Objective To explore the effects of finasteride therapy on the expressions of heat shock protein 70(HSP70)mRNA,heat shock protein 22 (HSP22)mRNA,tumor necrosis factor-alpha (TNF-a),interleukins-1β (IL-1β) in elderly patients with benign prostatic hyperplasia (BPH).Methods Totally 68 elderly men patients with BPH were divided into two groups:regular treatment group (n=34) and finasteride treatment group(n=34),30 elderly men patients without BPH were as control group.The blood samples of patients after treatment were collected for the detections of HSP70mRNA and HSP22mRNA with RT-PCR,meanwhile,the levels of TNF-α,IL-1β were measured by radioimmunoassay (RIA).Effect of treatment was evaluated in accordance with International Prostate Symptom Score (IPSS).Results The expressions of HSP70 mRNA and HSP22 mRNA,TNF-α,IL-1β in BPH patients were higher in finasteride group than in control group (P＜0.01).After 12 weeks therapy,the levels of HSP70mRNA,HSP22mRNA,TNF-α and IL-1β were decreased in finasteride group (P＜0.01 or P＜0.05) but no significantly difference in regular treatment group.Compared with regular treatment group,the expressions of HSP70 mRNA,HSP22mRNA and TNF-α were reduced in the finasteride group after treatment (P＜0.01 or P＜0.05),IPSS was more improved (P＜0.05),but IL-1β was not distingished between two groups (P＞0.05).IPSS was positively related to the levels of HSP70 mRNA,HSP22 mRNA,TNF-α and IL1β (r=0.4251,0.4976,0.4562,0.3627,P＜0.01 or P＜0.05).Conclusions The occurrence and development of BPH are closely related with immunoinflammatory factors.Finasteride not only affects the levels of dihydrotestosterone but also can inhibit the expression of immunoinflammatory factors to improve symptom by reducing the volume of benign prostatic hyperplasia.

Objective:To evaluate the efficacy of computed tomography (CT)-guided implantation of radioactive ~(125)I seeds in the treatment of residual foci of nasopharyngeal carcinoma after radiotherapy.Methods:Twenty-five nasopharyngeal carcinoma patients who had residual foci after radiotherapy were implanted radioactive ~(125)I seeds from January 2007 to January 2008 in our hospital. Three-dimensional treatment planning system (TPS) was used to calculate the quantity and total dosage of ~(125)I seeds. The radioactive ~(125)I seeds were implanted into residual foci under CT guidance. The dosage distribution of radioactive ~(125)I seeds were verified after surgery. The nasopharyngeal endoscopy CT scans were performed at 3, 6, and 12 months after surgery and the short-term efficacy and adverse reaction were evaluated. Results:Out of the 25 patients, 20 patients had complete response (CR), 2 patients had partial response (PR), 2 patients had no change (NC), and 1 patient had progressive diseases (PD). The overall effective rate (CR+PR) was 88.0%. All patients had no serious postoperative complications. Conclusion:CT-guided implantation of radioactive ~(125)I seed was an effective and safe method in the treatment of the residual foci of nasopharyngeal carcinoma after radiotherapy.

Objective Establishment and development of a novel Single-Nucleotide-Polymorphism TaqMan Real-Time PCR assay for rapid detection of rs12979860 that predicts HCV therapy response.Methods Human genomic DNA were extracted from solid tissues,secretion and plasma before allelic discrimination.With the property of minor groove binding protein (MGB) binding to minor groove of DNA with strong specificity and affinity,primers and MGB probes were particularly designed for differentiation of human genomic frequencies.MGB probe-based real-time PCR was established to increase allelic discrimination using two probes that only differ in one nucleotide of IL28B rs12979860.The specificity was evaluated by fluorescence signal emissions which were selected from two signal channels.And DNA sequencing was used to confirm the genomic polymorphisms.Results TaqMan probe-based SNP real-time PCR increased allelic discrimination using two probes that only differ by one nucleotide of amplicon,which indicated this assay was easily performed regardless of genomic DNA concentration and quality,minimizes sources of error.The sensitivity was as low as 1.5 ng/μl,the amplification efficacy was 97.6％.The genotype frequencies of CC,CT were remarkably different between Caucasian and Mongolian.The dominated genotype of Caucasian was CT,while most Mongolian was carried CC (26/40 vs.40/50,x-2 =18.75,P value ＜ 0.05).However,the genotype between two population showed no relationship with their virological clearance clinically (P value ＞ 0.05).Conclusions This TaqMan MGB assay shows highly specificity,which has potential as a routine diagnostic test for the detection of rs12979860 from various types of samples.This robust assay would be widely used clinically to predict patients' response before anti-HCV treatment in personalized medicine.

