1.Effect of FTY720 on the expression of hypoxia inducible factor-1 in rat renal ischemia reperfusion injury

Objective To investigate the role of FTY720 in the protection of rat renal ischemia reperfusion (I/R) injury and its influence on the expression of hypoxia inducible factor-1? (HIF-1?).Methods Ischemia/reperfusion kidney injury rat model was established. The rats were allocated into groups at random: sham group (Sham) and ischemia/reperfusion group (Control). The latter were sub-divided into FTY720 (1 mg/kg every day for 3 days before operation) group, MMF (20 mg/kg every day for 3 days before operation) group and CsA (10 mg/kg every day for 3 days before operation) group. Blood creatinine and the expression levels of HIF-1? mRNA and protein were observed 0 h, 6 h, 24 h, 72 h and 7 days after operation.Results At time points of 0 h , 6 h , and 24 h, Scr levels were higher in FTY720 group than in control group (P
2.Effect of mycophenolic acid on the proliferation and MCP-1 secretion in the glomerular mesangial cells

Objective To explore the inhibitory effect of mycophenolic acid (MPA) on the proliferation and monocyte chemoattractant protein-1 (MCP-1) secretion in mice glomerular mesangial cells (GMCs).Methods MPA with different concentrations ( 0.1 , 1, 5 and 10 ?mol/L ) was added into the cultured mice GMC line in vitro for 24 h. The GMCs proliferation level was detected by the 3-(4,5-Dimethylthiazol-2-yl)2,5-diphenyltetrazolium bromide (MTT) incorporation. The absorbance for counting the GMCs survival was read at 570 nm by using an automated microplate ELISA reader. The cells treated with MPA (0, 0.5 and 2.5 ?mol/L ) were stimulated by TNF-? (20 ng/ml), IL-1? (2 ng/ml), IFN-? (10 ng/ml). Four h later, the MCP-1 concentration in supernatant was determined by ELISA. At the same time, the lymphocyte migration was examined.Results MPA has a concentration -dependent antiproliferative effect on cultured GMCs. 4.Effect of Different Doses of Chemotherapeutic Agents on Sensitivity of Glioma U251 Cell Line to Chemotherapy.

Objective To study the effect of chemotherapeutic agents of different doses on sensitivity of glioma U251 cell line to chemotherapy.Methods The rates of cell growth of the U251 cell line under the effect of different chemotherapeutic agents(DOX、VCR、VP-16 and with verapamil)of a series of doses were detected by MTT assay.Results Using DOX or VCR or VP-16 singly,the rates of cell growth were detected at a lever with positively correlates with the concentrations of these chemotherapeutic agents.Moreover the rates of cell growth changed more obviously within some range of concentrations,while the change is limited with the high or low concentrations.The combination of pates of chemotherapeutic agents and verapamil show synergistic effect.Conclusions The relationship between the doses of chemotherapeutic agents and sensitivity of chemotherapy gas a basis for choosing appropriate clinical chemotherapy doses.
5.Comparative study on detection methods of mutations in hepatitis B virus precore and basic core promoter

The sequence of five clones from one serum which was identified precore mutation by mPCR-RFLP were all A1896 mutant strains.Another serum identified mixture infection by mPCR-RFLP , one clone was A1896 mutant strain and four were G1896 wild strains.The results of mPCR-RFLP were verified by cloning.Conclusions Comparison with sequencing, the mPCR-RFLP method is simple,accurate and can be used in large-scale surveys and clinical research.
6.Nasal primitive neuroectodermal tumor: a case report.

Primitive neuroectodermal tumor (PNET) is a small round cell malignancies, normally confined to the chest wall and soft tissues of children and young adults, and reports of this tumor existing in nasal cavity are scarce. We herein present a case of nasal PNET. A 35-year-old male patient presented with a chief complaint of stuffy nose was diagnosed with a primary nasal pPNET by the clinical, microscopic, immunohistochemical features. Since these tumors are highly aggressive and patient prognosis is typically very poor, early diagnosis, immediate surgical resection and re-resection if possible, early radiotherapy and chemotherapy and close follow-up are required. We also review literatures on pPNET.

