1.Research Progress on Antiviral Activity of Interferon-induced Transmembrane Proteins.

Interferon-induced Transmembrane Proteins (IFITMs) were identified through small interference RNA (siRNA) screening method in 1980s. The antiviral properties of the IFITMs were firstly discovered in 1996. Recently, its antiviral effect and mechanism have become a research hotspot. Many studies have shown that IFITM can inhibit the replication of multiple pathogenic viruses, including influenza A virus (IAV), Human Immunodeficiency Virus (HIV-1), hepatitis C virus (HCV), Ebola virus (EBOV), West Nile virus and so on. IFITMs inhibit the replication of virus in the early stage of the viral life cycle, which occurred before the release of viral genomes into the cytosol. Recent studies indicate that IFITM proteins could block viral replication by mediate viral membrane fusion. However, the mechanism is still under investigation. Here we review the discovery and characterization of the IFITM proteins, elucidate their antiviral activities and the potential mechanisms.

2.PREPARATION AND PRACTICAL APPLICATION OF MONOCLONAL ANTI-A ANTIDODIES

Balb/c mice were immunized by intraperitoneal iniection of group A erythrocytes. Immunized spleen cells were fused with Sp2/0 murine myeloma cells from the solid tumour. After 10~15 days of incubation at 37℃ CO_2, the supernatants were screened for the agglutinabal antibodies. Three hybridoma cell lines secreting high titer and specific monoclonal anti-A antibodies were established.These hybridoma cells have the ability of constant seceting antibodies which belong to the IgM class. The specificity monocloal anti-A sera was the same as the human anti-A sera.
3.Effects and mechanisms of bevacizumab-induced renal impairment

Bevacizumab has been widely used in tumor targeting therapy,while the most common adverse reaction is renal impairment,manifested as proteinuria. The main mechanisms may include interfering podocytes-endothelial vascular endothelial growth factor(VEGF)axis signals,increasing glomerular pressure caused by secondary hypertension,subacute renal thrombotic microangiopathy caused by endothelial damage and so on. Thrombotic microangiopathy is the main renal pathological type,and other rare types include glomer-ular lesions,renal interstitial disease,and benign renal arteriolar nephrosclerosis. Therefore,urine protein excretion and renal function should be closely monitored during bevacizumab treatment period for timely treat-ment,dose reduction or even withdrawal if necessary to ensure renal function.
4.A cross-sectional study on AIDS knowledge,attitude and behavior in medical students

Objective To understand the medical college students' knowledge,attitude and behavior of AIDS,and provide the basis for health education programmes.Methods 2 130 school students were recruited by cluster sampling method in one medical college,including 717 freshman,533 sophomore and 880 junior.Questionnaire was used to investigate AIDS-related knowledge,attitudes and behaviors to all of the 2 130 students.Results The correct answer rate was 80.72%,which was positively correlated with the grade,but the knowledge of non-AIDS transmission and detection techniques was still not very clear,such as mosquito bites,sharing towels and clothing with AIDS patients could spread the AIDS or not,the correct answer rate was just 36.7% and 54.3%.There was a certain attitude of discrimination in medical students on AIDS,79.9% of medical students were unwilling to shopping at the place of AIDS patients,and 76.4% were unwilling to have dinner with AIDS patients.Compared with the freshmen,high-grade students have higher knowledge level on HIV/AIDS,but their attitude to AIDS patients was poor.Conclusions Presently,medical students' recognizing on HIV/AIDS still have a long distance from the professional requirements,suggesting that AIDS health education in medical college students is necessary.
5.Building and use of the appraisal indicators system of hospitals brand effect

Literature review of the world on hospital brands, data collection in summary and comparison, and specialist consultancy were used to set the appraisal indicators, while the stratum analysis was used to calculate the weight coefficients of individual indicators, for building the appraisal indicators system for hospital brand effect. In addition, the indicators system and appraisal model were called into play to explore the use of hospital brands, offering scientific references for hospital management.
6.Analysis of diabetes prevalence and its influencing factors among residents in Yongchuan district of Chongqing

