1.Identification of the related substances in fasudil hydrochloride with hyphenated techniques.

The study aims to identify the related substances in fasudil hydrochloride by hyphenated techniques. A WondaSil C18 (250 mm x 4.6 mm, 5 microm) column was used for the separation of the related substances with a mixture of methanol and ammonium acetate buffer solution as the mobile phase by gradient elution. The structures of the related substances were speculated by electrospray positive ionization LC-TOF/MS accurate ion mass and MS/MS determination and elucidation, and verified further through synthesis and spectroscopic analysis. Fasudil hydrochloride and the related substances were separated under the established HPLC condition. Three related substances in fasudil hydrochloride were characterized by hyphenated techniques. The hyphenated LC-MS method is useful for the identification of related substances in fasudil hydrochloride and the results obtained are valuable for its manufacturing process and quality control.
2.The influence to present depth doses and peripheral doses by enhanced dynamic wedge and physical wedge

Purpose: To understand the influence to depth doses and peripheral doses by Enhanced Dynamic Wedge and physical wedge.Methods: The depth doses and peripheral doses were measurement for open field and Enhanced Dynamic Wedge field as well as physical wedge.Results: ⑴ depth doses and peripheral doses for Enhanced Dynamic Wedge fields was very similar to those for open fields ⑵ depth doses and peripheral doses for physical wedge fields was large than those for open fields. Conclusion: depth doses measured for open fields can also be used for Enhanced Dynamic Wedge fields dose calculation. Wedge depth dose must be used in the physical wedge fields dose calculation. It should be pay attend to peripheral doses for physical wedge fields in the clinic.
3.Effect of team-based tutorial system to nursing student in the order cultivation pattern

Objective To explore the effect of team-based tutorial system to nursing student in the order cultivation Pattern.Methods This study used the convenience sampling method.109 students were choosed from orientation training classes that 2012 and 2013 grade as the research subjects.The 59 students from 2013 grade as the test group,implement team-based tutorial system to them in the fourth semester.The 50 students from 2012 grade that had been implemented general management as the control group,then compare the fourth semester final grade,classes atmosphere,the evaluation of patients and clinical teachers over the same period.Results The test group and control group students' scores of their fourth semester final grade,classes atmosphere,the evaluation of clinical teachers and patients were (92.39±2.39),(3.45±0.64),(95.33±2.01),(94.75±2.38) points and (90.91±2.62),(3.14±0.63),(91.00±1.82),(90.63±2.34) points.There was difference between them,and the difference was statistically significant (t=-7.82 ~ 3.06,P < 0.05).Conclusions The implementation of team-based tutorial system to nursing student in the order cultivation pattern can improve students' academic performance,improve their class atmosphere and their communication skills so that patients get good evaluation of patients and clinical teachers.
4.Reliability and validity of Chinese version of the Client-Centred Care Questionnaire

Objective To translate the English version of the Client-Centred Care Questionnaire (CCCQ) into Chinese and to test the reliability and validity of the Chinese version of CCCQ.Methods The English version of CCCQ was translated into Chinese by the method of double translation-translation.Totally 320 chronic patients were recruited and they were investigated by the Chinese version of CCCQ.Results The Cronbach coefficient of the Chinese version of CCCQ was 0.925,P<0.01.The retest reliability was 0.807,P<0.01.The split-half reliability with r=0.883,P<0.01.The content validity index (CVI) was 0.956,and every purpose CVI values were above 0.8.Factor analysis got one factor,which explained 52.549% of the total variance,and each objective factor loading was above 0.4.Conclusions The Chinese version of CCCQ has been proved to be reliable and valid.It can be used as a valid tool for the chronic patients to measure their perceptions of the quality of nursing care.

6.The aetiology and treatment of 48 cases with neonatal hyponatremia

Objective To study the common aetiology and the principle of treatment of neonatal hyponatremia.MethodsThe clinical data of 48 neonatal hyponatremia cases,admitted to our hospital from June 2006 to January 2010,were analyzed retrospectively. ResultsThe main manifestations of neonatal hyponatremia were poor response ( 37.5% ) and anorexia ( 31.3% ).The common causes included anoxic diseases during perinatal period,improper feeding,effects of specific diseases,premature birth et al.After active treatment,the serum sodium returned to normal in 47 neonate within three days,except for 1 premature infants died of serious respiratory distress syndrome.Conclusion Neonatal hyponatremia shows no special clinical manifestation.The aetiologies are complicated,which should be treated in different methods correspondingly.The inquiry of feeding history,the treatment of primary illness,the early detection of serum sodium levels and timely correction of the serum sodium levels are very important to improve the successful rate and reduce sequela.
7.Experimental study on the accuracy of output factor calculation for rectangular inserts of electron beams

Objective To compare the accuracy of output factor calculation for rectangular inserts of electron beams by using three kinds of algorisms: square root, one dimensional formula and area periphery ration. Methods The differences between the values of output factor calculated by these three algorithms mentioned above and the results measured by ionization chamber for seven rectangular inserts were compared. Results When beam energy was fixed, the difference between calculated and measured output factors was larger for area periphery ration algorithm than that of the square root or one dimensional formula algorithms. The electron cone size, beam energy and rectangular inserts size all had some effect on the value of output factor calculated by the three algorithms. Conclusions The area periphery ration algorithm is not recommended for clinical use because of its great deviation. One dimensional formula and square root algorisms possess a good accuracy. One dimensional formula is superior to square root algorithm, but square root algorithm is more practical for clinical use.

