1.Determination of ?-sitosterol in Rhizoma Heterosmilacis Japonicae by HPLC

Objective To establish the method to determine the content of ?-sitosterol in Rhizoma Heterosmilacis Japonicae.Method ?-sitosterol in Rhizoma Heterosmilacis Japonicae was determined by HPLC.Chromatographic column was sinachrom ODS-BP 5 ?m column(4.6 mm?250 mm).Mobile phase was methanol-0.1% H3PO4 liquor.Detection wavelength was 210 nm.Flow rate was 1.0 mL/min.Result ?-sitosterol showed a good linear curve in the range of 0.22~8.8 ?g(r=1.000 0).The average recovery was 96.5% with RSD of 1.15%.Conclusion The method was accurate and reproducible,and can be used to control the quality of Rhizoma Heterosmilacis Japonicae.
2.An immunohistochemical study of the fibronectin in experimental viral myocarditis

For the purpose of establishment of a diagnostic method of mild viral myocarditis,an experimental mild viral myocarditis model was induced in Balb/c murine by coxsackie virus B 3.The cardiac tissue secfion were stained by LSAB-immunohistochemical method with anti fibronectin(Fn) antibody. The results demonstrated that the Fn deposition was found in the myocardium of mice with myocarditis and the mild degeneration of cardiomyocytes could be identified by Fn LSAB immunohistochemical staining.It is suggested that the Fn deposition in the myocardium tissue is one of the reliable marks of myocardium inflammation.
3.A postmortem semi-quantitative study on time-dependent changes of DNA content in chondrocytes in rat's costal cartilage

Objective To study the relationship between the DNA content of chondrocytes in the costal cartilage and postmortem interval in putrefactive rat cadavers.Methods Nuclear DNA wag visualized by modified Feulgen's staining method.DNA content of ehondrocytes in the costal cartilage was semi-quantita tively determined by a computerized image analysis system in rats within 35d postmortem.Results Staining intensity of the nuclei was gradually reduced within from 1d to 28d postmortem.The nuclej could not be detected at 35d.The DNA content of chondrocytes decreased time-dependently within 28 days after death as determined semi-quantitatively,which revealed a linear relationship between DNA content and postmortem interval.Conclusion DNA content of chondrocytes in the costal cartilage reduces time-dependently with the extension of postmortem interval.
4.Relationship between DNA of Rat Kidney and Post by Comet Assay

[Objective] To study the correlation between kidney cell DNA degradation and postmortem interval within the span of 6-48 hour after the subject rats′ death.[Methods] To select 18 healthy mature female SD rats and equally divide them into 6 groups.To execute the rats with cervical spine articulation and put the rats under the incubator temperature of 25.1℃ (the average temperature of the 5 previous Decembers in Guangzhou prefecture).Sample kidney tissue from the rat separately 0 hour,6 hours,12 hours,24 hours,36 hours,48 hours,and 60 hours after the rats′ execution to prepare monoplast suspension,which is committed to comet assay.The comet images were captured by fluorescence CCD.Kinetic Comet 4.0 software was used to analyze images.Relevant data were collected by kinetic Comet 4.0 software and were subjected to Kruskal-Wallis test.[Results] Within the postmortem interval of 6-48 h,the number of SD rat kidney cell DNA fragments increased as the postmortem interval lengthens.So did the comet tail length.The Oliver tail moment and tail DNA of comet also showed sign of increase in positive proportion to the postmortem interval (their values corresponding to 60-hour-postmortem-interval were not obtainable.Kruskal-Wallis test indicated:the discrepancies of TL among the 6 groups were all significant (P < 0.01).The difference of TM between 6 h group and 12 h group was not significant (P > 0.05).The difference of TM between 24 h and 36 h was significant (P < 0.05).The difference of TDNA among 6 h,12 h,and 36 h groups were not significant (P > 0.05).The difference of TDNA between 36 h and 48 h was significant (P < 0.05).[Conclusion] Degradation of nuclear DNA of the rat kidney cells increases as the postmortem interval lengthens and comet assay may provide important empirical evidence for determining the postmortem interval.

AIM: To study the changes of plasma atrial natriuretic peptide(ANP), endothelin-1 (ET-1) and serum cardiac troponin I(cTnI) levels and the effects of ANP and ET-1 on myocardium in crush injury rats. METHODS: Crush injury was produced in SD rats,the serum levels of cardiac enzyme and cardiac troponin I,plasma levels of ANP and ET-1 were studied by automated biochemical analyzer,automated chemiluminescence assay and radioimmunoassay. 6.Activation and apoptosis of spleen lymphocytes in patients with viral myocarditis

AIM: To explore the role of activation and apoptosis of splenic lymphocytes in the development of viral myocarditis (VMC). METHODS: Apotosis and MHC II antigen of splenic lymphocytes were detected in the VMC group (VMC, death, 8 cases) and control group (non-cardiac death, 4 cases) with TUNEL and immunohistochemistry methods.RESULTS: Compared with the control group, the increased expression of MHC II antigen and apoptosis were found in the splenic lymphocytes in the VMC group. CONCLUSION: These results suggest that the abnormality of the apoptosis/activation of splenic lymphocytes may be involved in the pathogenesis of VMC.
7.Relationship between the polymorphism of the AT2R gene 1675A/G and the effect of indapamide sustained release tablets in female hypertension patients

