1.Study of loss of imprinting of insulin-like growth factor Ⅱ and H19 genes in cervical carcinoma

Objective To study the loss of imprinting(LOI) of insulin-like growth factor Ⅱ(IGF2) and H19 genes in cervical carcinomas.Methods Polymerase chain reaction(PCR) method was used on DNA samples extracted from 40 cases of cervical carcinoma and 20 cases of normal control tissues to detect the heterozygosity of IGF2 and H19 genes.The LOI of IGF2 and H19 genes was detected by RT-PCR.Results The heterozygous frequency of IGF2 was 52% in cervical cancer samples.Comparatively,in 20 matched normal samples,13 cases showed the heterozygosity(65%).The biallelic expression of IGF2 was detected in 10 cases among the 21 informative cervical cancer samples(47.6%),however,1 case in 13 heterozygous samples of control group(7.7%)((P0.05).Conclusion LOI and LOH of IGF-2 and H19 genes are involved in cervical carcinomas.LOI of IGF2 and H19 may be involved in the initiation stage of carcinogenesis of cervical carcinoma.
2.Cyclooxygenase-2 inhibitors enhance the antileukemia activity of STI571

AIM: To investigate whether celecoxib,a cyclooxygenase-2(COX-2) inhibitor,potentiates the anti-leukemia activity of STI571 in K562 cells.METHODS: K562 cells were treated with STI571,celecoxib or combination of both at different concentrations in suspension culture.Cell proliferation was documented by MTT assay,and cell apoptosis was determined by flow cytometry and morphology.Meanwhile,RT-PCR was applied to analyze the probable mechanism underlying the effects of the drugs.RESULTS: The combination of STI571 and celecoxib dramatically suppressed the proliferation of K562 cells,in which 0.25 ?mol/L STI571 and 40.0 ?mol/L celecoxib enhanced the inhibiting rate to 76.1%?1.6%.Furthermore,the combining administration of drugs significantly promoted the apoptosis induced by STI571,which showed characteristic changes of morphologic features and increase in sub-G_1 cells.By using RT-PCR technique,the expression of COX-2 had no decline by single administration of celecoxib or STI571.However,a progressive down-regulation was caused by coadministration of two drugs.In contrast with COX-2,the expression of VEGF had no changes at any time.CONCLUSION: The administration of celecoxib alone only inhibits the proliferation of K562 cells.Combination treatment with STI571 and celecoxib promotes the apoptosis induced by STI571.
3.The expression of TM in endometrial carcinoma

Objective To study the expressi on of thrombomodulin (TM) in endometrial carcinoma and explore its relation to the infiltration degree and histological grading of endometrial carcinoma. Methods The expression of TM was detected by immunohistochemistry in 50 cases of endometrial adenocarcinoma, 10 cases of endometrial complex hyper plasia and 10 cases of normal endometrium. Results The posi tive rate of TM in endometrial carcinoma was significantly higher than that in c omplex hyperplasia and normal endometrium (P 0.05). Conclusion TM can be used as a new index for judging progre ssion and prognosis of endometrial carcinoma.
4.Effect of intrathecal administration of a mixture of butorphanol and ketamine on cAMP-PKA-CREB signal transduction pathway in spinal dorsal horn of rats with inflammatory pain

