1.Application of preoperative eye position training in FL-LASIK

Objective To investigate the clinical effects of preoperative eye position training on femtosecond laser-assisted laser in situ keratomileusis(FL-LASIK)? Methods One hundred and sixty-eight myopia patients(328 eyes)scheduled for selective FL-LASIK were randomly divided into the experiment group of 85 cases(166 eyes)and the control group of 83 cases(162 eyes)?The patients in the control group were given the routine preoperative education and the patients in the experiment group received the preoperative intervention of eye position besides routine preoperative education? The two groups were compared in terms of intraoperative changes of eye position,corneal topography and the postoperative visual acuity? Results The frequency of eye position adjustment and machine stopping in the experiment group was significantly smaller than that of the control group(P < 0?001)? But no significant differences were found between the two groups in corneal topography and the postoperative visual acuity(P > 0?05)? Conclusion Preoperative training of eye position may maintain ideal eye position and reduce the frequency of downtime due to eye position adjustment during FL-LASIK,which may ensure the successful completion of FL-LASIK?
2.Clinical Significance of Serum Antibodies in Inflammatory Bowel Disease

Background: There is no gold standard for the diagnosis and monitoring of inflammatory bowel disease (IBD).As immune system plays crucial role in the pathogenesis of IBD,some immune-specific serum antibodies are considered to be useful tools for the diagnosis and differential diagnosis of the disease.Aims: To investigate the clinical significance of serum antibodies,including anti-Saccharomyces cerevisiae antibody (ASCA) and perinuclear anti-neutrophil cytoplasmic antibody (pANCA) in IBD.Methods: Serum samples were obtained from 91 consecutive IBD patients from Feb.2015 to May 2016 at the First Affiliated Hospital of Soochow University.Of them,52 were Crohn's disease (CD) and 39 were ulcerative colitis (UC).Serum samples of 36 non-IBD patients were served as controls.ASCA-IgG and ASCA-IgA were detected by ELISA,and pANCA by indirect immunofluorescence assay.Using clinical diagnosis as gold standard,crosstabs statistics was performed to measure the diagnostic accuracy of ASCA and pANCA,and ROC curve,Pearson Chi-square test and Fisher's exact test were employed for analyzing the correlations of these two serum antibodies with IBD,CD,UC,and the location of the disease.Results: Both serum ASCA-IgG and IgA were correlated with CD (AUC=0.626 and 0.614),while UC was correlated with ASCA-IgA only (AUC=0.486).Serum pANCA had relevance to IBD (r=0.342),CD (r=-0.262) and UC (r=0.614);its sensitivity and specificity for IBD and UC were superior to those for CD (P<0.05).In CD patients,ASCA-IgG was associated with terminal ileal disease (P<0.05),and pANCA was associated with colonic involvement (P<0.05).In UC patients,both ASCA-IgG and IgA were correlated with terminal ileal disease (P<0.05).Conclusions: Serum ASCA and pANCA may be helpful for discrimination between CD and UC when the diagnosis of IBD is established.Furthermore,they are closely associated with the disease location,ASCA is related with the terminal ileal disease and pANCA is related with colonic involvement.
3.Effects of genistein on bone mineralization and osteoporosis in ovariectomized rats

