1.The evaluation of human albumin in patients with hypoproteinemia

Objective To analysis the rational use of human albumin in patients with hypoproteinemia in our hospital.Methods The drug evaluation criteria of human albumin were developed and the patients of different gen-dem,ages,departments,clinical diagnoses,human albumin species,administration,doses,course of treatment and pur-poses in our hospital were investigated.Results The using rate of human albumin in patients with hypoproteinemia was 53.1% in our hospital,20% human albumin accounted for the most(89cases),and the consumption of human albumin used in department of respiration was the most in our hospital (70cases).Diagnosis of 14 cases were not anastomosis with indications and 8 cases hadprolonged use and no indicative use and inappropriate medication. Conclusion The irrational use of human albumin are existed in our hospita1.We should develop the drug using standard of human albumin and strictly grasp the indications of drug use to avoid overuse of human albumin.
2.Clinical observation of nicorandil in treatment of 85 patients with angina pectoris

ObjectiveTo observe the therapeutic effect of nicorandil in treatment of angina pectoris. Methods155 cases of angina pectoris patients were randomly divided into the therapeutic group(85 cases)and control group(70 cases),both of which took normal doses of aspirin vitamin C effervescent tablets 0.1g/day,clopidogrel 50mg/day,elantan 50mg/day and simvastatin 40mg/day,where hypertensions or diabetics were given blood pressure and sugar control and heart failure patients were given proper diuretic therapy and other methods to release heart Ioad.Patients in the therapeutic group were additionally given 5 mg nicorandil,3 times/day.The two groups were both given one period of treatment about one month. ResultsIn therapeutic group,32 cases had significant therapeutic effectiveness,34 cases had effectiveness and 14 cases had no effectiveness with a total effective rate of 82.5%.In the control group,23 cases had significant therapeutic effectiveness,28 cases had effectiveness and 24 cases had no effectiveness with a total effective rate of 67.0%.The comparative difference of the total effectiveness of the twogroups had statistical significance(x2 =4.40,P<0.05).No other significant adverse reaction was found except several cases of light dizzy in the two groups. ConclusionNicorandil has good therapeutic effective in treatment of angina pectoris and no significant adverse reaction.
3.Studies on the differential expression of signal transduction related gene in human abdominal aortic aneurysms

ObjectiveTo study the differential expression of signal transduction related gene between the tunica media in human abdominal aortic aneurysms (AAA) and normal aorta, and inspect their role in the etiology of abdominal aortic aneurysms.MethodsGene chip technique was applied to study the differential expression of signal transduction related gene in human AAA, then screening some significant differential expressed genes for further research by RT-PCR, Western Blot, and immunohistochemistry.ResultsForty-five differentially expressed genes related to cellular signal transduction were detected accounting for 1.09% of the chip′s total 4?096 genes,among them 28 genes were up-regulated and 17 down-regulated in aortic aneurysm. ASK1, ERK1 gene were found up-requlated in aortic aneurysm.ConclusionsDifferential expression of signal transduction gene could play a key role in the occurrence of AAA.
4.Clinical Study of Partial Splenectomy in Pations Undergoing Traumatic Spleen Rupture

Objective To summarize the experience of partial splenctomy in patients with traumatic spleen rupture.Methods Patients were assigned to spleen conservation group in which 59 patients underwent partial splenectomy,control group Ⅰ in which 60 patients underwent splenectomy and control group Ⅱ in which 60 patients underwent gastroinestinal operation. 5.Preparation and evaluation of self-microemulsifying drug delivery systems with tanshinone Ⅱ_A

Objective The self-microemulsifying drug delivery system(SMEDDS) with tanshinone ⅡA was prepared in order to develop its new dosage forms.Methods Pseudo ternary phase diagrams were used to evaluate the self-microemulsification existence area under emulsifier,coemulsifer,and oil phase.The HPLC analysis in vitro was set up.Solubility in various vehicles was determined.The self-microemulsification efficiency was assessed,such as stability,particle size,and Zeta potential.Results The solubi-lity of tanshinone ⅡA in SMEDDS was about 2.5 mg/g,droplet size was within 20 nm,and the absolute value of Zeta potential was over 60 mV.The stability of SMEDDS with tanshinone ⅡA was better in centrifugal condition with high temperature but not good enough to the light.Conclusion The SMEDDS can make tanshinone ⅡA solublized in water,and is an optimum vehicle in new dosage forms of tanshinone ⅡA.
6.Role and significance of nuclear factor-?B activity in systemic inflammtory response syndrome