Objective To investigate the relationship of the1675A/G polymorphism of AT2 gene with the therapeutic effect of indapamide sustained release tablets in female patients with primary hypertension.Methods Two hundred and twenty female patients with primary hypertension were treated with Indapamide Sustained Release Tablets ( 1.5 mg · qd) for 8 weeks.The blood samples from the patients were collect to determine AT2 gene polymorphism by PCR combined with HRM and sequencing.Results Two hundred and five patients completed the test.In female patients,the therapeutic efficacy of indapamide sustained telease tablets among different AT2R genotypes( AA:70.6%,AG:71.6%,GG:71.4% ) showed no significant difference ( x2=2.53,P=0.49 ),neither do the decline of BP after therapy ( F=0.39 and 0.19 respecrively,P > 0.05).Conclusion The AT2 genotype was assumed to be not correlated to the blood pressure lowering response to Indapamide Sustained Release Tablets in female primary hypertension patients.
8.Effect of the expression of miRNA-106b-5p on the prognosis of non-small cell lung cancer patients treated by targeted therapy

Objective To investigate the effect of the expression of miRNA-106b-5p on the prognosis of non-small cell lung cancer (NSCLC) patients who received targeted therapy. Methods A total of 273 NSCLC patients who underwent targeted therapy from January 2012 to October 2013 in Dalian University Affiliated Xinhua Hospital were included. Fasting venous blood samples of all the hospitalized patients within 24 hours were collected, real time fluorescence quantitative polymerase chain reaction was used to measure the expression of miRNA-106b-5p, and their survival status were followed up. Kaplan-Meier method and log-rank test were used for survival analysis. Cox regression model was used to make multiple-factor analysis. Results There were 112 males and 161 females among 273 patients. The age was (61 ±12) years old. There were 40 patients having family history of cancer, 70 patients having smoking history. According to TNM stage, 77 patients were classified into stageⅠ, 39 patients were classified into stage Ⅱ, 107 patients were classified into stage Ⅲ and 50 patients were classified into stage Ⅳ. The relative expression level of miRNA-106b-5p was 1.72±0.67. Until October 2018, 27 out of 273 NSCLC patients lost follow-up. Of 246 follow-up patients, 185 patients (75.2%) died. The results of single factor survival analysis showed that family history of cancer (HR= 1.16, 95% CI 1.07-1.31), smoking history (HR= 2.17, 95% CI 2.08-3.26), advanced differentiation degree (HR=1.63, 95%CI 1.43-2.19), higher TNM stage (HR= 2.09, 95%CI 1.75-2.17), and high expression of miRNA-106b-5p (HR=2.37, 95%CI 1.63-3.63) were associated with the poor prognosis of NSCLC patients treated with targeted therapy. Multivariable Cox regression analysis showed that family history of cancer (HR= 1.15, 95%CI 1.09-1.27), smoking history (HR= 2.11, 95%CI 2.08-2.82), higher differentiation degree (HR= 1.42, 95%CI 1.31-1.89), higher TNM stage (HR= 2.07, 95% CI 1.83-1.95), higher miRNA-106b-5p expression (HR=2.21, 95%CI 1.81-3.25) were the independent risk factors affecting the prognosis of NSCLC patients treated with targeted therapy. Conclusion The high expression of miRNA-106b-5p is associated with the increased risk of death among NSCLC patients treated with targeted therapy.
9.Study on Quality Standard of Sanyuan Rupixiao Gel Paste

Objective To establish the quality standard for Sanyuan Rupixiao Gel Paste. Methods Sparganii Rhizoma, Gleditsiae Sinensis Fructus, Cyperi Rhizoma and Impatientis Semen were identified by TLC method. The content of tetrahydropalmatine was determined by HPLC. Waters symmetry column was used with the mobile phase of acetonitrile-0.1% phosphatic acid in a gradient manner (pH was adjusted to 6.4 by triethylamine) (55:45) at the detection wavelength of 280 nm. The flow rate was 1.0 mL/min at the column temperature of 30 ℃. Results The spots in TLC were clear without any interference;tetrahydropalmatine showed a good linear relation in the range of 0.092–1.84 μg;the average recovery was 100.15%with RSD of 1.58%(n=6). Conclusion The method is simple and accurate with high reproducibility, which can be used for the quality control of Sanyuan Rupixiao Gel Paste.
10.Imaging features of oblique vaginal septum syndrome and its diagnostic value

Objective To investigate the imaging findings of oblique vaginal septum syndrome and its diagnostic value.Methods Clinical and radiographic data of 13 patients with oblique vaginal septum syndrome were analyzed retrospectively and relevant literatures were reviewed.All 13 patients underwent ultrasonography examination preoperatively,and 8 of them underwent MRI examination,and 3 patients underwent CT examination as well.The position of the oblique septum,the size of the cavity in the rear of oblique septum, the positional relationship between the cavity in the rear of oblique septum and the cervix,and the concomitant changes of the uterine appendages and urinary system were analyzed.Results The ultrasonography and MRI examinations all manifested as uterus didelphys,cervix duplex,double vagina and obstructed hemivagina,and oblique septum originated from the middle of cervix duplex and attached to one side of the vaginal wall,shielding the ipsilateral cervix;oblique vaginal septum was on the right side in 4 cases and on the left side in 9 cases;the average volume of the cavity in the rear of oblique septum in 6 cases of type Ⅰ was 255 mL,and the average volume in 6 cases of type Ⅱ was 74 mL,and the volume in 1 case of type Ⅲ was 56 mL;2 cases were combined with ipsilateral hematosalpinx and 1 case was combined with a chocolate cyst of ipsilateral ovary;12 cases were accompanied with ipsilateral renal agenesis and 1 case was accompanied with ipsilateral renal hypoplasia.CT examinations of 3 cases all presented as uterus didelphys,a cystic hypodensity lesion under unilateral cervix and ipsilateral renal agenesis.Conclusion Imaging findings of oblique vaginal septum syndrome are quite characteristic. Ultrasonography,CT and MRI examinations have certain value for accurate diagnosis of the disease.
