To explore the detection of GNAS1 gene mutation from peripheral blood and non-involved bone marrow in patients with fibrous dysplasia of bone(FD). Both peripheral blood and non-involved bone marrow collected from six FD patients were used for genetic diagnosis by next generation sequencing and direct sequencing respectively. Two patients were found to have common gene mutation(positive) by next generation sequencing, accounting for 33.3% of total six people: p.R201H(CGT>CAT) was detected in both peripheral blood and bone marrow of patient No.3, and p.R201C(CGT>TGT) was detected in both tissues of patient No.6. While direct sequencing only found p.R201H(CGT>CAT) mutation in peripheral blood and bone marrow of patient No.3. Detection of GNAS1 gene mutation may contribute to the diagnosis of FD. Hot spot mutations of GNAS1 are present in peripheral blood and non-involved bone marrow samples from some Chinese FD patients, but the diagnostic value and sensitivity of GNAS1 gene mutation in non-involved tissues of FD patients is low by next generation sequencing or direct sequencing.

The early characterization of the diazepam-binding sites outside the brain led to their assignment as peripheralbenzodiazepine receptors, or PBRs, to distinguish them from the central benzodiazepine receptor.Although PBR is a widely used and accepted name in the scientific community,recent data regarding the structure and molecular function of this protein increasingly support renaming it to represent more accurately its subcellular role (or roles) and putative tissue-specific function (or functions). Translocator protein (18 ku,TSPO), is proposed as a new name, regardless of the subcellular localization of the protein. This review deals with the pharmacological, structural and molecular characterization of the PBR and its role in the neuropshcopharmacology.

Objective　To study the change of the serum lipoprotein(a)〔Lp(a)〕 and other lipid levels in the childhood simple obesity.Methods　The levels of serum Lp(a),cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-ch),low density lipoprotein cholesterol(HDL-ch),apoA-I and apoB were investigated in 42 simple obesity children and 30 control subjects.Results　The levels of serum Lp(a),TC,TG,HDL-ch，LDL-ch of obesity children were markedly higher than those of normal.Conclusion　Simple obesity children are at increased risk of the development of cardiovascular disease.

ObjectiveTo present the experiences of f re e graft transplantation two-stage urethroplasty for hypospadias repair.Methods58 cases of hypospadias including 10 subcoronal,36 pen ile shaft,9 scrotal and 3 perineal were treated with free full-thickness skin graft or(and) buccal mucosa graft transplantation two-stage urethroplasty.Of th e 58 cases,45 were new cases and 13 had had previous failed surgeries.Single for eskin graft was used in 35 cases,single postauricular skin graft in 8,single b uccal mucosa graft in 6,foreskin grafts combined with postauricular skin grafts in 6 and foreskin grafts combined with postauricular skin grafts and buccal muc osa grafts in 3.ResultsOf 58 cases after the first-st age operation,56 were successful(97%) with grafts healing well,2 foreskin grafts got gangrened.After the second-stage operation,5 cases failed with newly for med urethras ruptured due to infection,8 complicated by fistulas whereas 43 case s healed well(77%).ConclusionsFree graft transplantati on two-stage urethroplasty for hypospaedias repair is a kind of effective treat ment with broad indication,comparatively high success rate,less complications an d good cosmatic results,being indicated for different types of hypospadias repa ir.

Objective: To investigate the chemical constituents of monoterpenes in the fruits of Gardenia jasminoides. Methods: Various column chromatographies were used in the isolation and purification, and the physicochemical constant determination and spectral analysis were adopted to identify the chemical structures of monoterpenes. Results: Twelve monoterpenes were isolated from G. jasminoides, such as jasminoside B (1), jasminoside G (2), jasminodiol (3), crocusatin-C (4), (7S)-6-(hydroxymethyl)-1, 1, 5-trimethylcyclohex-3-enone (5), bornyl-6-O-β-D-xylopyranosyl-β-D-glucopyranoside (6), (10R, 11R)-gardendiol (7), (10S, 11S)-gardendiol (8), (5S, 9S)-gardenate A (9), (5R, 9R)-gardenate A (10), jasminoside E (11), and 5, 6-dihydroxymethyl-1, 1-dimethylcyclohex-4-enone (12). Conclusion: Compounds 5, 6, 8, 10, and 12 are first isolated from this plant.

