1.Effective components combination of Xiaoxuming Decoction on anti-Alzheimer’s disease

AIM: To apply high throughput screening methods to research Xiaoxuming Decoction's effective components combination and its action mechanism. METHODS: We studied the effects of 240 sequential components of Xiaoxuming Decoction (L1-L120,A1-A120) on anti-A? neurotoxicity、anti-H_2O_2 damage、anti-glutamic acid damage and ?-secretase activity. RESULTS: We evaluated these results of screening comprehensively,and found several components of Xiaoxuming Decoction (L1-L40,A30-A60,A100-A120) had the potential effects of listed before.So the combination of these components was regarded as the effective components combination of Xiaoxuming Decoction for anti-Alzheimer's disease. CONCLUSION: Xiaoxuming Decoction had effective protection against Alzheimer's disease through multi-component and multi-target.High throughput screening methods will push the development of Chinese traditional compound prescription greatly.
2.Effect of the group of effective components of Chinese medical prescription-Xiaoxuming Decoction on aging rats

AIM: To establish the model of aging rats and observe the effect of the group of effective components of Xiaoxuming Decoction(XXM)on aging rats. METHODS: D-galactose and rotenone were used to establish aging animal model.The ability of learning and memory were measured by using Morris water maze test.Then, the content of GSH and MDA,the activity of SOD,the activity of AchE and ChAT in rats'brain were measured. (RESULTS): D-galactose could induce aging like indicators in rats:decrease the content of GSH and the activity of SOD,and increase the content of MDA,but have no effect on AchE and ChAT.It showed that GEC could improve the ability of learning and memory and ameliorate biochemistry function in rats with aging induced by D-galactose. CONCLUSION: These results suggest that GEC of XXM could improve the responses of aging rats and these effects may be related to its ability of antioxidation.
3.A drug target for Alzheimers disease: ?-secretase

ABSTRACT Recent research suggests that the ?-amyloid peptide (A?) is central to the pathophysi-ology of Alzheimers disease (AD). Amyloid plaques, primarily composed of A?, have significant neurotoxic effects. So it is therapeutic strategies for AD to lower the production of A? in the brain. A? is a product of catabolism of amyloid precursor protein ( APP) by ?-secretase and y- secretase. BACE ( ?-secretase) exhibits all the properties of the ?-secretase, and as the key rate-limiting enzyme that initiates the formation of A?, BACE is an attractive drug target for AD. The recent research of ?-secretase is reviewed in this paper.

5.Study on the Determination of Aristolochic Acid by RP-HPLC

Objective To optimize the RP-HPLC procedure for the determination of aristolochic acid. Method Different extraction solvents and methods have been screened to get the most efficient way for sample preparation. The influence of different mobile systems on quantitation were compared to choose an approprate mobile system for determination. Besides,the minimum limits of different detecting waves were measured to give the method to detect trace amount of aristolochic acid. Result Refluxing with 70% methanol was better than other ways in sample preparation. Both methanol-1% acetic acid (70∶30) solution and 0.3% ammonium carbonate solution (pH=7.5)-acetonitrile (75∶25) with wavelength of 250 nm and 319 nm can be used for quantitation while 0.3% ammonium carbonate solution (pH=7.5)-acetonitrile (75∶25) with wavelength of 224 nm for trace detection. The minimum detectable amount was 0.02 ng. Conclusion The method established can be applied to determinate aristolochic acid and detect trace amout exactly.
6.Orthodontic treatment for AngleⅡ~2 malocclusion accompanying one side of TMJ click:contrast before and after treatment

Objective To investigate the imaging and condylar movements during mandibular opening/closing movement in AngleⅡ2 malocclusion accompanying one side of temporomandibular joint(TMJ)click before and after orthodontic treatment.Methods There were 20 AngleⅡ2 malocclusion patients with one side of TMJ click in treatment group,and 23 with the individual normal occlusion in control group.This study was performed taking the films of Sch?ller position before and after treatment,using CADIAX to record the condylar tracings during opening/closing movement and analyze the qualitative and quantitive changes.Results In AngleⅡ2 malocclusion patients,the condylar position of 58.33% patients recovered from retroposition to mesoposition.The condyle movement presented conspicuous verticality,the smoothing and symmetry were not well,and obvious improvements were observed after the treatment.All the index changed remarkably except the TCI and TCI of 5 mm.Conclusion It is suggested that orthodontic treatment can make the disc-condyle relationship return to normalization,and the condylar movement become much more coherent.
7.Neuroprotective effect of active components of Xiaoxuming decoction on focal cerebral ischemia/reperfusion injury in rats during early recovery period

Aim To explore the protective effect of ac-tive components of Xiaoxuming decoction ( XXMD ) on focal cerebral ischemia/reperfusion injury in rats dur-ing early recovery period .Methods The ischemia and reperfusion of middle cerebral artery model rats were established by nylon wire with 2 hours ischemic time on healthy male SD rats .The models of MCAO were eval-uated by Zea-Longa′s standard score .The model rats were randomly divided into sham operation group , the ischemia/reperfusion model group , the active compo-nents group of Xiaoxuming decoction and the positive group (extract of ginkgo biloba leaves EGb 761).Rats were orally administrated with different drugs 24 h after operation for up to 14 days, once a day.The effect of active components of Xiaoxuming decoction on behavior changes of MCAO rats in different recovery period was evaluated by a series of behavioral assessent methods such as modified neurological severity scores and cor-ner test.The infarct volume was observed by TTC stai-ning.Moreover, the contents of MDA and the activities of GSH-Px, SOD and NOS in the penumbra and core tissues of rat brain were detected by spectrophotometric method.Results Compared with the I/R model group, the active components group of XXMD could significantly alleviate the neurological deficit scores with prolonged administration . The motion function tended to be normal , stayed longer on the balance beam, sensory function gradually restored sensitivity , reduced the radio of turning to the right .Among them , compared with model group , the active components of XXMD could effectively improve the neurological dys-function after five days of administration ( P<0.05, P<0.01).Meanwhile, the active components of Xiaox-uming decoction could significantly reduce the infarct volume percentage in the cerebral tissue on post opera-tion day 5 and 14(P<0.01).In addition, the active components of XXMD could reduce the content of MDA and the activity of NOS , increase the activity of SOD and GSH-Px.Conclusion The active components of XXMD can generate neuroprotective effect in early stage of recovery and may play a major role in regula-ting the level of oxidative stress in rat brain .