BACKGROUND: Genistein is the main component of phytoestrogen soy isoflavone and its structure is similar to estrogen,which suggests that it might prevent or delay osteoporosis. Research on the effects of genistein on bone mineralization and calcium(Ca), phosphor(P), zinc(Zn) and magnesium (Mg) in ovariectomized rats are rare.OBJECTIVE: To investigate the effects of genistein on bone mineralization and Ca,P,Zn and Mg in ovariectomized rats to provide a theoretical gist for the prevention of osteoporosis by genistein.DESIGN: A controlled experiment based on experimental animals.SETTING: Department of Nutrition,General Hospital of Chinese PLA.MATERIALS: The study was conducted in the Institute of Hygiene and Environmental Medicine,Academy of Military Medical Sciences of PLA between February and June 2003. Ten-week old female Wistar rats [certification number: (military medical animal): D98014] with a body mass of(170±20) g were selected.INTERVENTIONS: Experimental animals were fed with normal feeding for 6 weeks and then the feeding was changed to AIN-93 compound. Animals were then randomly divided into ovariectomized group (n=40) and sham-operation group(n=7) based on bodyweight after 5 days. Ovariectomized group received ovariectomy and sham-operation group only received abdominal incision. After 5 days of recovery,the ovariectomized group was further randomly divided into 5 subgroups with 8 rats each including ovariectomized control subgroup,estrogen subgroup [diethylstilbestrol 20 μg/(kg · d)],genistein Ⅰ,Ⅱ,or Ⅲ subgroup[dose of 25,50 or 100 mg/(kg · d)]. After 3months of feeding,6 rats were randomly selected from each group for the detection of bone density and corresponding bone hismorphometric indicators.MAIN OUTCOME MEASURES: Bone density,corresponding parameters of bone mineralization,Ca,P,Zn and Mg contents in bone of mice in each group RESULTS: After ovariectomy,femoral bone density decreased [(0. 247± 0.007) g/cm2],mean osteoid width increased[(7. 04 ±0. 32)μm],bone mineralization delayed [(4.96±0.99) days],osteoid maturity prolonged [(26.99±7.70) days],and Ca[ (251.11± 5.31) mg/g],P[(115.08± 3.78) mg/g],Zn[ (299.69±37.1)μg/g] and Mg[(4. 32±0. 12) μg/g]were all significantly different from that of sham-operation group(P<0.05).After the application of genistein,femoral bone density had a tendency of improvement[ (0. 250±0. 007) g/cm2],mean osteoid width narrowed[ (4. 97±0.77) μm],bone mineralization delayed time[ (3.18±0.69) days] and osteoid maturity time[(14.53 ±3.84) days] shortened, contents of Ca [(270.00±5.65) mg/g],P[(124.25±2.37) mg/g] andMg[(4.61±0. 08) μg/g]elevated while Zn content had no significant changes.CONCLUSION: Genistein promotes osteoid mineralization,reduces the loss of Ca, P and Mg in the bone and prevent the generation of osteoporosis in unsexed rats
4.K-ras status and efficacy of cetuximab-containing regimen in the treatment of patients with metastatic colorectal cancer

Background and purpose: Cetuximab-containing regimen has been increasingly applied in the treatment of patients with metastatic colorectal cancer. Simultaneously, the predictive factors of outcome for cetuximab have been thoroughly researched. The aim of this study was to evaluate the relationship between K-ras status and efficacy of cetuximab containing regimen in the treatment of patients with metastatic eolorectal cancer. Methods: Between March 2006 and June 2008, twenty-seven patients with metastatic colorectal cancer were treated with cetuximab in combination with chemotherapy. The K-ras mutation status [wild-type (wt) or mutation (nat)] was examined by polymerase chain reaction (PCR) and direct sequencing. The influence of K-ras mutation status on efficacy of cetuximab-containing regimen was analyzed. Results: For 27 patiants in this cohort, K-ras wt was detected in 55.6% (15/27) cases and K-ras mt in 44.4% (12/27) cases. Statistically significant differences were found between the patients with K-ras wt and K-ras mt in terms of overall response rate (ORR) (66.7% vs 25.0%,P=0.035) and progression-free survival (PFS) (8 months vs 4 months, P=0.0028). However, there was no significant difference in overall survival (OS) (19 months vs 12 months, P>0.05). The most common treatment-related adverse effect was skin reaction, with incidence rate of 80.0% and 66.7% (P>0.05), respectively. No treatment related death was observed. Conclusion: K-ras mutation status is a predictive factor of good efficacy of cetuximab-containing regimen in the treatment of patients with metastatic colorectal cancer. Patients with K-ras wt could benefit from cetuximab in combination with chemotherapy.
5.Effect of medium-long-chain triacylglycerols on blood lipid and lipoproteins in hyperlipemic patients