Objective To investigate the effect of intrathecal administration of a mixture of butorphanol and ketamine on cAMP-PKA-CREB signal transductian pathway in the spinal dorsal ham of the rats with inflammatory pain. Methods Twenty-four male SD rats, weighing 240-280 g,in which intrathecal catheters were successfully placed, were divided into 4 groups randomly (n = 6 each): inflammatory pain group (group IP), butorphanol group (group B), ketamine group (group K), and butorphanol + ketamine group (group BK). The inflammatory pain was induced by injection of 5% formalin 50 μl into the plantar surface of left hind paw. Normal saline 10 μl, butorphannl 12.5 μg, ketamine 50 μg, and a mixture of butorphanol 12.5 μg and ketamine 50 μg was injected intrathecally 30 min before subcutaneous injection of formalin in group IP, B, K and BK respectively.Pain intensity score (PIS) was used to assess pain behavior every 5 min within an hour after subcutaneous injection of formalin. The animals were killed at 2 h after subcutaneous injection of formalin, and the L5 segment of the spinal cord was removed for determination of protein kinase A (PKA) and phosphorylated cAMP response element binding protein (p-CREB) expression using immunohistochemistry. Results Fonnahn administration induced pain behaviour expressed as two phases. PIS scores, PKA and p-CBEB expression, and staining scores were significantly lower during the fast and second phases in group BK than in group IP (P < 0.05 or 0.01), while no significant differences were found in the indices mentioned above between group B and IP and between group K and IP (P>0.05). Conclusion lntrathecal injection of a mixture of butorphanol and ketamine can reduce inflammatory pain in rats, and the mechanism may be related to the cAMP-PKA-CREB signal transduction pathway.
5.Practice and evaluation of creating magnetic nursing work environment in hospital

Objective To explore the moves that creating magnetic work environment in hospital, in order to improve the nurses job satisfaction and reduce the turnover rate then stable the nursing team. Methods Through a series of measures such as the establishment of scientific rational allocation of human resources, implementing fair performance evaluation and allocation mechanism, carrying out the nurse grading management system to create magnetic nursing work environment, the nurse job satisfaction and turnover rate were compared before and after the measures. Results In January 2014 a total of 520 questionnaireswere issued, 519 valid questionnaires, and the effective recovery rate was 99.81%(519/520);And 559 were valid of 577 issued questionnaires, which was 96.88% (559/577). The overall score of nurses′job satisfaction increased from (3.20±0.46) points in 2013 to (3.41±0.52) points in 2014 (t=9.40, P= 0.000); The nurse turnover rate fell to 1.97%(13/661) from 5.13% (30/585) (χ2= 10.27, P= 0.003). Conclusions That is very important for remain stable and high quality nursing team to carry out scientific and fine management according to the demand of nurses, so as to create a safe, harmonious, positive, cooperative magnetic nursing work environment which have development direction and space.
6.The effect of antisense bcr - abl oligonucleotides on the growth of K562 cells

AIM:To study the effect of bcr - abl gene antisense phosphorothioate oligonucleotides(Aspo) on K562 cell line and explore its significance in chronic myelogencous leukemia (CML) gene therapy. METHODS: Cells were exposed to oligomers, observed by inverted microscope. Cells inhibitory rate were determined by 0.4% trypan blue exclusion. CFU - K562 were cultured in 0.8% methylcellulose. P210 was measured by flow cytometry. RESULTS:K562 cells were treated with Aspo, they still grew in clone state and show antisense sequence specific and dose dependent. When the concentration of Aspo was more than 5?mol/L, the growth of cells was inhibited and P210 was down regulated or completely suppressed, and the greatest growth inhibition was at 120h. There was significant inhibition of cell proliferation in a rang of cells number from 1 ? 10-4/mL to 5 ? 10-4/mL after treatment with 10?mol/L Aspo. b2a2 Aspo was also effect on K562 cells which expressing b3a2 mRNA. CONCLUSION: bcr - abl Aspo has a specific growth inhibition effect on K562 cells, and worths further study in CML gene therapy.
7.Comparative study of volatile chemical constituents from shell and kernel of Caesalpinia minax Hance

