Thirty albino New Zealand rabbits were divided into three groups at random, ten in each group, which were N~6-2'-O-dibutyryladenosine-3', 5'-cyclic monophosphate (db-cAMP) + taurine (Tau), normal saline (NS) + Tau and db-cAMP+ NS. Tau, dbcAMP and NS were injected into the rabbit's ventricles by intracerebroventricular (ICV) rotate. The results showed that ICV Tau could inhibite the fever induced by ICV dbcAMP in the single long phase hyperthermic duration. 2.Clinical study of coronary artery perforation during percutaneous coronary interventions

Objective To summarize clinical characteristics of coronary artery perforation during percutaneous coronary interventions.Methods Retrospective analysis of consecutive 32 patients who had coronary artery perforation in Fu Wai Hospital from April,2004 to August,2006 was carried out.Results Thirty-two cases of coronary perforation occurred during 7102 PCI procedures performed within this period(incidence:0.5%).These cases are comparatively complicated lesions including 81.3% of type B2+C and 62.5% of chronic total occlusion(CTO)lesions.Ellis classification:typeⅠ:14(43.8%),type Ⅱ:10(31.2%),type Ⅲ:8(25.0%),type Ⅳ:0.Mechanism of coronary artery perforation:guiding wire:21(65.6%),predilatation:6(18.8%),postdilatation:3(9.4%),stent implantation:2(6.2%).Clinical consequence:death:3(9.4%),cardiac temponade:7(21.9%),acute myocardial infarction(AMI):9(28.1%).Treatment:reversal of heparin-induced anticoagulation by application of protamine:14(43.8%),prolonged balloon inflation:9(28.1%),percardiocentesis:7(21.9%),implantation of membrane covered stent:4(12.5%),bail-out surgical repair:2(6.2%).Conclusion Coronary perforation during PCI is a rare complication;type Ⅲ perforation is associated with significant morbidity and mortality,which needs urgent and intensive treatment.
3.Analysis of mechanism in acute myocardial infarction with previous percutaneous coronary intervention

Objective To observe the characteristics of culprit lesions in AMI with previous PCI.Methods Retrospective analysis was carried out in 61 consecutive AMI patients with previous PCI treated with primary PCI from April, 2004 to April, 2006 in Fuwai hospital.Results Location of culprit lesions in the 61 patients (62.1?10.0 yrs; male 88.5%) were: LAD 47.5%, RCA 39.5%, LCX 13.0%. Comparing the medication of the patients during the first year after the former PCI, besides the continous use of aspirin (93.8% vs 100%,P=0.113), all patients had stopped using clopidogrel, and the use of ?-blockers, ACEIs and statins also dropped significantly after 1 year (46.9% vs 75.0%, P= 0.001; 34.4% vs 70.8%, P= 0.001; and 28.1% vs 77.1%, P= 0.000, respectively). The characteristics of the culprit lesions included: acute and sub-acute in-stent thrombosis in 12 cases (19.7%), late and very late in-stent thrombosis in 6 cases (9.8%), plaque rapture in 41 cases (70.5%), but no restenosis was involved. The time of the recurrant AMI after the former PCI were: 13 cases (21.3%) within 1 month including 12 cases of acute or subacute in-stent thrombosis and 1 case of plaque rupture in anther coronary artery 3 days after primary PCI; 16 cases (26.2%) after the first month to 1 year including 12 cases of plaque rupture and 4 cases of late in-stent thrombosis; 32 cases (52.5%) 1 year including 30 cases of plaque rupture and 2 cases of very late in-stent thrombosis. Conclusion The major mechanism of recurrant AMI after preoious PCI is plaque rapture.
4.DNA damage of bone marrow cells in mice induced by radon exposure

Objective To study the DNA damage of bone marrow cells of mice after exposure to Radon.Methods Twenty-four mice were randomly divided into four groups, one control group and three experimental groups with the cumulative doses of radon at 27 WLM (low dose group), 52 WLM (middle dose group) and 105 WLM ( high dose group). DNA damage induced by radon in bone marrow of mice was detected by methods of single cell gel electrophoresis (SCGE), micronucleus(MN) and laser scanning confocal microscope (LSCM) observation. Results The DNA strand breakage, rate of MN and apoptosis increased significantly in the high dose group, but not in the middle and low dose groups. Conclusions Exposure to radon could induce DNA damage in bone marrow cells of mice at high levels.
5.A comparative study of in-stent restenosis after drug eluting stents

