1.Research on Synthesis Technology of Ligustilide Dipolymer

OBJECTIVE:To study the synthesis technology of Ligustilide dipolymer (compound Ⅰ). METHODS:Using phthalic anhydride(compoundⅡ)as raw materials,sodium borohydride was reduced to phthalide(compound Ⅲ),and then syn-thesize ligustilide dipolymer after 4 steps. Ligustilide dipolymer was characterized by 1H-NMR and 13C-NMR. The synthesis technol-ogy of fifth step was optimized by orthogonal test with catalyzer,reaction temperature and reaction time as factors using yield as in-dex. The validation test was conducted. RESULTS:The optimal synthesis technology of fifth step was as follows as aluminium mu-riate of catalyzer,reaction temperature of 150 ℃,reaction time of 5 h. The average yield of validation test was 56.16%(RSD=1.23%,n=3). Total reaction yield was 24.4%,and the purity of target product was 99.4%. CONCLUSIONS:Ligustilide dipoly-mer is synthesized successfully;synthetic process is stable,and its quality is controllable.
2.Simultaneous Determination of 4 Components in Compound Paracetamol and Amantadine Hydrochloride Tablet by GC

OBJECTIVE:To establish a method for the simultaneous determination of paracetamol,amantadine hydrochloride, caffeine,chlorphenamine maleate in Compound paracetamol and amantadine hydrochloride tablet. METHODS:GC was performed on the column of HP-5 sillica capillary,temperature programmed,detector was FID detector,with the temperature of 300 ℃,car-rier gas was nitrogen gas,the flow rate is 1.5 mL/min,the split ratio was 20:1 and injection volume was 1μL. RESULTS:The lin-ear range was 156.0-4990.4 μg/mL for paracetamol,125.7-4023.2 μg/mL for amantadine hydrochloride,19.14-612.4 μg/mL for caffeine and 2.515-80.48 μg/mL for chlorphenamine maleate(all r=0.9999);the limits of quantification were 1.4,0.5,1.1,0.9 ng,limits of detection were 0.4,0.2,0.3,0.3 ng;RSDs of precision,stability and reproducibility tests were lower than 2.0%;re-coveries were 99.59%-101.77%(RSD=0.8%,n=9),99.56%-101.80%(RSD=0.7%,n=9),98.44%-100.83%(RSD=0.7%,n=9) and 100.05%-101.91%(RSD=0.6%,n=9),respectively. CONCLUSIONS:This method is simple,rapid,accurate and reli-able,and suitable for the simultaneous determination of paracetamol,amantadine hydrochloride,caffeine,chlorphenamine maleate in Compound paracetamol and amantadine hydrochloride tablet.
3.Detection of Chemical Constituents in Antirheumatic Traditional Chinese Medicines

OBJECTIVE:To establish an HPLC method for the detection of Prednisone acetate, Hydrocortisone, Dexamethasone acetate, Piroxicam, Diclofenac sodium, Indometacin and Ibuprofen, the 7 chemical constituents in antirheumatic traditional Chinese medicine. METHODS: The chromatographic separation was performed on a Hypersil BDS C18 column (200 mm?4.6 mm,5 ?m) using methanol-1 g?L-1KH2PO4(58∶42) (adjusted to pH 4.0?0.05 with H3PO4) as the mobile phase at a flow rate of 1.0 mL?min-1. The detective wavelength was set at 220 nm. RESULTS: Seven chemical constituents were all well-separated with good linearity within respective range of sample size. CONCLUSION: The method is rapid and convenient, and can be used for the quantitative and qualitative analyses of 7 chemical constituents in antirheumatic traditional Chinese medicine and to detect whether there is addition of chemical constituents in antirheumatic traditional Chinese medicines.
4.Differential expression analysis of IL-8 and STAT3 in active tuberculosis patients and its value in adjuvant diagnosis