Objective:To compare the volatile chemical constituents from the shell and kernel of Caesalpinia minax Hance.Methods:The volatile chemical compositions of Caesalpinia minax Hance were obtained by organic solvent-wet distillation and were analyzed by GC-MS equipped with a elastic quartz capillary column HP-5MS 5%Phenyl Methyl Siloxane(30m?0.25mm?0.25?m).The constituents were identified by their mass spectra.The relative percentage of the volatile constituents was calculated from the GC peak areas.Results:One hundred and fifteen kinds of chemical constituents in the shell of Caesalpinia minax Hance were separated and sixty-three of them,which accounts for 54.7%of total volatile constituents, were characterized.There are five kinds whose relative contents were more than 2.0%.One hundred and two kinds of chemical constituents in the kernel of Caesalpinia minax Hance were separated and fifty-one of them,which accounts for 50.0%,were characterized.There are seven kinds whose relative contents were more than 2.0%.Form the shell and kernel of Caesalpinia minax Hance there were forty kinds of chemical constituents the same.The most relative contents were Limonene(5.31%,shell)and 1-Hexanol(12.94%,kernel).Conclusion:This paper reports,for the first time,the comparison of volatile constituents from shell and kernel of Caesalpinia minax Hance.It is helpful for controlling the quality and further researching of Caesalpinia minax Hance.

9.Availability of school health staff and health equipment in primary and secondary schools in Jinan

Objective:
To understand the situation of school health staff and the allocation of health equipment in primary and secondary schools in Jinan.
Methods:
A questionnaire survey was conducted in CDC and public primary and secondary schools to describe the situation of the school health staff and health equipment in primary and secondary schools.
Results:
There were 23 school health staffs in CDC, among which 26.1% were full time. The total staffing ratio of school doctors was 13.9%, with the compliance rate of 19.8%. The total allocation rate of health care teachers was 74.7%, 92.2% of them were part time. The total staffing ratio of school doctors in boarding schools was 20.0%, with a staffing compliance rate of 12.5%. The total allocation rate of school doctors and health care teachers in boarding schools was 66.7%. 16.8% of public primary and secondary schools and 23.3% of public boarding primary and secondary schools did not have any health staffs. 27.6% of public primary and secondary schools are equipped with infirmary, 70.8% with health rooms, and 10.3% without any. The configuration of necessary equipment in infirmary and health care rooms were less than 100%.
Conclusion
There is a serious shortage of school health staffs and health equipment in primary and secondary schools in Jinan. Attention and investment in school health should be strengthened, and resources should be rationally allocated to ensure the health of primary and secondary school students.
10.Remifentanil and fentanyl combined with propofol administered by target controlled infusion in neurosurgery.

OBJECTIVE:
To compare the hemodynamics and post-anesthetic recovery of total intravenous anesthesia (TIVA) with remifentanil or fentanyl combined with propofol administered by target controlled infusion (TCI) in neurosurgery.
METHODS:
A total of 80 patients undergoing selective neurosurgery were randomly divided into a remifentanil group (Group R, n=40) and a fentanyl group (Group F, n=40). In Group R,remifentanil and propofol was administered by TCI and the blood concentration were 3 approximately 5 microg/L and 3 approximately 5 mg/L each. In Group F, fentanyl was continuously infused at 2 approximately 3 microg/(kg.h) and propofol was administered by TCI with the same blood concentration as that in Group R.Vecuronium was injected at intervals to maintain muscle relaxant.Mean arterial pressure and heart rate during the anesthesia and post-anesthetic recovery were recorded.
RESULTS:
Mean arterial pressure of all the patients was decreased significantly from induction of anesthesia to termination of operation compared with that before the induction( P<0.01). The heart rate of Group R was increased obviously from recovery of respiration to extubation and heart rate of Group F was decreased obviously from fixed headframe to termination of operation compared with that before the induction (P<0.01). But there was no significant difference between the 2 groups (P>0.05). The eyes opened and extubed time of Group R were decurtated obviously and the scores of pain were increased significantly (P<0.01).
CONCLUSION
TIVA with remifentanil or fentanyl combined with propofol administered by TCI in neurosurgical operation can provided steadible hemodynamics. Resuscitation of remifentanil with propofol administered by target controlled infusion were more quickly but the scores of pain were more higher than that of fentanyl.
