Objective To evaluate the clinical application of autologous fat and acellular dermal matrix (ADM ) in vocal fold medialization .Methods A total of 16 New Zealand white rabbits of both sexes were divided into two study groups (8 rabbits in each group) ,after unilateral recurrent laryngeal nerve section ,autologous fat or ADM were injected into paralyzed vocal folds .Laryngoscopic exams were performed 2 ,4 ,8 ,and 12 weeks after implanta‐tion ;then larynx specimens were sampled and serially sectioned in axial plane from the false vocal fold to the sub ‐glottis .After section was stained with HE ,histologic behavior of the graft were evaluated by optical microscope , volume of remaining material were calculated by autoCAD software .Results The remaining volume of autologous fat and ADM reduced gradually ,there were no statistically significant differences between the 2 groups .In 2 weeks , fat cells survival was mass distribution ,as its necrosis and absorption ,intraimplant resulted in a plurality of cavitati‐on ,encapsulated by fibrous connective ,mixed with a little survival fat cells .In 12 weeks ,obvious capsule formation surrounded lipoma composed of fiber connective tissue .In the ADM group ,2 weeks after injection ,intra - implant fibroblast activity and neovascularization was noted ,later fibroblast infiltration increased gradually .In 12 weeks ,the implantation maintaun the basic outline and have compact organizational structure ,but not forming obvious bounda‐ries with the surrounding tissue .Conclusion Comparing degree of absorption ,there were no statistically significant differences between autologous fat and ADM in the study ,but histological outcome of ADM is closer to the original organization .In future ,ADM optimization or combination with other materials ,will have a larger development and clinical application value .
2.Study of reversal effect of PARP-1 inhibitor PJ34 on cisplatin-resistance in human lung adenocarcinoma cells
Chinese Pharmacological Bulletin 2015;(6):865-869,870
Aim To investigate the reversal effect of PARP-1 inhibitor PJ34 on cisplatin-resistance in hu-man lung adenocarcinoma A549/DDP cells and the mechanism. Methods A549/DDP cells were treated with PJ34 alone or combined with cisplatin. The effects of proliferation inhibition were assayed by MTT meth-
od. The apoptosis ratios of cells were analyzed by flow cytometry. The protein expression of PARP-1 and LRP, GST-π were measured by Western blot assay. Results PJ34 could inhibit the proliferation of A549/DDP cells alone. The non-toxic dose of PJ34 could signifi-cantly resensitize A 5 4 9/ DDP cells to cisplatin , induce
apoptotic, lower the expression of PARP-1 and resist-ance-associated protein LRP and GST-π. Conclusion PJ34 could inhibit the proliferation of A549/DDP cells and resensitize A549/DDP cells, partially reverse cisplatin-resistance in A549/DDP cells, with a proba-
ble mechanism relating to increased apoptotic rate, and lowered expression of PARP-1 and resistance-associat-ed protein LRP and GST-π.

Objective To investigate the inheritance principle of the expanded GAG repeat allele and the clinical features of spinocerebellar ataxias 3 (SCA3) in a consanguinity family with first cousin marriage. Methods The CAG repeats of SCA3 gene were amplified by means of polymerase chain reaction.Fragment analysis with laser-induced fluorescence in capillary electrophoresis were performed for the positive samples detected by agarose gel electrophoresis.Furthermore, the clinical features were analyzed carefully. Results Fragment analysis revealed that the proband carried 2 alleles with 56 and 72 CAG repeats separately. The proband' s father carried 28 and 66,and the expanded CAG repeat allele inherited from his grandfather. The proband' s mother carried 33 and 56,and the expanded CAG repeat allele inherited from his grandmother. The proband' s son carried 27 and 85 and presented with dystonia besides ataxia. Conclusions The proband' s parents have the common ancestors.Their alleles with expanded CAG repeats probably come from the same allele of their ancestor. The GAG repeat is more unstable in the paternal inheritance than in the maternal inheritance. The 71-year-old asymptomatic family member carry the allele with 56 CAG repeats, which indicates the 56 CAG repeats may be not associated with the disease. The patients within this family have variable clinical features, especially the juvenile-onset case presents with apparent dystonia.