BACKGROUND: Vascular endothelial growth factor receptor 2 (VEGFR2) is primarily involved in vascular endothelial growth factor (VEGF)-mediated signal transduction and plays a critical role in the pathological angiogenesis that occurs in a number of diseases, including leukemia. Besides, VEGF secreted by leukemia cells also induces its own expression which leads to an enhanced production of VEGFR2 which contributes to the survival and proliferation of leukemia cells.OBJECTrVE: To evaluate the inhibitive effect of Lenti6/shVEGFR2 on the VEGFR2 expression and leukemia growth in mouse.DESIGN: A randomized, parallelized, controlled and open trial.SETTING: Department of Pediatrics, the Second Affiliated Hospital of Sun Yat-sen University; Biotechnology Research Center, Sun Yat-sen University.MATERIALS: The experiment had been done in the laboratories for Medical Research Center of the Second Affiliated Hospital, Sun Yat-sen University and Biotechnology Research Center, Sun Yat-sen University from May 2004 to January Lentiviral RNAi Expression System was purchased from Invitrogen, Co.,Ltd.; human VEGFR2 Mcb (PE) was purchased from R&D; CD31 immunohistochemistry kit was purchased from Boster, Co.,Ltd.; CD33-PE fluorescence labeled antibody was purchased from BD, Co.,Ltd.transiently and expression clone (Lenti6/shVEGFR2) was constructed, then cotransfected with ViraPowerTM Packaging Mix pU6/shVEGFR2 entry clone and transducting with Lenti6/shVEGFR2 expression clone, the effect on the development of intravenous xenograft leukemia mouse model, the distribution of microvessels in mouse bone marrow was observed after leukemia model mouse injected with recombinant lentivirus (group B); leukemia model mouse injected with recombinant lentivirus and endothelial cell (group C); leukemia model mouse injected with endothelial cell (group D). Through detecting changes of CD33 positive cells and microvessel density (MVD) in bone marrow, observing peripheral blood cell (PBC)smear and slice of liver, spleen, the effect of Lenti6/shVEGFR2 recombinant lentivirus on mouse leukemia was evaluated.mediated with lentivirus on VEGFNEGFR2 paracrine and autocrine loops in leukemia mouse.effective in inhibiting HL60 cell. pU6/shVEGFR2 entry clone constructed according to it had cell inhibitory rate as high as after transfection of pU6/shVEGFR2 entry clone and transduction of Lenti6/shVEGFR2 expression clone: 48 hours after transfection of pU6/shVEGFR2 entry clone and transduction of Lenti6/shVEGFR2 expression clone, the cell growth inhibitive rates were similar. However, the cell growth inhibitive rate of entry clone descended rapidly after 48 hours (P<0.01); which of expression clone changed slowly, reaching the peak at 96 hours, dropped slightly, having no significance mouse: The amount of HL60 cells in bone marrow of groups A, B and C detected with flow cytometry were (25.8%±4.9)%, (14.3%±5.1)%, (8.4±2.6)%, respectively (P<0.05); MVD in group C was obviously less than that in group D (P<0.05); The amount of HL60 cells in leukemia model mouse injected with recombinant lentivirus and endothelial cell was the lowest as compared with the other groups.
10.Limb ischemic preconditioning protects human umbilical vein endothelial cells from oxidative stress induced by H2 O2

AIM: To investigate the effects of the sera from the rats after limb ischemic preconditioning (LIPC) on human umbilical vein endothelial cells (HUVECs) injured by hydrogen peroxide (H2O2).METHODS:The HUVECs were divided into 5 groups:the cells in control group were cultured without any intervention;the cells in model group (M) were damaged by 1 mmol/L H2O2 for 2 h;the cells in early preconditioning serum (EPS) group, delayed pre-conditioning serum (DPS) group or sham limb ischemic preconditioning serum (SPS) group were treated with the corre-sponding serum at 5%for 12 h, respectively , and then treaed with H 2 O2 for 2 h.The viability of the HUVECs was ana-lyzed by flow cytometry .The lactate dehydrogenase ( LDH) in the culture media was detected .The cell adhesion molecules in the HUVECs were detected by real-time PCR.The mRNA and protein expression of heme oxygenase-1 (HO-1) was also determined .RESULTS:The viability of HUVECs incubated with 1 mmol/L H2 O2 for 2 h significantly decreased compared with the control cells , which was accompanied with the augmentations of LDH in the medium and the cell adhesion mole -cules in cells , such as vascular cell adhesion molecule-1 ( VCAM-1 ) and intercellular adhesion molecule-1 ( ICAM-1 ) . Preincubation with EPS and DPS derived from the rats subjected LIPC attenuated these injuries .Furthermore, pretreatment with EPS and DPS increased the expression of HO-1 at mRNA and protein levels .CONCLUSION:LIPC protects the HU-VECs from H2O2-induced injury.
