1.Effect of malnutrition on rocuronium induced neuromuscular blockade

Objective It has been reported that nutritional status of the patients affects neuromuscular (N-M) blockade induced by succinylcholine, tubocurarine, atracurium and vecuronium. The purpose of this study was to investigate the effect of malnutrition on rocuronium induced N-M block. Methods Forty ASA Ⅰ -Ⅱ patients undergoing surgery under general anesthesia were divided into four groups of 12 patients each, based on body mass index (BMI): I normal group BMI = 18.5-25kg? m-2 ;Ⅱ mild malnutrition group BMI= 17-18.5 kg?m-2 ;Ⅲ moderate malnutrition groups BMI = 16-17 kg?m-2 and Ⅳ severe malnutrition group BMK90% the patient was intubated and mechanically ventilated. PETCO2 was maintained at 4.0-4.7kPa. Anesthesia was maintained with 0.5%-0.7% isoflurane +60% N2O. During operation when T1 returned to 25% of control rocuronium 0.15mg?kg-1 was supplemented. The onset time, degree of N-M block and duration of action of initial and supplemental dose were recorded.Results With initial dose the onset time was delayed in group Ⅲ [(184?58)s] and group Ⅳ [(252?62) s] as compared with that in group Ⅰ [(122?33 )s] (P0.05) .Conclusions The neuromuscular blockade induced by rocuronium is reduced in patients with moderate and severe malnutrition.
2.In vitro load test of biological prosthesis replacement after partial resection of femoral head

Objective To investigate effect of the biological prosthesis on load properties of the proximal femur after hemiarthroplasty with partial resection of the femoral head in order to assess stability of the prosthetic implant.Methods Twenty-four young fresh normal femur (with preservation of femoral head) specimens were assigned to prosthesis group (n =8),control group (n =8) and fatigue group (n =8).Specimens in prosthesis group underwent artificial femoral head replacement using self-designed prostheses.Stress at the femoral head with the model standing on one leg was performed.Biomechanical machine was used to detect the difference of load-stress and load-displacement before and after femoral head replacement under the load of 2000 N.Detection points included lateral superior base of femoral neck (A),medial side of lesser trochanter of femur (B),and inferior side of greater trochanter of femur (C).Results At the load of 2 000 N,the load-stress at inferior side of greater trochanter of femur and medial side of lesser trochanter of femur revealed no significance differences between the normal femur and femur with artificial femoral head replacement (P > 0.05),while the difference was significant at lateral superior base of femoral neck (P < 0.05).There were no significant differences of load-stress at A,B,and C points among fatigue group,prosthesis group and control group (P > 0.05).Conclusion After partial femoral head replacement,the biological prosthesis is effective in maintenance of the normal stress transfer in the proximal femur.
3.Construction and verification of adenovirus containing the PUMA gene under the control of the hTER promoter

Objective To construt the adenovirus vectors of Ad-hTERT-PUMA-EGFP containing human telomerase reverse transcriptase(hTERT) ,PUMA and enhanced green fluorescent protein (EGFP) .Methods By a series of molecular biology tech-niques ,PUMA gene was inserted into downstream of the hTERT promoter .And then EGFP was inserted into downstream of the PUMA in adenovirus vectors .Finally ,recombinant adenovirus vector ,Ad-hTERT-PUMA-EGFP was assembled into the adenovi-rus .Results Correct adenovirus laoding vectors was verified by viral genome PCR technology .After 12 h of transfection ,HEK293 cells showed green fluorescence in 72 h .Conclusion The correct structure of the adenovirus vectors containing PUMA gene and EGFP gene was identified by restriction enzyme analysis .
4.Effect of Psychologic Behavior Training on Cognitive Function of Community Chronic Schizophrenia Patients

Objective To observe the structure of cognitive injury and the effect of psychologic behavior training on cognitive function in community chronic schizophrenic patients. Methods80 healthy adults (control group) and 76 community chronic schizophrenia patients (research group) were assessed with World Health Organization-battery of cognitive assessment instrument for elderly (WHO-BCAI), mini-mental state examination (MMSE), and activities of daily living (ADL) pre- and post-psychologic behavior training. ResultsThere were significant differences in the scores of WHO-BCAI, MMSE, ADL between control group and research group (P<0.05 or P<0.01). 12 months after training, all the scores of research group significantly improved compared with that before training (P<0.05 or P<0.01). ConclusionCognitive disorders have occurred in the community chronic schizophrenia patients in wide range. Psychologic behavior training can improve their cognitive function.
5.Preparation,stability and in vitro activity of egg yolk immunoglobulin Y against human Sucrase