Objective To understand diabetes prevalence and its factors is to provide the suggestion for diabetes prevention and control in Yongchuan district .Methods Residents aged above 18 years old from 3 towns and 1 street were sampled by stratified clustered random sampling method .Residents were investigated with the questionnaire and physical examination and taken blood sample to test fasting glucose ,2‐h glucose .The data were analyzed to calculate diabetes prevalence by SPSS17 .0 .The factors of dia‐betes was compared by chi‐square and the independent influencing factors of diabetes were identified by step backward logistic re‐gression .Results A total of 1 202 were investigated .The rate of cognition about diabetes was below 30% .21 .46% of residents smoke every day .31 .78% of them do exercise .The prevalence of overweight ,obesity and diabetes was 24 .63% ,6 .07% and 10 .48% .The result of multi‐variant logistic regression indicated that the independent influencing factors of diabetes prevalence in‐cluded age(OR=1 .41 ,95% CI=1 .24-1 .62) ,the rate of cognition about the dose of salt intake per person per day recommended by Chinese guideline of diet(OR= 2 .12 ,95% CI= 1 .13 -3 .96) ,smoking at present(OR= 1 .44 ,95% CI= 1 .10 -1 .89) ,body weight(OR=1 .06 ,95% CI= 1 .00 -1 .12) ,blood pressure(OR=0 .76 ,95% CI=0 .59 -0 .98) .Conclusion Diabetes prevalence was high in Yongchuan district .The risk factors such as unhealthy diet ,smoking ,physical inactivity were common .Government should take the major role for diabetes prevention and control .
7.Analysis on hypertension prevalence and its influencing factors among residents in Yongchuan district of Chongqing municipality

Objective To understand the hypertension prevalence and its influencing factors in Yongchuan district of Chongqing to provide the suggestion for the prevention and control of hypertension .Methods The permanent residents aged above 18 years old were sampled from 3 towns and 1 street by adopting the stratified clustered random sampling method and performed the questionnaire survey ,physical examination and detection of fasting glucose and postprandial 2‐h glucose .The data were analyzed to calculate the hypertension prevalence by the SPSS16 .0 software .The influencing factors of hypertension were analyzed by chi‐square and the multivariate Logistic regression analysis .Results A total of 1 202 persons were investigated .The survey found that the cognition level on hypertension in the residents was below 30 .00% .21 .46% of residents smoked every day .31 .78% of them did exercise .The proportions of overweight and obesity were 24 .63% and 6 .07% respectively ,the hypertension prevalence rate was 23 .88% .The multivariate Logistic regression analysis results indicated that the independent influencing factors of hypertension prevalence included age (OR=1 .95 ,95% CI:1 .74-2 .17) ,cognition level on hypertension(OR=2 .11 ,95% CI:1 .18-3 .79) ,oil in‐take per person per day(OR=0 .80 ,95% CI:0 .64-0 .99) ,body weight(OR=1 .97 ,95% CI:1 .58-2 .46) and blood glucose(OR=0 .63 ,95% CI:0 .50 -0 .78) .Conclusion The hypertension prevalence among the residents in Yungchuan district is high .The knowledge about the prevention and treatment of hypertension is deficient .The high risk factors of smoking ,drinking ,irrational diet and insufficient body movement are common .The comprehensive prevention and treatment work of hypertension should be practi‐cally strengthened .
8.Effects of various prime-boost regimes on immunities in mice by using DNA and reocmbinant vaccin-ia-based H5N1 vaccines

Objective To develop an effective and broad immune protective vaccination strategy by using DNA and recombinant vaccinia-based H5N1 vaccines.Methods BALB/c mice were immunized with various prime-boost regimens by using different DNA ( pIRES-based or pVRC-based) and recombinant vaccinia (Tiantan strain, rTTV)-based H5N1 vaccines expressing multivalent antigens (HA, NA, M1 and M2).The differences of immunity induced by two DNA vaccines were compared between intradermal electro -poration (IDE) and intramuscular electroporation (IME) deliveries.Immune responses were analyzed by hemagglutination inhibition( HAI) assay, neuraminidase ( NA)-specific antibody measured by ELISA , mi-croneutralization assay and IFN-γELISPOT assay .Results High levels of humoral immunity and T cell re -sponses were induced in mice primed with DNA-based vaccine than those primed with rTTVb-ased vaccine . DNA priming by IDE resulted in higher levels of neutralizing antibody in mice than those by IME delivery . Higher levels of HAI and anti-NA antibodies as well as NA-specific T cell responses were induced by pVRC-based DNA prime than those by pIRES-based DNA prime .HA-specific T cell responses were also enhanced in mice primed with pVRC-based DNA than those primed with pIRES-based DNA by IME .Conclusion The prime-boost strategies by using DNA-based vaccine in combination with rTTV-based H5N1 vaccine could induce humoral and T cell responses in mice against multi-antigens .Immunities induced by vaccines in com-bination might be modulated by various prime regimes .The study provided references for the further develop-ment of novel H5N1 vaccine and the optimization of immunization programs of combined multi-antigen vac-cine candidates .
9.Characterization of immune response in mice induced by recombinant vaccinia virus(Tiantan) based multivalent H5N1 avian influenza vaccines