Objective To compare the long-term effect of three different drng-eluting stents (DES) for in-stent restenotic lesions. Methods From April 2004 to June 2006, 390 consecutive patients undergoing DES implantation including 187 Cypher (group C), 89 Taxus (group T) and 114 Firebird (group F) with resulting in-stent restenotic lesions were studied. A mean of 2 year clinical and 7-month angiographic follow-up was carried out. Results Baseline characteristics indicated that there were more unstable angina cases in T group and less left main disease and more triple vessel disease cases in F group. A mean of 2-year follow-up results showed no difference of major adverse cardiac events (MACE) rate among the three groups (23.0% vs 22.5% vs 13.2% , P = 0. 081) and no difference of cardiac death, non-fatal myocardial infarction (MI) and target vessel revascularization rate(1.1% vs 4. 5% vs 1.8% , P = 0. 210, 5.9% vs 2. 2% vs 2. 6% ,P =0. 226 and 2. 9% vs 2. 2% vs 0. 9% , P =0. 509). There was no difference of total stent thrombosis or its components among the three groups (total: 4. 8% vs 3.4% vs 2. 6%, P=0. 605, early: 0. 5% vs 0 vs 0. 9% ,P =0. 560, late: 1.6% vs 1.1% vs 0. 9% ,P =0. 849 and very late: 2. 9% vs 2. 2% vs 0. 9% , P =0. 509) according to Academic Research Consortium (ARC) standard definitions (definite + probable). 7-month angiographic follow-up indicated that there was a lower trend of both in-stent and in-segment trestenosis rate in C and F groups (17.9% vs 29. 4% vs 13.6% ,P = 0. 214 and 21.8% vs 35.3% vs 15.9%, P =0. 132) and in-stent and in-segment late loss was significantly smaller inCand F groups [(0.31±0.12) mm vs(0.75±0.24) mm vs(0.31±0.13) mm, P=0.000 and (0.33±0.18)mm vs (0.61±0.23)mm vs (0.31±0.14)mm, P=0.001]. Conclusions Results from this 2-year follow-up, single-center study showed comparable effectiveness and safety of Cypher, Taxus and Firebird DES for in-stent restnotic lesions, but Cypher and Firebird had better effect in reducing restenosis.
6.Association Rules of Chinese Herbal Compound Formula for Osteoporosis

OBJECTIVE:To discuss the medication regularity of Chinese herbal compound formula for osteoporosis. METHODS:A total of 104 Chinese herbal compound formula for osteoporosis (OP) published from 1995 to 2005 were retrieved from China Hospital Knowledge Database(CHKD); and a database was set up using EXCEL2000 software,with the drug name of each herbal medicine in the compound herbal formula included and a statistical analysis was carried out.The herbs which were used frequently were subjected to data mining and analysis using association rules. RESULTS:Of the total 104 Chinese herbal compound formula for osteoporosis,leading the first 12 places in terms of application frequency in turn were prepared Rehmannia glutinosa,Herba Epimedii,Dioscorea opposita and Eucommia ulmoides,etc.In the association rule set,there were respectively 7,6,6 and 8 rules which respectively consisted of 2,3,4 and(5,6,7) herbs.CONCLUSION:The relationship between the three factors(renal deficiency,splenic asthenia,and blood deficiency) and osteoporosis were verified in the study of the Chinese herbal compound formula for osteoporosis.

8.Protection of Tongxinluo Against Myocardial Ischemia/Reperfusion Injury Relates to Enhancing Endothelial Barrier in Diabetic Rats

Objective: To clarify that protection of Tongxinluo against myocardial ischemia/reperfusion injury relates to enhancing endothelial barrier in diabetic rats.
Methods: A total of 32 Zucker diabetic rats were randomized into 4 groups:Sham group, Model group, Insulin group and Tongxinluo group, n=8 in each group. In addition, there was a Control group containing 8 non-diabetic Zucker rats. Myocardial ischemia/reperfusion injury model was established by a 45-min ischemia and 3-h reperfusion protocol. The size of infarction was detected by pathological staining, the microvascular permeability was examined by Miles assay to obtain the lfuorescein isothiocyanate concentration in myocardial tissue, the Triton X-100 soluble and insoluble VE-cadherin was measured by membrane protein extraction and western blot analysis.
Results:The size of infarction in Model group was obviously higher than that in Control group (55.2 ± 1.4)%vs (36.2± 1.3)%,P<0.05 and the lfuorescein isothiocyanate concentration was also higher in Model group than Control group. Both insulin group and Tongxinluo group had the similar size of infarction with Model group (36.8 ± 1.2)%vs (38.7 ± 1.1)%, P>0.05. Both insulin group and Tongxinluo group got lower lfuorescein isothiocyanate concentration (all P<0.01) and VE-cadherin internalization (all P<0.05) than that in Model group.
Conclusion: Protection of Tongxinluo against myocardial ischemia/reperfusion injury in diabetic rats is as effective as
insulin, but the effect is independent of reducing blood glucose and may be related to enhancing endothelial barrier.
9.Study of Morphological Characteristics of Calcified Plaques and Luminal Stenosis of Coronary Artery at 64-slice Computed Tomographic Coronary Angiography