Objective To investigate the effect of oil with medium-long-chain triacylglycerols on blood lip- id and lipoproteins in hyperlipemic patients. Methods Totally, 112 patients with hypertriglyceridemia were en- rolled and randomly divided into MLCT group (consumed oil with medium-long-chain fatty acids) and LCT group (consumed oil with long-chain fatty acids) (both 25-30 g/d for 8 weeks). Patients in both two groups were also instructed to take exercises. Height and weight were measured at baseline and 8 weeks later. Blood glucose, serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), total cholesterols (TC), triglyc- erides (TG), high density lipoprotein cholesterol, low density lipoprotein cholesterol, apolipoprotein Al (ApoAl), ApoB, ApoA II , ApoC2, ApoC3, and ApoE were measured and compared. Results At the end of study, 101 subjects were included. There were 50 subjects left in LCT group and 51 subjects left in MLCT group, respectively. There was no significant difference in weight, ALT, AST, TC, and TG at baseline between two groups (P>0.05). Eight weeks later, weight, serum TG, ApoC2, and ApoC3 were significantly lower and ApoAl level was significantly higher than those at baseline in MLCT group (P < 0.05). At the end of study, the decreases in body weight and blood biochemical variables including TG, ApoB, ApoA II , ApoC2, ApoC3 were significantly much greater in MLCT groups than those in LCT group (P < 0. 05). Conclusion When the diet is reasonably controlled, oil of medium-long-chain triacylglycerols may reduce the concentration of TG and improve the levels of apolipoproteins.
6.EFFECT OF GENISTEIN ON BONE METABOLISM IN OVARIECTOMIZED RATS

Objective: To investigate the effects of genistein (Gen) on bone turnover in ovariectomized rats. Methods: Forty-seven Wistar rats were randomly allocated into six groups: sham-operated (sham), ovarietomized (ovx), ovarietomized plus diethyl stilbestrol [E, 20?g/(kg bw?d)] or Gen [25、50、100 mg/(kg bw?d)] administration. After the rats had been fed for 3 mo,bone mineral density (BMD), histomorphometric parameters, the serum concentration of osteocalcin (BGP) and urinary contents of pyridine and hydroxyproline (HYPRO) were analysed. Results: BMD, trabecular bone volume (TBV) and mean trabecular plate density (MTPD) of ovariectomized rats were significantly decreased, but serum concentrations of BGP,urinary contents of deoxypyridinoline and HYPRO and mean trabecular plate space (MTPS) increased compared with the sham. After 3 mo of Gen supplementation, TBV,MTPD and urinary deoxypyridinoline were improved significantly in ovariectomized rats. Conclusion: Genistein reduced bone loss in ovariecromized rats by suppressing bone metabolism and osteoclast activity.
7.Expression of Cdh1 and its downstream substrates in primary neurons after oxygen-glucose deprivation

BACKGROUND:Cdh1 has been shown to express in rat hippocampus and cortex in a large number. Moreover, in vitro test demonstrated that Cdh1 expression was higher in neurons than in neural stem cel s, which possibly associated with the differentiation of neural stem cel s into neurons. However, the effects of anaphase promoting complex Cdh1 on ischemic neuronal damage remain unclear.
OBJECTIVE:To investigate the expression of Cdh1 and its downstream substrate in primary cultured neurons with oxygen-glucose deprivation. METHODS:Primary neurons from cortex of postnatal 24-hour rat pups were cultured in vitro, and identified by immunofluorescence staining. The oxygen-and glucose-deprived models were established by three gas incubator fil ed with nitrogen in sugar-free Earle’s solution. After 1 hour of hypoxia, reoxygenation was conducted. Real-time fluorescent quantitative PCR was used to detect the mRNA expression of Cdh1 and its downstream substrates Skp2, Cyclin B1 before hypoxia, 6 hours, 1, 3, 7 days after oxygen glucose deprivation. RESULTS AND CONCLUSION:After oxygen glucose deprivation, the expression of Cdh1 and Cyclin B1 in primary neurons was increased (P<0.05), while Skp2 expression was decreased (P<0.05). Above data indicated that Cdh1 expression in neurons increased after oxygen-glucose deprivation. It may degrade Skp2 and participate in hypoxic neuronal apoptosis by ubiquitination.
8.EFFECTS OF MEDIUM-AND LONG-CHAIN FATTY ACID TRIACYLGLYCEROL ON BLOOD LIPID IN HYPERTRIACYLGLYEROLEMIC SUBJECTS