Objective: To study status of social support and its influencing factors in female migrant workers. Methods: Study participants were selected with a random cluster design from participants 16-60 years old in non-registered residents from 10 districts in one city. Social support revalued scale and general questionnaire were used to survey social support status of female migrant workers in August-December of 2016. Results: There was statistic difference in social support among different districts, occupations, and income of female migrant workers (P<0.05) . Results of multiple linear regression demonstrates that districts, occupations, and income of female migrant workers were associated with social support scale (district standarized β=0.03, occupations standarized β=-0.03, incorue standarized β=0.10, P<0.05) . Conclusion We should be attentive to self-employed and Low-income participants among female migrant workers.

Objective To investigate the effects of conductive education with routine rehabilitation on cerebral palsy. Methods 59 caseswere divided into the conductive group (n=38) and the control group (n=21). The conductive group received conductive education with routinerehabilitation and the control group received routine rehabilitation only. They were assessed with the gross motor function, intelligentand activity of daily living (ADL) before and after treatment. Results The improvement of gross motor was not significantly different betweenthese groups (P>0.05), while the intelligence, ADL improved more in the conductive group than in the control group (P=0.001). ConclusionConductive education can promote the development of the children in ADL and intelligent.

Objective To purify native and recombinant heparin-binding hemagglutinin(HBHA)protein,and investigate the activity of HBHA polyclonal antibody against aggregation of Bacillus CalmetteGuerin(BCG)induced by HBHA.Methods After growing BCG to the stationary phase in the 7H9 liquid medium,the native HBHA protein(nHBHA)was obtained by CL-6B column chromatography.At the same time,the HBHA gene fragment was cloned and expressed by transforming Escherichia coli BL-21.Then the polyclonal antibody against rHBHA was prepared by immunizing rabbit.Different comcentration of the HBHA protein was added to the BCG liquid medium,and the aggregation of the BCG was observed.Then,add the HBHA protein that incubated with anti-HBHA antibodies to the BCG culture medium and observe the aggregation of BCG.Results The purity of native HBHA was 99% and the concentration was 1.016 mg/ml.The expressed product contained 36% of total somtic protein.After purified,the purity of the recombinant HBHA protein was 97.1% and the concentration was 10.98 mg/ml.Both the rHBHA and nHBHA could induce the aggregation of BCG.When then concentration of nHBHA is 0.2μg/ml,BCG could be induced to aggregate,while the rHBHA concentration is 2μg/ml could induce the aggregation.Both aggregations could be suppressed by the polyclonal antibody against rHBHA.Conclusions The native and recombinant HBHA are successfully obtained.It is proved that the rHBHA could induce the aggregation of BCG similar as nHBHA,and polyclonal antibody against rHBHA could also suppress the activity of nHBHA.It suggested that rHBHA could be further used in clinical diagnosis and vaccination.

Advanced schistosomiasis is the most serious clinical type of schistosomiasis. Its diagnosis and treatment are relat?ed to many special departments,such as gastroenterology,general surgery,neurology,endocrinology,radiology,traditional Chinese medicine,blood purification,endoscopy,intervention,and ICU. It is necessary to apply a multidisciplinary treatment (MDT)mode. However,the mode has no universal standard and guide in practice. It is very important for the implementation of MDT mode of advanced schistosomiasis to form a treatment expert team,formulate the formal working procedures,and standard?ize the treatment schedules. The standardized implementation of MDT mode will be important to provide a more effective clinical decision on advanced schistosomiasis.

Objective: To investigate the mental health status of female migrant workers in a city. Methods: From August to December, 2016, cluster random sampling was used to select female migrant workers aged 16-60 years from 10 districts of this city, and Symptom Checklist-90 (SCL-90) and a questionnaire on general status were used. Results: Among these female migrant workers, the detection rates of obsessive-compulsion, hostility, and depression were 6.62%, 4.18%, and 4.10%, respectively. There was a significant difference in the detection rate of mental health issues between the female migrant workers from different districts, with different occupations, or with different education levels (P<0.05) . Districts, occupation, and education level were associated with mental health issues, after adjustment for age (P<0.05) . Conclusion Although female migrant workers in this city have a high mental health level, the issues such as obsessive-compulsion, hostility, and depression should be taken seriously. Targeted mental health surveys and comprehensive interventions should be implemented to improve the mental health level of female migrant workers.
