Journal of Chinese Physician 2021;23(3):359-365
Objective:To investigate the effect and mechanism of G protein-coupled receptor, class C, group 5, member A (GPRC5A) on the proliferation and apoptosis of laryngeal cancer cells (LCC).Methods:From June 2015 to December 2018, 22 patients with laryngeal cancer were selected from the People's Hospital of Xinjiang Uygur Autonomous Region. Tumor tissue samples and paracancerous tissue were collected. The expression of GPRC5A in laryngeal cancer tissues and laryngeal cancer cells was detected by real-time fluorescent quantitative polymerase chain reaction (qRT-PCR) and Western blot; pcDNA3.1-GPRC5A and control plasmid pcDNA3.1 were transfected into Hep-2 and AMC-HN-8 cells respectively. Methyl thiazolyl tetrazolium (MTT) assay was used to detect the effect of GPRC5A on the proliferation of laryngeal cancer cells; V-FITC/PI assay was used to detect the effect of GPRC5A on the apoptosis of laryngeal cancer cells; DCFH-DA was used to detect the level of reactive oxygen species (ROS) in laryngeal cancer cells; Western blot was used to detect the protein expression of vascular endothelial growth factor (VEGF), E-cadherin and vimentin in laryngeal cancer cells.Results:(1) The expression of GPRC5A in laryngeal carcinoma tissues and laryngeal carcinoma cells was lower than that in adjacent tissues and normal laryngeal epithelial cells ( P<0.05). (2) Overexpression of GPRC5A could inhibit the proliferation of laryngeal cancer cells and the expression of VEGF, E-cadherin and vimentin ( P<0.05); overexpression of GPRC5A could significantly increase the level of ROS, decrease the level of NAD + and adenosine triphosphate (ATP) ( P<0.05), increase the apoptosis rate ( P<0.05), and significantly increase the protein expression of Caspase-3 and Caspase-9 ( P<0.05). Overexpression of GPRC5A could inhibit the expression of signal transducer and activator of transcription 3/suppressor of cytokine signal transduction 3/myelocytomatosis oncogene (STAT3/SOCS3/C-MYC) pathway related proteins ( P<0.05); the expression of GPRC5A in 22 patients with laryngeal cancer were negatively correlated with STAT3 ( P<0.05). (3) STAT3 and C-MYC inhibitors significantly inhibited the expression of VEGF and E-cadherin in Hep-2 cells ( P<0.05), promoted apoptosis ( P<0.05), decreased the level of interleukin (IL)-6 in Hep-2 cells ( P<0.05), and significantly increased the level of ROS in Hep-2 cells. Conclusions:It suggests that GPRC5A inhibits proliferation and epithelial-mesenchymal transition (EMT), induces oxidative stress and apoptosis of LCC cells potentially by regulating STAT3/SOCS3/C-MYC signaling. These results provide a molecular basis for clinical treatment and diagnosis of laryngeal cancer.

Objective To evaluate clinical effects of integrated traditional Chinese and western medicine therapy for epilepsy after stroke. Methods Literatures about the treatment of epilepsy after stroke with the combination of traditional Chinese medicine and western medicine which come from the database such as CNKI, VIP, WanFang and Pubmed, were studied and all the randomized controlled trials on comparing therapeutic effects of treating epilepsy between combined treatment of TCM and western medicine and western medicine were enrolled. The Cochrane Collaboration's software RevMan 5 was used for meta-analysis. Results 7 articles were included in the study, there is homogeneity among these articles(P>0.05), The fixed effect model was used for the meta analysis. the combined results were OR=5.17, 95%CI(3.02, 8.82), the overall effect of test Z=6.01, P<0.01, the difference between the treatment group and the control groups was significant, that means the overall effect of the combination of traditional Chinese medicine and Western medicine treatment was better than western medicine treatment; Funnel plot graphic displayed asymmetrically, which indicated the literatures publication bias. Conclusion The current data indicate that the curative effect of TCM and west medicine combined treatment is better than pure western medicine therapy for epilepsy after stroke, but as the literature publication bias, clinical studies need more high quality large samples to prove this conclusion.

Professor Luo Lingjie is a national famous old doctor of traditional Chinese medicine (TCM). He has a set of effective thoughts and methods on health maintenance. The core of the thought is Tiao-Xin, that is to say the emotion regulation is the key to health. The idea and method include three aspects, which include emotion regulation, diet regimen and exercise regimen. The emotion regulation is divided into clean ears, optimistic mood, learn to adapt, maintain psychological balance, and comply with nature. Diet regimen is fell into pay attention to the health food, regular diet, dietotherapy as primary and followed by drug, comply with the four seasons, reasonable to choose the food. There are four principles of exercise regimen, which are the choice of sports according to individual person with the aim to relax the body, move to raise the shape which should be combined with static to repose, proceed in an orderly way and persevere it step by step. The idea and method can also be used for daily personal health and clinical nursing for health maintenance. It is important that the physical and spiritual needs be consistent with long-term practice.
