
Conclusion
ASD children generally have sensory abnormalities, which coincide with severity of behavioral problems. There is a positive correlation between sensory abnormalities and behavioral problems in ASD children. More attention should be paid to the abnormal sensory characteristics of children with ASD in the future diagnosis and treatment process.