Objective:To prepare the egg yolk immunoglobulin Y ( IgY) against human Sucrase and study its stability,in vitro activity. Methods:Hy-line laying hens were immunized with human Sucrase protein,IgY was isolated and purified from egg yolks of im-munized hens using water dilution and salting out method. Indirect ELISA was used to evaluate the titer and stability of IgY. The purity and specificity of IgY were analysed by SDS-PAGE and Western blot respectively. The inhibitory effects of IgY on α-glucosidase was studied by PNPG method. Results:Indirect ELISA results showed IgY could be detected on the tenth day after the first immunization, and the peak titer of IgY was 1:12 800 after the 40th day of immunization. SDS-PAGE showed that the heavy chain and light chain of IgY were 65 kD and 25 kD respectively, and the IgY against human Sucrase could specifically recognize the protein of human Sucrase. The IgY maintained primary titer when it was kept between 29-69℃ for 15 min,and pH 4-7,37℃,4 h. The titer of IgY was maintained 50% after digestion by pepsin and trypsin respectively for 2 hours. IgY had a higher resistence to pepsin than trypsin after longer digestion time. IgY showed an inhibitory effect on α-glucosidase in concentration dependent manner. The half inhibitory concentration (IC50) was 0. 540 mg/ml. Conclusion:The IgY against human Sucrase has been successfully obtained,which established foundations for its study of Type 2 diabetes mellitus rat models in vivo.
6.Ac-SDKP inhibits silicosis fibrosis in rats by regulating Gαs/Gαi signaling

ABSTRACT:Objective To observe whether N-acetyl-seryl-aspartyl-lysyl-proline (Ac-SDKP)can inhibit rat silicotic fibrosis by regulating stimulatory G proteinα(Gαs)/inhibitory G proteinα(Gαi)signal.Methods Male Wistar rats were randomly divided into three groups (n=1 0 ):control 1 6-w group,silicosis 1 6-w group,and Ac-SDKP pre-treatment group.The pathological changes of the lung tissue was observed by HE staining;the expressions ofα-smooth muscle actin (α-SMA),Collagen Ⅰ,fibronectin (Fn),Gαs,Gαi2 ,Gαi3 and cAMP were detected by Western blot.Immunofluorescence was performed on lung tissue sections to detect the coexpression ofα-SMA/Gαi3 . Results Within silicosis 16-w group,HE staining showed that the silicotic nodule volume increased,nodule fusion and the formation of interstitial fibrosis could be seen,and cell fibrous nodules were visible.Immunofluorescence staining showed the enhanced coexpression ofα-SMA/Gαi3 in fibrosis area.Compared with those in control group, the expressions ofα-SMA,Collagen Ⅰ,Fn,Gαi2 and Gαi3 significantly increased in silicosis 1 6-w group,but the expressions of Gαs and cAMP decreased.Compared with silicosis 1 6-w group,Ac-SDKP pre-treatment group had alleviated lung injury and decreased coexpression ofα-SMA/Gαi3 .The expressions ofα-SMA,Collagen Ⅰ,Fn,Gαi2 and Gαi3 protein significantly decreased in Ac-SDKP pre-treatment group,while the expressions of Gαs and cAMP increased obviously.Conclusion Ac-SDKP can regulate the expressions of Gαs and Gαi and promote the formation of cAMP,thus playing an effective role against silicotic fibrosis.
7.Study on HBV genotype distribution and clinical correlative indexes in patients with hepatitis B in Xi'an

Objective To study the relationship and clinical significance of hepatitis B genotype distribution in Xi'an City and clinical relative indicators.Methods The nested PCR was used to amplify HBV DNA in 389 serum samples from HBV infected pa-tients in Xi'an City ,then polymerase chain reaction restriction fragment length polymorphism (PCR-RLFP) was used to identify HBV genotypes.Results Among 389 cases of HBV Infection in Xi'an City ,58 cases of HBV B genotype accounted for 14.9% (58/389) ,326 cases of HBV C genotype accounted for 83.8% (326/389) and 5 cases of HBV B/C mixed type accounted for 1.2% (5/389).The gender ,HBeAg positive rate ,HBV DNA load level ,ALT and AST had no statistical difference between HBV genotype B and C with the patients(P>0.05);There was no statistically significant in the distribution of B and C genotypes between the pa-tients with chronic hepatitis B and HBV carriers (P>0.05) ,but it had statistical difference between liver cirrhosis and hepatocellu-lar carcinoma(P<0.05).HBV genotype C was easier to develop liver cirrhosis even liver cancer than genotype B.Conclusion The three kinds of HBV genotype B ,C and B/C exist in the HBV infected persons in Xi'an City ,with the genotype C as the main geno-type ,genotype B takes the second and mixed genotype B/C is minimal.The liver cirrhosis occurrence has a certain relationship with the genotype C ,which suggesting that the genotype C is more likely to develop liver cirrhosis.
8.Correlationship between level of bFGF in serum and synovia and severity of disease in X-ray patients with knee OA