Objective To examine the change and role of nuclear factor-kappa B(NF-?B) activity and tumor necrosis factor-alpha(TNF-?) level in patients with systemic inflammatory response syndrome(SIRS).Methods Eighty patients with SIRS were studied.The NF-?B activity in monocytes and the level of TNF-? in serum was measured.The state of illness was estimated by APACHEⅡ score.Results There was significant difference for the activity of NF-?B in patients with SIRS compared with the control group.The NF-?B activity,TNF-? level and APACHEⅡ score are higher in both resuscitation multiple organ dysfunction syndrome(MODS) group and death group than the non-MODS group and survival group correspondingly(P
7.Effects of rat interleukin-10 gene treatment on expression of collagen, MMP13 and TIMP1 in fibrotic rats

Objective:To study the effects of rat interleukin-10 (rIL-10) gene treatment on the expression of collagen , matrix metalloproteinase 13(MMP13) and their specific inhibitors the tissue inhibitor of metalloproteinase 1(TIMP1) in porcine serum in-duced liver fibrosis rats then to explore the anti-fibrotic effect of rL-10.Methods:Thirty SD rats were divided into normal control and fibrosis model group.Normal control group (group C) was intraperitoneally injected with 0.5 ml normal sodium twice a week for 8 week, while the fibrosis model group was injected with equal volume of pig serum for 8 week.At the beginning of the 5th week, fibrosis model group was further randomly divided into a fibrosis model subgroup ( group M ) , rIL-10 gene treatment subgroup ( group I ) and empty vector control subgroup(group P).Rats in group C and M were injected with Ringer’s solution as a reagent control via the tail vein weekly, rats in group I were injected with the rIL-10 plasmid pcDNA3-rIL-10, and rats in group P were injected with empty vector pcDNA3.All rats were sacrificed at the end of 8th week, and the liver tissue samples were collected to observe deposition of collegan in liver tissue by sirius red staining and detected the expression of MMP 13 and TIMP1 in the liver tissue by SP immunohistochemistry .Re-sults:Sirius red staining showed that the area of the collegan deposition was dramatically increased in fibrosis model subgroup and emp -ty vector control subgroup compared with the normal control group , and the area of the collagen deposition was dramatically decreased in rIL-10 gene treatment subgroup compared with the fibrosis model and empty vector control subgroup .Immunohistochemistry analysis showed that the expression of MMP 13 and TIMP1 in fibrosis model subgroup and empty vector control subgroup was significantly higher than the normal control group , but compared with normal control group , expression of MMP13 was significantly increased and expres-sion of TIMP1 was significantly decreased in rIL-10 gene treatment subgroup .Compared with fibrosis model subgroup and empty vector control subgroup, the expression of MMP13 and TIMP1 was dramatically decreased in rIL-10 gene treatment subgroup.Conclusion:rIL-10 gene treatment attenuates the area of collagen deposition in liver fibrosis rats associated with downregulation of TIMP 1.
8.Intravenous injection of exogenous rIL-10 plasmid DNA and its expression in rats

Aim To establish a method of hydrodynamic-based transfection(HBT) and provide a means in rats of the gene therapy of liver diseases,which allowed an efficient expression of rIL-10 gene in rat liver.Methods Using rIL-10 gene as a reporter gene,different volumes and doses of plasmid DNA solutions were rapidly injected into rat tail vein,then the serum and the tissue of liver,kidney,lung,spleen and heart in different time were collected and the expression of rIL-10 was detected by the methods of RT-PCR,ELISA and immunochemistry.Results Using rIL-10 gene as a reporter gene,the results demonstrated that an efficient transfer and expression of rIL-10 in rat liver could be achieved by a rapid injection of a large volume of rIL-10 DNA solution into rat via tail vein.Maintaining a stable expression of rIL-10 in serum could be assessed by repeated administration.Conclusion The HBT was a simple,convenient and efficient method of gene transfer and expression in rats,which could be used as an effective means to study further gene therapy of rIL-10 in liver diseases.
9.The observation of low or middle power microwave radiation on tissues adjacent to titanium alloy implants