To strengthen the medical ethics education in the clinical practice stage in the department ot obstetrics and gynecology meets the requirements of the modern medical education under new situation.Aiming at the status quo of medical ethics education in China and abroad,this paper put forward specific recommendations in terms of the setting of teaching contents,the design of teaching approaches,the construction of teaching staff,as well as the evaluation of teaching achievements,and thus to enhance the understanding of medical students and interns to medical ethics and cultivate their practical capacity of analyzing and solving problems in clinical ethical dilemmas.

Objective To establish method for the fingerprint of Radix et Rhizoma Gentianae using UFLC with the hierarchical cluster analysis.Methods Column:Shimadzu C18(3.0 mm?75 mm,1.7 ?m);mobile phase:Acetonitrile-0.4% phosphorylation(5.5∶94.5);flow rate:0.8 mL/min;detection wavelength:240 nm;temperature:40 ℃.Results This method had a good linearity in the range of 0.154 8—1.858 mg/mL,and the average recovery was 102.09% with RSD of 1.68% for swertiamarin.It had a good linearity in the range of 0.322—5.152 mg/mL,and the average recovery was 103.80% with RSD of 1.11% for gentiopicroside.The RSD of precision and reproducibility were lower than 2.1%.The result of fingerprint studying was the same with the hierarchical cluster analysis.Conclusion This method can be used as standardized implantation and quality control of Radix et Rhizoma Gentianae.

Objective:To explore the method of building the formative assessment (FA) system in diagnostics teaching and investigate its implementation effect.Methods:We planned and formulated the formative assessment system in diagnostics teaching of undergraduate curriculum. The correlation between the scores of the formative assessment and final exam were analyzed. A questionnaire survey on students' attitudes towards FA and an interview on teachers' perspectives were conducted. SPSS 17.0 was used for statistical analysis. Linear correlation analysis was used to compare the final exam scores with formative assessment scores, and Wilcoxon rank sum test was used to compare the scores of questionnaire items.Results:The scores of FA were significantly positive related to the scores of final exam ( r = 0.559, P < 0.001). The questionnaire analysis showed that students significantly valued the formative assessment superior to the summative assessment ( Z = -8.632, P<0.001). It was generally approved of the promotion of FA in students' self-direct learning and self-supervision and the advances in acquirement of knowledge and skills. Conclusion:It is feasible of carrying FA in diagnostics teaching which could provide better learning outcome and might have an extensive prospect of application.

ObjectiveTo evaluate the quantitative and qualitative changes of peripheral-type benzodiazepine recepors (PBRs) in brain mitochodria and in platelet membrane in aging rats. MethodsMale Sprague-Dawley rats were divided into 3- and 24-month groups. All animals were sacrificed by decapitation and the brains were immediately removed. Mitochondrial components from dissected cerebral cortex were isolated. The membrane of platelets from venous blood was prepared by the method of hypotonic hemolysis. The specific binding assay of the radioactive PBRs antagonist ［3H］PK11195 to membrane was performed. Scatchard analysis was performed to estimate the equilibrium dissociation constant (Kd) and the maximal binding site density (Bmax). ResultsA significant increase in ［3H］PK11195 binding activity in the mitochodria from cerebral cortex in 24-month rats was observed compared to that in 3-month rats(P<0-001). Meanwhile, the Scatchard analysis revealed that there was an increase in Bmax, with a significant increase in Kd in 24-month rats. The same change of ［3H］PK11195 binding activity was noted for platelet membrane in 24-month rats(P<0-001).ConclusionThe density of PBRs increases in cortex mitochondria in aging rats, but the binding affinity of PBRs decreases which may be attributable to the progressive pathogenesis of aging in rats. ［3H］ PK11195 binding activity of platelet membrane might reflect the change of PBRs in the brain tissue.