Abstract: Objective To compare the differences in the expression of interleukin-8 (IL-8) and signal transducer and activator of transcription 3 (STAT3) among three groups of individuals: patients with multi-drug resistant tuberculosis (MDR-TB), patients with drug-susceptible tuberculosis (DSP-TB), and healthy controls. By analyzing the differences between these groups, the study seeks to provide a reference basis for clinical auxiliary diagnosis of new type tuberculosis. Methods The differences in mRNA expression profiles of chip from peripheral blood mononuclear cells (PBMCs) among three groups were analyzed. MDR-TB and DSP-TB patients were selected as study subjects, with healthy individuals serving as a control group, and the serum IL-8 concentration of each group was measured using an ELISA assay to compare differences between groups. Additionally, PBMCs from all groups were isolated and total RNA was extracted for analysis of IL-8 and STAT3 mRNA expression using quantitative real-time PCR(Q-PCR). Differences in STAT3 protein expression among the groups were also compared by Western blot assay. Results The serum IL-8 concentration in the MDR-TB and DSP-TB groups was significantly higher than that in the healthy control group, with a statistically significant difference (P<0.05). The relative expression level of IL-8 mRNA in PBMCs of the MDR-TB group and DSP-TB group was also significantly higher than that of the healthy control group, with a statistical significance (P<0.05). Both Q-PCR and Western Blot results revealed that the expression of STAT3 in the healthy, DSP-TB and MDR-TB groups sequentially increased, presenting statistically significant differences between the groups (P<0.05), consistent with the mRNA chip results. Conclusions The expression of IL-8 in active tuberculosis patients (ATB) is significantly increased, which can be used as a marker to distinguish ATB from healthy populations. The changes of STAT3 concentration not only serve as a reference for the diagnosis of ATB but can also distinguish between DSP-TB and MDR-TB populations. The combined detection of IL-8 and STAT3 holds promise as a clinical auxiliary diagnostic basis for different tuberculosis patients.
5.Expression and significance of miR-21 and PDCD4mRNA in human colorectal cancer

Objective To investigate the expression of miR-21 and PDCD4mRNA in colorectal cancer and their correlation with clinicopathological characteristics,to elucidate relationship between PDCD4and miR-21 in vivo.Methods Expression of RNA (including miR-21and PDCD4mRNA) and PDCD4 protein were detected respectively by quantitative real-time reverse transcriptase-PCR and SP immunohistochemical staining in 43 cases of colorectal carcinoma and corresponding normal mucous membrane tissue.Results In colorectal carcinoma,expression of miR-21 was higher than that of the control ( P < 0.05 ),and expression of PDCD4mRNA lower than the control ( P < 0.05 ).Expression of miR21 was associated with lymph node metastasis and clinical stages ( Ⅰ + Ⅱ,Ⅲ + Ⅳ ) ( P < 0.05 ).On the other hand,no significant differences were observed regarding sex,tumor site,size,local invasion,distant metastases,clinical stages( Ⅰ,Ⅱ ) (P > 0.05 ).Expression of PDCD4mRNA was associated with local invasion,clinical stages ( Ⅰ,Ⅱ ) ( P < 0.05 ).While no significant differences were found concerning sex,site,size,lymph node metastasis,distant metastases,clinical stages( Ⅰ + Ⅱ,Ⅲ + Ⅳ ) ( P > 0.05 ).miR21 levels was negatively correlated with PDCD4 expression including nuclear and nuclear and cytoplasmic put together (P < 0.05 ),in contrast to PDCD4mRNA and PDCD4 expression in cytoplasmic ( P > 0.05 ).Conclusions ( 1 ) Abnormal expression of PDCD4 mRNA and miR-21 correlate with prognosis in colorectal cancer.(2) miR-21 suppress translation of PDCD4mRNA by binding to the 3'-untranslated region (3'-UTR)of target PDCD4mRNA.
6.Expression of RUNX3 and miR-130b in gastric carcinoma and clinical significance

ObjectiveTo explore the expression of miR-130b and RUNX3mRNA in human gastric carcinoma and the clinical significance. MethodsThe expression of miR-130b and RUNX3mRNA were detected by RT-PCR in 40 cases of gastric carcinoma and corresponding normal mucosa tissue. The expression of RUNX3protein was determined by immunohistochemistry SP method. ResultsThe expression of miR-130b was significantly up-regulated in gastric carcinoma than that in the adjacent normal gastric mucosa tissues (2.18 ± 3.75 ) vs.( 2.59 ± 3.45 ),P < 0.05 ; The expression of RUNX3mRNA in gastric carcinoma tissues was significantly lower than that in adjacent normal gastric mucosa tissues( 8.76 ±2.82) vs.( 7.58 ± 2.87 ),P < 0.05.The expression of miR-130b and RUNX3mRNA were positively correlated with lymph node metastasis and clinical stage ( P < 0.05 ) ; No significant association was found between the expression and age,gender,tumor size,distant metastasis and depth of tumor invasion ( P >0.05 ).The expression of miR-130b was negatively correlated with RUNX3 protein expression in nuclei and cytoplasm (P < 0.05 ).ConclusionsAbnormal expression of miR-130b and RUNX3mRNA correlates with prognosis of gastric carcinoma; Decreased RUNX3 protein expression may contribute to tumourigenesis.
7.Study of Knee Osteoarthritis Gait Index on Function Evaluation-Time Distribution at the Sole and the Vertical Rebound Force at Stand Phase

