1.Improvement of Glutamic Decarboxylase Radioassay and Its Apply

The radioassay of glutamic decarboxylase (GAD) was modified by taking NaOH as trapped agent instead of phenylethylamine. The results showed that the coefficient of variation (CV) within same sample was 9.6 % and the radioactivity remains stable after 72 hours if use NaOH as trap agent. It is significantly stable than use phenylethylamine as trap agent, which the CV was 31.9 % and the radioactivity decreased 47% within the first hour and decreased to background after 6 hours. The reabsorption experiment shows over 80 % of 14CO2 can be reabsorption by NaOH within 6 hours. It is suggested that NaOH is a much better trap agent than phenylethylamine and the sensitivity can increase 1.66 folds. Using this method the GAD activity in 0.39~ 17.8 rng of brain tissue can be measured and it is success in determine the GAD activity both in rat brain tissue and cultured neurons.
2.The study on the proliferation and the apoptosis factors in vitro of Kölliker organ supporting cells in the cochlea of newborn rat.

OBJECTIVE:
To study the apoptosis/proliferation of Kölliker organ supporting cells and to understand the prompting apoptosis factors in vivo in the supporting cells in the Kölliker organ by changing the environment of the cultured supporting cells in the Kliker organ in vitro, via the separation, culture and purification of the supporting cells in the K6lliker organ.
METHOD:
A combinatorial approach of enzymatic digestion and mechanical separation was employed to isolate and culture in vitro pure Kölliker organ supporting cells. The purity was tested by flow cytometry assay. And K6lliker organ supporting cells were harvested to detect the rate and cycle of apoptosis by flow cytometry after Annexin V/PI staining, to test the cell growth curve by MTT assay, and to observe the differential expressions of the Bcl-2, Caspase-3, Caspase-8 and Caspase-9 through the Realtime PCR and Western blot. The calcium, potassium and glutamate concentrations in the culture medium of these cells in vitro were changed to detect the survival rate of cells by MTT assay.
RESULT:
The purity of K6lliker organ supporting cells by flow cytometry assay was 96. 56%. And these cells showed no significant difference in apoptosis, but an evident linear growth. The results of Realtime PCR and Western blot showed that the expression of Bcl-2, Caspase-3, Caspase-8 and Caspase-9 mRNA and protein in all different time points kept stable. Furthermore, the elevation of extracellular Ca2+ might contribute to decrease the cell viability of supporting cells. And K+ participated regulation of cell viability in a concentration-depending way. However, glutamate appeared to be a protective factor in high concentration.
CONCLUSION
There is no significant apoptosis in vitro of the supporting cells in the Kölliker organ of rats, showing a linear growth. The Ca2+ in high concentration might contribute to the apoptosis factor of these cells. However, the K+ and glutamate appear to be protective factors in high concentration.

5.In vitro evaluation of the antibacterial activities of different irrigants against Enterococcus faecalis

90 extracted single rooted human teeth were inoculated with E.faecalis.The cannals of the teeth were irrigated with MTAD, 5.25% NaClO,compound chlorhexidine gargle,tinidazole mouthwash,distilled H2 O(infection control)and autoclave followed by distilled H2 O(autoclaving control),respectively(n =1 5).Then E.faecalis infection of the samples were examined.The results showed that MTAD had significantly greater antibacterial effects than 5.25% NaClO,tinidazole mouthwashes,compound chlorhexidine gargle and tinidazole mouthwash(P <0.05).There was no significant difference between each 2 of the other 3 irrigants.
6.Study on EEG of childhood absence epilepsy

Objective To investigate the characteristics of electroencephalograms ( EEGs ) of patients with childhood absence epilepsy ( CAE ) .Methods EEG of 46 patients with CAE were analyzed .Results Of 46 patients,there were 44 cases (95.6%) with guided bilateral symmetry 3Hz spike-and-wave.16 cases were character-ized by multiple spike-and-wave on the 3Hz spike and slow wave;and the centrotemporal spikes was found in 1 case. Additionally,the 3Hzδactivity in occipital region has been traced in 34 cases of these 46 patients.Conclusion In general,the EEGs of CAE patients have typical characteristics ,and few of them showed atypical manifestation ,which must be verified by clinical attention .

9.Operation and Monitoring of Rational Use of Drug Based on the Wuhu Model of Separation of Drug and Medical Care

OBJECTIVE:To provide reference for rational use of drugs in hospital under the condition of the separation of drug and medical care. METHODS:The volumes of business,income from drug and income condition of medical staff were compared before and after implementation of separation of drug and medical care. The chance for pharmaceutical work under the condition of the separation of drug and medical care were analyzed to put forward advice for improvement of drug use in medical institutions. RESULTS & CONCLUSION:After conducting Wuhu model in 8 medical institutions,some achievement and experience have been obtained. It should be propelled to implement effective administration for rational use of drugs to resolve the problems on over expensive medical service.