Objective To study the correlation between morphological characteristics of coronary artery calcified plaques and luminal stenosis of local coronary artery segments with 64-slice computed tomography.Methods One hundred and eleven patients who had undergone 64-slice computed tomographic coronary angiography(CTCA)and conventional coronary angiography(CAG)were retrospectively analyzed.The calcified plaques were classified as punctate,nodular,strip-like and nubbly in long-axis view of coronary artery lumen,and were classified as crescent,semilunar,round moon and circinate in short-axis view.The morphologic characteristics of these calcified plaques on CTCA were retrospectively analyzed and compared with luminal stenosis of CAG results.Results Among 528 calcified coronary segments which were analyzed in 111 patients,there were 383(72.5%)punctate calcified plaque segments and 145(27.5%)of non-punctate plaques.There were 34(23.4%,34/145)non-punctate calcified plaques which caused severe stenosis(≥75%),including 4(11.8%)nodular,8(23.5%)stripe-like and 22(64.7%)nubbly calcified plaques on the long-axis view,and 0(0.0%)cresent,8(23.5%)semilunar,18(52.9%)round moon and 8(23.5%)circinate calcified plaques on the short-axis view.The ratios of different morphological coronary artery calcifications which caused severe stenoses were significantly different with each other(all P<0.01).Conclusion Different figures of coronary artery calcified plaques demonstrate different degrees of stenoses of local coronary artery lumen.Severe stenoses were mostly caused by nubbly calcified plaque on long-axis view,round moon and circinate calcified plaque on short-axis view.

Objective To construct the recombinant yeast expressing PreS2 120-146-hepatitis B surface antigen (HBsAg),and to evaluate the immune effects of whole yeast cells.Methods PreS2 120-146 and HBsAg gene sequence were optimized according to the yeast cell codon preference,and were recombined and cloned into pPIC3.5K yeast expression vector to construct pPIC3.5K/PreS2 120-146 plasmid.After digested and linearized by Bgk Ⅱ restriction enzyme,pPIC3.5K/PreS2 120-146-HBsAg recombinant plasmid was electrotransformed into GS115 strain to screen PreS2 120-146-HBsAg-recombinant Pichiapastoris .The expression of PreS2 120-146-HBsAg was identified by sodium doclecyl sulfate polyacrylamide gel electrophogesis (SDS-PAGE),Western blot and enzyme linked immunosorbent assay (ELISA)analysis. BALB/c mice were vaccinated by inactivated whole recombinant yeast cells expressing target protein. Specific antibodies to HBsAg were detected by ELISA.Cytotoxic T lymphocyte (CTL)response induced by interferon (IFN)-γ was detected by reverse transcription-polymerase chain reaction (RT-PCR)when immune spleen cells of mice were stimulated by CTL epitope on HBsAg.Independent sample t test was used. Results Data of PCR detection,restriction enzyme digestion and sequencing analysis showed that recombinant pPIC3.5K/PreS2 120-146-HBsAg plasmid was successfully constructed.SDS-PAGE,Western blot and ELISA verified the expression of PreS2 120-146-HBsAg in the lysate of the recombinant Pichiapastoris induced by methanol.Levels of specific anti-HBsAg IgG antibodies produced by inactivated yeast cells vaccinated mice were comparable to purified HBsAg immunization (t =0.946,P =0.381 ). Analysis of HBsAg-specific CTL responses revealed that the level of IFN-γwas significantly higher when the immune spleen cells of mice were stimulated by CTL epitope peptides on HBsAg (t =2.305 ,P =0.044).Conclusions PreS2 120-146-HBsAg target protein is successfully expressed by construction of recombinant Pichiapastoris . The specific humoral and cellular immune responses are induced by recombinant whole yeast cells vaccinated mice.