Objective To observe the influence of low or middle power microwave radiation on tissues adja-cent to titanium alloy implants. Methods Titanium alloy internal fixation plates were implanted into the upper femur of the right side of New Zealand white rabbits, the left femurs served as the control. Then the femurs were exposed to 20, 40, 60, or 80 W of microwave radiation at a frequency of 2450 MHz for 30 minutes, and real-time temperature of the implants and its adjacent muscles were recorded. The nerve conduction study of the sciatic nerve was performed by stimulating the nerve and recording from gastrocnemius in both groups. Finally, all the rabbits were sacrificed and histo-logical studies were performed on nerve and muscle sections. Results Compared to the control limbs, temperatures of limbs with titanium alloy implants increased significantly when exposed to microwave radiation at 60 and 80 W ( P<0.05) but no significant differences were revealed when exposed to radiation at 20 and 40 W ( P<0.05) . For the sciatic nerve conduction, significantly prolonged distal latency of CMAP ( P<0.05) and slower nerve conduction velocity ( P<0.05) were observed in the experimental limbs when exposed to microwave radiation at 60 and 80 W, but the decrease of CMAP amplitude was not significant (P>0.05). Pathological examination showed obvious congestion and bleeding in the intercellular spaces of skeletal muscle cells in the experiment group when exposed to radiation at 60 W, with no ob-vious pathological changes found in the sciatic nerve. However, when exposed to radiation at 80 W, vacuoles appeared in myocytes, the cellular structure was destroyed and the myelin sheath of the sciatic nerve swelled. Conclusion Ex-posure to microwave irradiation at 20 and 40 W, but not 60 and 80 W exerts no adverse effects on nerves and muscles adjacent to titanium alloy internal fixation plates. Consequently, continuous wave microwave irradiation at low powers (20~40 W) is a safer auxiliary treatment for limbs with titanium alloy implants.
10.The protective effects of pretreatment with Lipo-PGE1 on fiver injury after hemorrhagic shock and resuscitation in rats

Objective To observe the effects and mechanism of pretreatment in rats with prostaglandin E1 on liver after hemorrhagic shock and resuscitation(HSR).Method In total,32 male SD rats were randomly(random number)divided into four groups(n=8):group A(sham group),group B(shock group),group C(HSR group)and group D(Lipo-PGEl+HSR).In group B,rats were sacrificed 90 min after shock,and in group C,rats were anesthetized and then subjected to hemorrhagic shock followed by resuscitation.In group D,rats were pretreated with Lipo-PGEI one hour before HSR.Liver function,NO and ET.1 were measured,and pathological changes of liver tissue in each group were observed,and the expres8ions of iNOS and ET.1 of liver tissue were measured by using immunohistochemistry 6 hours after HSR.Data were analyzed by analysis of variance,and P<0.05 was considered as significantly different in statitistics.Results The levels of liver iNOS and ET-I increased in HSR group compared with shock group [(O.225±0.080)vs.(0.082±0.021)and(0.292±0.047)vs.(0.082±0.035),P<0.05].Pretreatment with Lipo-PGEl markedly reduced the damage of Liver function,and lowered the levels of NO and ET-I.which were consistent with decrease in iNOS and ET-16 hours after HSR[(0.116±0.034)vs.(0.225±0.080)and(0.198±0.041)vs.(0.292±0.047),P<0.05].Conclusions Pretreatment with Lipo-PGEl could reduce liver injury after HSR.The mechanisms might be attributed to inhibiting iNOS and ET-1,regulating the balance of NO/ET-I.