Objective To explore the correlationship between level of bFGF (basic fibroblast growth factor) in serum and synovia and X-ray severity (Kellgren-Lawrence Grading System) in patients with knee OA. Method 68 patients with knee OA were enrolled into this study. Knee OA grading was evaluated according to the Kellgren-Lawrence classification. bFGF levels in both serum and synovia were examined using enzyme-linked immunosorbent assay. Results The level of bFGF in serum and synovial fluid in patients with knee OA were positively correlated with radiographic severity (r = 0.619, P < 0.001 and r = 0.603, P < 0.001, respectively). Further analysis revealed that there was a positive correlation between the level of bFGF in serum and in synovia fluid (r = 0.688, P < 0.001). Conclusions Levels of bFGF in serum and synovial fluid were significantly increased in patients with OA, and levels of bFGF were positively correlated with radiographic severity. These findings indicate that bFGF levels may be a biomarker of disease severity and could play an crucial part in the pathophysiology of degenerative process in OA.
9.Identification of differentially expressed genes in two new human bladder carcinoma cell lines

Objective To screen and identify differentially expressed genes in two new human urothelial carcinoma cell lines, BLS-211 and BLX. Methods Suppression subtractive hybridization (SSH) was used to createa subtracted library, and clones were sequenced. Results Totally 13 over-expressed genes in BLX and 9 in BLS-211 cells were obtained, respectively. Among them, 18 were known genes and 4 were new ESTs (Expressed Sequence Tag), and were collected by GenBank dbEST database (The access number was EB390424-7). Conclusion SSH is a powerful method for the identification of differentially expressed genes. The differential expression of some BCG-associated genes in different cells may be related to the different responses to clinical BCG therapy. The identified new ESTs can be cloned for full length to further study their functions.
10.Quantitative study of cardiac MR T 1 mapping/iECV in patients with aortic insufficiency

Objective:To explore the clinical value of T 1 mapping/indexed extracellular volume fraction (iECV) quantified with cardiac MR (CMR) parameters, and its correlation with traditional indicators of myocardial dysfunction in aortic insufficiency (AI) patients. Methods:A total of 36 patients clinically and radiologically diagnosed with chronic AI in our hospital between May 2012 and February 2016 were retrospectively selected. All AI patients underwent conventional CMR protocol, native and post T 1 mapping. CMR parameters, such as aortic regurgitant fraction (RF), late gadolinium enhancement (LGE) mass fraction, myocardial extracellular volume fraction (ECV) and iECV. Based on the values of aortic RF, AI patients were divided into mild AI group (9 cases), moderate AI group (14 cases) and severe AI group (13 cases). The clinical characteristics were teased from the patients′ electronic medical records. Univariate analysis of variance was used to compare the measurement data of native T 1 mapping, post-contrast T 1 mapping, ECV, and iECV. LSD test was used for pair wise comparison between the mild AI, moderate AI and severe AI groups. Data about cardiovascular history, New York Heart Association (NYHA) heart function classification, and LGE were compared by chi-square test or Fisher exact test. The correlation between left ventricle ejection fraction (LVEF) and iECV was evaluated by Spearman correlation analysis. Results:There was no difference in age, sex, cardiovascular history among the three groups. Comparison of patients with different severity of AI in the three groups: (1) There was statistically significant difference in the LGE positive rate among the three groups ( P=0.023), while the myocardial replacement of fibrosis increased with the grade of aortic regurgitation. (2) There was no statistically significant difference in the measurement data of native T 1 mapping, post-contrast T 1 mapping, ECV among the three groups ( H=1.815, 0.929, 2.496, all P values>0.05), while the diffuse myocardial fibrosis tended to increase with the degree of aortic regurgitation. There was statistically significant difference in iECV among the three groups ( H=16.725, P<0.001). The measurement data of iECV in the severe AI group was significantly higher than those in the other two groups ( P<0.05). LVEF value was inversely correlated with iECV ( r=-0.649, P<0.001). Conclusions:Quantitative T 1 mapping/iECV can serve as a parameter to noninvasively identify diffuse myocardial fibrosis in AI patients of different severities. It changes with LVEF and can manifest the reversible stage of left ventricular decompensation.