Objective To develop an effective and broad immune protective H5N1 vaccine.Methods We first developed two recombinant vaccinia ( Tiantan strain) virus ( rTTV ) based H5N1 vaccines, which consisted of bicistron expressing the hemagglutinin(HA) and matrix protein 2(M2), or bicistron expressing the neuraminidase(NA) and matrix protein 1 (M1). The expression of H5N1 protein in rTTVs was confirmed. We immunized the BALB/c mice twice with two kind of dose ( 104 PFU, 107 PFU)using different combination. Subsequently, we assessed the humoral and cellular immune response in vaccinated mice. Results Our data showed that rTTV-based H5N1 vaccine induced rapidly robust HA- and NAspecific antibody level and IFN-γ secreting form cell(SFC) with either single dose of 107 PFU or twice dose of 104 PFU or 107 PFU. We also detected significant neutralizing antibody and matrix-specific immune response. In addition, we found that immunization with two kind of rTTV-based H5N1 vaccines induced much high level of M2-specific antibody than that with single of rTTV-based H5N1 vaccine. Conclusion rTTVbased H5N1 vaccines in this study elicited board array of immunity and our study offers a promising alternative H5N1 vaccine candidates with favorable potential to prevent various H5N1 pandemic.
10.Clinical efficacy and experiences of laparoscopic hepatectomy for segment Ⅶ and Ⅷ liver tumors

Objective To investigate the clinical efficacy and experiences of laparoscopic hepatectomy (LH) for segment Ⅶ and Ⅷ liver tumors.Methods The retrospective cross-sectional study was conducted.The clinicopathological data of 94 patients who underwent LH for segment lⅦ or Ⅷ liver tumors in the Sir Run Run Shaw Hospital Affiliated to Zhejiang University School of Medicine from June 2010 to August 2016 were collected.The operating space for operation was built under laparoscopy.According to liver cirrhosis grading,tumor size,adjacent relationship with major blood vessels and residual liver volume,non-anatomical and anatomical hepatectomies were selected by patients.During the operation,tumors were precisely pinpointed and plane of liver resection was determined,and then proper instruments of liver partition and techniques of hepatic inflow occlusion were selected.Observation indicators:(1) surgical and postoperative recovery situations;(2) postoperative pathological examination;(3) follow-up and survival situation.Follow-up using outpatient examination and telephone interview was performed to detect the patients' survival up to July 2017.Measurement data with normal distribution were represented as (x)±s.Measurement data with skewed distribution were described as M (interquartile range).Survival rate was caculated by the Kaplan-Meier method.Results (1) Surgical and postoperative recovery situations:all 94 patients received successful operations,without perioperative death,including 73undergoing non-anatomical hepatectomy and 21 undergoing anatomical hepatectomy.Fourteen patients had conversion to open surgery and 27 received hepatic inflow occlusion.The median operation time,median volume of intraoperative blood loss and cases with intraoperative blood transfusion were respectively 187.5 minutes (75.0minutes),200 mL (200 mL) and 15.Eighteen patients had postoperative complications,including 6 with pleural effusion,6 with abdominal effusion,1 with wound infection,1 with abdominal infection,1 with venous thrombosis,1 with bleeding,1 with coagulation disorders and 1 with hepatic insufficiency.Clavien-Dindo classification of complications:11,1,5 and 1 patients were detected in grade Ⅰ,lⅡ,Ⅲ and Ⅳ,respectively.All complications were improved by symptomatic treatment.The median duration of hospital stay was 7 days (6 days).(2) Postoperative pathological examination:results of tumor pathological examination showed that 45,5,9 and 35 patients were respectively confirmed as hepatocellular carcinoma,cholangiocarcinoma,metastatic hepatic carcinoma and benign liver tumor.(3) Follow-up and survival situation:59 patients with malignant tumors were followed up for 6.0-52.0 months,with a median time of 42.6 months.Postoperative 1-and 3-year overall survival rates of 59 patients with malignant tumors were 98.3% and 84.7%,respectively.Conclusions LH for segment Ⅶ and Ⅷ liver tumors which is conducted in experienced medical center is safe and feasible,with definite effects.Building operating space for operation under laparoscopy,determining precise positioning of the tumor and plane of liver resection,and selecting proper instruments of liver partition and techniques of hepatic inflow occlusion are the key points of successful operation.