Objective To investigate time distribution of pressure of footplate in each stage and vertical anti-force from ground of patients with knee osteoarthritis (KOA).Methods 40 KOA patients, 10 aged people, and 10 common saplings were selected to test time distribution of pressure of footplate in each stage and vertical anti-force from ground in period of monopodia support.Results To unilateral KOA patient, there was not obvious difference to be observed in initial contact phase and forefoot push off phase ( P>0.05), but there was a significant difference in forefoot contact phase and foot flat phase between health side and sick side ( P<0.001), and there was obvious difference decreased buffer time and buffer force coefficient between them ( P<0.01).Conclusion To KOA patients, the time decreases in weight-bearing phase and absorption shocks, and amortization coefficient also decreases. These indexes can be used as the walking index of functional assessment and the objective evidence of curative effect.
8.Detection of a BRCA1 c.2013_2014ins GT variant an ethnic Han Chinese pedigree affected with breast cancer.

OBJECTIVE:
To detect potential variant in an ethical Han Chinese pedigree affected with breast cancer.
METHODS:
The proband and her relatives were subjected to next-generation sequencing using a target capture sequencing kit containing 121 cancer-related genes. Candidate variants were selected by analysis of their type, frequency in population, and segregation with the phenotype. Candidate variant was verified by Sanger sequencing and TA cloning.
RESULTS:
A c.2013_2014ins GT variant was detected in the BRCA1 gene among all breast cancer patients from this pedigree but not among healthy females. The variant was not recorded in the 1000 Genome Project database or the Exome Aggregation Consortium (ExAC) database. The frameshifting insertion was predicted to form an premature stop codon in gene transcript and can give rise to a truncated protein.
CONCLUSION
The BRCA1 c.2013_2014ins GT variant probably underlies the pathogenesis of breast cancer in this Chinese pedigree.

9.Reliability Thermal Design,Simulation and Experimental Verification of Electronic Monitoring Unit for Magnetic Resonance System

The monitoring unit used in the nuclear magnetic resonance system,as an important unit of the system,faces a high thermal risk during its entire life cycle.This paper ensures the high efficiency and reliability of the thermal design of the product module from the two dimensions of structural design and device derating design.In order to reduce the risk of thermal design of electronic modules and comprehensively verify the effectiveness of thermal design of electronic modules,the design verification is carried out by combining simulation and experiment.In the simulation process,by establishing a thermal simulation model at the circuit board level,the crustal temperature of the core device is numerically calculated,and the index is compared with the thermal design index value and the test value,on the one hand,to verify the correctness of the simulation model.On the other hand,the validity of thermal design is verified.In the testing process,a thermal test platform for product modules is built,and the thermal characteristics test values of the core components of the module under extreme electrical conditions are obtained,and the corresponding conversion methods are used to predict the thermal performance and thermal design margin of the product at different altitudes.The results show that the electronic module can meet the thermal design requirements in terms of structural design and derating design of core components,and can ensure that the product module can work safely and reliably during the entire life cycle of the NMR system.
10.Effect of Synthesized Rehabilitation on Detoxification Addicts

ObjectiveTo explore the effect of synthesized rehabilitation on detoxification addicts.Methods100 detoxification addicts were randomly divided into the rehabilitation group and control group with 50 cases in each group. Cases of the rehabilitation group were treated with "ten-step" rehabilitation treatment, but that of the control group did not. The rehabilitating treatment included cognitive psychotherapy, behavioral therapy, family treatment, social skill training and etc. The effects of two groups were evaluate with Symptom Checklist-90 (SCL-90) scale.ResultsExcept hostility, other factorial scores of the rehabilitation group including positive items, somatization, obsession, interpersonal sensitivity, depression, phobia, paranoid and psychotic factors were significantly higher than that of the control group (P<0.05~0.01).ConclusionThe synthesized rehabilitation can obviously improve the psychology of detoxification addicts.