Objective To investigate the effects of medium-and long-chain fatty acid triacylglycerols on blood lipid in hyperltriacylglycerolemic subjects. Method A case-control clinical trial was carried out,in which 112 subjects with hypertriacylglycerolemia were enrolled and divided into two groups. These two groups were randomized to long-chain fatty acid triacylglycerol oil(LCT) ,and medium-and long-chain fatty acid triacylglycerol oil(MLCT) . All subjects were requested to ingest fixed energy intake and perform physical activity daily during the whole study. Both of the oils were asked to consume at 25-30g daily for consecutive 8w. Anthropometric measurements and blood biochemical variables were measured at the initial and final time of the study. Results Eleven subjects were excluded from the study because of various reasons. There were 50 and 51 subjects left in LCT and MLCT group,respectively. No difference of sex distribution was noted between two groups. There were also no significant differences in daily intake of energy,protein,fat and carbohydrate,as well as daily physical activity time between two groups at 0,2,4,6 and 8w. The subjects consuming MLCT demonstrated significant decreasesin body weight,BMI,WC,HC,WHR,body fat,body fat % compared with the initial values after 8w. A greater extent of decrease in body weight,BMI,WC,body fat and body fat% was found in MLCT group than in LCT group. The levels of TG,ApoB,ApoAII,ApoC2,ApoC3 in MLCT group were significantly lower than those in LCT group after 8 w,and the extent of decrease in these indicators was much greater in the former than in the latter. Conclusion Consumption of medium-and long-chain fatty acid oil may help to reduce body weight,body fat and concentration of blood triacylglycerol and improve apolipoprotein metabolism in hypertriacylgly cerolemic subjects under an appropriate dietary regime.
9.Study of Xiaoyao Tablet on hepatoprotective action

Objective: To study the protective effect of Xiaoyao Tablet(XYT)(Radix Bupleuri, Radix Angelicae Sinensis, Rhizoma Atractylodis Macrocephalae, Radix Paeoniae Alba, Poria, Radix Glycyrrhizae, Herba Menthae, Roasted ginger) on liver. Methods: Using the acute hepatic injury model induced by Tetrachloride and D-galactosamine (CD-GaIN) in rats or mice, we observed the influence of XYT on ALT and AST level in the serum, MDA level and glutathione-s-transferase(GST) activity in the liver homogenates. The pain-killing effect was tested by body twisting model induced by glacial acetic acid and by hot plate model in mice. Results: The XYT reduced the serum ALT level significantly in the acute hepatic injury model in rats and mice. In the CCl 4 reduced acute hepatic injury mice model, XYT decreased the MDA level and increased the GST activity in liver significantly, also it reduced the MDA level in the serum. The XYT reduced twisting number in mice induced by glacial acetic acid. In the hot plate test, the pain threshold was increased. Conclusion: The drug has the similar hepatoprotective function to Xiaoyao Pills.
10.Expression of APC-Cdh1 Protein after Cerebral Ischemia-reperfusion Damage in Rats

ObjectiveTo investigate the expression of APC-Cdh1 protein after cerebral ischemia-reperfusion injury.Methods60 male Sprague-Dawley rats were randomly divided into Sham-operated group(SH) and ischemia-reperfusion group(IR). The rats of ischemia-reperfusion groups were induced by four-vessel occlusion (4-VO). At different times after injury, the expression of APC-Cdh1 of rat hippocampus was observed by Western blotting and immunohistochemistry.ResultsCompared with sham-operated group, the expression of Cdh1 protein significantly decreased 1 day and increased obviously 3 days, but decreased again 7 days after injury in ischemia-reperfusion group. The immuno-staining showed that APC-Cdh1 was highly cerebral cortex and hippocampus in ischemia-reperfusion group. ConclusionAPC-Cdh1 may be involved in the central nervous system injury.