7.L-type calcium channels of human mesenchymal stem cells

Objective:To investigate L-type calcium channels of human mesenchymal stem cells(hMSCs) cultured in vitro.Methods:hMSCs were isolated,cultured by Percoll gradient centrifugation,adherence to plastic flask and monoclonal culture.Immunophenotypes of hMSCs were detected by immunofluorescence and flow cytometry techniques.Whole cell patch-clamp technique was used to observe the change of L-type calcium channels of human mesenchymal stem cells(hMSCs).Results:High homogenous hMSCs had been isolated and cultured in vitro.hMSCs had unique immunophenotypes and they were positive for CD44,CD29,c-kit but negative for CD34,CD31,CD54.Calcium currents could be found in 40 percent of hMSCs,peak currents of calcium(I Ca-Peak ) were (102.67?19.06)pA from +20 mV to +30 mV.The currents could be inhibited by Cd~ 2+ (20 ?mol/L).Conclusion:Undifferentiated hMSCs express less L-type calcium channes,regulation of the chanels might contribute to directional differentiation.
8.Reversal effect of RNA interference targeting Fascin1 on migration and invasion of glioma cell line U87 MG

Objective:To detect the functional role of Fascin1 and its related molecular mechanisms in migration and invasion capacity of glioma cells,we utilized gene specific small interference RNA of Fascin1 in cell line U87 MG. Methods:Fascin1-siRNA or negative siRNA was transfected into U87 MG cells of control group or experiment group. Transwell method was employed to assess the migration and invasion capacity of glioma cells. Western blot analysis was used to detect the protein expression of Fascin1,pAKT and pSTAT3. The impact of PI3K/AKT pathway and STAT3 pathway on migration and invasion of U87 MG cells was verified,via applying LY294002 and LY294002,which was inhibitor of the two pathways respectively. Results:As compared to control groups,the migration and invasion capacity of transfected glioma cells were attenuated about 52% or 43%(P<0. 05),accompanied with the decreased phos-phorylation of AKT and STAT3. As utilizing the inhibitors of AKT and STAT3,attenuated migration and invasion capacity of U87 MG cells were observed. Conclusion:Down-regulated expression of Fascin1 could suppress the migration and invasion capacity of U87 MG cells by inhibiting the phosphorylation of PI3K/AKT pathway and STAT3 pathway.
9.The cross-sectional study on diabetic peripheral neuropathy among the middle-aged and old-aged population with type 2 diabetes mellitus in a community

Objective To study the prevalence and the related risk factors of diabetic peripheral neuropathy(DPN) after good blood glucose well controlled among in middle-aged and old-aged population with type 2 diabetes mellitus in a community of Beijing.Methods DPN was screened by Michigan neuropathy screening instrument (MNSI),and common blood biochemical parameters were tested,among over 45 years of age registered population patients with type 2 diabetes mellitus in Shuangjing community of Chaoyang District of Beijing.Results The blood glucose control rate was 90.36% (253/280),and the prevalence of DPN was 25.36% (71/280) with male of 24.47 % (23/94) and female of 25.81% (48/186).Univariate analysis showed that both age and HbA1c in DPN group were higher than that of non-DPN group,and the differences were statistically significant(P =0.000,0.008).The level of plasma low density lipoprotein cholesterol(LDL-C) of the DPN group was lower than that of the non-DPN group,and the difference was statistically significant (P=0.017).The hypertension prevalence rate of the DPN group was 87.32%(62/71),higher than that of the nonDPN group(75.6% (158/209)),and the difference was statistically significant (P =0.037).The multivariate logistic regression analysis showed that the estimate of parameter for age was 0.143 (P =0.0001),OR was 1.153,95%CI was from 1.029 to 1.219,for HbA1c was 0.529(P=0.03),OR was 1.698,95%CI was from 1.053 to 2.738,and for LDL-C was-O.919 (P =0.018),OR was 0.399,95% CI was from 0.186 to 0.854.Conclusion The prevalence of DPN is still above 25% among middle-aged and old-aged in a community,even though the well controlled rate of blood glucose control rate is above 90%.It is possible that old age and HbA1c are related to DPN,apart from blood glucose.
10.Protective effect of allopurinol in kainic acid-induced epileptic rats

Objective To investigate the protective effect of allopurinol in kainic acid-induced epileptic rats and to explore new ideas and methods for the clinical treatment of epilepsy. Methods 120 Wistar rats were randomly divided into sham group, KA epilepsy group and allopurinol groups. Six rats of each group were randomly selected and were given electrodes into their left frontal and hippocampal regions. After injection, behavior changes were observed in all rates without electrodes. 24 h later, MDA level and SOD enzymatic activity of the left hippocampi were measured. One week later, the EEGs were recorded in rates with electrode, as well as total time of seizures /30 min and numbers of seizures / 30 min. Results Compared with the KA model group, latency period of the epilepsy in the allopurinol group was longer (P < 0.05) and the extent was lighter (P < 0.05); the MDA level was significantly lower (P < 0.01), the SOD enzymatic activity was significantly higher (P < 0.01). The total time of seizures / 30min and numbers of seizures / 30 min in allopurinol group reduced significantly (P < 0.01). Conclusion Allopurinol has potential antiepileptic and antioxidative activities in kainic acid-induced epileptic rats.
